Sex-based differences in the severity of radiation-induced arthrofibrosis.

As cancer survivorship increases, so does the number of patients that suffer from the late effects of radiation therapy. This includes arthrofibrosis, the development of stiff joints near the field of radiation. Previous reports have concentrated on skin fibrosis around the joint but largely ignored the deeper tissues of the joint. We hypothesized that fat, muscle, and the joint tissues themselves would play a more significant role in joint contracture after radiation than the skin surrounding the joint. To address this hypothesis, we irradiated the right hind flanks of mice with fractionated

National Cancer Institute Workshop on Proton Therapy for Children: Considerations Regarding Brainstem Injury.

PURPOSE: Proton therapy can allow for superior avoidance of normal tissues. A widespread consensus has been reached that proton therapy should be used for patients with curable pediatric brain tumor to avoid critical central nervous system structures. Brainstem necrosis is a potentially devastating, but rare, complication of radiation. Recent reports of brainstem necrosis after proton therapy have raised concerns over the potential biological differences among radiation modalities. We have summarized findings from the National Cancer Institute Workshop on Proton Therapy for Children convened i

Pharmacological alteration of the lung vascular response to radiation.

The role of endothelial cell damage in the development of radiation injury in the lung was investigated in rats. Vascular permeability-surface area product (PS) was measured as an indicator of the degree of endothelial cell damage in lungs of rats exposed to single dose hemithorax irradiation. Hemithorax irradiation was chosen to simulate clinical radiotherapy, in which only a portion of the lung is irradiated. In addition, it provided a control lung to compare to the irradiated lung. Radiation is postulated to lead to activation of several different biochemical pathways that result in lung in

Management of postirradiation sepsis.

Ionizing radiation depresses the immune defenses and enhances susceptibility to local and systemic infection due to endogenous or exogenous microorganisms. Exposure to a lethal dose of ionizing cobalt-60 gamma radiation induces a dose-related reduction in the number of both aerobic and anaerobic bacteria from 10(10-12) to 10(4-6) per gram of stool within 4 days. Whereas the number of anaerobic bacteria stays low, the number of Enterobacteriaceae per gram of stool increases significantly--up to 10(9) by the 12th day following irradiation. This increase is associated with bacterial translocation

An injectable Ce-MnCo LDH nanozyme gel with cascade catalytic activity for acute radiation proctitis in rats.

Acute radiation proctitis (ARP) is a common complication of pelvic radiotherapy driven by direct DNA damage and excessive reactive oxygen species (ROS) induced oxidative stress. Currently, effective clinical treatments that can systematically intervene in ARP are lacking. In this study, we developed an injectable, multicenter cascade nanozyme gel (Ce-MnCo LDH gel) for intrarectal administration to combat radiation-induced intestinal injury. By atomically integrating cerium (Ce) into the lattice of manganese-cobalt layered double hydroxides (MnCo LDH), we constructed a complex multicenter elect

Thrombopoietin Receptor Agonist Mitigates Hematopoietic Radiation Syndrome and Improves Survival after Whole-Body Ionizing Irradiation Followed by Wound Trauma.

Ionizing radiation combined with trauma tissue injury (combined injury, CI) results in greater mortality and H-ARS than radiation alone (radiation injury, RI), which includes thrombocytopenia. The aim of this study was to determine whether increases in numbers of thrombocytes would improve survival and mitigate H-ARS after CI. We observed in mice that WBC and platelets remained very low in surviving RI animals that were given 9.5 Gy 60Co-γ-photon radiation, whereas only lymphocytes and basophils remained low in surviving CI mice that were irradiated and then given skin wounds. Numbers of RBC a

Cardiovascular Complications of Cranial and Neck Radiation.

Cancer survivors who receive head and neck radiation are at increased risk for cerebrovascular events. This is mediated via damage to the hypothalamus-pituitary axis leading to the metabolic syndrome and extracranial arterial injury leading to carotid artery stenosis. Head and neck radiation can also lead to intracranial injury that can present as moyamoya, especially in children. Survivors require lifelong periodic follow-up for the development of pan-hypopituitarism or its individual components as well as for dyslipidemia and obesity. Aggressive control of traditional cardiovascular risk fac

Risk of carotid blowout after reirradiation of the head and neck: a systematic review.

PURPOSE: Carotid blowout (CB) is a rare but frequently fatal complication of head-and-neck (H&N) cancer or its treatment. We sought to determine the reported rate of CB in patients receiving salvage reirradiation for H&N cancer. METHODS AND MATERIALS: A literature search identified 27 published articles on H&N reirradiation involving 1554 patients, and a pooled analysis was performed to determine the rate of CB. Treatment parameters, including prior radiation dose, interval from prior radiation, dose and fractionation of reirradiation, use of salvage surgery, and chemotherapy, were abstracted

Targeting Kupffer Cell CD44-Mediated Ammonia Death via the ELAVL1-GLS Metabolic Circuit in Radiation-Induced Liver Disease.

Radiation-induced liver disease (RILD) remains a major dose-limiting toxicity in liver cancer radiotherapy. However, the underlying mechanisms, particularly the immunometabolic reprogramming that sustains inflammatory amplification following irradiation, are still poorly understood. Here, we report that Kupffer cell (KC) depletion significantly attenuated radiation-induced liver inflammation and injury. Irradiation markedly upregulated CD44 expression in KCs, and single-cell RNA sequencing revealed a CD44⁺ macrophage subset associated with injury, inflammatory activation, and metabolic stress
