Alcohol intake and ovarian cancer risk: a pooled analysis of 10 cohort studies.

Alcohol has been hypothesized to promote ovarian carcinogenesis by its potential to increase circulating levels of estrogen and other hormones; through its oxidation byproduct, acetaldehyde, which may act as a cocarcinogen; and by depletion of folate and other nutrients. Case-control and cohort studies have reported conflicting results relating alcohol intake to ovarian cancer risk. We conducted a pooled analysis of the primary data from ten prospective cohort studies. The analysis included 529 638 women among whom 2001 incident epithelial ovarian cases were documented. After study-specific re…

Determination of toxic carbonyl compounds in cigarette smoke.

Toxic carbonyl compounds, including formaldehyde, malonaldehyde, and glyoxal, formed in mainstream cigarette smoke were quantified by deriva … mg/cigarette(-1), respectively. The general decreasing order of the carbonyl compounds yielded was acetaldehyde (1110-2101 microg/cigarette(-1)) > diacetyl (301-433 microg/cigarette(-1)), acrolein (238-468 microg/cigarette(-1)) > formaldehyde (87.0-243 microg/cigarette(-1)), propanal (87.0-176 microg/cigarette(-1)) > malonaldehyde (18.9-36.0 microg/cigarette( …

Risk factors and mechanisms of hepatocarcinogenesis with special emphasis on alcohol and oxidative stress.

Hepatocellular cancer is the fifth most frequent cancer in men and the eighth in women worldwide. Established risk factors are chronic hepat … ation of survival factors and interference with retinoid metabolism. Direct DNA damage results from acetaldehyde, which can bind to DNA, inhibit DNA repair systems, and lead to the formation of carcinogenic exocyclic DNA etheno adducts. Finally, chronic alcohol abuse interferes with methyl group transfer and may thereby alter gene expression.

[Alcoholic liver disease--established treatment and new therapeutic approaches].

Alcoholic liver disease is the most frequent organ damage encountered in chronic alcoholics and the annual death rate attributed to alcohol-induced end-stage liver disease exceeds that of car accidents. Alcoholic liver damage occurs mainly due to the toxicity of its first metabolite acetaldehyde, and due to interactions with numerous macro- and micronutrients. Established treatment options comprise psychotherapy aiming to achieve abstinence, nutritional therapy, management of hepatological complications, and liver transplantation in selected individuals. Since these therapeutic approaches are …

Hepatocyte injury resulting from the inhibition of mitochondrial respiration at low oxygen concentrations involves reductive stress and oxygen activation.

By correlating lactate/pyruvate ratios and ATP levels, cytotoxicity induced by the mitochondrial respiratory inhibitors or hypoxia:reoxygena … s (eg. fructose, dihydroxyacetone, glyceraldehyde) or the NADH utilising substrates acetoacetate or acetaldehyde which decreased the reductive stress and prevented intracellular iron release. These results suggest that liver injury resulting from insufficient respiration involves reductive stress which releases intracellular Fe, converts xanthine dehydrogenase …

Inhibition of cell-free oxidative bactericidal activity by erythrocytes and hemoglobin.

Sickle cell anemia and other chronic hemolytic anemias are associated with an increased frequency of bacterial infections. There is evidence … d human hemoglobin (43.8 versus 17.4 nmol per tube, P = 0.000001). A cell-free bactericidal system, acetaldehyde and xanthine oxidase with or without myeloperoxidase and Cl-, was markedly inhibited by column-purified hemoglobin. For example, after 2 h of incubation, surviving numbers of Staphylococcus aureus were: control (buffer only), 2.5 X 10(6)/ml; bacteri …

Glycolaldehyde-derived advanced glycation end products suppress STING/TBK1/IRF3 signaling via CD36.

AIMS: We have previously reported that advanced glycation end products derived from incubation of albumin with glycolaldehyde (glycol-AGE), lead to suppression of the toll-like receptor 4 (TLR4) signaling response to lipopolysaccharide. Glycol-AGE-induced suppression of TLR4 signaling is involved in the downregulation of CD14, which is an adaptor protein necessary for transferring lipopolysaccharide to TLR4. Therefore, glycol-AGEs impair the innate immune response through suppression of the upstream process in TLR4 signaling. However, the effect of glycol-AGEs on intracellular signaling relate

Solar photolysis kinetics of disinfection byproducts.

Disinfection byproducts (DBPs) discharged from wastewater treatment plants may impair aquatic ecosystems and downstream drinking-water quality. Sunlight photolysis, as one process by which DBPs may dissipate in the receiving surface water, was investigated. Outdoor natural sunlight experiments were conducted in water for a series of carbonaceous DBPs (trihalomethanes, haloacetic acids, halopropanones, and haloacetaldehydes) and nitrogenous DBPs (nitrosamines, halonitromethanes, and haloacetonitriles). Their pseudo-first-order rate constants for photolytic degradation were then used to calibrat…

Impact of Temperature Regimes and Smart Packaging on Volatilome Evolution During the Shelf Life of Agaricus bisporus.

Agaricus bisporus is highly valued for its sensory and nutritional aspects, but it is highly perishable because of its intense postharvest m … respiratory surge and significantly delayed the rise of spoilage-related markers like methanethiol, acetaldehyde, and hexanal. At the same time, SB preserved freshness indicators such as 1-octen-3-one and benzaldehyde, which normally fade as mushrooms age. OVOCs drive radical cycling and ozone formation in background air.

Surface ozone pollution is a critical global environmental challenge driven by the complex, nonlinear photochemical cycling of RO x radical … ical modeling. We find that models constrained with only the three most common OVOCs (formaldehyde, acetaldehyde, and acetone) overestimate hydroxyl radical concentrations by 50 %-100 %, whereas comprehensive constraints align simulations with observations. This discrepancy is caused by complex offsetting errors, including the severe overestimation of isoprene …

Direct Observation of Radical-Mediated Oxide-to-Alloy Transformation in Solution Under Pulsed Laser Irradiation.

Pulsed laser irradiation (PLI) is widely used to synthesize metal-based functional materials in solution, yet the molecular-level mechanism … oxyethyl radical; the oxygen is subsequently released as water and the alcohol is dehydrogenated to acetaldehyde. Even though the initial hydrogen transfer is facile, the release of lattice oxygen is entropy-driven and becomes accessible only at temperatures of order 103 K, so that the extent of reduction is governed by the transient local temperature and henc …

PDC1 deficiency results in 2-deoxyglucose sensitivity through inhibition of Pdc2 activity in yeast.

Pyruvate decarboxylase (PDC) catalyzes the decarboxylation of pyruvate to yield acetaldehyde, using thiamine pyrophosphate as a coenzyme. Saccharomyces cerevisiae has two functional PDCs, Pdc1 and Pdc5, whose reactions constitute the first step in the alcoholic fermentation pathway. PDC1 is abundantly expressed in wild-type cells, whereas PDC5 is markedly repressed when cells are cultured in a glucose medium. PDC5 expression is induced in a Pdc1 deletion mutant, a phenomenon referred to as autoregulation, and this induction depends on the transcriptional regulator Pdc2. Therefore, it has been …

Possible regulatory network and associated pathways governing the expression of ADH2 in Saccharomyces cerevisiae.

With the day to day increase in energy consumption due to increase in urbanization production of bioethanol is highly in demand. At this point where the traditional methods are not able to suffice the demands due to its high cost and low productivity, new methods need to be developed. This review aims to understand the importance and the regulation of ADH2 in Saccharomyces cerevisiae because Adh2p is the only enzyme that initiates the reaction for the conversion of ethanol, the end product of fermentation to acetaldehyde. The effect of glucose on regulatory mechanisms of Alcohol dehydrogenase …
