Suspended particles for omnidirectional template sacrifice for rapid vascular patterning within engineered tissues.

Engineered tissues offer great promise for therapeutic organ repair. Yet, biofabrication of organ-scale tissues can take hours or days for large constructs with geometric complexity, during which living cells in the construct lack vascular support. To address this, we developed an approach to accelerate biofabrication by embedding microreservoirs of evacuation reagents throughout geometrically patterned sacrificial agents. We call this approach Suspended Particles for Omnidirectional Template Sacrifice (SPOTS). We show that microparticles can serve as both structural impurities as well as depo

Modeling ventricular tachycardia in genetic long QT syndrome using engineered cardiac tissues.

Congenital long QT syndrome (LQTS) predisposes patients to malignant ventricular tachycardias, yet existing in vitro models do not fully capture the clinical arrhythmic phenotypes that determine patient outcomes. We report the development of an engineered platform with mature, fully hiPSC-derived human cardiac tissues that reproduces genotype-specific ventricular tachycardia under physiologically relevant triggers. Using patient-derived and corrected hiPSCs, we engineered electromechanically matured cardiac tissues to model LQT8 and LQT2 syndromes. LQT8 tissues exhibited spontaneous early afte

Mid-Term Durability of Tissue-Engineered Vascular Graft Patches for Pulmonary Artery Reconstruction.

Autologous pericardium is commonly used for pulmonary artery (PA) reconstruction in congenital heart surgery but is not always available. Synthetic materials lack growth potential and may cause calcification or infection. Tissue-engineered vascular grafts (TEVGs), created by in-body tissue architecture, may overcome these limitations. This study evaluated mid-term outcomes of PA reconstruction using TEVG patches. From July 2014 to December 2024, TEVG molds were implanted in 19 patients; seven underwent PA reconstruction using 15 patches. Diagnoses included pulmonary atresia with ventricular se

Engineering heterogeneous tissues and organs via multi-material bioprinting: Advances, challenges, and opportunities.

Native tissues and organs exhibit intrinsic heterogeneity in their structural, compositional, and functional properties, which poses significant challenges towards the development of effective functional substitutes. Multi-material three-dimensional (3D) bioprinting has emerged as a promising strategy to address these challenges by enabling enhanced spatial control over the deposition of biomaterials, signaling factors, and diverse cell types. This capability facilitates the fabrication of heterogeneous tissue constructs that aim to approach the complexity of native tissues, although achieving

From injury to recovery: investigating wound healing in a 3D tissue-engineered cornea equivalent.

The semi-automatic generation of a multilayer cornea model was to study the process of wound healing and the influence of substances modulating wound healing. We generated in vitro corneal human tissues including epithelium and a semi automatedly produced stroma equivalent based on a hydrogel matrix. The maturation was evaluated using impedance spectroscopy, histology and immunofluorescence over 21 days. Inflicting corneal wounds by clinical excimer laser, injury was observed under treated with gentamicin and non-treated for 14 days by histology and optical coherence tomography (OCT). The semi

Short- and Medium-Term Surgical Outcomes of Tissue-Engineered Pulmonary Valve Replacement in Sheep.

BACKGROUND: Tissue-engineered pulmonary valves (TEPVs) hold considerable potential for improving outcomes in valve replacement surgeries. We investigated the surgical outcomes of TEPVs replacement in sheep, specifically examining the effects of valve type (decellularized versus adipose-derived stem cell-seeded valve [ADSC]) and the animal's age at the surgery. The primary goals were to assess survival rates, postoperative complications, and the effects of cardiopulmonary bypass (CPB) on homeostasis. METHODS: Nineteen juvenile and adult sheep were randomly assigned to orthotopic pulmonary valv

Development and functional characterization of a tissue-engineered blood-air barrier model for in vitro applications.

BACKGROUND: The blood-air barrier (BAB) of the lung is a critical interface responsible for gas exchange and protection against external attempts, and acts as a selective barrier. Developing in vitro models that replicate its structural and functional properties is essential in studying pulmonary diseases and their therapy. METHODS: In this study, a model consisting of alveolar epithelial (A549) and primary endothelial (pHUVEC) cells seeded on opposite sides of a thin (11 ± 4 μm), electrospun poly(ε-caprolactone) mesh of nanofibers (140-800 nm) to represent the basal membrane, and the interst

Method for Targeted Cellular Seeding of Tubular Tissue-Engineered Scaffolds for Tracheal Regeneration Approaches.

Effective tracheal tissue engineering benefits from scaffolds that mimic the native structure of the tissue, provide mechanical stability, and support spatially controlled cell seeding to encourage tissue regeneration. This study presents a novel approach for fabricating tubular scaffolds for tracheal regeneration that integrates a 3D-printed polycaprolactone (PCL) backbone with a freeze-dried collagen-hyaluronic acid (CHyA) layer. Two scaffold geometries (tubular and c-shaped) were produced and mechanically characterized, and it was demonstrated that PCL reinforcement significantly enhanced s…

Non-invasive longitudinal monitoring of vascularization in tissue-engineered grafts by ultrasound localization microscopy.

Tissue engineering is a rapidly advancing field aimed at regenerating damaged tissues by combining cells, biomaterials, and bioactive molecules. Adequate vascularization is a critical determinant of successful tissue regeneration, as it supports both the regenerative process and the maintenance of physiological functions in the newly formed tissue. Traditional methods for evaluating vascularization, such as histological and immunohistochemical techniques, are inherently invasive and limited to endpoint analysis. In this study, we innovatively explored the potential of ultrasound localization m…

Regeneration Process of an Autologous Tissue-Engineered Trachea (aTET) in a Rat Patch Tracheoplasty Model.

The treatment of long-tracheal lesion is difficult because there are currently no viable grafts for tracheal replacement. To solve this problem, we have developed an autologous Tissue-Engineered Trachea (aTET), which is made up of collagenous tissues and cartilage-like structures derived from rat chondrocytes. This graft induced successful long-term survival in a small-animal experiment in our previous study. In this study, we investigated the regeneration process of an aTET to attain reproducible success. We prepared an aTET by using a specially designed mold and performed patch tracheoplasty

Repair of complex ovine segmental mandibulectomy utilizing customized tissue engineered bony flaps.

Craniofacial defects require a treatment approach that provides both robust tissues to withstand the forces of mastication and high geometric fidelity that allows restoration of facial architecture. When the surrounding soft tissue is compromised either through lack of quantity (insufficient soft tissue to enclose a graft) or quality (insufficient vascularity or inducible cells), a vascularized construct is needed for reconstruction. Tissue engineering using customized 3D printed bioreactors enables the generation of mechanically robust, vascularized bony tissues of the desired geometry. While…

Development of tissue-engineered tracheal scaffold with refined mechanical properties and vascularisation for tracheal regeneration.

Introduction: Attempted tracheal replacement efforts thus far have had very little success. Major limiting factors have been the inability to efficiently re-vascularise and mimic the mechanical properties of native tissue. The major objective of this study was to optimise a previously developed collagen-hyaluronic acid scaffold (CHyA-B), which has shown to facilitate the growth of respiratory cells in distinct regions, as a potential tracheal replacement device. Methods: A biodegradable thermoplastic polymer was 3D-printed into different designs and underwent multi-modal mechanical assessment.

Exploring the Pathophysiology of Delirium: An Overview of Biomarker Studies, Animal Models, and Tissue-Engineered Models.

Delirium is an acute brain disorder associated with disorganized thinking, difficulty focusing, and confusion that commonly follows major surgery, severe infection, and illness. Older patients are at high risk for developing delirium during hospitalization, which may contribute to increased morbidity, longer hospitalization, and increased risk of institutionalization following discharge. The pathophysiology underlying delirium remains poorly studied. This review delves into the findings from biomarker studies and animal models, and highlights the potential for tissue-engineered models of the b

Functional tissue-engineered microtissue formed by self-aggregation of cells for peripheral nerve regeneration.

BACKGROUND: Peripheral nerve injury (PNI) is one of the essential causes of physical disability with a high incidence rate. The traditional tissue engineering strategy, Top-Down strategy, has some limitations. A new tissue-engineered strategy, Bottom-Up strategy (tissue-engineered microtissue strategy), has emerged and made significant research progress in recent years. However, to the best of our knowledge, microtissues are rarely used in neural tissue engineering; thus, we intended to use microtissues to repair PNI. METHODS: We used a low-adhesion cell culture plate to construct adipose-der…
