Wharton's Jelly mesenchymal stromal cell-derived extracellular vesicles promote nucleus pulposus cell anabolism in an in vitro 3D alginate-bead culture model.

BACKGROUND: Intradiscal transplantation of mesenchymal stromal cells (MSCs) has emerged as a promising therapy for intervertebral disc degeneration (IDD). However, the hostile microenvironment of the intervertebral disc (IVD) may compromise the survival of implanted cells. Interestingly, studies reported that paracrine factors, such as extracellular vesicles (EVs) released by MSCs, may regenerate the IVD. The aim of this study was to investigate the therapeutic effects of Wharton's Jelly MSC (WJ-MSC)-derived EVs on human nucleus pulposus cells (hNPCs) using an in vitro 3D alginate-bead culture

Alginate lyases, which are important in both basic and applied sciences, fall into ten polysaccharide lyase (PL) families. PL36 is a newly established family that includes 39 bacterial sequences and one eukaryotic sequence. Till now, the structures or catalytic mechanisms of PL36 alginate lyases have yet to be revealed. Here, we characterized a novel PL36 alginate lyase, Aly36B, from Chitinophaga sp. MD30. Aly36B is a polymannuronate specific endolytic alginate lyase. To probe the catalytic mechanism of Aly36B, the structures of wild-type Aly36B and its mutants (K143A/Y185A in complex with alg

An injectable responsive exosome-releasing hydrogel based on sodium alginate restores motor and bladder function by alleviating the injury microenvironment and facilitating distal nerve repair.

In the early stage of spinal cord injury (SCI), excessive reactive oxygen species (ROS) accumulate at the injury site to establish a microenvironment favoring complex secondary injuries, including cavity formation, and impacting the distal lumbosacral spinal cord. Currently, no definitive clinical treatment is available for SCI. Here, we created an responsive and injectable hydrogel composite (GEL-EXO) by modifying and cross-linking biological macromolecules sodium alginate (SA) and gelatin in the form of embedded exosomes. This GEL-EXO composite integrated the bioactivity of exosomes with the

Unsaturated alginate oligosaccharides alleviate insulin resistance and fatty liver in non-obese NAFLD mice by regulating bile acid metabolism through gut microbiota.

Despite the rising global incidence of non-obese non-alcoholic fatty liver disease (NAFLD), this condition has received limited attention. Unsaturated alginate oligosaccharides (UAOS) have shown promising potential in the management of metabolic diseases. In this study, we used liver-specific growth hormone receptor (GHR) knockout mice as a model for non-obese NAFLD and treated them with UAOS. Our findings demonstrated that UAOS effectively ameliorated insulin resistance and hepatic steatosis in GHR-deficient mice. Additionally, UAOS improved intestinal barrier integrity, altered gut microbiot

Acellular, bioresorbable, ultra-purified alginate gel implantation for intervertebral disc herniation: Phase 1/2, open-label, non-randomized clinical trials.

Discectomy is the current surgical procedure for lumbar intervertebral disc (IVD) herniation. Discectomy was performed to remove the IVD material and relieve the pain inflicted by nerve root compression and axonotoxic effects, such as inflammatory cytokines in the IVD material; however, defects within the IVD caused by discectomy may impair tissue healing and predispose patients to subsequent IVD degeneration. Given that viable cells with the capacity for IVD regeneration are scarce, discectomy alone is not conducive to tissue repair. Here, we report the use of an acellular, bioresorbable, ult
