Serrated Polyposis Syndrome: A Narrative Review from Clinical Recognition to Risk-Adapted Cancer Prevention.

Serrated polyposis syndrome (SPS) is a clinically defined colorectal polyposis syndrome characterized by multiple and/or large serrated lesions and an increased risk of colorectal cancer (CRC). Its diagnosis remains challenging because it depends on cumulative endoscopic and histopathological findings rather than a single germline marker, and it remains under-recognized in routine practice. The biological basis of SPS is heterogeneous: serrated tumorigenesis commonly involves BRAF or KRAS activation, CpG island methylation, and MLH1 silencing, yet most cases are not explained by classic Mendel

Timing-Dependent Spiking Neural Network: Board-Level Hardware Implementation with Photoelectroactive Van der Waals Synapses.

The rapid growth of unstructured data in applications such as autonomous systems and edge AI underscores the urgent need for energy-efficient, real-time computing exemplified by biological brains, where synaptic weights are adjusted according to the timing of neural spikes, known as spike-timing-dependent plasticity (STDP). This work presents the first experimental realization of a multi-channel timing-dependent spiking neural network (TD-SNN) at the board-level by integrating photoelectroactive synaptic devices with an analog leaky integrate-and-fire (LIF) neuron circuit. The synaptic devices

3D spatial distribution of Sost mRNA and sclerostin protein expression in response to in vivo tibia loading in female mice.

Bones adapt to external mechanical loads through a process known as mechanoadaptation. Osteocytes are the bone cells that sense the mechanical environment and initiate a biological response. Investigating the changes in osteocyte molecular expression following mechanical loading has been instrumental in characterizing the regulatory pathways involved in bone adaptation. However, current methods for examining osteocyte molecular expression do not preserve the three-dimensional structure of the bone, which plays a critical role in the mechanical stimuli sensed by the osteocytes and their spatial

Testosterone, fathers as providers and caregivers, and child health: Evidence from fisher-farmers in the Republic of the Congo.

Males in vertebrate species with biparental care commonly face a life history trade-off between investing in mating versus parenting effort. Among these males, testosterone is frequently elevated during mating and competition and reduced when males help raise offspring. These physiological patterns may be adaptive, increasing males' fitness through investments in young. However, for some species, including humans, indirect parenting often benefits young but can also involve male competition and risk-taking behavior and may be facilitated by elevated testosterone. Despite potential adaptive fun

Environmental discontinuity hypothesis: Buffer dysfunctions as a source of human disease.

Adaptive physiologic buffers enable organisms to respond to environmental variation with appropriate plasticity. Modern humans have substantially remodeled their environment such that many interactions with the environment have become relatively discontinuous functions compared to the past. Examples include sunlight, temperature, and altitude. We propose that environmental discontinuity represents a Darwinian maladaptation and may promote disease by inducing buffer dysfunctions. Skin pigmentation is an adaptive, dynamic buffer that normalizes sunlight exposure to balance the potential harm of

A biohybrid system to interface peripheral nerves after traumatic lesions: design of a high channel sieve electrode.

Peripheral nerve lesions lead to nerve degeneration and flaccid paralysis. The first objective in functional rehabilitation of these diseases should be the preservation of the neuro-muscular junction by biological means and following functional electrical stimulation (FES) may restore some function of the paralyzed limb. The combination of biological cells and technical microdevices to biohybrid systems might become a new approach in neural prosthetics research to preserve skeletal muscle function. In this paper, a microdevice for a biohybrid system to interface peripheral nerves after traumat

Differences in biological characteristics of two strains of the cestode Hymenolepis diminuta.

Biological characteristics of infectivity, growth rate and fecundity of Hymenolepis diminuta isolated from wild Rattus rattus in Japan were compared with parasites of Texas origin maintained for several generations in this and many other laboratories in laboratory bred Rattus norvegicus. The timing of development and maturation was similar in parasites from both sources, but the mean parasite dry weight was less and the mean egg production lower in Japanese parasites in both single and multiple infections. The differences persisted over 10 weeks in single infections, and were unaffected by rat

Therapeutic reprogramming of natural killer cells for solid tumors: expanding frontiers into rare and ultra-rare malignancies.

Rare and ultra-rare solid tumors collectively account for approximately one-quarter of all cancer diagnoses yet remain associated with poor clinical outcomes. Delayed diagnosis, limited biological characterization, few therapeutic options, and the logistical challenges of conducting adequately powered clinical trials underscore the potential for adaptable cellular immunotherapies that can be applied across diverse tumor histologies. Natural killer (NK) cells have emerged as a promising platform for adoptive cell therapy because of their ability to recognize and eliminate malignant cells indepe

Reframing liver test abnormalities after AAV gene therapy: a mechanistic diagnostic framework.

Hepatotoxicity remains one of the most frequent and clinically important safety concerns in adeno-associated virus (AAV) gene therapy. Yet the laboratory patterns observed after vector administration are biologically complex and often difficult to interpret. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) elevations are non-specific, as they may arise from AAV-induced innate and adaptive immune recognition of vector capsid, endoplasmic reticulum stress, mitochondrial strain, cholestasis, or background muscle turnover, especially in neuromuscular disorders where ALT and AST

A dual-time-scale cooperative memory mechanism for spiking neural networks.

Spiking neural networks (SNNs), as core models of brain-inspired computing, simulate the computational mechanisms of biological neural systems. However, existing models lack the division of labor and collaboration mechanisms found in biological fast and slow neural populations, making it difficult for single-timescale neural populations to simultaneously capture transient events and maintain long-term dependencies. Furthermore, these models are deeply coupled with specific data and tasks. Therefore, a spiking neural network with dual-timescale cooperative memory is proposed to simulate the div

Mapping cell density and hypoxia in glioblastoma using time-dependent diffusion MRI: improved cell density assessment compared to conventional diffusion metrics.

Objective.Tumor heterogeneity, including differences in cell density and hypoxic fraction (HF), can impact the efficacy of radiation therapy (RT). Quantitative imaging biomarkers (QIBs) are required to assess spatial tumor heterogeneity to allow personalization of RT with biologically guided adaptive RT. Time-dependent diffusion MRI (TDD-MRI) derived metrics are promising QIBs, as they are potentially able to characterize different aspects of tissue microstructure better than conventional diffusion-weighted MRI (DW-MRI). Time-dependent diffusion contrast (TDDC), a recently proposed TDD-MRI met

Biology of teeth and implants: Host factors - pathology, regeneration, and the role of stem cells.

In chronic periodontitis and peri-implantitis, cells of the innate and adaptive immune systems are involved directly in the lesions within the tissues of the patient. Absence of a periodontal ligament around implants does not prevent a biologic process similar to that of periodontitis from affecting osseointegration. Our first focus is on factors in the biology of individuals that are responsible for the susceptibility of such individuals to chronic periodontitis and to peri-implantitis. Genetic factors are of significant importance in susceptibility to these diseases. Genetic factors of the h

Hypoxia-induced transcriptional reprogramming in sugar beet seedlings: Linking energy conservation and metabolic maintenance.

Sugar beet (Beta vulgaris L.), as a major sugar crop, faces a significant threat to its yield and industrial development due to hypoxia stress, a critical environmental factor limiting its growth. Here, we investigate the response mechanisms of sugar beet leaves and roots to hypoxia stress at different developmental stages (first and third true leaf stages). Hypoxia significantly reduced plant height, leaf area, and root area. Key photosynthetic parameters, including net photosynthetic rate, stomatal conductance, and intercellular CO2 concentration, decreased, leading to impaired photosyntheti

Hard-object feeding adaptations infer relative predator-prey size relationships in Devonian placoderms.

Devonian placoderms exhibit exceptional diversity in jaw and dental morphology, making them an ideal system for examining early gnathostome feeding mechanisms. Here, we investigate inferognathal (mandible) function in eight eubrachythoracid arthrodire placoderms from Australia's Late Devonian Gogo Formation. A combination of finite element analysis and quantification of dental surface complexity revealed distinct, size-dependent strategies for processing mechanically resistant materials (durophagy). Taxa with broad, flat dental surfaces experienced low strain during simulated bites, consistent

Childhood asthma heterogeneity at the era of precision medicine: Modulating the immune response or the microbiota for the management of asthma attack.

Exacerbations are a main characteristic of asthma. In childhood, the risk is increasing with severity. Exacerbations are a strong phenotypic marker, particularly of severe and therapy-resistant asthma. These early-life events may influence the evolution and be involved in lung function decline. In children, asthma attacks are facilitated by exposure to allergens and pollutants, but are mainly triggered by microbial agents. Multiple studies have assessed immune responses to viruses, and to a lesser extend bacteria, during asthma exacerbation. Research has identified impairment of innate immune

Relation of the pdxB-usg-truA-dedA Operon and the truA Gene to the Intracellular Survival of Salmonella enterica Serovar Typhimurium.

Salmonella is the genus of Gram-negative, facultative intracellular pathogens that have the ability to infect large numbers of animal or human hosts. The S. enterica usg gene is associated with intracellular survival based on ortholog screening and identification. In this study, the λ-Red recombination system was used to construct gene deletion strains and to investigate whether the identified operon was related to intracellular survival. The pdxB-usg-truA-dedA operon enhanced the intracellular survival of S. enterica by resisting the oxidative environment and the usg and truA gene expression

Repeated in-vitro and in-vivo exposure leads to genetic alteration, adaptations, and hypervirulence in Salmonella.

Adaptation is an important phenomenon for the survival of any organism in various in-vitro and in-vivo conditions leading to their better survival. We studied Salmonella, for the evolutionary adaptation under different conditions. When Salmonella was passaged for 12 rounds in Luria Bertani (P12-STM) media, F media (F12-STM) and Caenorhabditis elegans (Ce12-STM), we found certain phenotypic variations with respect to unpassaged strain. The passaged strains showed a significant increase in motility, body size and number of flagella they possessed, increased tolerance to ROS and RNS under in-vitr

The Sinocyclocheilus cavefish genome provides insights into cave adaptation.

BACKGROUND: An emerging cavefish model, the cyprinid genus Sinocyclocheilus, is endemic to the massive southwestern karst area adjacent to the Qinghai-Tibetan Plateau of China. In order to understand whether orogeny influenced the evolution of these species, and how genomes change under isolation, especially in subterranean habitats, we performed whole-genome sequencing and comparative analyses of three species in this genus, S. grahami, S. rhinocerous and S. anshuiensis. These species are surface-dwelling, semi-cave-dwelling and cave-restricted, respectively. RESULTS: The assembled genome si

Natural transformation and genome evolution in Streptococcus pneumoniae.

Streptococcus pneumoniae is a frequent colonizer of the human nasopharynx that has the potential to cause severe infections such as pneumonia, bacteremia and meningitis. Despite considerable efforts to reduce the burden of pneumococcal disease, it continues to be a major public health problem. After the Second World War, antimicrobial therapy was introduced to fight pneumococcal infections, followed by the first effective vaccines more than half a century later. These clinical interventions generated a selection pressure that drove the evolution of vaccine-escape mutants and strains that were

Chromosome phylogenies of malaria mosquitoes.

Malaria mosquitoes often belong to complexes of sibling species, members of which are morphologically and genetically similar to each other. However, members within these complexes can vary significantly in their ecological adaptations and abilities to transmit the malaria parasite. The high degree of genetics similarity among sibling species makes the reconstruction of phylogenetic relationships within species complexes difficult. This paper reviews studies that infer the ancestral--descendant relationships among sibling species using molecular markers and chromosomal inversions. A methodolog
