Steam pretreatment and fermentation of the straw material "Paja Brava" using simultaneous saccharification and co-fermentation.

Pretreatment, enzymatic hydrolysis and simultaneous saccharification and fermentation (SSF) of the South American straw material Paja Brava were investigated. Suitable process conditions for an SO₂-catalyzed steam pretreatment of the material were determined and assessed by enzymatic digestibility of obtained fiber slurries for 72 h at a water insoluble solids (WIS) content of 2%. The best pretreatment conditions obtained (200 °C, 5 min holding time and 2.5% SO₂) gave an overall glucose yield following enzymatic hydrolysis of more than 90%, and a xylose yield of about 70%. Simultaneous sacchar

Determinants of forage quality in Pensacola bahiagrass and Mott elephantgrass.

Our objective was to determine animal responses and forage characteristics that could explain the greater forage quality of 'Mott' dwarf elephantgrass (Pennisetum purpureum Schum.) than that of 'Pensacola' bahiagrass (Paspalum notatum Flugge). Both grasses were harvested after 5 wk of regrowth in June and September. Sheep fed Mott hays had greater (P = .001) voluntary intake of digestible OM (DOM) than did those fed Pensacola, but voluntary intake of ash-free NDF (NDFa) did not differ (P = .21) between grasses. When hays were fed at equal NDFa intakes, sheep fed Mott chewed at a greater (P = .

Paradigm of engineering recalcitrant non-model microorganism with dominant metabolic pathway as a biorefinery chassis.

The development and implementation of microbial chassis cells have profound impacts on circular economy. Non-model bacterium Zymomonas mobilis is an excellent chassis owing to its extraordinary industrial characteristics. Here, the genome-scale metabolic model iZM516 is improved and updated by integrating enzyme constraints to simulate the dynamics of flux distribution and guide pathway design. We show that the innate dominant ethanol pathway of Z. mobilis restricts the titer and rate of these biochemicals. A dominant-metabolism compromised intermediate-chassis (DMCI) strategy is then develope

Sphingolipids contribute to acetic acid resistance in Zygosaccharomyces bailii.

Lignocellulosic raw material plays a crucial role in the development of sustainable processes for the production of fuels and chemicals. Weak acids such as acetic acid and formic acid are troublesome inhibitors restricting efficient microbial conversion of the biomass to desired products. To improve our understanding of weak acid inhibition and to identify engineering strategies to reduce acetic acid toxicity, the highly acetic-acid-tolerant yeast Zygosaccharomyces bailii was studied. The impact of acetic acid membrane permeability on acetic acid tolerance in Z. bailii was investigated with pa

Changing leaf litter feedbacks on plant production across contrasting sub-arctic peatland species and growth forms.

Plant species and growth forms differ widely in litter chemistry, which affects decay and may have important consequences for plant growth v … compared with untreated plants, particularly litters with a high decomposition rate or low initial lignin/P ratio. In the second year, however, litter effects were neutral or positive, and related to initial litter N concentration (positive), C/N, polyphenol/N and polyphenol/P ratios (all negative), but not to decomposability. Differences in effect size …

Independent origins of endodermis in lycopsid and euphyllophyte roots.

The endodermis is a vital barrier tissue in plants, but its origin, evolution, and homologies across land plants remain ambiguous. Here, we implement a multifaceted investigation of the endodermis in lycopsids to shed light on its evolution in roots. We confirmed the presence of an endodermis with Casparian strips (CSs) in two of the three living lineages of lycopsids, while endodermoid layers with polar lignin caps were identified in the Lycopodiaceae. Although lignified tissues, including CSs, can be preserved in fossils, we did not find evidence for either an endodermis or an endodermoid in…

Biological Activities of p-Hydroxycinnamic Acids in Maintaining Gut Barrier Integrity and Function.

It is well established that p-Hydroxycinnamic acids (HCAs), including ferulic, caffeic, sinapic, and p-coumaric acids, possess a characteristic phenylpropanoid C6-C3 backbone and account for about one-third of the phenolic compounds in our diet. HCAs are typically associated with various plant cell wall components, including mono-, di-, and polysaccharides, sterols, polyamines, glycoproteins, and lignins. Interestingly, enzymes produced by intestinal microbes liberate HCAs from these associations. HCAs are completely absorbed in their free form upon ingestion and undergo specific reactions upo…

Multifunctional Flame-Retardant, Thermal Insulation, and Antimicrobial Wood-Based Composites.

Wood has been used in a variety of applications in our daily lives and military industry. Nevertheless, its flammability causes potential fire risks and hazards. Improving the flame retardancy of wood is a challenging task. Herein, a phytic acid-based flame retardant (referred to as AMPA) was synthesized based on supramolecular reactions between melamine and p-amino-benzene sulfonic acid followed by a reaction with phytic acid using deionized water as the solvent. A composite wood was prepared by removing lignin to tailor the unique mesoporous structure of the material, followed by coating AMP…

Rapid Release of Halocarbons from Saline Water by Iron-Based Photochemistry.

Methyl halides play important roles in stratospheric ozone depletion, but their formation mechanisms are not well defined. This study demonstrated that iron-based photochemistry significantly enhanced alkyl halide production by promoting the reaction of the representative monomer of lignin with halide ions in saline water under solar light irradiation. The methyl chloride (CH3Cl) emission from the light/Fe(III) process was 2 orders of magnitude higher than dark treatment and in the absence of iron. In addition, bromide and iodide showed better reactivity in the formation of the corresponding m…
