Effects of anxiogenic drugs on the emission of 22- and 50-kHz ultrasonic vocalizations in adult rats.

RATIONALE: Adult rat 22-kHz vocalizations are often associated with alarm or distress, whereas a subset of 50-kHz calls is preferentially emitted in response to amphetamine and other rewarding stimuli. Whether any 50-kHz calls reflect anxiety is unknown. OBJECTIVE: To determine the effects of anxiogenic drugs on 50-kHz call rate and call subtype profile, in comparison with D-amphetamine. METHODS: Adult male rats received systemic amphetamine (1 mg/kg) three times several days before testing. Ultrasonic vocalizations were then recorded after acute intraperitoneal injection of amphetamine or o…

Effects of drug discrimination history on the generalization of pentobarbital to other drugs.

In pigeons trained to discriminate between pentobarbital and saline, pentobarbital, amobarbital and diazepam substituted for pentobarbital, whereas phencyclidine (PCP) substituted in part for pentobarbital and d-amphetamine, morphine and drug vehicles did not substitute. After morphine replaced pentobarbital as the training drug (group A), morphine, pentobarbital and diazepam substituted, PCP substituted in part, but not d-amphetamine, haloperidol and vehicles. After d-amphetamine replaced pentobarbital as the training drug (group B), d-amphetamine, pentobarbital and diazepam substituted, PCP …

Retention of sequential drug discriminations under fixed-interval schedules for long time periods without training.

The experiments showed that sequential drug discriminations can be learned and retained under a fixed-interval (FI) schedule for more than 18 months without additional training under a complex three-choice procedure. Pigeons were trained to discriminate among 5 mg/kg pentobarbital, 2 mg/kg D-amphetamine, and saline. After responding stabilized, dose-response curves were determined for other drugs. Subsequently, pentobarbital was replaced with 5 mg/kg morphine as a training drug, and D-amphetamine was replaced with 30 mg/kg caffeine. After the pigeons learned these new discriminations, dose-res…

Phenylalkylamine abuse among opiate addicts attending a methadone treatment programme in the Republic of Ireland.

Since the early 1990s ring-substituted derivatives of amphetamine have been abused widely in the Republic of Ireland. The main ring-substituted amphetamines being abused include methylenedioxyamphetamine (MDA), methylenedioxymethamphetamine (MDMA) and methylenedioxyethamphetamine (MDEA). A newer illicit synthetic analogue, which has been seized to a lesser extent by Irish police, is N-methyl-1-(3,4-methylenedioxyphenyl)-2-butanamine (MBDB). The work presented here involved the determination of the type of ring-substituted amphetamines being abused by a group of recovering opiate abusers partic…

Striatal and extrastriatal dopamine release measured with PET and [(18)F] fallypride.

The amphetamine challenge, in which positron emission tomography (PET) or single photon emission computed tomography radioligand binding following administration of amphetamine is compared to baseline values, has been successfully used in a number of brain imaging studies as an indicator of dopaminergic function, particularly in the striatum. [(18)F] fallypride is the first PET radioligand that allows measurement of the effects of amphetamine on D2/D3 ligand binding in striatum and extra-striatal brain regions in a single scanning session following amphetamine. We scanned 15 healthy volunteer …

Thyroid status regulates CART but not AgRP mRNA levels in the rat hypothalamus.

We examined the effects of thyroid status on cocaine- and amphetamine-regulated transcript and agouti-related peptide expression in the rat hypothalamus. Hypo- and hyperthyroidism were induced in adult male rats, and the mRNA content of cocaine- and amphetamine-regulated transcript and agouti-related peptide was determined using in situ hybridization. Hyperthyroidism induces a reduction in cocaine- and amphetamine-regulated transcript mRNA levels in the paraventricular nucleus, without any change in the arcuate and dorsomedial nuclei and in the lateral hypothalamic area. On the other hand, hyp…

Some central effects of opioid antagonists. Part I.

This study concerned the influence of opioid antagonists: naloxone, naltrexone and diprenorphine on amphetamine hyperactivity in mice and rats, apomorphine hyperactivity in rats, amphetamine and apomorphine stereotypy in rats, and stereotyped gnawing induced by methylphenidate in mice. Naloxone, naltrexone and, in higher doses, diprenorphine attenuated the amphetamine hyperactivity in rats. In mice in the same test all antagonists at some doses produced attenuation. However they did not affect the apomorphine hyperactivity in rats. Both naloxone and naltrexone attenuated amphetamine and apomor…

The effect of antimotion sickness drugs on habituation to motion.

The effect of antimotion sickness drugs on habituation was studied. Subjects were rotated once a day for 5 d to the malaise III end-point after receiving placebo, 1 mg scopolamine, 10 mg d-amphetamine, or the combination of 0.6 mg scopolamine with 5 mg of d-amphetamine. The placebo scores had a Spearman coefficient of correlation of 0.88 with the initial untreated tests. This demonstrated a high reliability for the M-III end-point and that little habituation resulted from the test design. The combination of 0.6 scopolamine with 5 mg amphetamine produced the fastest rate of habituation closely …

Behavioral aspects of serotonin-dopamine interaction in the monkey.

The effects of serotonin (5-hydroxytryptamine; 5-HT) antagonists and 5-HT uptake inhibitors on the behavioral response to amphetamine and haloperidol in monkeys (cercopithecus aethiops) were investigated. Amphetamine increased locomotor activity and reactivity and induced repetitive movements of head, limbs and trunk, but no oral hyperkinesia. Haloperidol induced dystonia and parkinsonism. Pretreatment with the 5-HT antagonists cyproheptadine and mianserin increased amphetamine-induced locomotor activity, reactivity and repetitive movements and decreased haloperidol-induced dystonia and parkin…

Effect of dopamine agonists and fenfluramine on discriminative behavior in obese and lean Zucker rats.

Dopamine agonists and fenfluramine were used as pharmacological probes to investigate the possible difference in sensitivity and time course of drug action in genetically obese Zucker rats and their lean littermates. All rats were trained to discriminate between the stimulus properties of 0.6 mg/kg d-amphetamine and its vehicle in a two-lever, food-motivated operant task. Once trained, both groups of rats showed a dose-related decrease in discriminative performance with lower amphetamine doses. Analysis of the dose-response curves indicated an ED50 for the obese rats of 0.17 mg/kg and for the …

[Delay in formation of phenamine tolerance after epiphysectomy in rats].

Amphetamine-induced stereotyped behavior is a nonsteady oscillatory process with minute fluctuations rate of head turnings. After acute amphetamine stereotypy have lower amplitude and limited rhythmicity in pinealectomized rats as compared with intact animals. Pinealectomy also delayed tolerance after chronic administration of amphetamine (5 mg/kg/day, 14 days) without usual decrease number of short period (2-3 min) fluctuations in time course of stereotypy.

Changing Drug Consumption Patterns Among Clients Attending the Medically Supervised Injecting Centre, Sydney: Implications for Service Provision and Harm Reduction Response.

INTRODUCTION: The Uniting Medically Supervised Injecting Centre (MSIC) in Sydney (Australia) provides a supervised space for people to inject pre-obtained drugs. We investigated injecting patterns among clients over time and the impact on overdose risk and client behaviours. METHODS: MSIC holds information on visit-specific drug injected, overdose, and sanctions for disruptive behaviour. Data for May 2001 to December 2024 were analysed. Where amphetamines or opioids were injected, the study record was marked accordingly. Data were aggregated to an annual visit summary and drug categorised int…

Psychostimulant-induced plasticity of intrinsic neuronal excitability in ventral subiculum.

Psychostimulant drugs such as amphetamine are prescribed to increase vigilance, suppress appetite, and treat attention disorders, but they powerfully activate the dopamine system and have serious abuse potential. Repeated psychostimulant exposure induces neuronal plasticity within the mesolimbic dopamine system. Here we present evidence that repeated amphetamine exposure results in a suppression of intrinsic neuronal excitability in the ventral subiculum, a hippocampal region that activates dopamine neurotransmission. We used patch-clamp recordings from brain slices obtained at different times…

Lithium increases nerve growth factor levels in the rat hippocampus in an animal model of mania.

Pharmacological studies suggest that neurotrophins may play a role in the effects of lithium and valproate on mood regulation. In this study, we tested the hypotheses that lithium and valproate would reverse and prevent the behavioral and biochemical effects of amphetamine, using a rat model of mania. In the reversal treatment, male Wistar rats were first administered D-amphetamine or saline for 14 days, and then, between days 8-14, rats were treated with lithium, valproate or saline. In the prevention treatment, rats were pretreated with lithium, valproate or saline, and then, between days 8-…

Dynamic changes in sensitivity occur during the acute response to cocaine and methylphenidate.

RATIONALE: We have previously shown that during the acute response to amphetamine, a stimulant that released dopamine, behavioral sensitivity to the drug undergoes dynamic changes, as evident in the altered behavioral profile expressed to the subsequent administration of a low dose of the drug. OBJECTIVE: The present studies were designed to determine if these dynamic changes in sensitivity occur with amphetamine-like stimulants that act primarily by blocking dopamine uptake. METHODS: Groups of animals were primed with 40 mg/kg cocaine or 30 mg/kg methylphenidate, then during the acute respo…

[Effect of enkephalinamide on the sensitivity of neurons of the lateral hypothalamus to phenamine in the rat].

The influence of opiatotropic drugs on the sensitivity of lateral hypothalamic neurons to amphetamine in the Wistar rats was studied microinotophoretically . D-met2-pro5-enkephalinamide reinforced the amphetamine reactions of most units studied and nalorphine had no effect. A suggestion is made that differences observed in the action of D-met2-pro5-enkephalinamide and nalorphine on the neuronal responses to amphetamine are due to activation of different opiate receptors.

Fenfluramine: An Uncommon Cause of False Positive Urine Drug Testing: A Case Report.

BACKGROUND: Fenfluramine is an antiseizure medication approved by the Food and Drug Administration for the treatment of Dravet syndrome in patients older than 2 years. Fenfluramine is an amphetamine derivative. It cross-reacts with amphetamines in urine drug screen immunoassays. PATIENT PRESENTATION: A 2-year-old patient with Dravet syndrome had a cardiorespiratory arrest and tested positive for amphetamines in a urine drug screen, raising concerns of child abuse. Liquid chromatography-mass spectrometry confirmed the presence of fenfluramine but did not detect amphetamines. CONCLUSION: Fenfl…

Effects of aripiprazole on operant responding for a natural reward after psychostimulant withdrawal in rats.

RATIONALE: Withdrawal from repeated amphetamine administration has been shown to decrease the motivation to work for a natural reward in rats, a phenomenon thought to be associated with hypofunction of the mesolimbic dopamine system. OBJECTIVES: We tested whether aripiprazole, a partial dopamine receptor agonist, can restore the animals' responding for reward pellets after amphetamine withdrawal. MATERIALS AND METHODS: Rats were trained to lever-press for food pellets under a progressive ratio-schedule of reinforcement. After reaching a stable breakpoint, i.e., the highest ratio completed, o…
