Chemotaxis-promoting and adhesion properties of human tonsillar follicular dendritic cell clusters.

Lymph follicles are globular and compact due to aggregation of lymphoid cells on follicular dendritic cells (FDC). To probe the mechanisms underlying this accumulation of cells, we analyse here the role played by FDCs in attracting and binding cells. FDCs prepared from human tonsils by mild separation techniques appeared in the form of clusters (FDC clusters), where, via cytoplasmic extensions, they enveloped lymphoid cells. Using Boyden's chambers, we demonstrated that these FDC clusters produced one or more chemoattractants capable of inducing chemotaxis of lymphoid cells. Supernatants of FD

Follicular dendritic cells adhere to fibronectin and laminin fibers via their respective receptors.

The expression of adhesion molecules on human tonsillar follicular dendritic cells (FDCs) in the secondary lymphoid follicle (LF) in vivo and in vitro was investigated using cryostat sections and cytospin preparations of FDCs isolated with a magnetic cell sorter, respectively. FDCs were immunochemically positive for Mac-1 (CD11b), sialyl-Lex (CD15s), CD22, integrin beta 1 (CD29), CD40, very late activation antigen (VLA)-alpha 3 (CD49c), VLA-alpha 5 (CD49e), VLA-alpha 6 (CD49f), intercellular adhesion molecule (ICAM)-3 (CD50), ICAM-1 (CD54), B7 (CD80), and vascular cell adhesion molecule (VCAM)

Does dendritic growth underly recovery from neonatal occipital lesions in rats.

Rats were given lesions of visual cortex on postnatal day 4 or 10, or in adulthood. Ninety days later they were trained on a horizontal-vertical stripes discrimination task and a visual-spatial navigation task. None of the operated rats acquired the tasks. The brains were processed for Golgi-Cox staining and dendritic arborization was quantified in the layer III pyramidal cells in somatosensory cortex. Relative to normal control brains, the neurons of the day 4 and adult operates showed a reduction in dendritic branching, whereas the neurons of the day 10 operates had a significant increase in

Imaging of calcium variations in living dendritic spines of cultured rat hippocampal neurons.

1. Cultured rat hippocampal neurons were loaded with the Ca2+ indicator fura-2 through micropipettes and visualized with an inverted microscope equipped with a high power objective and a cooled CCD camera. The responses of dendritic spines and their parent dendrites to stimuli which evoke a rise of [Ca2+]i were monitored. 2. NMDA caused a rapid and transient rise in [Ca2+]i, which was more evident in the spine than in the parent dendrite. The recovery in both compartments had the same time course, and was dependent on normal [Na+]o. 3. Application of alpha-latrotoxin, which causes release of n…

The identification of germinal centres and follicular dendritic cell networks in rheumatoid synovial tissue.

We document here the identification of germinal centres with dark and light zones, a follicular dendritic cell network and clonal expansion in the synovium of rheumatoid arthritis patients. Synovial tissue from 24 patients suffering from rheumatoid arthritis or the polyarticular form of juvenile rheumatoid arthritis were screened for the presence of lymphoid follicles. The synovial tissues of 14 patients contained follicles and four of these had germinal centres and a follicular dendritic cell network. There was a statistically significant association between follicles in the synovium and the

Usefulness of follicular dendritic cell pattern in classification of peripheral T-cell lymphomas.

Classification of peripheral T-cell lymphomas based on morphological criteria can present problems due to overlap in histological features amongst the subtypes. An immunohistochemical study was designed to study the follicular dendritic cell patterns in 21 cases of peripheral T-cell lymphoma which had been classified using the updated Kiel classification. Three patterns of distribution were observed: 1 follicular dendritic cells not detected (3 cases); 2 follicular dendritic cells restricted to remnant follicle centres (7); 3 follicular dendritic cells present as an expanded network of cells e

In vivo activation of mouse dendritic epidermal T cells in sites of contact dermatitis.

Adult mouse epidermis contains a population of dendritic, Thy-1+, CD3+, CD4-, CD8- and T cell receptor (TcR) V gamma 3/V delta 1+ leukocytes termed dendritic epidermal T cells (DETC). DETC isolated from skin and placed into culture will proliferative vigorously in response to T cell mitogens and T cell growth factors. In the present study, we examined whether DETC can be activated in situ by modulating their epidermal environment. Ear skin of CBA mice was painted with the chemical irritant, croton oil, and the epidermal cells (EC) isolated from such sites were then tested for proliferative res

Dendritic architecture of nucleus ambiguus motoneurons projecting to the upper alimentary tract in the rat.

The motor innervation for palatal, pharyngeal, laryngeal, and esophageal muscles originates within the nucleus ambiguus. Although the viscerotopic organization of the upper alimentary tract in the nucleus ambiguus has been extensively studied, little information concerning the dendritic arborization of nucleus ambiguus motoneurons is available. The neural tracer cholera toxin-horseradish peroxidase, which is particularly effective at revealing dendrites of retrogradely labeled neurons, was used to determine the dendritic architecture and organization of nucleus ambiguus motoneurons. In 72 rats…

Synapses, axonal and dendritic patterns of GABA-immunoreactive neurons in human cerebral cortex.

Gamma-aminobutyric acid (GABA) containing neurons were characterized in human association cortex by a combination of Golgi impregnation and immunohistochemistry. Neurons were Golgi impregnated, gold toned, drawn and then classified on the basis of their dendritic and axonal arborization in layers I-VI. An antiserum to GABA was used to determine which of the impregnated neurons were immunopositive. Twenty-four GABA-positive cells were Golgi impregnated: 7 were bitufted with their dendrites predominantly radially oriented, and 17 were multipolar stellate cells. Three of the multipolar cells with…

Ultrastructural characteristics of the interdigitating dendritic cell in dermatopathic lymphadenopathy of mycosis fungoides patients.

The ultrastructure of interdigitating dendritic cells in dermatopathic lymphadenopathy from patients with mycosis fungoides was investigated. The most remarkable findings were: a marked hypertrophy of the smooth endoplasmic reticulum with a peculiar concentric arrangements of the cisternae around lipid droplets, an abundance of cytoplasmic lipid droplets and the presence of paranuclear chromatoid-like bodies. It is suggested that these ultrastructural features may correspond to some unknown functional capability of the interdigitating dendritic cell.

Isolated frog olfactory cilia: a preparation of dendritic membranes from chemosensory neurons.

The recently introduced frog olfactory cilia preparation (Chen and Lancet, 1984; Pace et al., 1985) has been useful for studies of molecular chemosensory mechanisms. Here we describe in detail the properties of this cilia preparation. The "calcium shock" procedure leads to a complete removal of the cilia from the olfactory epithelial surface. Isolated cilia constitute segments of proximal regions with 9 X 2 + 2 microtubular arrangement and a large proportion of membrane vesicles, probably derived from the ciliary distal segments. Polypeptides unique to the olfactory cilia preparation, compared

Feed-forward dendritic inhibition in rat hippocampal pyramidal cells studied in vitro.

1. Intracellular recordings from CA1 pyramidal cells in the rat hippocampal slice preparation have been used to study the neuronal pathways … roduced by GABA released from neuronal elements in the dendritic field which acts on pyramidal cell dendrites.6. The depolarizing i.p.s.p. is strongly temperature-dependent and increases in amplitude and duration progressively as slices are cooled from 37 to 22 degrees C.7. Depolarizing i.p.s.p.s can be produced by orthodromic stimulation in s. oriens as we …

Dendritic-cell-targeting adeno-associated virus as potent cancer vaccine carriers.

BACKGROUND: The clinical efficacy of therapeutic cancer vaccines is critically limited by the inefficient in vivo delivery of tumor antigens to dendritic cells (DCs), the key regulators of T-cell immunity. METHODS: To address this issue, we developed a dendritic-cell-targeting adeno-associated virus (AAV) vector through rational capsid design. This was achieved by site-specifically inserting the high-affinity Clec9a-targeting peptide CBP-12, optimizing its flanking sequences, and introducing T491V/S662V gain-of-function mutations to synergistically enhance DC transduction. RESULTS: The resul

Dataset for small RNA sequencing of murine dendritic cell-derived exosomal miRNAs.

This data article describes a murine dendritic cell-derived exosomes (DEXs) small RNA sequencing dataset generated to support reuse of exosomal miRNA profiles in studies of immune regulation and osteoarthritis-related mechanisms. Data collection began with BMDC isolation, culture and characterization via flow cytometry; DEXs were purified from imDC supernatant using a CP-100WX Ultracentrifuge (HITACHI, Japan) with a standardized ultracentrifugation protocol. The dataset contains six paired-end FASTQ files from three small RNA sequencing libraries, DEXs1, DEXs2 and DEXs3, prepared from the DEX
