Acylglycerol recycling from triacylglycerol to phospholipid, not lipase activity, is defective in neutral lipid storage disease fibroblasts.

Neutral lipid storage disease (NLSD) is an autosomal recessive disorder in which excess triacylglycerol (TG) accumulates in most cells. Although it has been hypothesized that the TG accumulation is caused by a functional defect in cytosolic lipase activity, we were able to expose TG hydrolysis in NLSD cells by using triacsin C, an inhibitor of acyl-CoA synthetase that blocks the reincorporation of hydrolyzed fatty acids into glycerolipids. Our data suggest that TG lipolysis in NLSD cells is masked by rapid TG resynthesis, occurring because released acylglycerols cannot be used for phospholipid

Effects of growth temperature, 47-megadalton plasmid, and calcium deficiency on the outer membrane protein porin and lipopolysaccharide composition of Yersinia pestis EV76.

The expression of several virulence determinants of Yersinia pestis is known to be dependent on the in vitro growth temperature. One of these, calcium dependence, is associated with the presence of a 47-megadalton plasmid. We have examined the effects of incubation temperature, calcium in the growth medium, the presence of the 47-megadalton plasmid on the outer membrane protein, and the lipopolysaccharide composition of Y. pestis EV76. When cells were grown at 37 degrees C as opposed to 26 degrees C, a change in lipopolysaccharide composition and a decrease in the amount of an outer membrane p

Longitudinal changes in fatty acid metabolism and in the mitochondrial protein import system in overconditioned and normal conditioned cows: A transcriptional study using microfluidic quantitative PCR.

This study investigated the effect of body condition around calving on the hepatic mRNA expression of genes involved in fatty acid (FA) metabolism and mitochondrial protein import system of dairy cows during the transition period. Fifteen weeks before their anticipated calving date, 38 multiparous Holstein cows were selected based on their current and previous body condition scores (BCS) and allocated to either a high or a normal BCS group (19 cows each). They received different diets to reach targeted differences in BCS and backfat thickness (BFT) until dry-off. At dry-off, normal BCS (NBCS)

[Rapid and simultaneous determination of 11 ergot alkaloids in cereals and their products by ultra performance liquid chromatography-tandem mass spectrometry combined with Captiva EMR-Lipid column purification].

Ergot alkaloids (EAs) are mycotoxins produced by Claviceps and are present in cereals and their products; their residues pose significant threats to human health through food consumption, resulting in ergotism and sickness. Herein, a sensitive and rapid method for the determination of 11 EAs in cereals and their products using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) were developed. Eleven EAs were extracted with 20 mL acetonitrile-200 mg/L ammonium acetate solution (80∶20, v/v) for 15 min using the vortex shock method followed by 15 min of ultrasonication.

Double-stranded DNA unzipping: A fundamental and experimental investigation of fluid-induced stress in ultra-high pressure liquid chromatography systems and slalom columns.

Recently reintroduced for the separation of large (> 1 MDa) double-stranded DNA (dsDNA) and RNA biopolymers, slalom chromatography (SC) exploits the entropic elasticity of long nucleic acid polymers subjected to hydrodynamic drag in packed columns. Using a bioinert Slalom Column (4.6 × 300 mm) packed with 2.5μm particles operated with a 40 mM Tris-acetate-EDTA mobile phase at 30 °C, we examine the unzipping of the dsDNA concatemer strands as they undergo repeated coil-to-stretch transitions under intense fluid stress throughout the ultra-high pressure chromatography system (open tubing and con

Long-chain fatty acid homeostasis contributes to survival of uropathogenic E. coli during copper toxicity.

Uropathogenic Escherichia coli (UPEC) is the leading causative agent of urinary tract infection (UTI). Copper (Cu) is a host effector mobilized to the bladder during the innate immune response activated by acute UTI. E. coli precisely regulates Cu homeostasis via efflux systems because excess Cu is toxic. Long-chain fatty acid (LCFA) homeostasis is critical for the survival of E. coli during envelope stress. However, the interaction between LCFA metabolism and survival during Cu stress, which is also known to induce envelope stress, is not known and is the subject of this report. We hypothesiz

Serine supplementation suppresses hypoxia-induced pathological retinal angiogenesis.

Rationale: Pathological retinal angiogenesis with irregular and fragile vessels (also termed neovascularization, a response to hypoxia and dysmetabolism) is a leading cause of vision loss in all age groups. This process is driven in part through the energy deficiency in retinal neurons. Sustaining neural retinal metabolism with adequate nutrient supply may help prevent vision-threatening neovascularization. Low circulating serine levels are associated with neovascularization in macular telangiectasia and altered serine/glycine metabolism has been suggested in retinopathy of prematurity. We her

Evaluation of oxidative stress and genetic instability among residents near mobile phone base stations in Germany.

Human exposure to radiofrequency electromagnetic fields (RF-EMF) is restricted to prevent thermal effects in the tissue. However, at very low intensity exposure "non-thermal" biological effects, like oxidative stress, DNA or chromosomal aberrations, etc. collectively termed genomic-instability can occur after few hours. Little is known about chronic (years long) exposure with non-thermal RF-EMF. We identified two neighboring housing estates in a rural region with residents exposed to either relatively low (control-group) or relatively high (exposed-group) RF-EMF emitted from nearby mobile phon

Protocol for a cross sectional study of cancer risk, environmental exposures and lifestyle behaviors in a diverse community sample: the Community of Mine study.

BACKGROUND: Physical inactivity and unhealthy diet are modifiable behaviors that lead to several cancers. Biologically, these behaviors are linked to cancer through obesity-related insulin resistance, inflammation, and oxidative stress. Individual strategies to change physical activity and diet are often short lived with limited effects. Interventions are expected to be more successful when guided by multi-level frameworks that include environmental components for supporting lifestyle changes. Understanding the role of environment in the pathways between behavior and cancer can help identify w

Effect of a lifestyle-focused electronic patient support application for improving risk factor management, self-rated health, and prognosis in post-myocardial infarction patients: study protocol for a multi-center randomized controlled trial.

BACKGROUND: Cardiac rehabilitation (CR) programs addressing risk factor management, educational interventions, and exercise contribute to reduce mortality after myocardial infarction (MI). However, the fulfillment of guideline-recommended CR targets is currently unsatisfactory. eHealth, i.e., the use of electronic communication for healthcare, including the use of mobile smartphone applications combined with different sensors and interactive computerized programs, offers a new array of possibilities to provide clinical care. The present study aims to assess the efficacy of a web-based applicat

Movement through Active Personalised engagement (MAP) - a self-management programme designed to promote physical activity in people with multimorbidity: study protocol for a randomised controlled trial.

BACKGROUND: Multimorbidity, defined as two or more concurrent chronic diseases within the same individual, is becoming the clinical norm within primary care. Given the burden of multimorbidity on individuals, carers and health care systems, there is a need for effective self-management programmes. Promoting active participation within their clinical care and following a healthy lifestyle will help empower patients and target lifestyle factors that are exacerbating their conditions. The aim of this study is to establish whether a tailored, structured self-management programme can improve levels

A-to-I edited SNHG3 promotes non-small cell lung cancer metastasis by promoting fatty acid oxidation and resisting ferroptosis.

Adenosine-to-inosine (A-to-I) RNA editing is a critical post-transcriptional modification that enhances tumor genome diversity and contributes to cancer progression. In non-small cell lung cancer (NSCLC), while specific A-to-I editing events have been identified, their functional mechanisms and clinical relevance remain poorly understood. Here, through whole-transcriptome analysis of NSCLC specimens, we discovered a hyper-editing event at position c.1746 in the long non-coding RNA SNHG3 (c.1746 A > I), which correlates with advanced metastatic stages and reduced patient survival. Functional st

Oncoprotein HBXIP Modulates Abnormal Lipid Metabolism and Growth of Breast Cancer Cells by Activating the LXRs/SREBP-1c/FAS Signaling Cascade.

Abnormal lipid metabolism is a hallmark of tumorigenesis. Accumulating evidence demonstrates that fatty acid synthase (FAS, FASN) is a metabolic oncogene that supports the growth and survival of tumor cells and is highly expressed in many cancers. Here, we report that the oncoprotein, hepatitis B X-interacting protein (HBXIP, LAMTOR5) contributes to abnormal lipid metabolism. We show that high expression of HBXIP in 236 breast cancer patients was significantly associated with decreased overall survival and progression-free survival. Interestingly, the expression of HBXIP was positively related

Neuroprotective effect of atorvastatin in an experimental model of nerve crush injury.

BACKGROUND: Statins have therapeutic benefits for the management of several disorders. A short-term course of a high-dose statin pretreatment has demonstrated neuroprotective effects against neurological diseases. However, the molecular basis underlying the neuroprotective action of statins remains unclear. OBJECTIVE: We investigated whether a short-term course of high-dose atorvastatin pretreatment has beneficial effects in protecting sciatic nerve from crush injury. METHODS: Atorvastatin (5 mg/kg) or saline was given orally to Sprague-Dawley rats for 7 days before injury. The rats were sub
