Why are the most lethal pathogens the simplest? Lack of D-amino acid usage, coherent fractal morphology, and Hz-level biological oscillations prevalent in beneficial pathogens.

This paper examines a biochemical paradox in microbial pathogenicity: the most lethal human pathogens (viruses, protozoa, prions) systematically lack D-amino acid usage, coherent fractal morphology, and Hz-level biological oscillations; these characteristics prevail in beneficial and treatable bacterial pathogens. Systematic analysis of 47 major human pathogens and quantitative fractal dimension analysis of 40 cryo-electron microscopy structures unveil refined hierarchical patterns within universally low structural complexity. Bacterial pathogens employ D-amino acids in peptidoglycan synthesis

Manipulation of nonclassical ubiquitin signaling by pathogenic bacteria.

Manipulation of the host ubiquitin signaling system is emerging as a common and critical mechanism of virulence among pathogenic bacteria. In some cases, such as the intracellular pathogens Legionella pneumophila and Shigella flexneri, ubiquitin regulation is the focus of a significant proportion of the bacterial repertoire of secreted effectors (10% and 30%, respectively). While many of these bacterial effectors hijack classical aspects of the ubiquitin system, such as proteasomal degradation and immune signaling, others target nonclassical ubiquitin signals that are less well understood. The

Where metabolism and virulence meet-sulfoxide reductases as determinants of bacterial survival.

Bacterial metabolism is increasingly recognised as a driver of virulence in pathogens, and this includes the reduction of sulfoxides, which has traditionally been linked to anaerobic energy generation and maintenance of cellular proteins. Sulfoxide reduction is catalysed by two major enzyme groups, the Msr peptide methionine sulfoxide reductases that repair damaged proteins, and several types of mononuclear molybdenum enzymes that either convert small molecular S- and N-oxides or repair damaged proteins. Where these enzymes support virulence, they appear to be regulated by exposure to oxidisin

Candida auris skin colonization: mechanisms, microecological interactions, and emerging decolonization strategies.

Candida auris, as an emerging multidrug-resistant pathogen, has become a major threat to global public health due to its high mortality rate, strong transmissibility, and propensity to cause outbreaks in healthcare settings. Persistent skin colonization is a key factor contributing to the high transmissibility of C. auris and represents one of its important distinguishing features from other Candida species. However, the mechanisms underlying C. auris skin colonization remain poorly understood, and relevant prevention and control strategies are still underdeveloped. This article aims to system

Functional and Structural Studies of Alphavirus-Receptor Interactions.

Alphaviruses are mosquito-borne viruses that can cause disease in humans and animals. Alphaviruses bind to proteinaceous receptors found on the surface of host cells in the first step of the virus life cycle. Multiple studies have revealed that alphavirus interactions with cellular receptors are plastic, with receptor-binding patterns that vary across viral species even for the same receptor and that also vary in different infected hosts. We discuss recent progress on the functional and structural understanding of alphavirus interactions with cellular receptors, with implications for understan

Towards an Omics-Guided Framework for Microbial Biological Control of Fungal and Oomycete Diseases in Cannabis sativa: Integrating Host and Pathogen Genomics and Microbiomes.

The intensification of Cannabis sativa cultivation has increased the need for effective and sustainable plant-health strategies. Pathogen control is particularly challenging because pesticide residues may compromise product safety and quality, while repeated chemical treatments can favour the emergence of fungicide-resistant populations. Microbial biological control agents (BCAs) represent a promising component of integrated disease management, although their discovery and validation remain largely empirical. This narrative review examines the current literature on the biological control of fu

Critical role of the inflammatory rheostat in influenza-associated pulmonary aspergillosis.

Respiratory infections due to influenza A virus remain a major cause of morbidity and mortality. In the past decade, influenza-associated pulmonary aspergillosis (IAPA) has emerged as a major clinical issue. However, the mechanisms involved in linking host-virus-fungal interactions remain poorly understood. Increasing experimental and clinical evidence is converging to underline the impact of inflammatory imbalance in the pathogenesis of IAPA.

Molecular Pathways Driving Corneal Neovascularization in Herpes Simplex Keratitis.

Herpes simplex keratitis (HSK) is classically described as an immunopathological disease driven by recurrent herpes simplex virus type 1 (HSV-1) infection and chronic inflammation. So far, immune-mediated tissue damage has not fully explained the molecular mechanisms governing disease progression toward corneal neovascularization (CNV), a major cause of corneal blindness and vision loss worldwide. Increasing evidence indicates that CNV results from complex interactions that extend beyond leukocyte-driven inflammation, as the host cell machinery, including key pathways and molecular markers, is

Cellular Biology: Virus-Host Interactions and RNA Structure and Function.

Dengue is the most important viral disease transmitted to humans by mosquitoes and is caused by the dengue virus (DENV), which belongs to the Orthoflavivirus genus within the Flaviviridae family. The DENV RNA genome encodes ten proteins: three structural (C, prM, and E) and seven nonstructural (NS1-NS5) proteins. A comprehensive understanding of DENV interactions with both mammalian and mosquito cells is essential for the development of specific antivirals, which are currently lacking. In this chapter, we provide an overview of the viral replicative cycle, with a strong emphasis on the viral R

Orientia tsutsugamushi Alters Mitochondrial Function and Selectively Associates with VDAC.

Orientia tsutsugamushi is an obligate intracellular alphaproteobacterium and the causative agent of the potentially fatal rickettsiosis, scrub typhus. During infection, O. tsutsugamushi replicates exclusively in the eukaryotic cytosol near mitochondria and alters host metabolic pathways governed by mitochondria. We report that O. tsutsugamushi induces mitochondrial enzymatic impairment and structural abnormalities without altering mitochondrial abundance or the levels of proteins that maintain mitochondrial homeostasis. Confocal and structured illumination microscopy revealed a selective spati

Candida albicans, HIV And Immune System in a Holobiont Perspective.

Holobiont describes all types of relationships between hosts and their associated microbial communities, which can also attenuate the virulence of pathogens by the host microbiome. The cross-kingdom interactions between HIV, Candida albicans, and the human immune system have presented intriguing biological aspects. The human immunodeficiency 1 (HIV-1) virus is responsible for causing the acquired immune deficiency syndrome (AIDS) by infecting CD4+ cells. HIV-1 internalization also depends on binding to coreceptors in the cell membrane, such as CCR5 and CXCR4. The expression of these chemokine

Untangling the effects of within-host and between-host natural selection in viruses with applications to evolution of HIV-1 co-receptor usage.

The fitness of a pathogen can be characterized at multiple levels corresponding to different life cycle stages. Typically, epidemiological studies for measuring pathogen fitness compute a single selection coefficient for pathogen variants that encompasses the entire lifecycle. However, evolution towards greater fitness within hosts can potentially compromise transmissibility. To address this, we develop a population-genetic model and software for the inference of multi-level selection coefficients. Our approach builds on population-genetic models previously applied to macro-evolution, whereby

IFITM3 at the viral fusion-pore checkpoint: membrane energetics and entry-route-dependent outcomes.

Interferon-induced transmembrane proteins (IFITMs) are small interferon-stimulated membrane proteins that can limit viral entry, but their effects depend on the virus, entry route, and experimental context. This Mini Review focuses on IFITM3 at the viral fusion-pore checkpoint, defined here as the transition at which hemifusion either proceeds to fusion-pore opening and viral-content release or stalls before productive entry. Work on influenza virus supports a model in which IFITM3 remodels late-endosomal membranes, prolongs or kinetically stabilizes hemifusion-like intermediates, and raises t

From Periodontal Pocket to Systemic Diseases: Mechanistic Insights Into the Role of Porphyromonas gingivalis in Host Pathology.

Porphyromonas gingivalis (P. gingivalis), a keystone pathogen in periodontitis, has been increasingly recognised as a mechanistic bridge linking periodontal infection to pathological destruction in distant organs. P. gingivalis virulence factors, including gingipains, lipopolysaccharides (LPS), and outer membrane vesicles (OMVs), mediate complex host-pathogen interactions. In this review, we critically evaluated recent experimental studies which demonstrate the effects of P. gingivalis oral infection on systemic diseases, including cardiovascular disease (CVD), diabetes mellitus (DM), adverse …

Exosome-mediated bidirectional immune dysregulation in tuberculosis: proteomic profiling reveals strain-specific strategies of virulent H37Rv and attenuated H37Ra.

INTRODUCTION: Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a global health crisis, with drug resistance and immune evasion complicating control efforts. Mtb subverts macrophage function to establish persistent infection, but the role of exosomes in immune regulation remains poorly understood. METHODS: This study employed iTRAQ-based proteomics to dissect strain-specific immune modulation strategies of virulent H37Rv (RV) and attenuated H37Ra (RA) through macrophage and exosome profiling. RESULTS: We revealed distinct survival strategies of Mtb in Macrophages: RV mai

Lipid metabolism in the resuscitation of dormant mycobacterium tuberculosis: molecular resilience, host hijacking, and clinical opportunities.

Latent infection and reactivation of Mycobacterium tuberculosis (Mtb) represent key obstacles to the global strategy to end tuberculosis. The 2024 ICE-TB international consensus redefined tuberculosis as a continuum of pathological states that includes subclinical forms, with lipid metabolism being the core mechanism driving Mtb's transition across different disease stages. This review systematically elucidates the molecular mechanisms by which Mtb maintains survival through lipidome remodelling under restrictive microenvironments (phosphate deprivation, oxidative stress), with a focus on the

MicroRNA response in insect salivary glands to plant virus infection.

UNLABELLED: Most arboviruses rely on insect vectors for transmission, with salivary glands serving as a critical gateway for viral spread to new hosts. MicroRNAs (miRNAs) are key regulators of gene expression, yet their roles in salivary glands during virus infection remain poorly understood. Using the small brown planthopper (SBPH)-rice stripe virus (RSV) system, we sequenced small RNAs from salivary glands of nonviruliferous and viruliferous insects and identified 5,909 known miRNAs from 201 families. Of these, 1,143 miRNAs were differentially expressed upon RSV infection, including 1,090 up

Crosstalk Between Viral Envelop-E Protein and Host Innate Immune System: Exploring Pharmacological Targets and Agents.

The Envelope (E)-protein is a key structural element of enveloped viruses that plays a significant role in host-pathogen interactions, viral growth, and hijacking of host innate immune system. Due to lack of antiviral agents and significant adverse effects and less affordability of vaccines, the E-protein targeted drug development is gaining critical attention among the researchers. The present review explores the structural and genomic diversities of E-protein among animal viruses with special interest to flaviviruses, coronaviruses, and herpesviruses. E-protein's viroporin activity damages h…
