Causal relationship between sleep characteristics and thyroid function: A bidirectional Mendelian randomization study.

BACKGROUND: Previous researches have revealed some links between thyroid function and sleep characteristics, however it remains unclear which one causes the other. The purpose of this study was to investigate the potential causal relationship between hyperthyroidism, hypothyroidism, and sleep characteristics. METHODS: We utilized aggregated data from published genome-wide association studies (GWAS) to select genetic instruments for sleep variables. The 5 sleep-related traits (chronotype, short sleep duration, long sleep duration, daytime sleepiness, and insomnia) were associated with distinct

Causal association between common rheumatic diseases and arrhythmia: a Mendelian randomization study.

BACKGROUND: Observational studies have suggested a link between rheumatic diseases and arrhythmias. However, these studies have been limited by confounding factors and reverse causality, leaving the causal relationship between rheumatic diseases and arrhythmias uncertain. This study addresses this inquiry using genetic evidence. METHODS: Selected single nucleotide polymorphisms (SNPs) from genome-wide association study (GWAS) data were employed as instrumental variables. Inverse variance weighting (IVW), MR-Egger regression, and the weighted median method were utilized in the two-sample Mende

Sleep disturbance and psychiatric disorders: a bidirectional Mendelian randomisation study.

AIMS: Sleep disturbance is an important factor in the pathophysiology and progression of psychiatric disorders, but whether it is a cause, or a downstream effect is still not clear. METHODS: To investigate causal relationships between three sleep-associated traits and seven psychiatric diseases, we used genetic variants related to insomnia, chronotype and sleep duration to perform a two-sample bidirectional Mendelian randomisation analysis. Summary-level data on psychiatric disorders were extracted from the Psychiatric Genomics Consortium. Effect estimates were obtained by using the inverse-v

Causal relationships between dyslexia and the risk of eight dementias.

Observational and genetic studies have reported the relationship between dyslexia and Alzheimer's disease (AD). Until now, the causal effect of dyslexia on AD risk has remained unclear. We conducted a two-sample univariable Mendelian randomization (MR) analysis to determine the causal association between dyslexia and the risk of AD, vascular dementia (VD), Lewy body dementia (LBD), and frontotemporal dementia (FTD) and its four subtypes. First, we selected 42 dyslexia genetic variants from a large-scale genome-wide association studies (GWAS) dataset and extracted their corresponding GWAS summa

No evidence for schistosome parasite fitness trade-offs in the intermediate and definitive host.

BACKGROUND: The trematode parasite Schistosoma mansoni uses an aquatic snail intermediate and a vertebrate definitive host to complete its life cycle. We previously showed that a key transmission trait-the number of cercariae larvae shed from infected Biomphalaria spp. snails-varies significantly within and between different parasite populations and is genetically controlled by five loci. We investigated the hypothesis that the success of parasite genotypes showing high propagative fitness in the intermediate snail host may be offset by lower reproductive fitness in the definitive vertebrate h

Genotype-phenotype correlation in the spectrum of frontotemporal dementia-parkinsonian syndromes and advanced diagnostic approaches.

The term frontotemporal dementia (FTD) refers to a group of progressive neurodegenerative disorders characterized mainly by atrophy of the f … r palsy (PSP) and cortico-basal syndrome (CBS). Furthermore, FTD-related genes are characterized by genetic pleiotropy and can cause, even in the same family, pure motor phenotypes, findings that underlie the clinical continuum of the spectrum, which has pure cognitive and pure motor phenotypes as the extremes. The genotype-phenotype correlation of the spectrum, FTD …

Assessing Causal Relationship Between Human Blood Metabolites and Five Neurodegenerative Diseases With GWAS Summary Statistics.

Background: Neurodegenerative diseases (NDDs) are the leading cause of disability worldwide while their metabolic pathogenesis is unclear. Genome-wide association studies (GWASs) offer an unprecedented opportunity to untangle the relationship between metabolites and NDDs. Methods: By leveraging two-sample Mendelian randomization (MR) approaches and relying on GWASs summary statistics, we here explore the causal association between 486 metabolites and five NDDs including Alzheimer's Disease (AD), amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), Parkinson's disease (PD), and m

Impact of fetal and maternal genetically predicted birth weight on cardiometabolic risk: a Mendelian randomization study of cytokine mediation in Europeans.

BACKGROUND: Birth weight (BW) is influenced by both fetal and maternal genetic factors and is correlated with cardiometabolic outcomes later in life. Investigating these factors can clarify whether the association between BW and health risk arises from fetal, maternal, or shared genetic factors. Inflammation likely plays a key role in cardiometabolic risk related to BW. This study examined the causal effects of fetal and maternal genetically predicted BW on cardiometabolic outcomes, focusing specifically on the mediating role of inflammatory cytokines. METHODS: We used a two-sample Mendelian

Hyperthyroidism Is Genetically Associated With Reduced Risk of Parkinson's Disease: A Mendelian Randomization Analysis.

Parkinson's disease (PD) is a progressive neurodegenerative disorder whose aetiology involves an intricate interplay of genetic, immune, metabolic and environmental factors. Endocrine dysfunction-particularly disturbances of thyroid hormone signalling-has been proposed as a contributor to neurodegeneration, but conventional observational studies have produced inconsistent results, and prior Mendelian randomization (MR) work has largely focused on continuous thyroid biomarkers rather than clinically defined hyperthyroid disease states. To clarify this relationship, we performed a two-sample bid

Mendelian randomization and transcriptomic data analysis reveals the causal association between gout and breast cancer.

The association between gout and cancer risk has garnered significant interest, particularly in relation to breast cancer, which is the most prevalent cancer among women globally. Nevertheless, the coincidental link between gout and breast cancer, along with its underlying pathogenesis, remains inadequately elucidated. This study utilized publicly available genome-wide association study data from individuals of European ancestry, with sample sizes drawn from multiple genome-wide association study studies, covering genetic variations related to gout and breast cancer. Additionally, transcriptom

Causal effects of physical activity and frailty on pneumothorax risk: a Mendelian randomization study.

INTRODUCTION: Pneumothorax is the presence of air in the pleural cavity, often resulting in respiratory distress and impaired lung function. Although clinical observations have shown that high intensity physical activity and frail state affect respiratory health, their causal role in pneumothorax development is unclear. This study is the first to use a two-sample Mendelian randomization method (TSMR) to investigate the causal relationship between physical activity and frailty and the risk of pneumothorax, with a view to providing new insights into the pathogenesis of pneumothorax and preventiv

Investigating the Gene Relation Between Cervical Spondylosis and Depression: Bidirectional Mendelian Randomization Study.

BACKGROUND: Previous observational studies have suggested a potential link between depression and cervical spondylosis (CS). While it is known that depression and CS can coexist, the specific relationship between them is not fully understood. We hypothesize that there may be connections between the two conditions, but the independent causal relationship of depression as a risk factor for CS, remains uncertain. This particular study has important implications for the future clinical treatment of depression and cervical spondylosis because Mendelian randomization has not been widely used in this
