General PCR.

The primary purpose of polymerase chain reaction (PCR) is to rapidly make many copies of a specific region of DNA or RNA so that it can be adequately detected, often by agarose gel electrophoresis. PCR is routinely used to amplify, modify, and clone genes for expression studies. There are many other applications for PCR, including paternity testing, biological relationships, mouse genotyping, diagnosing genetic diseases, forensics, and finding bacteria and viruses.

Identical twin parents; research reviews: twin pregnancy risk factors, a new twin type and a school legislation update; twin parents and twin researchers in the news.

Some legal and social complications that arise when identical twins have children are examined. The specific case presented concerns assignment of paternity. This section is followed by a review of recent studies of multiple birth pregnancy risk factors, namely mother to infant HIV transmission and congenital hypothyroidism. Honors given to recently recognized twin researchers are noted, as is the birth of twins to an older mother in the United States.

A triploid partial mole placenta from paternal isodisomy with a diploid fetus derived from one sperm and one oocyte may have caused angiogenic imbalance leading to preeclampsia-like symptoms at 19 weeks of gestation.

Partial hydatidiform mole with a normal fetus is extremely rare. A 30-year-old woman presented at 19 weeks gestation with clinical manifestations of severe preeclampsia. The fetus revealed a normal 46,XX karyotype and the placenta revealed triploid 69,XXX from paternal isodisomy. Microsatellite analysis revealed that the fetus and the triploid partial mole were derived from one sperm and one oocyte, followed by duplication of paternal chromosomes in only a trophectodermal cell. The maternal serum levels of angiogenic factors were extremely high compared with those reported in preeclampsia, sug

"Crying on the Bus": First Time Fathers' Experiences of Distress on Their Return to Work.

There is increasing research interest in the experiences of new fathers taking paternity leave, but less insight into men's experiences of returning to work after the birth of their first baby. For many men in the UK context, this could take place immediately after the birth or after one or two weeks of paternity leave. This paper utilizes data from a UK-based study whilst also drawing on international literature and policy contexts. A constructivist grounded theory method was adopted to generate theory from the data gathered. Twelve new fathers shared their experiences in this study by partic…

Experimental extra-pair copulations provide proof of concept for fertility insurance in a socially monogamous bird.

Extra-pair paternity is common among socially monogamous birds, but whether females benefit from having extra-pair copulations remains unresolved. In this study, I staged single extra-pair copulations in captive pairs of socially monogamous Japanese quail (Coturnix japonica) and used extra-pair males of a different colour to identify extra-pair fertilizations. This eliminated among-female variation in extra-pair copulations as a source of variation in extra-pair fertilizations and tested whether a single extra-pair copulation would insure against infertility of the social male. Overall, the pr…

Risks of preterm birth and growth restriction in second births after a first-born male infant.

PURPOSE: A first-born male infant may lead to an exaggerated immune response in the mother in subsequent pregnancies, increasing the risk of preterm birth (PTB) and growth restriction. This study investigated whether risks of PTB and growth restriction are greater among infants preceded by a first-born male infant and evaluated if the associations differ by paternity change or the second-born's sex. METHODS: We conducted a population-based cohort study using 2003-2014 Washington State birth certificate data and included mothers and birth year frequency-matched second live-born infants precede…

Female haemophilia A in a family with seeming extreme bidirectional lyonization tendency: abnormal premature X-chromosome inactivation?

We studied a female child with mild classical haemophilia A, presenting with a F VIII deficiency similar to that detected in her maternal gr … m the hemizygous grandfather to both his daughters and further to the haemophilic female child. The paternity of the child was analyzed using various red cell and HLA antigens and RFLP by p29C, a probe detecting polymorphic hypervariable TaqI and PstI fragments in the pseudoautosomal areas of the X- and Y-chromosomes. All results obtained were concordant wi …

Longer and faster sperm exhibit better fertilization success in Japanese quail.

In birds, sperm storage tubules (SST) located in the utero-vaginal junction are thought to be a site of sperm selection; however, the exact mechanism of sperm selection is poorly understood. Here, we investigated sperm entry into the SST and subsequent fertilization success under a competitive situation created by artificial insemination of a sperm mixture obtained from 2 males. We employed 2 quail strains, a wild-type and a dominant black (DB) type, as this allows easy assessment of paternity by feather coloration. We found paternity of embryos was biased toward DB males when a sperm mix with…

Extreme fertilization bias towards freshly inseminated sperm in a species exhibiting prolonged female sperm storage.

The storage of sperm by females across successive reproductive cycles is well documented in internal fertilizers, yet the fate of stored spe … fects (e.g. behaviourally mediated cryptic female choice) and specifically test for post-copulatory paternity biases that favour either stored or fresh sperm when they compete to fertilize eggs. Our paternity analyses revealed the almost complete dominance of freshly inseminated sperm over stored sperm, supporting previous studies reporting similar patterns …

Reproduction and marriage among male survivors of cancer in childhood, adolescence and young adulthood: a national cohort study.

BACKGROUND: Increased survival after cancer in young age has made long-term follow-up studies of high external validity important. In this national cohort study, we explored the impact of cancer in young age on reproduction and marital status in male survivors. METHODS: Hazard ratios (HRs) and relative risks (RRs) of reproductive and marital outcomes were studied for male survivors of cancer in young age (<25 years) and cancer-free male comparisons, born during 1965-1985, by linking compulsory national registries in Norway. RESULTS: Male cancer survivors (n=2687) had reduced paternity (HR: 0…

Two affected siblings with nuclear cataract associated with a novel missense mutation in the CRYGD gene.

PURPOSE: To identify the disease locus for nuclear congenital cataract in a nonconsanguineous family with two affected members. METHODS: One family with two affected members with congenital cataract and 170 normal controls were examined. DNA from leukocytes and bucal swabs was isolated to analyze the CRYGA-D cluster genes and microsatellite markers D2S325, D2S2382, and D2S126, and to discard paternity through gene scan with several highly polymorphic markers. RESULTS: DNA sequencing analysis of the CRYGA-D cluster genes of the two affected members showed a novel heterozygous missense mutatio…

Molecular pathology in forensic medicine--Introduction.

Techniques of molecular biology have improved diagnostic sensitivity, accuracy and validity in forensic medicine very much, especially in the field of identification (paternity testing, stain analysis). Since more than 10 years these techniques - meanwhile well established in clinical disciplines - are used also for other applications in forensic medicine: determination of cause and manner of death, tissue identification by mRNA and microRNA, examination of gene expression levels (survival time, time since death, cause of death), toxicogenetics.

Evidence against the structural gene encoding type II collagen (COL2A1) as the mutant locus in achondroplasia.

The structure of the locus encoding the major cartilage collagen gene (COL2A1) was studied in a total of 19 cases of achondroplasia. No gross rearrangements were seen. The segregation of COL2A1 was examined in three affected kindreds using restriction site and length variants as genetic markers. In two kindreds discordant segregation between the achondroplasia and COL2A1 loci was demonstrated. Paternity/maternity was confirmed using a 'minisatellite' core sequence probe which reveals cross hybridising polymorphic loci.

Fertility in cryptorchidism. Does treatment make a difference?

The testis which remains nonscrotal beyond puberty will not produce sperm. While much of the data presented imply that therapy of cryptorchi … efore, verification is needed to determine whether or not fertility is decreased in cryptorchidism. Paternity is a better index for verification than sperm counts since it is known that men with subnormal sperm counts may have normal paternity rates. Such verification using paternity may require more than fathering a child, such as the age of the father at …

Risk of non-reproductive cancer in men from couples conceiving through assisted reproduction: a Swedish nation-based register study.

Whereas the link between impaired male fertility and risk of testicular and prostate cancer is well established, studies investigating non-reproductive cancer risk among men with impaired reproductive function are scarce and show conflicting results. The aim of the study was to compare the risk of developing non-reproductive cancers in men achieving paternity through assisted reproduction as compared to those conceiving naturally. All first-time fathers n = 1 137 829 in Sweden during the period January 1994 to December 2014 were included and followed from the time of conception until date of c…

Fertility of Cryptorchid Testis-An Unsolved Mistery.

Cryptorchidism (undescended testis) is one of the most common diagnoses in the pediatric urologist office. Even in the modern era, there still are a lot of debates regarding the optimal time for surgery related to the expected results in relation with the testicular function, including fertility. The review below intends to clarify issues regarding the impact of cryptorchidism on testicular histology and function, semen analysis, the relation between hormonal and surgical treatment, future fertility, and paternity rate.

Cryptorchidism and Fertility.

Congenital cryptorchidism, that is, undescended testis, is one of the most common urogenital abnormalities observed in newborn boys. In addition to the congenital form, there is also acquired form of cryptorchidism. Fertility potential of patients with cryptorchidism has been evaluated by testicular histology and volume, semen quality, reproductive hormone levels, time to conception, and paternity rates. Cryptorchidism is associated with abnormalities in testicular development, and early treatment is recommended to optimize the fertility potential of the patients.

The controversy regarding the need for hormonal treatment in boys with unilateral cryptorchidism goes on: a review of the literature.

Hormonal treatment for unilateral undescended testes continues to be recommended in some countries. We reviewed the literature in favor and against this recommendation. Since the paternity rate of men with a history of unilateral undescended testes only treated with surgery is normal, the effectiveness of hormonal treatment to produce testicular descent is low, the cost is considerable, and there are potential adverse effects, hormonal treatment for boys with unilateral undescended testes should no longer be recommended.

Long-term follow-up of fertility in cryptorchid patients.

Sixty-four patients who had unilateral or bilateral orchiopexy for cryptorchism between 1950 and 1960 responded to a questionnaire. Marriage rates were the same as in a control group. Unilaterally cryptorchid patients had normal paternity rates and sperm concentrations on semen analysis. Only 2 of 15 bilaterally cryptorchid patients had fathered children, while severe oligospermia was present in all 10 who had semen analysis performed. Thirty per cent of pexed testes had a cosmetically poor result.
