Cardiovascular effects of dopamine after experimental coronary occlusion in the dog.

Hemodynamic effects of dopamine were studied on 30 open-chest dogs with experimental myocardial infarction. These dogs were divided into 3 groups. Three different doses of intravenous dopamine, 5, 10, and 20 mug/Kg/min, were administered for 30 min. Heart rate and myocardial contractility were increased dose-dependently. Total peripheral vascular resistance was reduced with 5 or 10 mug/Kg/min of dopamine. With 20 mug/Kg/min of dopamine, it became the lowest 5 min after the initiation of infusion but increased above the control value 20 min after the start of administration. Renal vascular resi

Left ventricular mechanical dispersion in a middle-aged population: associations with subclinical cardiac remodelling and cardiovascular risk.

BACKGROUND: Heterogeneous myocardial contraction is common in myocardial disease and linked to ventricular arrhythmias, but its frequency and relevance in the general population remain unclear. This study aimed to investigate the frequency of increased left ventricular mechanical dispersion (LVMD) and to examine its clinical and imaging correlates in a population-based cohort. METHODS: LVMD was assessed using speckle-tracking echocardiography in 3997 participants aged 50-64 years (51% women) from the Swedish Cardiopulmonary Bioimage Study. The upper limit of normal (ULN) for LVMD was defined …

Dexmedetomidine Exerted Anti-arrhythmic Effects in Rat With Ischemic Cardiomyopathy via Upregulation of Connexin 43 and Reduction of Fibrosis and Inflammation.

BACKGROUND: Persistent myocardial ischemia post-myocardial infarction can lead to fatal ventricular arrhythmias such as ventricular tachycardia and fibrillation, both of which carry high mortality rates. Dexmedetomidine (Dex) is a highly selective α2-agonist used in surgery for congenital cardiac disease because of its antiarrhythmic properties. Dex has previously been reported to prevent or terminate various arrhythmias. The purpose of the present study was to determine the anti-arrhythmic properties of Dex in the context of ischemic cardiomyopathy (ICM) after myocardial infarction. METHODS

Effect of septal ablation on myocardial relaxation and left atrial pressure in hypertrophic cardiomyopathy an invasive hemodynamic study.

OBJECTIVES: The objective of this study was to examine the effects of septal ablation on diastolic function with the use of invasive hemodynamics. BACKGROUND: Septal ablation is an alternative therapy for patients with obstructive hypertrophic cardiomyopathy (HCM). However, its beneficial effect on diastolic function, by relieving the systolic contraction load, may be countered by adverse effects from the infarction on left ventricular mechanics. METHODS: Using high-fidelity, micromanometer-tipped catheters, we examined 40 HCM patients by taking direct measurements of the left ventricular ou

Isoform expression of the sarcoplasmic reticulum Ca2+ release channel (ryanodine channel) in human myocardium.

The Ca2+ release channel of the sarcoplasmic reticulum (SR) is essential for the release of Ca2+ from intracellular stores and is expressed widely in various excitable cells. It plays a key role particularly in excitation contraction coupling in myocytes in skeletal and cardiac muscle. Three isoforms of the SR Ca2+ release channel have been cloned. Recently coexpression of different isoforms was reported in different animal species and various tissues. In human cardiac tissue, however, isoform expression is not yet established. Therefore the aim of this study was to characterize isoform expres

Disentangled representational learning for anomaly detection in single-lead electrocardiogram signals using variational autoencoder.

Wearable technology enables the unsupervised recording of electrocardiogram (ECG) signals. Analyzing these high-dimensional ECG data poses challenges regarding statistical approaches and explainability. This work investigates the feasibility of medically explainable anomaly detection through disentangled representational learning of ECGs and personalization to mitigate inter-subject variations. Five open-source ECG datasets were converted into a set of denoised one-second traces of lead I signal, each covering individual features such as wave morphologies and pathologies. A beta total correlat

Assessment of right ventricular functions in acromegaly: comparison of active disease with remission.

Aim    Cardiac involvement in acromegaly is defined as acromegalic cardiomyopathy, an insidious and chronic disease. Previous research on acromegalic cardiomyopathy was largely focused on morphological and functional assessment of the left heart. Since the literature data regarding right heart function in acromegalic patients are limited, we aimed to evaluate the structure and function of the right heart in such patients.Material and Methods    We included 43 adult participants as the acromegaly group and 42 individuals as the control group. All patients underwent echocardiographic evaluation.

Differences in regional diastolic function between restrictive and normal right ventricular physiology in adult patients late after tetralogy of Fallot repair.

BACKGROUND: Restrictive right ventricular physiology (RRVP) is a common finding in adult patients after tetralogy of Fallot (TOF) repair. Despite many attempts to relate this phenomenon to various factors, its clinical importance and the underlying myocardial pathologies are still enigmatic. AIM: We sought to evaluate the regional diastolic right ventricular (RV) function in patients after TOF repair with and without RRVP. METHODS: A group of 112 consecutive patients with repaired TOF underwent transthoracic echocardiography with tissue Doppler imaging, cardiopulmonary exercise test, cardiac

Alternagin-C (ALT-C), a disintegrin-like protein from Rhinocerophis alternatus snake venom promotes positive inotropism and chronotropism in fish heart.

Alternagin-C (ALT-C) is a disintegrin-like protein purified from the venom of the snake, Rhinocerophis alternatus. Recent studies showed that ALT-C is able to induce vascular endothelial growth factor (VEGF) expression, endothelial cell proliferation and migration, angiogenesis and to increase myoblast viability. This peptide, therefore, can play a crucial role in tissue regeneration mechanisms. The aim of this study was to evaluate the effects of a single dose of alternagin-C (0.5 mg kg(-1), via intra-arterial) on in vitro cardiac function of the freshwater fish traíra, Hoplias malabaricus, a

Sexual dimorphism in the control of skeletal muscle interstitial Po2 of heart failure rats: effects of dietary nitrate supplementation.

Sex differences in the mechanisms underlying cardiovascular pathophysiology of O2 transport in heart failure (HF) remain to be explored. In HF, nitric oxide (NO) bioavailability is reduced and contributes to deficits in O2 delivery-to-utilization matching. Females may rely more on NO for cardiovascular control and as such experience greater decrements in HF. We tested the hypotheses that moderate HF induced by myocardial infarction would attenuate the skeletal muscle interstitial Po2 response to contractions (Po2is; determined by O2 delivery-to-utilization matching) compared with healthy contr

Dose-dependent effects and mechanisms of exercise-like stimulation on cardiac injury and contractile function: outcomes of the MICRO-ATHLETE study.

Observational studies revealed that exercise produces cardiac troponin release and cardiac fatigue in athletes. However, human in vivo studies offer limited insights into the underlying mechanisms of these potential deleterious effects. This study investigated exercise-induced cardiac troponin release, cardiac fatigue, and its potential mechanisms in a versatile in vitro Engineered Heart Tissue (EHT) model. EHTs were subjected to exercise-like electrical pulse stimulation (EL-EPS) at 2.5 Hz (150 bpm) for 2 or 4 h and compared to non-stimulated controls and doxorubicin (DOX)-exposed tissues. Hi

Mechanistic diversity within RLC-dependent myosin ATPase inhibitors differentiates EDG-7500, a diastolic-selective cardiac sarcomere modulator.

Small molecules that modulate myofibril ATPase activity via the myosin regulatory light chain (RLC) display a broad spectrum of activity in their ability to enhance relaxation and slow contraction. EDG-7500 exhibits features consistent with a 'diastolic-selective' cardiac sarcomere modulator (d-CSM), and its ability to treat HCM was explored in engineered human tissue (EHT), human HCM cardiac strips, and an R403Q mutation swine model. In fibers, EDG-7500 preferentially inhibited myofibril ATPase activity and force at diastolic calcium levels, retained length-dependent force activation, acceler

Standardized Isolation, Culture, and Imaging of Contracting Cardiomyocytes from Neonatal Mouse Hearts.

Cardiomyocyte maturation represents the final stage of functional development in the heart. During this critical process, fetal cardiomyocytes gradually acquire adult phenotypes and become capable of sustaining billions of contraction-relaxation cycles throughout an organism's lifespan. This transition is driven by gene expression changes that remodel cardiomyocyte structure, metabolism, and functions. In mice, cardiomyocyte maturation takes place during embryonic day (E)18.5 to postnatal two weeks, during which the heart loses its regenerative capacity in response to injury. Hence, characteri

The new life of myectomy in the era of myosin inhibitors.

The management of symptomatic obstructive hypertrophic cardiomyopathy remains a therapeutic clinical challenge. Surgical septal myectomy has long been established as the gold-standard intervention for patients with persistent symptoms despite optimal medical therapy, offering durable symptom relief with low perioperative risk in experienced centers. Recently, the introduction of cardiac myosin inhibitors, such as mavacamten and aficamten, has reshaped the treatment paradigm for obstructive hypertrophic cardiomyopathy. By selectively inhibiting cardiac myosin ATPase activity, these agents reduc

Myosin-binding protein H-like nonsense variants exhibit impaired sarcomere incorporation and alter contractility.

Heart function depends on cardiomyocyte contractile apparatus and proper sarcomere protein expression. Variants in sarcomere genes cause inherited forms of cardiomyopathy and arrhythmias, including atrial fibrillation. Recently, a sarcomere component, myosin-binding protein-H like (MyBP-HL), was identified. MyBP-HL is mainly expressed in cardiac atria and is homologous to the last three C-terminal domains of cardiac myosin-binding protein-C (cMyBP-C). The MYBPHL R255X nonsense variant has been linked to atrial enlargement, dilated cardiomyopathy, and arrhythmias. Similar nonsense mutations in
