Chemopreventative effect of an inducible nitric oxide synthase inhibitor, ONO-1714, on inflammation-associated biliary carcinogenesis in hamsters.

The present study was designed to investigate whether an inducible nitric oxide synthase (iNOS)-specific inhibitor, ONO-1714 [(1S, 5S, 6R, 7R)-7-chloro-3-imino-5-methyl-2-azabicyclo[4.1.0] heptane], could prevent inflammation-associated biliary carcinogenesis in bilioenterostomized hamsters. Syrian golden hamsters underwent choledochojejunostomy and then received subcutaneous injections of the chemical carcinogen N-nitrosobis(2-oxopropyl)amine every 2 weeks at a dose of 10 mg/kg body wt, starting 4 weeks after surgery and continuing for 18 weeks. The hamsters were divided into two groups accor

[Industrial noise and presbyacusia in the determination of hearing damage: studies in chemical industry workers].

An audiometric investigation was carried out on 710 workers of a chemical plant in order to ascertain the role of industrial noise and presbysocio-acusia in causing hearing loss. The workers underwent an audiometric test both aerial and via bone, in a sound-proof chamber, after a 16-hour rest from noise. The subjects also answered a questionnaire on nonoccupational noise exposure and previous hearing and dysmetabolic disorders. Noise exposure was assessed on the basis of a cumulative exposure index obtained for each subject by multiplying the mean daily exposure level by seniority in the job.

Mice lacking liver-specific β-catenin develop steatohepatitis and fibrosis after iron overload.

BACKGROUND & AIMS: Iron overload disorders such as hereditary hemochromatosis and iron loading anemias are a common cause of morbidity from liver diseases and increase risk of hepatic fibrosis and hepatocellular carcinoma (HCC). Treatment options for iron-induced damage are limited, partly because there is lack of animal models of human disease. Therefore, we investigated the effect of iron overload in liver-specific β-catenin knockout mice (KO), which are susceptible to injury, fibrosis and tumorigenesis following chemical carcinogen exposure. METHODS: Iron overload diet was administered to

Pericardial Injection of Kainic Acid Induces a Chronic Epileptic State in Larval Zebrafish.

Epilepsy is a common disorder of the brain characterized by spontaneous recurrent seizures, which develop gradually during a process called epileptogenesis. The mechanistic processes underlying the changes of brain tissue and networks toward increased seizure susceptibility are not fully understood. In rodents, injection of kainic acid (KA) ultimately leads to the development of spontaneous epileptic seizures, reflecting similar neuropathological characteristics as seen in patients with temporal lobe epilepsy (TLE). Although this model has significantly contributed to increased knowledge of ep

Suppressive effects of dietary curcumin on the increased activity of renal ornithine decarboxylase in mice treated with a renal carcinogen, ferric nitrilotriacetate.

Curcumin, a natural, biologically active compound extracted from rhizomes of Curcuma species, has been shown to act as a biological response modifier in various disorders. We have reported previously that the dietary supplementation of curcumin enhances the activities of antioxidant and phase II metabolizing enzymes in mice (M. Iqbal, S.D. Sharma, Y. Okazaki, M. Shenqi Yanshen Decoction protects against renal and bone injury in chronic kidney disease-mineral and bone disorder with potential involvement of Wnt5a/RhoA/Pkn3 signaling.

ETHNOPHARMACOLOGICAL RELEVANCE: Shenqi Yanshen Decoction (SQYSD) is a traditional Chinese herbal formula that is used for treating chronic kidney disorders and kidney-related bone complications in the clinical setting under the traditional concept that "the kidney governs the bone". However, experimental evidence for its coordinated protective effects on renal and skeletal injury in chronic kidney disease-mineral and bone disorder (CKD-MBD) is limited. AIM OF THE STUDY: In this study, the therapeutic effects of SQYSD on renal dysfunction, mineral metabolism disorder, and bone injury in CKD-MB

Phenothiazine and butyrophenone intoxication in children.

Antipsychotic agents are widely used for the treatment of psychotic disorders as well as for the acute treatment of nausea and vomiting, cough and cold treatments, and as supplementary agents for sedation for minor surgical or diagnostic procedures. There are many different circumstances in which the clinician may encounter a child who has experienced antipsychotic drug toxicity, such as from an acute accidental ingestion or as a side effect from therapeutic use. The phenothiazines and butyrophenone drugs have many pharmacologic actions. Thus, a wide range of clinical symptoms and signs may be

Challenges facing providers caring for HIV/HCV-coinfected patients.

Despite the high prevalence of hepatitis C virus (HCV) infection among injection drug users also infected with human immunodeficiency virus (HIV), and the synergistic adverse effect of the two diseases on patients' health and survival, research on the clinical management of these patients and particularly the low uptake of HCV therapy is limited. We conducted qualitative interviews with 17 HIV providers from two urban public hospitals. We discovered that the limitations of the current state of medical knowledge, the severe side effects of HIV and HCV therapies, and the psychosocial vulnerabili

[Tuberculous meningitis: a comparative study in relation to concurrent human immunodeficiency virus infection].

INTRODUCTION: This study assesses the epidemiological and clinical data, as well as therapy and evolution in a recent series of patients with tuberculous meningitis (MT). A comparative study was conducted between adult MT patients with and without concurrent human immunodeficiency virus (HIV) infection. METHODS: From 1987 to 2000, 75 episodes of MT were diagnosed, 39 of them (52%) in patients with prior HIV infection. A comparative study was performed of variables related to the presence or absence of HIV and MT coinfection. RESULTS: MT was more frequent in HIV patients (6.4% versus 1.2%, p

Nonocular manifestations of parenteral drug abuse.

The nonocular manifestations of parenteral drug abuse are presented to familiarize the ophthalmologist with this ever growing, medical and psychosocial problem. The ophthalmologist, like other primary care physicians, may be the first professional to encounter these patients whose substance abuse may lead to a wider spectrum of physical and psychiatric illness. The variety, history, and manifestations of abused substances are discussed.

Heme oxygenase-1: function, regulation, and implication of a novel stress-inducible protein in oxidant-induced lung injury.

Accumulating evidence suggests that oxidative stress plays a central role in the pathogenesis of many pulmonary diseases including adult respiratory distress syndrome, emphysema, asthma, bronchopulmonary dysplasia, and interstitial pulmonary fibrosis. The morbidity and mortality of these diseases remain high even with optimal medical management. In our attempts to devise new therapies for these disorders, it is crucial to improve our understanding of the basic mechanism(s) of oxidant-induced lung injury. A major line of investigation seeks to characterize the cellular and molecular responses o

Si-Wu-Tang ameliorates chemotherapy-induced premature ovarian insufficiency by regulating a signaling pathway in endoplasmic reticulum stress.

BACKGROUND: Premature ovarian insufficiency (POI) is a clinically refractory reproductive system disorder. Our previous studies demonstrated that the traditional Chinese herbal formula Si-Wu-Tang (SWT) might ameliorate POI in animal models by improving ovarian angiogenesis, yet the underlying mechanisms remained unclear. PURPOSE: To explore the mechanisms of the traditional Chinese medicine (TCM) formula SWT on human umbilical vein endothelial cells (HUVECs) and animal models of POI. By studying its mechanisms, the research hopes to reveal the connotation of 'nourishing blood and activating b

Berberine contributes to protecting against the cadmium-induced pancreatic damage: role of intestinal microbiome modulation and barrier function.

Cadmium (Cad) is a worldwide heavy metal pollutant associated with global health challenges. Alteration of the intestinal microbiome, due to chemicals' exposure, plays a vital role in the pathogenesis of gastrointestinal diseases such as pancreatic disorders. Hence, modulation of the gut microbiota might be a targeted approach to manage pancreatic diseases. Using murine modeling, this study consisted of two dependent experiments to investigate the curative potential of berberine (BBR) in a Wistar rat model of Cad-provoked pancreatic toxicity and the possible contribution of gut microbiota to B

Modulation of steroidogenesis by Actaea racemosa and vitamin C combination, in letrozole induced polycystic ovarian syndrome rat model: promising activity without the risk of hepatic adverse effect.

BACKGROUND: Complementary remedies such as the Chinese herb 'Sheng Ma' (Black cohosh; Actaea racemosa 'AR') are being sought to overcome the shortcomings of conventional hormonal and surgical therapies developed for the treatment of polycystic ovary syndrome (PCOS). However, AR-induced hepatotoxicity necessitates a cautionary warning to be labeled on its products as recommended by the United States Pharmacopeia, where four out of seven hepatotoxic cases in Sweden were possibly associated with black cohosh products. METHODS: We investigated the effects, safety, and molecular targets of black c
