The role of the invasive, placental trophoblast in human pregnancy.

During early pregnancy the placenta-derived extravillous trophoblast starts to invade the maternal uterus in order to regulate adequate blood flow and nutrient supply to the growing fetus. A unique set of events including plugging and remodelling of maternal vessels, regulation of oxygen levels, as well as the crosstalk with maternal decidual cells are thought to be precisely controlled by the invading extravillous trophoblasts. However, defects in these processes can lead to severe complications during pregnancy threatening the well-being of both the mother and the developing fetus. For insta

Lindane (gamma-HCH) causes reproductive failure and fetotoxicity in mice.

Lindane (gamma-Hexachlorocyclohexane) was orally given to pregnant Swiss female mice at various stages of pregnancy. During early pregnancy … en was given together with lindane at early pregnancy, implantation was normal, although subsequent fetal development was adversely affected. Progesterone, unlike estrogen, could not correct lindane-induced failure in implantation. On the other hand, when estrogen and progesterone were simultaneously given to lindane-fed mice during early pregnancy, both implantati …

Contributions of androgen and estrogen to fetal programming of ovarian dysfunction.

In female mammals, including humans, deviations from normal androgenic or estrogenic exposure during fetal development are detrimental to subsequent adult ovarian function. Androgen deficiency, without accompanying estrogen deficit, has little apparent impact on ovarian development. Fetal estrogen deficiency, on the other hand, results in impaired oocyte and follicle development, immature and abnormal adult ovaries, and excessive ovarian stimulation from endogenous gonadotropins ultimately generating hemorrhagic follicles. Complete estrogen deficiency lasting into adulthood results in partial …

Magnetic resonance imaging of the fetal brain and spine: an increasingly important tool in prenatal diagnosis: part 2.

Fetal MR imaging is an increasingly available technique used to evaluate the fetal brain and spine. This is made possible by recent advances in technology, such as rapid pulse sequences, parallel imaging, and advances in coil design. This provides a unique opportunity to evaluate processes that cannot be approached by any other current imaging technique, and it affords a unique opportunity for studying in vivo brain development and early diagnosis of congenital abnormalities inadequately visualized or undetectable by prenatal sonography. This 2-part review summarizes some of the latest develop

Impact of Ionizing Radiation Exposure on Placental Function and Implications for Fetal Programming.

Accidental exposure to high-dose radiation while pregnant has shown significant negative effects on the developing fetus. One fetal organ which has been studied is the placenta. The placenta performs all essential functions for fetal development, including nutrition, respiration, waste excretion, endocrine communication, and immunological functions. Improper placental development can lead to complications during pregnancy, as well as the occurrence of intrauterine growth-restricted (IUGR) offspring. IUGR is one of the leading indicators of fetal programming, classified as an improper uterine e…

A journey through the pituitary gland: Development, structure and function, with emphasis on embryo-foetal and later development.

The pituitary gland and the hypothalamus are morphologically and functionally associated in the endocrine and neuroendocrine control of other endocrine glands. They therefore play a key role in a number of regulatory feedback processes that co-ordinate the whole endocrine system. Here we review the neuroendocrine system, from the discoveries that led to its identification to some recently clarified embryological, functional, and morphological aspects. In particular we review the pituitary gland and the main notions related to its development, organization, cell differentiation, and vasculariza

The sex-chromosome constitution and early postimplantation development of diandric triploid mouse embryos.

Diandric triploid mouse embryos were produced by standard micromanipulatory techniques, using eggs isolated from female mice with a normal chromosome constitution that had been mated to homozygous Rb(1.3)1Bnr males (which carry a large metacentric "marker" chromosome, viz., a Robertsonian translocation involving chromosomes 1 and 3). The tripronucleate embryos were transferred to the oviducts of pseudopregnant mice, which were subsequently autopsied at about midday on the 10th day of gestation. Although a relatively small number of the isolated conceptuses consisted of morphologically abnormal

Exogenous progesterone supplementation: a strategy to enhance conceptus development in sheep and pigs?

ABSTRACT: The inability of animals to get pregnant, pregnancy loss and weak or stillborn offspring are significant economic burdens to livestock producers worldwide. Progesterone, the hormone of pregnancy, has a crucial role in the establishment of pregnancy, and it has been suggested that progesterone supplementation may be a promising strategy to improve pregnancy outcomes and conceptus development. This review article describes the existing literature on progesterone supplementation in sheep and pigs in relation to pregnancy outcomes and conceptus development. LAY SUMMARY: Pregnancy loss a

Pancreas deficiency modifies bone development in the ovine fetus near term.

Hormones have an important role in the regulation of fetal growth and development, especially in response to nutrient availability in utero. Using micro-CT and an electromagnetic three-point bend test, this study examined the effect of pancreas removal at 0.8 fraction of gestation on the developing bone structure and mechanical strength in fetal sheep. When fetuses were studied at 10 and 25 days after surgery, pancreatectomy caused hypoinsulinaemia, hyperglycaemia and growth retardation which was associated with low plasma concentrations of leptin and a marker of osteoclast activity and collag

The current understanding of cleft lip malformations.

Cleft lip with or without cleft palate is one of the most common congenital malformations. Epidemiology differentiates between cleft malformations connected with syndromes and the more common nonsyndromic forms not associated with other deformities. Although many syndromes with cleft lip with or without cleft palate are known, the majority of orofacial clefts are of the nonsyndromic form. These are known to be of multifactorial origin, with both environmental and genetic factors in their etiology, most of which remain to be fully investigated. Recent literature reveals that wide ethnical and r

Children and passive smoking: a review.

BACKGROUND: For the past 40 years, evidence has been accumulating on the effects of passive smoking on the fetus and on children. Over this period, research methods have become more precise and accurate, with confounding factors controlled for and actual exposure to smoke measured and validated by cotinine tests on body fluids. This review follows the progress of these studies and assesses the weight of evidence for various health risks of passive smoking on children from before birth to adolescence. METHODS: Nearly 200 research papers published worldwide were reviewed. RESULTS: Effects of m

NR2F2 regulation of interstitial cell fate in the embryonic mouse testis and its impact on differences of sex development.

Testicular fetal Leydig cells produce androgens essential for male reproductive development. Impaired fetal Leydig cell differentiation leads to differences of sex development including hypospadias, cryptorchidism, and infertility. Despite fetal Leydig cells are thought to originate from proliferating progenitor cells in the testis interstitium, the precise mechanisms governing the interstitial cells to fetal Leydig cell transition remain elusive. Using mouse models and single-nucleus multiomics, we find that fetal Leydig cells arise from a Nr2f2-positive interstitial population. Embryonic del

The effects of prenatal azithromycin exposure on offspring ovarian development at different stages, doses, and courses.

Azithromycin, a commonly used macrolide antibiotic for treating chlamydial infections during pregnancy, has sparked investigations into its potential effects on offspring development. Despite these inquiries, there remains uncertainty about the specific impact of prenatal azithromycin exposure (PAzE) on offspring ovarian development and the precise "effect window". Pregnant mice, following clinical guidelines for azithromycin dosing, were orally administered azithromycin at different gestational stages [(gestational day, GD) 10-12 or GD 15-17], doses (50, 100, or 200 mg/kg·d), and courses (sin

Benzene-induced growth restriction and compensatory rebound in mouse fetuses and placentas.

Benzene is a ubiquitous environmental toxicant and carcinogen, yet its effects on early placental and fetal development remain poorly defined. In our previous work, repeated mid-gestation benzene exposure increased fetal and placental size at gestational day (GD) 19, indicating compensatory growth responses. Here, we investigated the immediate consequences of the same exposure paradigm to determine how these effects unfold over time. Pregnant CD-1 mice received intraperitoneal injections (200 mg/kg) of either benzene or a vehicle control on GD8, 10, 12, and 14, and fetal and placental outcomes…

Fetuses with deceleration of growth improve their growth following maternal rest.

OBJECTIVE: The purpose of this study was to determine if fetuses with deceleration of growth velocity resulting in an EFW <10th percentile increase their growth above the 10th percentile following 2 weeks of maternal rest in the left lateral recumbent position. METHODS: This was a retrospective observational study of 265 fetuses with the prenatal diagnosis of an EFW <10th percentile. Fetuses were classified by four definitions of abnormal growth velocity: (1) a growth velocity less than 20 g/day, (2) 30 percentile decrease in the EFW, (3) 50 percentile decrease in the EFW, and (4) abnormal gr

Scaling the EVERREST of severe, early-onset fetal growth restriction.

Severe, early-onset fetal growth restriction is a leading cause of medically indicated preterm birth and substantially increases the risk for perinatal death or disability. No treatments exist to improve fetal growth or safely prolong pregnancy. Furthermore, wide-ranging phenotypes limit the accurate prediction of pregnancy outcome. In this issue of the JCI, Spencer and colleagues combine a discovery-science approach with ultrasound parameters to identify the most discriminative models for predicting either the primary outcome of fetal or neonatal death, or a secondary outcome of death or deli
