Applications of biotechnology in parasitic diseases in Malaysia.

Biotechnological tools are being used in malaria, filariasis and dengue research. The main emphasis has been on the production of reagents for immunodiagnosis and research. In this respect monoclonal antibodies (McAbs) against various species and stages of the above pathogens have been produced. It is hoped that these McAbs will be useful not only in immunodiagnosis but also for seroepidemiological applications. A DNA probe against Brugia malayi has been tested in Malaysia and was found to be sensitive and specific.

Uptake and development of Wuchereria bancrofti in Aedes aegypti and Haitian Culex quinquefasciatus that were fed on a monkey with low-density microfilaremia.

Colonized mosquitoes of Culex quinquefasciatus (Haitian strain) and Aedes aegypti (Liverpool strain) were blood fed on a patas monkey (Erythrocebus patas) that had been experimentally infected with the Haitian strain of Wuchereria bancrofti and harbored a consistently low microfilaremia (1-3 mf per 20 mm3). Both species ingested more than twice the expected number of microfilariae (mf), i.e. 1.9 and 0.77 mf per mosquito, respectively. However, at 10-16 hours post ingestion only 4.2% of the mf had migrated from the blood meal in Cx. quinquefasciatus versus 20.7% in Ae. aegypti. Subsequently, on

Cloned antigen genes of Brugia filarial parasites.

Human lymphatic filariasis is a chronic, potentially debilitating disease caused by Brugia and Wuchereria species of parasitic nematodes. The spectrum of clinical manifestations appears to be related to the immune response of individuals to invasive larvae, adult worms and circulating first-stage larvae (microfilariae). Potential immunopathological outcomes place constraints on vaccine development, emphasizing the need to understand the basis of immunity and pathology. Clones coding for a number of distinct antigenic proteins of Brugia pahangi and Brugia malayi have been isolated via immunolog

Identification and partial characterization of a parasite antigen in sera from humans infected with Wuchereria bancrofti.

The purpose of this study was to identify and characterize soluble parasite antigens present in sera from humans infected with the filarial nematode Wuchereria bancrofti. Affinity chromatography and immunoblot methods were used to demonstrate a 200,000 m.w. circulating parasite antigen in sera from infected humans which corresponded to an antigen released by adult W. bancrofti during in vitro culture. Two monoclonal antibodies were produced to this antigen by immunizing mice with antigens from Dirofilaria immitis, a filarial parasite that is closely related to W. bancrofti, and screening cell

Genital filariasis in Minnesota.

Genitourinary filariasis is a well-known entity in most tropical areas. However, filariasis cannot be eliminated from the differential diagnosis of testicular, epididymal, or spermatic cord masses in nontropical climes. We report a case of filariasis of the spermatic cord that manifested as an unexplained scrotal mass in a patient in Rochester, Minnesota.

Effectiveness of a simple lymphoedema treatment regimen in podoconiosis management in southern ethiopia: one year follow-up.

BACKGROUND: Podoconiosis is a non-filarial elephantiasis caused by long-term barefoot exposure to volcanic soils in endemic areas. Irritant silicate particles penetrate the skin, causing a progressive, debilitating lymphoedema of the lower leg, often starting in the second decade of life. A simple patient-led treatment approach appropriate for resource poor settings has been developed, comprising (1) education on aetiology and prevention of podoconiosis, (2) foot hygiene (daily washing with soap, water and an antiseptic), (3) the regular use of emollient, (4) elevation of the limb at night, an…

Construction of bacterial artificial chromosome libraries from the parasitic nematode Brugia malayi and physical mapping of the genome of its Wolbachia endosymbiont.

The parasitic nematode, Brugia malayi, causes lymphatic filariasis in humans, which in severe cases leads to the condition known as elephantiasis. The parasite contains an endosymbiotic alpha-proteobacterium of the genus Wolbachia that is required for normal worm development and fecundity and is also implicated in the pathology associated with infections by these filarial nematodes. Bacterial artificial chromosome libraries were constructed from B. malayi DNA and provide over 11-fold coverage of the nematode genome. Wolbachia genomic fragments were simultaneously cloned into the libraries givi…

Cross-sectional study to assess filarial infection among the never treated individuals in selected districts in India: a study protocol.

INTRODUCTION: With the global lymphatic filariasis (LF) elimination goal set to 2030, it is necessary to address challenges hindering the last-mile efforts. Never treated individuals are those who self-report that they have never taken the drugs for LF during any mass drug administration (MDA) rounds. Hence, it is necessary to identify these individuals and assess if they can be potential reservoirs of infection and understand the reasons for non-compliance. METHODS AND ANALYSIS: This mixed method study, proposed for a period of 2 years, will assess the filarial infection status of never trea

First ever programmatic implementation of triple drug therapy in the prevention of lymphatic filariasis in Haiti: lessons learned in improving the quality and coverage of locally led programming in lymphatic filariasis-persistent and conflict-affected endemic communes.

Although significant strides have been made towards lymphatic filariasis (LF) elimination in Haiti, which has experienced ongoing multidimensional crises, 19 out of 140 communes still require mass drug administration (MDA) despite multiple rounds using the two-drug regimen of diethylcarbamazine (DEC) and albendazole. This includes the commune of Limonade, which has experienced persistently high LF prevalence despite nine rounds of two-drug MDAs since 2010. In September and October 2023 and July 2024, the Haitian Ministry of Public Health and Population, with support from IMA World Health and t

Piloting lymphoedema management for lymphatic filariasis: experiences and perceptions of patients and stakeholders from Kwale county, coastal Kenya.

BACKGROUND: The global program to eliminate lymphatic filariasis (GPELF) was started in 2003 with two strategies: mass drug administration (MDA) to interrupt disease transmission and the morbidity management and disability prevention (MMDP) to provide basic hygienic care to filariasis lymphoedema patients. METHODS: We conducted a mixed methods study with pre-intervention, intervention and post-intervention phases without control groups among 106 participants from two lymphatic filariasis-endemic wards of Lunga Lunga sub-county, Kwale county on the coastal Kenya between 2019 and 2021. During t

Phenotypical characterization, and antibiotics susceptibility patterns of skin bacteria found in podoconiosis patients in the North West Region of Cameroon.

BACKGROUND: Podoconiosis, a non-infectious disease originating from long-term exposure of bare feet to irritant red clay soil is a lifelong, disabling disease with no specific diagnostic tool, classified into 5 stages based on the severity of leg swelling (lymphoedema). Secondary bacterial infections have been suggested to cause acute dermatolymphangioadenitis (ADLA) attacks and drive disease progression. Although the North West Region of Cameroon has a proven history of podoconiosis endemicity, the bacterial composition of lymphoedema due to this condition has not been studied. Thus, this stu

Pharmacokinetics, feasibility and safety of co-administering azithromycin, albendazole, and ivermectin during mass drug administration: A review.

INTRODUCTION: Traditionally, health ministries implement mass drug administration programmes for each neglected tropical disease (NTD) as separate and distinct campaigns. Many NTDs have overlapping endemicity suggesting co-administration might improve programme reach and efficiency, helping accelerate progress towards 2030 targets. Safety data are required to support a recommendation to undertake co-administration. METHODOLOGY: We aimed to compile and summarize existing data on co-administration of ivermectin, albendazole and azithromycin, including both data on pharmacokinetic interactions a
