Hypoxic-Ischaemic Encephalopathy and the Blood-Brain Barrier in Neonates.

This review aims to highlight a possible relationship between hypoxic-ischaemic encephalopathy (HIE) and the disruption of the blood-brain b … e is increased understanding of the contribution of the components of the NVU, e.g., astrocytes and pericytes, to the maintenance of this physiological barrier. We also explore the development of therapeutic options in HIE, such as harnessing the transport systems in the BBB, to enable the delivery of large molecules with molecular Trojan horse technology, …

Extracellular vesicles of the blood-brain barrier: Role in the HIV-1 associated amyloid beta pathology.

HIV-infected brains are characterized by increased amyloid beta (Aβ) deposition. It is believed that the blood-brain barrier (BBB) is critic … red to controls. Interestingly, brain endothelial cell-derived ECV transferred Aβ to astrocytes and pericytes. Infusion of brain endothelial ECV carrying fluorescent Aβ into the internal carotid artery of mice resulted in Aβ fluorescence associated with brain microvessels and in the brain parenchyma. These results suggest that ECV carrying Aβ can be success …

Chronic Hypertension Leads to Neurodegeneration in the TgSwDI Mouse Model of Alzheimer's Disease.

Numerous epidemiological studies link vascular disorders, such as hypertension, diabetes mellitus, and stroke, with Alzheimer's disease (AD). Hypertension, specifically, is an important modifiable risk factor for late-onset AD. To examine the link between midlife hypertension and the onset of AD later in life, we chemically induced chronic hypertension in the TgSwDI mouse model of AD in early adulthood. Hypertension accelerated cognitive deficits in the Barnes maze test (P<0.05 after 3 months of treatment; P<0.001 after 6 months), microvascular deposition of β-amyloid (P<0.001 after 3 months o

Angiogenic factors in bone local environment.

Angiogenesis plays an important role in physiological bone growth and remodeling, as well as in pathological bone disorders such as fracture repair, osteonecrosis, and tumor metastasis to bone. Vascularization is required for bone remodeling along the endosteal surface of trabecular bone or Haversian canals within the cortical bone, as well as the homeostasis of the cartilage-subchondral bone interface. Angiogenic factors, produced by cells from a basic multicellular unit (BMU) within the bone remodeling compartment (BRC) regulate local endothelial cells and pericytes. In this review, we discu…

Morphological aspects of the euphonic voice.

The high quality of a euphonic voice is the result of complex interactions between many organs and systems. Vibrating vocal folds play a cru … roma. Unchanged epithelial cells were situated on the basal membrane with folds. Moreover, numerous pericytes, vessels with multiplication of basal membranes, scanty collagenous fibers, plasmatic cells and lymphocytes were seen. Morphological changes with signs of atrophy and polypoid degeneration of the vocal fold mucosa were found in only 3 (15%) patients …

Angiogenesis and experimental hepatic fibrosis.

Angiogenesis is a basic change occurring during repair by granulation tissue. This process seems to precede fibrosis formation in most types … hepatica model due to circumstances to be discussed. Special attention was paid to the presence of pericytes and to their tendency to become detached from the vascular wall and be transformed into myofibroblasts, which is a sequence of events that explains the decisive role angiogenesis plays in fibrosis.

Thalidomide stimulates vessel maturation and reduces epistaxis in individuals with hereditary hemorrhagic telangiectasia.

Hereditary hemorrhagic telangiectasia (HHT) is an inherited disorder characterized by vascular malformations. Many affected individuals deve … y reversed by pharmacological or genetic interference with PDGF signaling from endothelial cells to pericytes. Biopsies of nasal epithelium from individuals with HHT treated or not with thalidomide showed that similar mechanisms may explain the effects of thalidomide treatment in humans. Our findings demonstrate the ability of thalidomide to induce vessel m …

Vascular development: genetic mechanisms and links to vascular disease.

Vertebrate development depends on the formation of intricate vascular networks at numerous sites and in precise patterns; these vascular networks supply oxygen and nutrients to the rapidly expanding tissues of the embryo. Embryonic blood vessels are composed of endothelial cells and pericytes that organize and expand into highly branched conduits. Proper development of the vasculature requires heterogeneity in the response of endothelial cells to angiogenic cues provided by other tissues and organs. The pathogenesis of vascular diseases results from genetic mutations in pathways that provide t…

[Pathogenesis of diabetic retinopathy].

Diabetic retinopathy (DR) develops in patients with both type 1 and type 2 diabetes and is the major cause of vision loss and blindness in t … PK or NF-kappaB. In diabetes, damage to the retina occurs in the vasculature (endothelial cells and pericytes), neurons and glia, pigment epithelial cells and infiltrating immunocompetent cells: monocytes, granulocytes, lymfocytes. These activated cells change the production pattern of a number of mediators such as growth factors, vasoactive agents, coagula …

Nerve growth factor prevents both neuroretinal programmed cell death and capillary pathology in experimental diabetes.

BACKGROUND: Chronic diabetes causes structural changes in the retinal capillaries of nearly all patients with a disease duration of more than 15 years. Acellular occluded vessels cause hypoxia, which stimulates sight-threatening abnormal angiogenesis in 50% of all type I diabetic patients. The mechanism by which diabetes produces acellular retinal capillaries is unknown. MATERIALS AND METHODS: In this study, evidence of programmed cell death (PCD) was sought in the retinas of early diabetic rats, and the effect of nerve growth factor (NGF) on PCD and capillary morphology was evaluated. HLA DR and DQ distribution in normal human ocular structures.

HLA DR and DQ distribution was investigated in normal human ocular tissues, together with class I antigens and immunocompetent cell subsets, … the sites of endothelial cells. The characterization of those cells, which could be hypothetized as pericytes needs further studies but suggests close relationships between neuroretina and the immune system. This study may provide insight in the implication of the immune system in many poorly understood ocular diseases.

[Transmission electron microscopic studies of the organization of the ependymocytes in the central canal of the spinal cord of Cercopithecus nigroviridis (Pocock 1907), (Platyrrhina, Cercopithecoidea)].

The ependyma of the central canal of the spinal cord of the monkey Cercopithecus nigroviridis was examined by transmission electron microscopy. In the lumbar region and in the filum terminale, many cytoplasmatic protrusions are visible. They are irregular in size and shape and display many microvilli. They are extending into the lumen of the central canal. The basal parts of the ependymocytes occasionally have a very close association with the ependymal blood vessels. The pericapillary space, the pericapillary structures like pericytes and collagen fibrils, and the basal lamina are absent. Opp…

[Kaposi's sarcoma. A vascular neoplasm of presumably viral origin. Histologic and ultrastructural characteristics].

Kaposi's sarcoma is a multiform mesenchymal neoplasm of uncertain histogenesis but of possible viral origin. Several anatomo-clinical forms have been described the cutaneous nodular lesion being the most common and at highest frequency in equatorial Africa. Eighteen of such typical lesions were reported; their morphology was uniform and associated: 1) spindle cells, 2) an excess of vascular spaces composed of intratumoral intercellular slits and of intra and peritumoral well structured vessels, 3) a lymphoplasmocytic inflammatory infiltration. The lesions of the other and less common forms of

Transmitter histochemistry of the rat olfactory bulb III. Autoradiographic localization of [3H]GABA.

The distribution of [3H]gamma-aminobutyric acid (GABA) labeled elements in rat olfactory bulb was studied by light and electron microscopic autoradiography. [3H]GABA was strongly taken up into glial cells and pericytes in all layers of the bulb. The neuronal uptake of [3H]GABA was mainly seen in certain types of nerve terminals. About one-third of the granule dendritic terminals, some nerve endings of short axon cells, and certain nerve endings of extrabulbar origin showed a strong labeling. Labeling was seen in a small population of the periglomerular, short axon and granule cell bodies. Most…

The effect of injury on two hydrolases in the hypoglossal nucleus, with quantitative data on N-acetyl-beta-glucosaminidase.

N-acetyl-beta-glucosaminidase and acid phosphatase were studied histochemically in sections of hypoglossal nuclei of normal adult male albin … eurones at 18 days after axotomy. There was no change in the amount of reaction product per cell in pericytes, astrocytes, or non-astrocyte glia. Sections scanned quantitatively showed that normal and operated nuclei had a similar amount of reaction product, thus the neuronal diminution must be balanced by the increase elsewhere, either in the elevated numb …

Near-Infrared-Triggered "Bridge-and-Attack" Strategy via a Bioinspired Copper-Polyphenol Nanoarchitectonics Platform for Vascular Normalization-Enhanced Cuproptosis-Immunotherapy of Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) presents formidable treatment barriers due to dysfunctional vasculature and an immunosuppressive microenvironment. To address these challenges, we engineered a bioinspired near-infrared (NIR)-responsive copper-polyphenol nanoplatform, SCP, to implement a NIR-triggered "bridge-and-attack" therapeutic strategy. This nanoassembly was constructed through the coordination of salvianolic acid B (SAB) with copper ions and further stabilized by a polydopamine (PDA) shell. Upon NIR irradiation, SAB and Cu2+ were co-released from SCP, enabling simultaneous vascular r

Inflammation-centered neurovascular-immune-metabolic remodeling in ischemic stroke: stage-dependent mechanisms, regulated cell death, and therapeutic translation.

Ischemic stroke evolves beyond arterial occlusion through an inflammation-centered network linking neurovascular dysfunction, immune remodeling, metabolic reprogramming, and regulated cell death. These interactions are organized across the hyperacute, acute, subacute, and chronic phases. Hyperacute energy failure, excitotoxicity, thromboinflammation, pericyte contraction, and capillary stalling can sustain microcirculatory no-reflow despite recanalization. Acute injury is characterized by blood-brain barrier disruption, innate immune amplification, mitochondrial stress, as well as ferroptotic,
