Occupational exposure to wood dust and formaldehyde and risk of nasal, nasopharyngeal, and lung cancer among Finnish men.

Controversy exists over whether or not occupational inhalation exposure to wood dust and/or formaldehyde increases risk for respiratory cancers. The objective of this study was to examine the risk of nasal, nasopharyngeal, and lung cancer in relation to occupational exposure to wood dust and formaldehyde among Finnish men. The cohort of all Finnish men born between the years 1906 and 1945 and in employment during 1970 was followed up through the Finnish Cancer Registry for cases of cancers of the nose (n = 292), nasopharynx (n = 149), and lung (n = 30,137) during the period 1971-1995. The subj…

Application of a broad-range resequencing array for detection of pathogens in desert dust samples from Kuwait and Iraq.

A significant percentage of the human population is exposed to high levels of naturally occurring airborne dusts. Although the link between airborne particulate inhalation and a variety of respiratory diseases has long been established, little is known about the pathogenic role of the microbial component of the dust. In this study, we applied highly multiplexed PCR and a high-density resequencing microarray (RPM-TEI version 1.0) to screen samples of fine topsoil particles and airborne dust collected in 19 locations in Iraq and Kuwait for the presence of a broad range of human pathogens. The re…

Concentrations of dust, allergens and endotoxin in stables, living rooms and mattresses from cattle farmers in southern Bavaria.

Agricultural work is considered to be a major risk factor for occupational diseases. In particular, allergic reactions to cow dander cause numerous cases of airway disorders. We measured the concentration of allergens (e.g. Bos d2, Der p1) and endotoxin in the stables, living-rooms and mattresses of 46 farmers with a diagnosis of occupational asthma or allergic rhinitis caused by cow dander allergen. The concentration of cow dander allergen was highest in stables (median 20,400 microg/g) but also noticeable in dust samples from living-rooms (median 155 microg/g) and mattresses (median 195 micr…

Adsorption of lipoproteins onto mineral dust surfaces: a possible factor in the pathogenesis of particle-induced pulmonary fibrosis?

We compare the adsorption behavior of high density lipoproteins (HDL) and low density lipoproteins (LDL) on "fibrogenic" and "nonfibrogenic" mineral dusts. The adsorption tests with bovine lipoprotein concentrate and human serum produced the following results: 1) All seven examined fibrogenic dusts (SiO2 DQ12, SiO2 F600, silica, graphite, TiC, kaolin, talc) adsorbed significantly more high density lipoproteins (HDL), than the five examined nonfibrogenic (inert) dusts (TiO2, SnO2, Al2O3, Fe2O3, Fe3O4). This different behavior was particularly conspicuous in the presence of competing adsorbates …

The measurement and health impact of endotoxin contamination in organic dusts from multiple sources: focus on the cotton industry.

Endotoxin is derived from Gram-negative bacterial membranes, and its inflammatory effects following inhalation are well characterized. The significance of this fact becomes apparent when the wide-ranging environments containing high levels of this microbial product are considered. Endotoxin is present in numerous industrial environments, especially where organic fibers are processed. Microbial contamination of these fibers mainly occurs at the agricultural stage. Materials such as flax and hemp are affected in this way, but the most important product in this context is cotton, from which chron…

Additive effect of smoking and cotton dust exposure on respiratory symptoms and pulmonary function of cotton textile workers.

One hundred and sixty-nine and 175 cotton textile workers (CTWs) were enrolled in the first (1991) and second (1996) surveys to investigate the prevalence of byssinosis. The synergistic effect of smoking on cotton dust exposure was also evaluated. Although the difference in prevalence of abnormal pulmonary function between the first (38.5%) and second study (38.9%) was not statistically significant, smokers had significantly higher frequency than nonsmokers in both surveys. A significant trend existed between the cotton dust levels and the frequency of abnormal lung function. The significant t…

Ferruginous bodies in cases with occupational and non-occupational exposure to inorganic dusts. A study with 40 matched cases.

In this study we evaluated the usefulness of ferruginous bodies (FB) quantification in lung digest and histologic sections from 40 patients with a history of exposure to inorganic dusts. The patients were matched with 40 cases without occupational exposure. The samples of peripheral lung tissue were digested and all FB were quantified. Lung sections were also evaluated in order to confirm the presence of FB. The concentrations of FB in the patients were higher than the controls (Wilcoxon test, p < 0.001). The patients group showed a median concentration of 194.5 FB/g of dry lung tissue (FB/g) …

[The determination of allergen-specific IgE and IgG antibodies in patients sensitized to the house dust mite Dermatophagoides pteronyssinus].

45 patients, hypersensitive to house-dust mites, were examined by the method of skin tests to D. pteronyssinus allergen. Besides, in their blood sera the levels of allergen-specific IgE antibodies were determined in the radioallergosorbent test and allergen-specific IgG antibodies, in the enzyme immunoassay. These tests revealed that in 91% of the patients the results of skin tests were positive, in 68% an elevated level of specific IgE antibodies and in 93% of the patients an elevated level of specific IgG antibodies were detected. All patients showed the positive result in one of the above-m…

[Histological studies and clinical observations of the effect of various types of dust on the organ of vision of small laboratory animals].

For seven days, three times a day, female rats were instilled into their conjunctival sacs suspensions of power plant dusts, soil and clay. Clinically observed were inflammatory changes of the conjunctiva and cyclids margins as well as traumatisation of the corneal and conjunctival epithelia, appearing at different stages on the experiment. Cytoenzymatic examinations indicated disturbances of oxidation and reduction processes, changes in dialysis through physiologic membranes and dissociation of protein synthesis.

Sauna, dust and migration as risk factors in lung cancer among smoking and non-smoking males in Finland.

To study possible causes of the high lung cancer incidence among Finnish males, the Finnish Cancer Registry and the Cancer Registry of Norway in 1962 performed a population survey covering smoking habits, occupational and residential history and, in Finland, sauna habits. The cohort of 4,475 Finnish men has now been followed up for incidence of lung cancer in 1964-1979. The relative importance of 3 factors--sauna baths, occupational exposure to dust, and migration to urban areas--was studied in different smoking categories. Among smokers of greater than or equal to 25 g/day, takers of frequent…

Role of histamine in the aetiology of byssinosis. I Blood histamine concentrations in workers exposed to cotton and flax dusts.

The formation or the accumulation, or both, of histamine in the lungs may be potentiated by agent(s) present in cotton dust at higher level(s) than in flax dust and negligible in cottonseed dust. It has been suggested that such potentiation may be due to the activation of the ability of the lung to produce histamine and/or produce or recruit mast cells; this may present an acceptable explanation of the mechanism by which the propagation of the chronic effect of the dust proceeds in cotton and flax workers. Histamine accumulated in the lung over the weekend is released on exposure to dust causi…

Comparative study of the smooth muscle contractor activity of airborne dusts and of dustiness in cotton, flax, and jute mills.

A bioassay technique using isolated guinea-pig ileum was employed to compare the smooth muscle contractor activity of various dusts from mills in which the prevalence of byssinosis was known. The activity of dust from a mill spinning a coarse grade of cotton was several times greater than that in dust from a mill processing a fine grade of cotton. There was a similar order in the difference of the prevalence of byssinosis in these mills. However, the activities of fine cotton, flax, and jute dusts were very similar to each other, in spite of marked differences in the prevalence of byssinosis i…

SiO2 dust induces the transformation of lung fibroblasts to myofibroblasts by activating the S1P-RhoA-YAP signaling pathway.

Silicosis, a prevalent and severe form of pneumoconiosis, is characterized by silicotic nodule formation and extensive pulmonary fibrosis. Caused by prolonged inhalation of free silica (SiO₂) dust, its pathogenesis involves complex regulatory mechanisms that remain unclear. Using a co-culture model of THP-1 macrophages and HFL-1 cells exposed to SiO₂, we investigated the role of the S1P-RhoA-YAP pathway in fibroblast-to-myofibroblast transformation. Intervention with fingolimod or si-RhoA was employed. Results showed that SiO₂ exposure significantly reduced HFL-1 cell viability, induced morpho…

The impact of mineralogy on coal dust toxicity: potassium as a proxy for illite and surface iron-driven oxidative stress.

Epidemiological evidence suggests that the chemical composition of coal dust influences its toxicity, although the underlying mechanisms remain unclear. We previously identified potassium content as a strong predictor of lung cell responses, likely reflecting the presence of potassium alumino-silicate minerals. In this study, we investigated the role of potassium and potassium alumino-silicates in coal dust-induced cellular responses and explored the mechanisms driving these effects. Forty-four coal samples and four potassium alumino-silicate minerals were assessed for their effects on cytotox…

Impact of Biomass Fly Ash on the Performance of Diatomite and Iron Dust Powder-Based Alkali-Activated Binder.

This study investigates the influence of biomass fly ash (BFA) produced from high-temperature combustion of woody biomass fuels typical of Lithuanian energy plants on the mechanical performance, microstructure, and reactivity of alkali-activated binders based on diatomite and iron dust. Diatomite was used as a reactive silica source, while iron dust served as a matrix-modifying component enhancing binder density and strength. The role of BFA (10-30%) was evaluated in terms of compressive strength, softening factor, water resistance, and structural development using XRD and FTIR analyses. The r…
