Stressor controllability, social interaction, and benzodiazepine systems.

Effects of benzodiazepine receptor-active compounds on inescapable shock-produced changes in social interaction were studied in the rat. Inescapably shocked animals exhibited less social interaction in a novel situation than did escapably shocked or unshocked rats 24 h after shock. Administration of the selective benzodiazepine receptor antagonist flumazenil at the time of shock prevented the decrease in social interaction. Social interaction was unaffected by the same treatment at the time of measurement. Reduction in social interaction induced by inescapable stress endured for 48-72 h follow

Evaluation of depression in rats exposed to chronic (unpredictable) electric shock.

The present study was undertaken to evaluate the applicability of a proposed behavioral stress paradigm as an animal model for depression. Rats were trained to press a lever under a fixed ratio (FR) 5 schedule in a Skinner box for 10 days and were subsequently exposed to a daily regimen of 20 cycles of FR 5 and 10 cycles of variable ratio (VR) 10 for about a week. This exposure resulted in a reduction of the number of lever presses and successful escapes compared to the level achieved after training. In addition, weight gain was significantly suppressed compared with other treatments. Acute an

Neurohypophysial responses to emotional stress after deafferentation of sino-aortic baroreceptors in rats.

In an attempt to test the possibility that sino-aortic baroreceptors may mediate the previously reported stress response in hypothalamic magnocellular neurosecretory cell activity in rats, effects of deafferentation of sino-aortic baroreceptors on plasma levels of vasopressin and oxytocin after fear-related emotional stress were studied in male rats 28-33 days after the surgery. An alpha 1-adrenergic receptor agonist, phenylephrine (1 mg/kg) injected i.p. under anesthesia increased arterial blood pressure in the rats that had received surgical operation of sino-aortic denervation (SAD) and in

Conditioned freezing (generalized motor inhibition) in several rat strains: its usefulness in assessing somato-vegetative responses to nociceptive stress.

Conditioned initial freezing duration was timed in the light-dark box test. Four genetically diverse groups of rats (Wistar, Long Evans, Brattleboro heterozigous for diabetes insipidus, Brattleboro homozygous for diabetes insipidus) were employed. 0.6 mA and 1.8 mA footshocks were administered as nociceptive stressors. The results showed that freezing duration easily and conveniently measures the animal's capacity of responding to stress. This finding is discussed in terms of genetically determined hormonal make-up and of CNS receptorial mechanisms. Initial freezing duration is proposed as a t

Biochemical and pharmacological evidence for central cholinergic regulation of shock-induced aggression in rats.

Acetylcholinesterase (AChE) activity was estimated in brain and heart homogenates and plasma of 'aggressive' and 'nonaggressive' rats. Brain homogenates of 'nonaggressive' rats hydrolyzed significantly more substrate when compared to the 'aggressive' rats. Such differences were not seen in the heart homogenates or plasma of these two groups of rats. Acute DFP (0.1, 0.3 and 1.0 mg/kg) attenuated shock-induced aggression (SIA) 2 hr after treatment but facilitated SIA 24 hr and 48 hr after drug administration. Long-term DFP (0.3 mg/kg x 10 days), on the other hand, induced a significant enhanceme

Shock-induced modulation of lymphocyte responsiveness and natural killer activity: differential mechanisms of induction.

The present study was designed to determine the influence of signaled shock on splenic natural killer (NK) activity and nonspecific T-lymphocyte mitogenic responsiveness. Furthermore, experiments were conducted to examine possible mechanisms mediating this suppression. The results demonstrate that a single session of signaled shock induces suppression of splenic NK activity and T-cell response to the mitogens concanavalin A (Con A) and phytohemagglutinin (PHA). However, the suppression of mitogenic responsiveness was attenuated after five daily sessions of shock, while NK activity remained sup

Behavioral variability within the Brattleboro and Long-Evans rat strains.

Samples of genetically diabetes insipidus (DI) and normal (NO) rats were obtained from American suppliers (Rochester (RO)/DI and NO) and from the colony maintained at Charing Cross Hospital in London (Charing Cross (CC)/DI and NO) to test the hypothesis that the behavior of vasopressin-deficient Brattleboro (DI) and possibly normal Long-Evans rats may vary significantly between different colonies. DI rats of both colonies exhibit longer latencies to emerge into an open field than do NO rats. RO/DI and CC/DI rats acquire goal-approach behavior in a straight runway at similar rates. Differential effect of posttraining naloxone, beta-endorphin, leu-enkephalin and electroconvulsive shock administration upon memory of an open-field habituation and of a water-finding task.

Rats were trained and tested in an open field for habituation of rearing responses, for a water-finding task, or for both tasks simultaneously. Training-test interval was 24 hr. The water-finding task consisted of locating a metal tube in one of the walls of the box, which was attached to a water bottle on the outside; animals were water deprived between training and testing. Retention was estimated by measuring the latency to lick from the tube on the test session. Rats learned this task either with or without water deprivation, also prior to training. Habituation learning (reduction of the n

Neuropeptide modulation of the behavioral effects of drugs.

Studies employing conditioned operant behavior of squirrel monkeys, rabbits and pigeons have demonstrated that the neuroactive peptides thyrotropin-releasing hormone (TRH), substance P (SP) and neurotensin (NT) produce marked behavioral effects under a wide range of procedures. These compounds appear to be particularly active when given in combination with other drugs. The co-administration of TRH with chlordiazepoxide, pentobarbital and ethanol potentiates the effects of these agents on punished behavior. Under other conditions SP can reverse the effects of chlordiazepoxide, whereas NT enhanc

Investigating benzodiazepine receptor function in vivo using an intravenous infusion of DMCM.

A method for measuring seizure thresholds using an intravenous infusion of a convulsant beta-carboline benzodiazepine receptor ligand (DMCM) is reported. Seizure thresholds to DMCM are elevated by the benzodiazepine receptor agonist, flurazepam and antagonist, Ro 15-1788, and the proconvulsant beta-carboline, FG 7142. Various other anticonvulsant also antagonise DMCM seizures. Selectivity for the benzodiazepine/GABA receptor complex is demonstrated since bicuculline and pentylenetetrazol lowered thresholds whereas strychnine and N-methyl DL-aspartate did not.

Effects of instruction on acquisition and extinction of electrodermal responses to fear-relevant stimuli.

In the present study we examined the hypothesis that electrodermal responses conditioned to fear-relevant stimuli are insensitive to verbal instructions. In the first experiment, different groups of subjects were conditioned to fear-relevant and fear-irrelevant control stimuli in a long interstimulus interval differential paradigm with shock as the unconditioned stimulus. Then half of the subjects were informed that no more shocks would be presented, and a number of extinction trials followed. The instruction completely abolished responding to fear-irrelevant stimuli, while leaving responses t

Morphine action at central nervous system sites in rat: analgesia or hyperalgesia depending on site and dose.

Morphilne was injected via fine-gauge cannulas permanently implanted in various subcortical sites in the rat brain. In this way the blood-brain barrier was avoided and precise quantities of the drug were delivered to the intended sites. Ten micrograms of morphine in the posterior hypothalamus resulted in siginificant analgesia, while the same dose injected into the medial septum, the caudate, or the periaqueductal gray matter yielded hyperalgesia. The morphine-produced hyperalgesia at the last-mentioned site was accompanied by stereotyped violent circular leaps, an effect of morphine not previ

(2R,6R)-HNK attenuates negative emotional behaviors and synaptic plasticity deficits via GluA1 in rats exposed to single prolonged stress and foot shock.

BACKGROUND: Post-traumatic stress responses are debilitating psychiatric conditions triggered by exposure to traumatic events. Dysregulation of the glutamatergic system, particularly in circuits governing negative emotional responses, plays a central role in pathogenesis of stress-related disorders. However, the specific contribution of glutamate receptor subtypes remains poorly defined. METHODS: This study examined the role of GluA1-containing AMPA receptors (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors) within the hippocampus (HIP), prefrontal cortex (PFC), and amygdala (A

Sex differences in the impact of trauma on alcohol self-administration.

Challenging encounters can lead to escalated alcohol consumption, as evidenced by individuals grappling with posttraumatic stress disorder (PTSD). However, the precise influence of prior stress experiences, as opposed to acute stressors, on alcohol intake remains incompletely understood. This study sought to simulate the enduring repercussions of trauma using a stress paradigm involving foot shocks presented in an odor-enriched environment. The presence of a scent throughout contextual fear conditioning is more likely to cause stress effects to generalize across various environments, and this

Enhanced noradrenergic activity in the amygdala contributes to hyperarousal in an animal model of PTSD.

Increased activity of the noradrenergic system in the amygdala has been suggested to contribute to the hyperarousal symptoms associated with post-traumatic stress disorder (PTSD). However, only two studies have examined the content of noradrenaline or its metabolites in the amygdala of rats previously exposed to traumatic stress showing inconsistent results. The aim of this study was to investigate the effects of an inescapable foot shock (IFS) procedure (1) on reactivity to novelty in an open-field (as an index of hyperarousal), and (2) on noradrenaline release in the amygdala during an acute

Stress potentiates early and attenuates late stages of visual processing.

Stress can fundamentally alter neural responses to incoming information. Recent research suggests that stress and anxiety shift the balance of attention away from a task-directed mode, governed by prefrontal cortex, to a sensory-vigilance mode, governed by the amygdala and other threat-sensitive regions. A key untested prediction of this framework is that stress exerts dissociable effects on different stages of information processing. This study exploited the temporal resolution afforded by event-related potentials to disentangle the impact of stress on vigilance, indexed by early perceptual a
