Material-Encoded Synchronization of Immunogenic Cell Death With Adenosine A2A Receptor Blockade Reprograms the Tumor Microenvironment.

Adenosine rapidly suppresses antitumor immunity through the adenosine A2A receptor (A2AR). Immunogenic cell death (ICD) releases extracellular ATP, which can be rapidly converted into immunosuppressive adenosine. We therefore designed BSCS@PHY to synchronize ICD induction with local A2AR blockade in the same spatiotemporal window, aiming to protect antigen priming from adenosine-mediated suppression. BSCS@PHY integrates a bismuth-copper diselenide core for photothermal heating, glutathione depletion, and chemodynamic hydroxyl-radical generation; hyaluronic-acid-modified phase-change materials

CAR T cells engineered against LAG-3: Unleashing potency in the tumor microenvironment of breast cancer.

Chimeric antigen receptor (CAR) T cell therapy has achieved remarkable success in hematologic malignancies but faces significant challenges in solid tumors such as breast cancer. A primary obstacle is the immunosuppressive tumor microenvironment (TME), which drives T-cell exhaustion and limits therapeutic efficacy. Lymphocyte-activation gene 3 (LAG-3) is a key mediator of this exhaustion, suppressing antitumor immunity upon engagement with ligands such as MHC class II. This review examines the rationale for targeting the LAG-3 pathway to enhance CAR T cell potency within the breast cancer TME.…

Tunable Technologies for the Glioma Tumor Microenvironment: A Comprehensive Review on Bench-to-Bedside Neurosurgical Advances.

Gliomas remain among the most treatment-resistant malignancies of the central nervous system. Glioblastoma (GBM), the most aggressive adult-type diffuse glioma, is associated with persistently poor survival despite maximal safe resection followed by chemoradiation. Gliomas do not grow in isolation. Work over the past twenty years has dismantled the older tumor-centric view of glioma biology, replacing it with a model in which malignant cells operate within a tumor microenvironment (TME) composed of immune, vascular, stromal, and neural elements that together govern disease behavior. What makes…

Unraveling the role of glutamine metabolism in cancer: from cell death mechanisms to tumor microenvironment modulation.

Glutamine, the most abundant amino acid in human serum, serves not only as a key nutrient for maintaining normal cellular biological functions but also as an essential metabolic substrate for the proliferation and survival of tumor cells. Tumor cells exhibit a pronounced characteristic of "glutamine addiction" through metabolic reprogramming. This trait endows tumor cells with the dual capacity to exhibit both adaptability and vulnerability when confronted with regulated cell death (RCD) under metabolic stress, thereby opening up potential avenues for cancer therapy. This paper systematically

Urinary proteome and metabolome uncover tumor microenvironment and cellular metabolism changes of renal clear cell carcinoma.

BACKGROUND: Clear cell renal carcinoma (ccRCC) is the most frequent form of kidney tumors with high recurrence and progression rates. Early diagnosis of ccRCC could significantly improve survival rate. Liquid biopsies could capture molecular information which would not only shed more light on the signatures of the onset of ccRCC, but also discover potential biomarker for early diagnosis. METHOD: We applied LC-MS to profile the urine proteome and metabolome of 314 ccRCC, 341 healthy control and 49 kidney benign disease enrolled from three cohorts. Further cell origin annotation and protein-pro

Assessment of Tumor Microenvironment Inflammatory Biomarkers in Metastatic Renal Cell Carcinoma Treated With Immune Checkpoint Inhibitors.

INTRODUCTION: Therapies using immune checkpoint inhibitors (ICI) are standard of care in metastatic renal cell carcinoma (mRCC). Currently, no accepted standardized biomarkers are available to predict treatment response in mRCC. We aimed to identify the predictive value of immunomodulatory markers interacting within the tumor microenvironment. PATIENTS AND METHODS: We included 45 untreated and pretreated patients treated with ICI, divided by their progression free survival (PFS) into groups of good (> 18 months), intermediate (6-18 months) or poor treatment response (< 6 months). Tumor specim…

NFE2L2-Associated Ferroptosis Resistance Reshapes the Tumor Immune Microenvironment and Guides Therapeutic Strategies in Prostate Cancer.

Prostate adenocarcinoma (PRAD) poses a significant challenge due to therapy resistance and an immunosuppressive tumor microenvironment (TME). Ferroptosis has emerged as a therapeutic vulnerability, yet its immunomodulatory role in PRAD remains elusive. Here, we employed a multi-omics approach-integrating bulk RNA-seq (498 tumors), single-cell RNA-seq (68,322 cells), and spatial transcriptomics (19,483 spots)-to decode the ferroptosis-immune landscape. We derived a robust 16-gene ferroptosis signature that predicted biochemical recurrence (C-index = 0.76) and validated it in two independent coh…

The tumor microenvironment of cutaneous squamous cell carcinoma in high-risk patient groups: A scoping review.

Cutaneous squamous cell carcinoma (cSCC) is a prevalent skin cancer in the general population that poses so far unresolved challenges in high-risk groups such as organ transplant recipients and individuals with recessive dystrophic epidermolysis bullosa. Although most cases of cSCC respond well to standard treatments, these 2 groups often face more aggressive disease, characterized by higher rates of metastasis and cancer-specific mortality. The tumor microenvironment plays a pivotal role in cSCC progression, influencing tumor growth, immune evasion, and therapy response. Therefore, this scopi…

Distinguishing Breast Cancer Subtypes via Ratiometric Photoacoustic Sensing of pH and Cu2+ in the Tumor Microenvironment.

Triple-negative breast cancer (TNBC) is characterized by a lack of effective therapeutic targets, high invasiveness, and a propensity for metastasis, making the development of highly specific and sensitive early diagnostic and personalized treatment strategies particularly urgent. Leveraging the distinct features of the TNBC tumor microenvironment─specifically, acidic pH and abnormal Cu2+ enrichment─this study designed and constructed a pH/Cu2+ dual-responsive ratiometric photoacoustic (PA) nanoprobe, termed LPCR NP, for precise in vivo PA imaging and the differential diagnosis of TNBC. The na…

NAD(P)H: quinone oxidoreductase 1-mediated oxidative stress resistance and tumor microenvironment remodeling in glioblastoma.

BACKGROUND: Glioblastoma (GBM) is a highly aggressive brain tumor, with glioblastoma stem cells (GSCs) occupying the pinnacle of a complex tumor microenvironment (TME), conferring therapeutic resistance. The TME plays a role in tumor development by creating a niche rich in reactive oxygen species (ROS) through oxidative stress (OS). Here, we identified NAD(P)H: quinone oxidoreductase 1 (NQO1) as an essential regulatory factor in antioxidant stress response, which is key to maintaining GSCs and the immunosuppressive TME. METHODS: Proteomics analysis and epigenetic profiling using H3K27ac ChIP-…

SOX30 Facilitates Triple-Negative Breast Cancer Metastasis via TNFR2-NF-κB Signaling and Tumor Microenvironment Remodeling.

Triple-negative breast cancer (TNBC) remains therapeutically challenging due to its aggressive metastatic potential, high recurrence rates, and lack of effective targeted therapies. Herein, we identified that SOX30 is significantly upregulated in TNBC and correlates with poor clinical prognosis. Functionally, SOX30 specifically drives TNBC metastasis and activates the TNFR2-mediated downstream NF-κB signaling cascade. Mass cytometry profiling of the tumor immune microenvironment revealed that SOX30 facilitates the recruitment of tumor-associated macrophages (TAMs) within pulmonary metastases,

Tumor Microenvironment-Responsive Intelligent Hydrogel Co-Delivery System Enhances Ferroptosis for Triple-Negative Breast Cancer Therapy.

PURPOSE: Triple-negative breast cancer (TNBC) lacks effective therapeutic targets. Aurisin A (AA), a natural sesterterpenoid, shows anti-TNBC potential but is limited by poor water solubility. This study aimed to develop a tumor microenvironment (TME)-responsive co-delivery hydrogel to enhance AA efficacy. METHODS: AA was complexed with hydroxypropyl-β-cyclodextrin (HP-β-CD) to form AA@HP-β-CD, which was co-loaded with acetazolamide (ACZ) into a triple-responsive injectable hydrogel (PCG). The anti-TNBC mechanism was investigated using HCC1937 cells, focusing on ferroptosis induction. In vivo…

Mechanical regulation of m6A-dependent autophagy sustains cancer stemness and chemoresistance in a viscoelastic tumor microenvironment.

The mechanical properties of the tumor microenvironment fundamentally influence cancer progression, while how viscoelastic cues interface with epitranscriptomic regulation to control cell fate remains poorly understood. Here, a pair of hyaluronic acid-based supramolecular hydrogels with tunable network dynamics are engineered to mimic the viscoelastic properties of tumor ECM. Human osteosarcoma (HOS) cells encapsulated in highly dynamic (HD) hydrogels form compact spheroids, display elevated stemness markers, and exhibit enhanced metabolic activity compared to low-dynamic (LD) matrices. Mechan…

Obesity promotes aggressiveness of endometrial cancer via metabolic reprogramming and intercellular crosstalk in the tumor microenvironment.

Endometrial cancer is one of the most prevalent malignancies of the female reproductive system, and the global rising prevalence of obesity has further increased its incidence and poor clinical prognosis. Although obesity is recognized as a critical modifiable risk factor for endometrial cancer, it remains unclear whether and how obesity drives tumor initiation and progression by modulating the tumor microenvironment at single-cell resolution. Using a genetic mouse model with pten conditional knockout in the uterine endometrium, clinical samples from endometrial cancer patients, and single-cel…

Regulatory T cells in the bladder cancer tumor microenvironment: mechanisms of immune suppression and therapeutic opportunities.

Bladder cancer is characterized by a highly dynamic tumor microenvironment (TME) that critically influences tumor progression, immune evasion, and therapeutic responsiveness. Among the immune populations in the TME, regulatory T cells (Tregs) play a central role in maintaining immune tolerance but also suppress effective antitumor immunity. Increasing evidence suggests that Tregs accumulate in bladder tumors and are associated with disease progression and reduced response to immunotherapies. The bladder cancer TME provides multiple signals that promote Treg recruitment, expansion, and function…
