Alterations in secondary lipids are associated with neuroinflammation in the brain of Neu1-deficient mice.

Neu1 (lysosomal sialidase 1) is essential for removing sialic acid from oligosaccharides and glycoconjugates. Neu1 deficiency impairs lysosomal digestion, leading to sialidosis and sialoglycoprotein accumulation. It also increases lipids, including gangliosides GM3, GD3, GM4, and LM1, in the kidney, liver, and spleen. Neu1-/- mice display symptoms resembling Type II sialidosis, including enlarged spleen and liver, kidney issues, neurological problems, spinal defects, and oligosaccharide buildup. The study examined secondary lipid alterations and inflammation in the cortex and cerebellum of the…

Microfluidic Nano-Assembly of Red-Blood-Cell (RBC) Lipids and Components for Engineering Extracellular Vesicles.

A central challenge in nanomedicine is the scalable and precise engineering of nanocarriers that are simultaneously tunable, immune-compatible, and capable of encapsulating diverse bioactive cargos. Extracellular vesicles (EVs), including native red blood cell-derived EVs (RBCEVs), offer biocompatibility but remain limited by heterogeneous composition, low yield, poor scalability, and limited control in cargo loading. Here, we introduce a bottom-up strategy for developing engineered red blood cell extracellular vesicles (eRBCEVs) through microfluidic diffusional mixing of purified RBC lipids a…

Plasma branched-chain amino acids in chronic kidney disease: associations with atherogenic lipids and mortality.

Branched-chain amino acids (BCAAs) are essential nutrients that promote muscle protein anabolism but also associate with cardiometabolic diseases; however, their role in chronic kidney disease (CKD) remains unclear. We investigated plasma BCAA levels and their associations with metabolic parameters and survival in CKD patients. Plasma BCAA levels, along with clinical and laboratory parameters, were measured in 328 patients with CKD stage 5 (median age 54 years, 60% males). BCAA concentrations in 83 community-dwelling controls (median age 51 years, 66% males) served as comparators. Multivariate

Immune effects of lipids isolated from Oncorhynchus mykiss eggs on cyclophosphamide-induced immunosuppression in mice.

Rainbow trout (Oncorhynchus mykiss) eggs are a rich source of polyunsaturated fatty acids (PUFAs), with levels exceeding those found in fish … ted mice (p < 0.05). Consequently, these results suggest that OM-PEG6000, prepared by incorporating lipids isolated from O. mykiss eggs into PEG6000, contributes to immune system activation and has potential as a functional food ingredient due to its immune-enhancing efficacy.

Predictive Lung- and Spleen-Targeted mRNA Delivery with Biodegradable Ionizable Lipids in Four-Component LNPs.

Background/Objectives: Lipid nanoparticles (LNPs) are leading mRNA delivery vehicles, with ionizable lipids (ILs) as their key component. However, the relationship between the IL structure and LNP endogenous organ-targeting is not well understood. In this study, we developed a novel library of biodegradable ILs featuring beta-propionate linkers, which, when incorporated into a four-component LNP formulation, show excellent extrahepatic selectivity and high protein expression. Methods: We explored the impact of structural modifications in the hydrophobic chains and polar-head groups in the ILs …

Ugi-reaction-derived ionizable lipids with cyclic tertiary amine heads enable spleen-targeted mRNA delivery.

Lipid nanoparticles (LNPs) have become an important platform for nucleic acid delivery. However, LNP-mediated delivery to non-hepatic organs and specific cell types remains a non-negligible challenge. As a key component of LNPs, ionizable lipids were rationally designed to adjust the LNPs properties to achieve organ-targeted delivery. The use of the Ugi four-component reaction (Ugi-4CR) as a one-pot multicomponent synthesis strategy to construct ionizable lipid molecules offers advantages, as it enables multidimensional structural diversity of ionizable lipids. We use isocyanides with cyclic t…

Modulation of Doxorubicin-Induced ROS Accumulation in Cardiomyocytes Using Ibuprofen-Conjugated Synthetic Lipids as Carriers.

Conjugation of an NSAID such as ibuprofen to the head group of oxanorbornane-based lipids and the use of their aggregates as carriers for doxorubicin (Dox) are discussed here. These conjugates were characterized by various spectroscopic techniques, including 2D-NMR, and insights into their assembly were gathered through PXRD, AFM, SEM, DLS, and qNano techniques. Free lipids as well as their formulations (lipid:cholesterol:Dox in a 3:1.5:2 molar ratio) showed a high tendency to form solid lipid particles, which was verified by TEM analysis. The presence of the ibuprofen unit led to an increase …

Mannich reaction-based combinatorial libraries identify antioxidant ionizable lipids for mRNA delivery with reduced immunogenicity.

The immunogenicity of lipid nanoparticles (LNPs) used for the delivery of nucleoside-modified messenger RNA limits the levels and durability of expression of the encoded protein. Here, by leveraging the Mannich reaction for ionizable lipid synthesis, and via the in vitro and in vivo screening of six combinatorial libraries of synthesized lipids, we report the identification of an antioxidant ionizable lipid, C-a16, exhibiting reduced immunogenicity. When incorporated into LNPs for mRNA delivery, C-a16 mitigated the generation of intracellular reactive oxygen species, thereby extending the dura…

Synthesis and Screening of Bispecific Linker-based Combinatorial Libraries Identified Thiosuccinimide Lipids for mRNA Delivery.

Ionizable lipids are essential for lipid nanoparticle-mediated mRNA delivery. The discovery of novel ionizable lipids relies on combinatorial chemistry synthesis and screening. However, current libraries mainly focus on optimizing amine cores and hydrophobic tails, often neglecting linker optimization, which restricts novel ionizable lipid discovery. To address this, a one-pot, two-step, three-component reaction based on N-hydroxysuccinimide-ester-maleimide bispecific linkers is first developed for constructing combinatorial libraries. Two libraries containing 344 thiosuccinimide lipids are sy…

Nanostructuring and Antioxidant Activity of Nanotherapeutics Designed by Self-Assembly of Natural Lipids and Phytochemicals.

Lyotropic liquid crystalline nanostructures formed by self-assembly in an aqueous medium are of fundamental interest and crucial for therapeutic applications, encapsulation of nutraceuticals, tissue engineering, and diagnostics. The biomimetic lipid bilayer building blocks impart biodegradable properties and low toxicity of the created nanoassemblies. The question of synergistic or quenching effects on the resulting bioactivity arises from the coencapsulation of multiple antioxidants (e.g., vitamin E (VitE), curcumin (CU), or coenzyme Q10) in nanocarriers of mixed nonlamellar-phase lipids (e.g…

Significant Interplay Between Lipids, Cytokines, Chemokines, Growth Factors, and Blood Cells in an Outpatient Cohort.

Cardiovascular disease (CVD) remains the leading global cause of morbidity and mortality, largely driven by atherosclerosis, a chronic inflammatory process involving lipids and immune cells. Although traditional lipid biomarkers such as low-density lipoprotein (LDL) and high-density lipoprotein (HDL) are well-established in CVD risk stratification, the interplay between cytokines, chemokines, growth factors (CCGFs), lipid metabolism, and hematological parameters in non-cardiac populations remains underexplored. We investigated associations between plasma cytokines and lipid-related biomarkers …

Supramolecular Lipid Nanoparticles Based on Amine β-CD Host-Guest Lipids: Design, Mechanisms, and Biosafety.

Background/Objectives: Lipid nanoparticles (LNPs) have demonstrated notable clinical success as advanced drug delivery systems. However, the development of novel covalently bonded ionizable lipids faces substantial technical challenges, as their modification is difficult and they have a high molecular weight. To address this issue, we report the use of host-guest complexes in supramolecular chemistry as functional lipid motifs for constructing LNPs. Methods: Ionizable amine β-cyclodextrin (amine β-CD)-derived host-guest amphiphilic lipid molecules (HGLs) were designed for the construction of m…

Oxidative stress in alcoholic liver disease, focusing on proteins, nucleic acids, and lipids: A review.

Oxidative stress is one of the important factors in the development of alcoholic liver disease. The production of reactive oxygen species and other free radicals is an important feature of alcohol metabolism in the liver and an important substance in liver injury. When large amounts of ROS are produced, the homeostasis of the liver REDOX system will be disrupted and liver injury will be caused. Oxidative stress can damage proteins, nucleic acids and lipids, liver dysfunction. In addition, damaging factors produced by oxidative damage to liver tissue can induce the occurrence of inflammation, t…

Milk fat globule membrane and its polar lipids: reviewing preclinical and clinical trials on cognition.

In most parts of the world, life expectancy is increasing thanks to improved healthcare, public health policies, nutrition, and treatment. This increase in lifespan is often not accompanied by an increase in health span, which severely affects people as they age. One notable consequence of this is the increasing prevalence of neurodegenerative diseases such as mild cognitive impairment, dementia, and Alzheimer's disease. Therefore, dietary and pharmaceutical measures must be taken to reduce the burden of such pathologies. Among the different types of nutrients found in the diet, lipids and esp…

Complex actions of sodium glucose transporter-2 inhibitors on lipids, calcific atherosclerosis, and bone density.

PURPOSE OF REVIEW: Inhibitors of sodium-glucose cotransporter-2 (SGLT2) lower renal glucose reabsorption and, thus, are used to treat patients with type 2 diabetes mellitus. Clinical trials coincidentally showed that SGLT2 inhibitors also benefitted patients with heart failure. This review explores the impact of SGLT2 inhibitors on other aspects of cardiovascular disease and skeletal health. RECENT FINDINGS: In some, but not all, clinical and preclinical studies, SGLT2 inhibitors are found to reduce serum levels of free fatty acids and triglycerides. Their effects on total and low-density lip

Lipids in the American Alligator stratum corneum provide insights into the evolution of vertebrate skin.

In terrestrial vertebrates, the outermost layer of the skin, the stratum corneum (SC), provides a durable and flexible interface with the environment and is comprised of corneocytes embedded in lipids. However, the morphology and lipid composition of the SC varies throughout evolutionary history. Because crocodilians and birds phylogenetically bracket the Archosaurian clade, lipid composition in crocodilian SC may be compared with that of birds and other vertebrates to make inferences about broader phylogenetic patterns within Archosaurs while highlighting adaptations in vertebrate skin. We id…

Protocol for building synthetic protocell membranes that sense redox using synthetic phospholipids and natural lipids.

Sensing is a critical function of artificial cells; however, this is challenging to realize using bottom-up approaches. Here, we present a protocol for building protocell membranes that sense cues important for redox biochemistry and signaling by combining synthetic phospholipids and natural lipids. We detail procedures for building giant unilamellar vesicles as protocell models that fluoresce in response to the biologically significant redox agents peroxynitrite, hydrogen peroxide, and hydrogen sulfide. For complete details on the use and execution of this protocol, please refer to (i) Gutier…
