Characterization of the WAVE1 knock-out mouse: implications for CNS development.

Developing neurons must respond to a wide range of extracellular signals during the process of brain morphogenesis. One mechanism through which immature neurons respond to such signals is by altering cellular actin dynamics. A recently discovered link between extracellular signaling events and the actin cytoskeleton is the WASP/WAVE (Wiscott-Aldrich Syndrome protein/WASP-family verprolin-homologous protein) family of proteins. Through a direct interaction with the Arp2/3 (actin-related protein) complex, this family functions to regulate the actin cytoskeleton by mediating signals from cdc42 as…

Characterization and expression analyses of the mouse Wiskott-Aldrich syndrome protein (WASP) family member Wave1/Scar.

Characterization of multiprotein complexes involved in actin remodeling and cytoskeleton reorganization is essential to understand the basic mechanisms of cell motility and migration. To identify proteins implicated in these processes, we have isolated the mouse Wave1/Scar gene, a member of the Wiskott-Aldrich syndrome protein (WASP) family. The mouse Wave1 gene was physically localized on chromosome 10 and spans over 12 Kb comprising eight exons and seven introns. The mouse Wave1 complementary DNA encodes a predicted 559 amino acid protein, with a SCAR homology domain, a basic domain, a proli…

Ezrin and moesin expression within the developing human cerebrum and tuberous sclerosis-associated cortical tubers.

The ERM (ezrin, radixin, and moesin) proteins belong to the band-4.1 superfamily of membrane-cytoskeleton-linking proteins which bind to the actin cytoskeleton via their C-terminal sequences and bind ERM binding membrane proteins (ERMBMPs). We investigated the immunohistochemical expression of two of the ERM proteins (ezrin and moesin) in developing human cerebral cortex and in cortical tubers from patients with tuberous sclerosis (TSC), to assess possible consequences of TSC gene product malfunction or inactivation in the developing brain in relation to ERM protein expression. Ezrin is abunda…

Expression of a truncated keratin 5 may contribute to severe palmar--plantar hyperkeratosis in epidermolysis bullosa simplex patients.

Epidermolysis bullosa simplex are dominant disorders of skin fragility characterized by intraepidermal blistering upon mild mechanical trauma. Skin fragility is caused by expression of either an abnormal keratin 5 or an abnormal keratin 14 protein, which compromises the structure and function of the keratin cytoskeleton of basal cells. We report an epidermolysis bullosa simplex patient with a novel single base substitution (A-->T1414) that changes the lysine residue at amino acid 472 to a non-sense codon (K472X). This change predicts the synthesis of a truncated keratin 5, missing 119 amino ac…

Stage-specific apoptotic patterns during Drosophila oogenesis.

In the present study we demonstrate the existence of two apoptotic patterns in Drosophila nurse cells during oogenesis. One is developmentally regulated and normally occurs at stage 12 and the other is stage-specific and is sporadically observed at stages 7 and 8 of abnormally developed follicles. The apoptotic manifestation of the first pattern begins at stage 11 and is marked by a perinuclear rearrangement of the actin cytoskeleton and the development of extensive lobes and engulfments of the nurse cell nuclei located proximal to the oocyte. Consequently, at late stage 12 (12C), half of the …

Tyrosine phosphorylation regulates the SH3-mediated binding of the Wiskott-Aldrich syndrome protein to PSTPIP, a cytoskeletal-associated protein.

Wiskott-Aldrich syndrome is an X-linked hematopoietic disease that manifests itself in platelet deficiency and a compromised immune system. Analysis of hematopoietic cells from affected individuals reveals that mutations in the Wiskott-Aldrich syndrome protein (WASP) result in structural and functional abnormalities in the cell cortex, consistent with the suggestion that WASP is involved with regulation of the actin-rich cortical cytoskeleton. Here we report that WASP interacts with a recently described cytoskeletal-associated protein, PSTPIP, a molecule that is related to the Schizosaccharomy…

Ischemia-induced phosphorylation and translocation of stress protein alpha B-crystallin to Z lines of myocardium.

It is becoming clear that stress proteins play a role in various aspects of postischemic myocardial recovery and that the cytoskeleton of cardiac myocytes is an important determinant for cellular survival during ischemia and energy depletion. In the present study, we addressed the question of whether the cytoskeleton-binding stress protein alpha B-crystallin may be involved in early cellular responses of rat and porcine myocardium to ischemia. Immunostaining and subcellular fractionation revealed a rapid ischemia-induced redistribution of alpha B-crystallin from a cytosolic pool to intercalate…

Cytokeratins 8 and 18 in smooth muscle cells. Detection in human coronary artery, peripheral vascular, and vein graft disease and in transplantation-associated arteriosclerosis.

During development of atherosclerotic lesions, vascular smooth muscle cells (SMCs) undergo changes both phenotypically and in their cytoskeleton composition. An expression of cytokeratins 8 and 18 in SMCs in plaques of the human superficial femoral artery and of cytokeratin 8 in lesions of the aorta was recently described. Since cytokeratins are epithelial markers generally not found in normal adult vascular SMCs, we performed a detailed immunofluorescence microscopy study using a large panel of antibodies against the various cytokeratin polypeptides and other elements of the cytoskeleton. We …

Differences in hippocampal protein expression at 3 days, 3 weeks, and 3 months following induction of perinatal asphyxia in the rat.

Perinatal asphyxia (PA) was induced in Sprague-Dawley rats; pups were sacrificed 3 days, 3 weeks, and 3 months following the asphyctic insult, and hippocampal protein levels were determined by a gel-based proteomic method. Levels of antioxidant, metabolic, cytoskeleton, signaling, channel, proteasomal, chaperone, splicing, and synaptosomal proteins were dysregulated depending on the age following induction of PA. These proteins are proposed to be potential markers or pharmacological targets for PA.

Rafting toward a depression biomarker.

The journey from the cytoskeleton and G protein signaling to a framework that might provide a simple blood diagnostic for depression as well as a harbinger of individual antidepressant response, has been convoluted, occasionally frustrating, but ultimately rewarding. Herein, I provide a travelogue of sorts on our progress toward this end, and also chart a course toward the goal of providing a simple, quantitative and objective, biomarker-driven assessment tool that reflects the neurobiology of depressed mood.

Neurofibromatosis 2: loss of merlin's protective spell.

Schwannomas and meningiomas occur as multiple tumors in sufferers of neurofibromatosis 2 (NF2) and as solitary tumors in the general population due to the inactivation of a gene at chromosome 22q12. In 1993, a location cloning approach revealed this tumor suppressor, dubbed merlin, as a novel member of a family of proteins that link elements of the cytoskeleton and the cell membrane. Subsequent investigations have confirmed merlin's role in tumor formation, but have yet to reveal its mechanism of action.

[Inherited abnormalities of the epidermis caused by mutation of keratins].

The recent identification of keratin mutations as a cause of hereditary disorders of keratinization stresses the importance of an intact cytoskeleton of keratinocytes. Four disorders have reported to be caused by keratin mutations so far: epidermolysis bullosa simplex, bullous congenital ichthyosiform erythroderma, ichthyosis bullosa and epidermolytic palmoplantar hyperkeratosis. Molecular genetic diagnosis of keratin disorders is being introduced into the clinical routine and prenatal diagnosis is possible after 10 weeks of gestation.

Zyxin deficiency aggravates podocyte injury in hypertensive nephropathy.

Podocyte injury and detachment are early cellular events in hypertensive nephropathy, yet their underlying mechanisms are not well clarified. Zyxin, a mechanotransducer located at focal adhesions, regulates actin cytoskeleton remodeling and exhibits diverse biological functions. However, its role in podocytes is poorly understood. In this study, we constructed a hypertensive nephropathy model in podocyte-specific zyxin knockout mice to explore the role of zyxin in hypertensive conditions. In vitro, mechanical stretch and Angiotensin II (AngII) were used to stimulate podocytes. Western blot, re…

SARS-CoV-2 S assembly into virions facilitated by host ERM proteins.

The host cell cytoskeleton plays a critical role in the SARS-CoV-2 life cycle, though the underlying mechanisms remain poorly understood. This study investigates the interaction between the SARS-CoV-2 spike (S) protein and the cytoskeleton-associated ezrin-radixin-moesin (ERM) proteins through biochemical and structural characterization. A previously unidentified ERM-binding motif on the SARS-CoV-2 S protein is identified, revealing that S-ERM interactions are specifically conserved among highly pathogenic coronaviruses, including SARS-CoV, MERS-CoV, and SARS-CoV-2. Functionally, these interac…
