BACKGROUND: Myocardial dysfunction is a major determinant of mortality after cardiac arrest, yet the molecular events driving post-resuscitation injury remain incompletely understood. Nitric oxide (NO) has been proposed as a cardioprotective adjunct during extracorporeal life support (ECLS), but its mechanistic impact on myocardial recovery is unclear. We investigated whether NO supplementation during ECLS modulates oxidative stress, metabolic pathways, and apoptotic signaling in the post-cardiac arrest heart.
METHODS: Male Sprague Dawley rats underwent hypothermic cardiac arrest followed by

Marfan syndrome (MFS), caused by fibrillin-1 (FBN1) mutations, leads to thoracic aortic aneurysm formation through mechanisms in which redox imbalance plays an important role. Perivascular adipose tissue (PVAT) modulates vascular tone, but its role in aortic reactivity in MFS is unknown. To address this, we examined thoracic aortic reactivity using wire myography in ascending and descending aortic segments from 2 to 3-month-old male and female wild-type and Fbn1C1041G/+ mice with or without PVAT. PVAT-mediated modulation of α1-adrenergic phenylephrine-induced contractions was preserved in the

Berberine (BER), a benzylisoquinoline alkaloid, has garnered attention for its multifaceted pharmacological properties, including pronounced antioxidant, anti-inflammatory, and neuroprotective effects. Despite its therapeutic potential in neurodegenerative disorders, including Parkinson's disease, cerebral ischemia, and epilepsy, its clinical translation in Alzheimer's disease (AD) is hindered by poor aqueous solubility, limited systemic bioavailability, and restricted blood-brain barrier (BBB) permeability. This study aimed to overcome these limitations by formulating a BER-loaded nanoemulsio

Autism spectrum disorder (ASD) is a multifactorial neurodevelopmental condition characterized by impaired sociability, repetitive behaviors, and communication deficits. Animal models have been instrumental in elucidating the mechanisms underlying ASD, with prenatal exposure to valproic acid (VPA) being one of the most widely validated approaches. However, most studies rely on intraperitoneal administration, which poorly reflects human exposure. Here, we investigated the effects of oral prenatal VPA exposure in Wistar rats, focusing on behavioral outcomes, biochemical alterations, and sex-depen

BACKGROUND: Persistent oxidative stress disrupts intrahepatic immune homeostasis, leading to cytokine dysregulation and fibrogenic progression in the liver. This study investigated the immunomodulatory and antifibrotic effects of methanolic extracts of Amaranthus spinosus (leaf, stem, and root) in a thioacetamide (TAA)-induced murine model of hepatic fibrosis.
METHODS: Swiss albino mice were administered TAA (150 mg/kg, intraperitoneally, thrice weekly for 8 weeks) to induce fibrosis, followed by oral treatment with plant extracts (250 mg/kg) for 15 consecutive days. Hepatocyte-specific cytok

Background/Objectives: Neutrophils express the receptor for advanced glycation end products (RAGE), yet its role in antibacterial responses remains incompletely defined. This study aims to elucidate the dual functionality of RAGE as a membrane-bound signaling sensor and a source of soluble RAGE (sRAGE) in human neutrophils challenged with Salmonella typhimurium, a clinically relevant Gram-negative pathogen. Methods: Human peripheral neutrophils from healthy donors were isolated and stimulated with S. typhimurium, LPS, or fMLP. Calcium flux, ROS/RNS production, and phagocytosis were assessed us

UNLABELLED: Converging evidence proposed NMDA-receptor dysfunction as a real challenge underlying excitotoxicity implicated in neurological changes of autism spectrum disorder (ASD). Nevertheless, the role of NMDA receptors in relation to toll-like receptor-4 (TLR-4), NOD-like receptor-3 (NLRP3), and microglia/astrocyte activity in autism-related neuroinflammation has not been investigated hitherto.
METHODS: The present study was designed to explore the potential role of NMDA-receptor blockade in autism by chronic memantine (MEM) treatment (20 mg/kg/d, i.p.) in male Wistar rats, prenatally ex

Metformin is the first-line treatment for type 2 diabetes mellitus. Type 2 diabetes mellitus is associated with decreased nitric oxide bioavailability, which has significant metabolic implications, including enhanced insulin secretion and peripheral glucose utilization. Similar to metformin, nitric oxide also inhibits hepatic glucose production, mainly by suppressing gluconeogenesis. This review explores the combined effects of metformin and nitric oxide on hepatic gluconeogenesis and proposes the potential of a hybrid metformin-nitric oxide drug for managing type 2 diabetes mellitus. Both met

INTRODUCTION: Doxorubicin (Dox), a potent chemotherapeutic agent, is associated with cardiovascular complications. Dox-induced cardiotoxicity involves accumulation of Nε-(carboxymethyl)lysine (CML) and activation of its receptor, RAGE (receptor for advanced glycation end-products) in the cardiac microvasculature, potentially driven by NADPH oxidase (NOX)-mediated oxidative stress. NOX5 is a critical contributor to endothelial dysfunction. But its role, alongside extracellular CML, in Dox-induced vascular dysfunction remains unclear. This study investigated the impact of Dox and CML-bovine seru

INTRODUCTION: Temporomandibular disorders have a multifactorial etiology including biological, biomechanical, neuromuscular, and biopsychosocial factors. Current research on temporomandibular disorders focuses on identifying clinically relevant biomarkers thus creating a new way of thinking about this dysfunction. The aim of the study was to determine the relationship between salivary/blood concentrations of oxidative/nitrosative stress biomarkers and biopsychosocial findings in patients with temporomandibular disorder-myofascial pain with referral.
METHODS: The sample enrolled a total of 26

The anticancer, antioxidant, and immunomodulatory properties of carotenoids from saffron or apocarotenoids (e.g., crocin, safranal, crocetin, and picrocrocin) have prompted research into their benefits. Apocarotenoids seem to be effective compound for the treatment of chronic diseases, such as neurodegenerative, cardiovascular, cancer, respiratory, and metabolic disorders. Endoplasmic reticulum (ER) is an essential organelle found in the cytoplasm of eukaryotic cells that participates in the biosynthesis of proteins, lipids, and steroid hormones. BACKGROUND: Pemetrexed is combined with cisplatin to treat cancer. Whether pemetrexed-cisplatin combination chemotherapy exacerbates cisplatin nephrotoxicity is unclear. Here, we investigated kidney injury in mice administered a non-lethal low-dose regimen of pemetrexed or cisplatin alone and compared it with a pemetrexed-cisplatin combination.
METHODS: Mice were randomly divided into four groups and administered intraperitoneally the experimental drugs solubilized in captisol (sulfobutylether β-cyclodextrin). Group 1 received captisol, Group 2 pemetrexed (10 mg/kg), Group 3 cisplatin (1 mg/k

The role of nitric oxide (NO) in the pathogenesis of cerebral malaria and its cognitive sequelae remains controversial. Cerebral malaria is still the worst complication of Plasmodium falciparum infection, which is characterized by high rates of morbidity and mortality. Even after recovery from infection due to antimalarial therapy, the development of cognitive impairment in survivors reinforces the need to seek new therapies that demonstrate efficacy in preventing long-lasting sequelae. During disease pathogenesis, reactive oxygen and nitrogen species (RONS) are produced after the established

BACKGROUND: Increasing evidence suggests that glaucoma affects the ocular surface. We aimed to investigate the cellular mechanisms underlying the glaucoma-associated corneal alterations in an animal model.
METHODS: Wistar rats underwent the cauterization of two episcleral veins of the left eye to elevate the intraocular pressure (ipsilateral, G-IL). Control animals received a sham procedure (C-IL). Contralateral eyes did not receive any procedure (G-CL or C-CL). Enzymes related to the redox status, oxidative damage to macromolecules, and inflammatory markers were assessed in corneal lysates.

Tobacco use is a major cause of preventable morbidity and mortality globally. Tobacco products, including smokeless tobacco (ST), generally contain tobacco-specific N-nitrosamines (TSNAs), such as N'-nitrosonornicotine (NNN) and 4-(methylnitrosamino)-1-(3-pyridyl)-butanone (NNK), which are potent carcinogens that cause mutations in critical genes in human DNA. This review covers the series of biochemical and chemical transformations, related to TSNAs, leading from tobacco cultivation to cancer initiation. A key aim of this review is to provide a greater understanding of TSNAs: their precursors

This study targets on-site/real-time taxonomic identification and metabolic profiling of seven different Candida auris clades/subclades by means of Raman spectroscopy and imaging. Representative Raman spectra from different Candida auris samples were systematically deconvoluted by means of a customized machine-learning algorithm linked to a Raman database in order to decode structural differences at the molecular scale. Raman analyses of metabolites revealed clear differences in cell walls and membrane structure among clades/subclades. Such differences are key in maintaining the integrity and

Cortical synucleinopathies, including dementia with Lewy bodies and Parkinson's disease dementia, collectively known as Lewy body dementia, are characterized by the aberrant aggregation of misfolded α-synuclein (α-syn) protein into large inclusions in cortical tissue, leading to impairments in proteostasis and synaptic connectivity and eventually resulting in neurodegeneration. Here, we show that male and female rat cortical neurons exposed to exogenous α-syn preformed fibrils accumulate large, detergent-insoluble, PS129-labeled deposits at synaptic terminals. Live-cell imaging of calcium dyna

Cerebral ischemia is the primary basis of stroke, both sharing common pathogenic origins leading to irreversible brain damage if blood supply is not restored promptly. Existing evidence indicates that carbonic anhydrase (CA) inhibitors (CAIs) may impart therapeutic benefits against ischemia-reperfusion (I/R) pathology via the adenylyl cyclase-cyclic adenosine monophosphate (cAMP) pathway. We hypothesize that CAI and cAMP activation may enhance the therapeutic outcome against I/R conditions. In this investigation, the potential of dichlorphenamide (CAI) and the role of cAMP against ischemia-rep
