A rapid electrochemical assay of lecithin in amniotic fluid using a fluoride ion-sensitive electrode.

An electrochemical method is described for the determination of lecithin in rat and human amniotic fluid. Choline is released from lecithin enzymatically by phospholipase D and the hydrogen peroxide released by the action of choline oxidase is quantitatively determined by peroxidase-catalyzed rupture of the covalent C-F bond of 4-fluorophenol. The concentration of F- ions in solutions is determined by a fluoride sensitive electrode from the resulting cell potential difference recorded before and 10 min after addition of a solution containing phospholipase D, choline oxidase and horseradish per

Stabilization of microtubules by inorganic phosphate and its structural analogues, the fluoride complexes of aluminum and beryllium.

In order to elucidate how the elementary reactions of GTP cleavage and subsequent inorganic phosphate (Pi) release, which accompany microtubule assembly, regulate microtubule dynamics, the effect of Pi and of its structural analogues AlF4- and BeF3- on the stability of GDP-microtubules has been investigated. Inorganic phosphate binds to microtubules with a low affinity (KD = 25 mM) and slows down the rate of GDP-subunit dissociation by about 2 orders of magnitude. AlF4- and BeF3- exhibit phosphate-like effects with 1000-fold higher affinity. Evidence has been obtained for direct binding of BeF

Effects of acid-base status and fluoride on the composition of the mineral in developing enamel and dentine in the dog.

Developing molar teeth of the dog were sectioned, embedded in copper containing polymethyl methacrylate, polished and their Ca/P and Ca/Na molar ratios investigated with the electron microprobe. The teeth were obtained at 30 days from 9 pups fed regimes of different acid-base status with or without fluoride supplementation from birth to sacrifice at 30 days. No clear trends in their Ca/P or Ca/Na ratios with variation in the diet were observed. However, evaluation of the Ca/P ratio of the enamel as a function of depth revealed that this ratio was 0.80 +/- 0.15 at the mineralization front. This

Dentine hypersensitivity: effect of interactions between metal salts, fluoride and chlorhexidine on the uptake by dentine.

A large number of compounds may be applied to the teeth for preventive or therapeutic purposes, most notably in dentine hypersensitivity. The uptake of any one compound by tooth substances may clearly be affected by another to produce synergistic, additive or antagonistic effects. This study determined whether uptake interactions occurred between fluoride, chlorhexidine, strontium, tin and zinc. All compounds, in particular tin, showed considerable affinity for dentine. Fluoride reduced the uptake of zinc but not tin, strontium or chlorhexidine. Chlorhexidine increased the uptake of zinc, but

Effects of calcium, fluoride and magnesium supplementations on tissue mineralization in calcium- and magnesium-deficient rats.

There is considerable uncertainty about the interrelated effects of calcium (Ca), magnesium (Mg) and fluoride (F) on hypocalcification of the skeleton and Ca accumulation in vital soft tissues. This paper describes experiments with rats fed a diet deficient in Ca and Mg, the latter deficiency being accentuated by a low potassium content. For different groups the drinking water was supplemented with Ca, Mg, F, Ca + F or Mg + F; Ca and F were supplied as chemically compatible compounds. At the end of the 54-d experiment, plasma Ca was strongly reduced in the Ca-deficient groups. Plasma Mg was re

The relationship between fluoride effects on bone histology and on bone mass in patients with postmenopausal osteoporosis.

Twenty-one patients with postmenopausal osteoporosis were studied for 4 years after initiation with sodium fluoride (NaF), 20-25 mg b.i.d., elemental calcium, 1.0 g/day, and vitamin D2, 50,000-100,000 I.U. weekly. Histomorphometry was carried out on bone biopsies taken prior to and while on NaF treatment. The total bone mineral mass of the central third of the skeleton was measured by neutron activation analysis at 0.5-1.0 year intervals, and the result expressed as the Calcium Bone Index (CaBI), which normalizes the mineral mass to values for normal subjects of the same size. Twelve patients

Enhanced transmembrane proton conductance in Streptococcus mutans GS-5 due to ionophores and fluoride.

The uptake of protons by intact cells of Streptococcus mutans GS-5 was measured directly by assessing the pH increase in lightly buffered cell suspensions as protons moved across the plasma membrane after acid pulses. The barrier function of the cell membrane for protons was disrupted completely by 5% (vol/vol) butanol. This function was compromised severely by the ionophores valinomycin and gramicidin and by the uncoupler carbonylcyanide-m-chlorophenyl hydrazone. Proton conduction across the membrane was also enhanced by fluoride. It appeared that HF traversed the membrane and acted as a carr

Effect on caries of self-application of a zirconium silicate paste containing 9% stannous fluoride.

A 2-year clinical trial was conducted using an adolescent population from a low-fluoride area to evaluate the anticaries effect of self-applications of zirconium silicate paste containing 9% stannous fluoride. tthree study groups totaling 464 children, with an initial average of 12.8 years, completed the study. The control group brushed with a non-fluoride zirconium silicate paste (tzircate Prophylaxis Paste) once each year: the first experimental group brushed with zirconium silicate paste containing 9% stannous fluoride (Zircate Treatment Paste) once each year; and the second experimental gr

A randomized trial of sodium fluoride (60 mg) +/- estrogen in postmenopausal osteoporotic vertebral fractures: increased vertebral fractures and peripheral bone loss with sodium fluoride; concurrent estrogen prevents peripheral loss, but not vertebral fractures.

Postmenopausal Caucasian women aged less than 80 years (n = 99) with one or more atraumatic vertebral fracture and no hip fractures, were treated by cyclical administration of enteric coated sodium fluoride (NaF) or no NaF for 27 months, with precautions to prevent excessive stimulation of bone turnover. In the first study 65 women, unexposed to estrogen (-E study), age 70.8 +/- 0.8 years (mean +/- SEM) were all treated with calcium (Ca) 1.0-1.2 g daily and ergocalciferol (D) 0.25 mg per 25 kg once weekly and were randomly assigned to cyclical NaF (6 months on, 3 months off, initial dose 60 mg

Oral hygiene grade and quality of life in children with chemotherapy-related oral mucositis: a randomized study on the impact of a fluoride toothpaste with salivary enzymes, essential oils, proteins and colostrum extract versus a fluoride toothpaste without menthol.

AIM: The aim of this study was to assess the impact of the use of a fluoride toothpaste (Bioxtra® , Biopharm, Milan, Italy) with salivary enzymes, essential oils, proteins and colostrum extract versus a fluoride toothpaste without menthol on the oral hygiene grade and on the quality of life (QoL) of children with oral mucositis (OM) grade 1 or 2 receiving chemotherapy for Acute Lymphoblastic Leukaemia (ALL). METHODS: Patients between 6 and 14 years with OM were randomly assigned to two groups, group A (Bioxtra® toothpaste) and group B (fluoride toothpaste without menthol). The patients were i
