Effect of citrate on the reactions of cytochrome c with reductants and cyanide.

The rate of reduction of cytochrome c by ascorbate is decreased in the presence of anions. This decrease is due to two factors: (a) nonspecific changes in ionic strength which occur when the total ion concentration or the charge on the anion is altered and (b) "specific" binding of anions to a site on cytochrome c which directly inhibits the reaction. The reaction between cyanide ion and cytochrome c is also affected by anions: increasing the ionic strength decreases the apparent association rate constant for cyanide binding. Substitution of citrate for morpholinopropane sulphonate in isoionic

[Effect of thyroxine on oxidative enzymatic activity in the rat aorta].

The activity of hexokinase, dehydrogenases of pentose cycle, total lactate dehydrogenase, and cytochrome-C-oxidase level was determined in the aorta of rats with hyperthyroidism caused by the intraperitoneal administration of L-thyroxin. In the administration of hormone doses approaching the physiological ones there was an increase in the activity of cytochrome-C-oxidase and a tendency to the increase of 6-phosphogluconate dehydrogenase. Elevation of the activity of all the enzymes under study, excluding lactate dehydrogenase, and in increase of the amount of extractable protein occurred in th

The lung of the copper-deficient rat. A model for developmental pulmonary emphysema.

Based on the hypothesis that cross-linked elastin is critical for normal lung structure, lung tissue from copper-deficient rats was studied. Copper deficiency was induced in the second generation by feeding dams a milk-based diet low in copper (less than 1 ppm) during gestation and lactation. The weanlings were fed the same diet until they showed severe signs of deficiency between 6 and 10 weeks of age. Controls animals received the basal diet supplemented with 10 ppm copper. Liver cytochrome oxidase activity, which served as the chief index of deficiency, decreased from a normal level of appr

Fluorocitrate ototoxicity. A morphologic and cytochemical model for primary neural degeneration in the guinea pig cochlea.

Fluorocitrate, an inhibitor of the tricarboxylic acid cycle at the aconitase reaction, produces a time and dose related neural dystrophy in the guinea pig cochlea. There is direct inhibition of succinic dehydrogenase activity but not nicotinamide adenine dinucleotide dehydrogenase and cytochrome oxidase via cytochrome c activities. The dystrophic neural changes morphologically are similar to those noted in primary neural degeneration and neural presbycusis in man. Neural degeneration in aging appears to be the result of a dissociation of biochemical reactions preventing the proper utilization

Isolation of synaptic junctional complexes of high structural integrity from rat brain.

A new method has been developed for isolating synaptic junctional complexes (SJC) of high structural integrity. The major step in the isolation involves homogenization of a synaptosomal membrane (SM) fraction in a biphasic system consisting of Freon 113 and an aqueous phase containing 0.2% Triton X-100. Well-preserved SJCs, along with membrane vesicles, were recovered in the aqueous phase after low-speed centrifugation of the homogenate. The membranes were subsequently separated from the SJCs by centrifugation on a discontinuous sucrose density gradient. The purity and identity of subcellular

Phosphorylation of purified thyroid plasma membranes incubated with [32-P]ATP.

Plasma membranes have been prepared from porcine thyroid glands using sucrose gradients. The fractions having a density in sucrose of 1.18 g/ml mainly contained plasma membranes and were moderately contaminated with other subcellular components as shown by marker enzyme data. Purified plasma membranes incubated in the presence of [32-P]gamma ATP incorporated 32-P. Kinetics of incorporation of 32-P into endogenous substrates studied in various buffers and with increasing ATP concentration suggest a phosphodephosphorylating system related to cAMP-dependent protein kinase and phosphoprotein phosp

Quantitative evaluation of electron transport system proteins in mitochondrial encephalomyopathy.

The levels of mitochondrial electron transport system proteins cytochrome c oxidase (COX) and complex III were measured in muscle fibers of patients with mitochondrial encephalomyopathy using quantitative immunoelectron microscopy. In a patient with Leigh's encephalopathy, immunoreactive COX protein was decreased to 20% of the normal mean value in all muscle fibers examined, while the amount of complex III was within the normal range. In a patient with fatal infantile COX deficiency, the level of COX protein was found to be decreased to 27-40% of the normal value in all muscle fibers examined.

Secondarily reduced cytochrome c oxidase activity in various neuromuscular disorders.

Decreased cytochrome c oxidase (CCO) activity was found in various neuromuscular diseases, including infantile spinal muscular atrophy (SMA) and Fukuyama type congenital muscular dystrophy (FCMD), that was thought to result from a secondarily induced mitochondrial defect. To determine whether or not the enzyme activities in the mitochondrial electron transport system can be secondarily changed, we measured the enzyme activities in denervated rat muscles. The CCO activity decreased progressively to 38-47% of the control value in 3 weeks after denervation, the NADH-cytochrome c reductase and suc

Fimbriarioides intermedia and F. lintoni (Cestoda: Hymenolepididae): molecular phylogeny, species status, and transmission in Arctic coastal ecosystems.

Cestodes of the genus Fimbriarioides (Hymenolepididae, Fimbriariinae) parasitise waterfowl and shorebirds in the Holarctic. Using molecular and morphological methods, we studied two Fimbriarioides spp. from sea ducks of the European North of Russia: the type species F. intermedia from eiders Somateria mollissima and S. spectabilis 1758 and a closely related species F. lintoni from a previously unrecognised host, Velvet Scoter Melanitta fusca 1758. Our analysis also involved metacestodes Fimbriarioides sp. from barnacles Semibalanus balanoides of the Barents Sea and the Sea of Okhotsk. Metacest

Phylogeny of two accacoeliid species (Digenea: Hemiuroidea) ex Mola mola (Linnaeus, 1758) (Tetraodontiformes: Molidae) from Northwest Pacific, with first molecular data on Odhnerium Yamaguti, 1934.

Accacoeliid digeneans associated with fish of the family Molidae exhibit enigmatically high taxonomic diversity. However, the phylogenetic relationships between species within this digenean taxon are poorly understood. In the present study, the first nuclear 28S rRNA gene, ITS2 region of nuclear DNA, and mitochondrial cox1 gene sequence datasets were obtained for two members of the Accacoeliidae, a type and only species of the genus Odhnerium Yamaguti 1934 and an unidentified Accacladocoelium sp. collected from Mola mola (Linnaeus, 1758) off Iturup Island. Analyses of molecular differentiation

N-acetylcysteine and cysteamine bitartrate prevent azide-induced neuromuscular decompensation by restoring glutathione balance in two novel surf1-/- zebrafish deletion models of Leigh syndrome.

SURF1 deficiency (OMIM # 220110) causes Leigh syndrome (LS, OMIM # 256000), a mitochondrial disorder typified by stress-induced metabolic strokes, neurodevelopmental regression and progressive multisystem dysfunction. Here, we describe two novel surf1-/- zebrafish knockout models generated by CRISPR/Cas9 technology. While gross larval morphology, fertility, and survival into adulthood appeared unaffected, surf1-/- mutants manifested adult-onset ocular anomalies and decreased swimming activity, as well as classical biochemical hallmarks of human SURF1 disease, including reduced complex IV expre

Hemiuroidean trematodes from freshwater Patagonian fishes: description of a new species, distribution and molecular phylogeny.

A new trematode species, Derogenes lacustris Tsuchida, Flores, Viozzi, Rauque et Urabe n. sp. (Derogenidae: Derogeninae), from freshwater fishes is described using morphological and molecular approaches in Argentinean Patagonia. D. lacustris is the most common hemiuroidean species in the Limay River basin and parasitizes almost all the native and introduced Patagonian freshwater fish. This new species could be considered as the unique freshwater species in the genus Derogenes Nicoll, 1910. Another hemiuroidean species, Thometrema patagonica Szidat (Archiev Hydrobiol 51: 542-577, 1956) Lunaschi

Evidence of a Putative Novel Species of Avian Schistosome Infecting Planorbella trivolvis.

Freshwater gastropods of the genera Lymnaea Lamarck, 1799, Physa Draparnaud, 1801, Gyraulus Charpentier, 1837, Radix Montfort, 1810, and Stagnicola Jeffreys, 1830 are considered suitable intermediate hosts for avian schistosomes. A large trematode biodiversity survey performed across 3 yr on 6 lakes in Alberta confirmed 3 already-reported snail hosts for 7 North American avian schistosomes; however, the cytochrome c oxidase subunit 1 (COI) nucleotide sequence from 1 cercarial sample (from a single specimen of Planorbella trivolvis) was distinct from all other COI schistosome sequences. Molecular characterization of Oxyspirura mansoni and Philophthalmus gralli collected from the eyes of domestic chickens in Bangladesh.

A variety of helminths have been found in domestic chickens in Bangladesh, but little is known about their gene sequences. Here, parasitic nematodes and trematodes were collected from the eyes of domestic chickens and analyzed for their morphological and morphometric characteristics, and characterized molecularly. The helminths were identified as Oxyspirura mansoni and Philophthalmus gralli. The ITS1 and ITS2 sequences of O. mansoni were 532 bp and 306 bp in length, respectively, and showed low identity (50.7-62.7%) with those of O. petrowi and O. conjunctivalis. Furthermore, the O. mansoni CO

The amnestic agent anisomycin disrupts intrinsic membrane properties of hippocampal neurons via a loss of cellular energetics.

The nearly axiomatic idea that de novo protein synthesis is necessary for long-term memory consolidation is based heavily on behavioral studies using translational inhibitors such as anisomycin. Although inhibiting protein synthesis has been shown to disrupt the expression of memory, translational inhibitors also have been found to profoundly disrupt basic neurobiological functions, including the suppression of ongoing neural activity in vivo. In the present study, using transverse hippocampal brain slices, we monitored the passive and active membrane properties of hippocampal CA1 pyramidal ne
