A synthetic microbial community for the enhanced removal of complex emerging pollutants from domestic wastewater.

Pharmaceutical and personal care products (PPCPs) residues pose ecological and health risks due to their persistence and promotion of antibiotic resistance genes (ARGs) dissemination. Here, we constructed a synthetic microbial community (SMC) comprising two bacterial strains (Sphingopyxis sp. GC21 and Ochrobactrum sp. TCC-2) and one fungal strain (Apiotrichum sp. IB-1) for the enhanced simultaneous removal of three representative PPCPs: chloramphenicol (CAP), triclocarban (TCC), and naproxen (NPX) in simulated wastewater (SW) and real wastewater (RW). Under aerobic conditions, the removal effi

Molecular and Biosafety Perspectives of Bacterial Self-Healing Concrete: From Sporulation and Biomineralization to Public Health Implications.

Bacterial self-healing concrete has emerged as a bio-based strategy to enhance the durability of cementitious materials and reduce the environmental impact associated with premature infrastructure deterioration. Its functional principle relies on microbially induced calcium carbonate precipitation (MICP), through which bacterial metabolism promotes CaCO3 deposition within cracks. However, self-healing efficiency cannot be explained solely by mineral precipitation capacity. Concrete is a restrictive microbial environment characterized by alkalinity, desiccation, osmotic stress, nutrient limitat

Psoriasis risk allele function in activated Th1/17 cells with "memory" to antigen exposure.

Most causal variants for complex diseases are expected to affect gene regulation in a cell- and context-specific manner. Hence, identification of such dynamically functioning variants requires functional readouts in disease-relevant tissues and context. In this study, we prioritized causal variants for psoriasis by adding functional annotations from disease-relevant cells. We demonstrate that disease-relevant immune cells, unlike most other tissues, possess functional annotations that match candidate causal SNPs. Specifically, we identified an eQTL, rs4672505, that reduces B3GNT2 gene expressi

Lactobacillus in autoimmune thyroid diseases: benefit or risk?

Autoimmune thyroid diseases (AITDs), mainly Hashimoto's thyroiditis and Graves' disease, are common organ-specific autoimmune disorders driven by complex interactions among genetic susceptibility, immune dysregulation, environmental exposure, and microbial factors. Increasing evidence supports the relevance of the gut-thyroid axis, in which gut microbiota may influence thyroid hormone metabolism, intestinal barrier integrity, systemic inflammation, and thyroid-directed autoimmunity. Among gut commensals, Lactobacillus-related taxa have attracted attention because selected strains can modulate

Antibiotic-Driven Gut Microbiome Dysbiosis: Resistome Dynamics, Metabolic Disruption, and Paths to Restoration.

The gut microbiome is a dynamic ecosystem that plays essential roles in host metabolism, immune regulation, colonization resistance, and maintenance of intestinal homeostasis. Antibiotic exposure profoundly disrupts this ecosystem by reducing microbial diversity, depleting beneficial commensals, reshaping microbial metabolic functions, and remodeling the gut resistome through the selection and dissemination of antibiotic resistance genes (ARGs). Increasing evidence from longitudinal metagenomic, multi-omics, and experimental studies indicates that these perturbations may persist long after ant

Unlock the Potential of Pichia in Coffee: Knowledge Transfer From Fermented Beverages.

Considerable research has focused on non-Saccharomyces yeasts to explore their biochemical potential and genetic diversity beyond the capabilities of the conventional starter culture, Saccharomyces cerevisiae. Among these, species of the genus Pichia have emerged as promising candidates in diverse food fermentation systems. Although studies on the application of Pichia species in coffee fermentation remain limited, existing evidence indicates positive outcomes, including improved regulation of fermentation kinetics and enhanced coffee bean quality. This review critically examines the occurrenc

Serum proteomic and metabolomic analyses from patients with IBD identify biological pathways associated with treatment success with anti-integrin therapy.

Crohn's disease (CD) and ulcerative colitis (UC) are chronic inflammatory diseases of the gastrointestinal tract believed to arise from an imbalance between its epithelial, immune and microbial components. It has been shown that biological differences (e.g. genetic, epigenetic, microbial, environmental) exist between patients with IBD. It is also known that there is important heterogeneity in the response to therapies that target very specific biological pathways (e.g. TNF-alpha signaling, IL-23R signaling, immune cell trafficking). The aim of this study was to identify potential biological di

Enteric glia regulate Paneth cell secretion and intestinal microbial ecology.

Glial cells of the enteric nervous system (ENS) interact closely with the intestinal epithelium and secrete signals that influence epithelial cell proliferation and barrier formation in vitro. Whether these interactions are important in vivo, however, is unclear because previous studies reached conflicting conclusions (Prochera and Rao, 2023). To better define the roles of enteric glia in steady state regulation of the intestinal epithelium, we characterized the glia in closest proximity to epithelial cells and found that the majority express the gene Proteolipid protein 1 (PLP1) in both mice

Metagenome-Based Functional Differentiation of Gut Microbiota and Ecological Adaptation Among Geographically Distinct Populations of Przewalski's Gazelle (Procapra przewalskii).

Przewalski's gazelle (Procapra przewalskii) is an endangered ungulate endemic to the Qinghai-Tibet Plateau, with a small population size and exposure to multiple ecological pressures. Its gut microbiota may play a crucial role in host environmental adaptation. To investigate the functional divergence of gut microbial communities, we performed high-throughput metagenomic sequencing on 105 wild fecal samples collected from 10 geographic regions around Qinghai Lake. The results revealed significant regional differentiation in key functional modules related to metabolism, antibiotic resistance mec

Recombinant Komagataella phaffii cells loaded with diaphorase-mimicking nanozymes for formaldehyde detection, detoxification and electricity generation from wastewater.

Formaldehyde (FA) is a toxic, volatile compound commonly used in industry and frequently found in polluted environments. Its rapid and dependable identification is vital for both environmental assessment and health protection. Microbial biosensors enhanced with nanomaterials offer promising platforms for FA sensing due to their sensitivity, cost-efficiency, and operational simplicity. However, unstable gene expression often limits the practical application of genetically modified microorganisms. To address these challenges, we developed a hybrid biosensing system based on Komagataella phaffii

The influence of depth on the global deep-sea plasmidome.

Plasmids play a crucial role in facilitating genetic exchange and enhancing the adaptability of microbial communities. Despite their importance, environmental plasmids remain understudied, particularly those in fragile and underexplored ecosystems such as the deep-sea. In this paper we implemented a bioinformatics pipeline to study the composition, diversity, and functional attributes of plasmid communities (plasmidome) in 81 deep-sea metagenomes from the Tara and Malaspina expeditions, sampled from the Pacific, Atlantic, and Indian Oceans at depths ranging from 270 to 4005 m. We observed an a

Prevotella intermedia Synergistically Exacerbates Pneumonia Induced by Oral Streptococci.

BACKGROUND: The precise mechanisms of respiratory infection caused by oral anaerobic bacteria remain elusive. Unexpectedly, bacterial microbiota analysis with 16S rRNA revealed "hidden" mixed infections of anaerobic bacteria and commensal oral Streptococcus species in patients with community-acquired pneumonia. The study aimed to elucidate the mechanisms by which Prevotella intermedia exacerbates oral streptococcal pneumonia. METHODS: Oral streptococci were oropharyngeally administered with the culture supernatants of P intermedia (PiSup) in mice to assess survival, microbial burden, inflamma

Microbes, macrophages, and melanin: a unifying theory of disease as exemplified by cancer.

It is widely accepted that cancer mostly arises from random spontaneous mutations triggered by environmental factors. Our theory challenges the idea of the random somatic mutation theory (SMT). The SMT does not fit well with Charles Darwin's theory of evolution in that the same relatively few mutations would occur so frequently and that these mutations would lead to death rather than survival of the fittest. However, it would fit well under the theory of evolution, if we were to look at it from the vantage point of pathogens and their supporting microbial communities colonizing humans and muta

Microbiota and antibiotic exposure in sarcoidosis.

BACKGROUND: Sarcoidosis is a multisystem granulomatous disorder characterized by excessive immune activation in genetically predisposed individuals. Despite decades of investigation, its etiology remains unresolved. Emerging evidence indicates that disruptions in host-microbiota homeostasis may contribute to immune dysregulation and disease persistence, challenging the traditional view of sarcoidosis as a purely immune-mediated condition. PERSPECTIVE: Recent findings have revealed alterations in both respiratory and intestinal microbiota among patients with sarcoidosis, suggesting that microb

Epigenetic Mechanisms in Aging: Extrinsic Factors and Gut Microbiome.

BACKGROUND/OBJECTIVES: Aging is a natural physiological process involving biological and genetic pathways. Growing evidence suggests that alterations in the epigenome during aging result in transcriptional changes, which play a significant role in the onset of age-related diseases, including cancer, cardiovascular disease, diabetes, and neurodegenerative disorders. For this reason, the epigenetic alterations in aging and age-related diseases have been reviewed, and the major extrinsic factors influencing these epigenetic alterations have been identified. In addition, the role of the gut microb

Application of dissimilatory iron-reducing bacteria for the remediation of soil and water polluted with chlorinated organic compounds: Progress, mechanisms, and directions.

Chlorinated organic compounds are widely used as solvents, but they are pollutants that can have adverse effects on the environment and human health. Dissimilatory iron-reducing bacteria (DIRB) such as Shewanella and Geobacter have been applied to treat a wide range of halogenated organic compounds due to their specific biological properties. Until now, there has been no systematic review on the mechanisms of direct or indirect degradation of halogenated organic compounds by DIRB. This work summarizes the discussion of DIRB's ability to enhance the dechlorination of reaction systems through di

Potential clinical implications of molecular mimicry-induced autoimmunity.

BACKGROUND: Molecular mimicry is hypothesized to be a mechanism by which autoimmune diseases are triggered. It refers to sequence or structural homology between foreign antigens and self-antigens, which can activate cross-reactive lymphocytes that attack host tissues. Elucidating the role of molecular mimicry in human autoimmunity could have important clinical implications. OBJECTIVE: To review evidence for the role of molecular mimicry in major autoimmune diseases and discuss potential clinical implications. METHODS: Comprehensive literature review of clinical trials, observational studies,

Microbial applications for sustainable space exploration beyond low Earth orbit.

With the construction of the International Space Station, humans have been continuously living and working in space for 22 years. Microbial studies in space and other extreme environments on Earth have shown the ability for bacteria and fungi to adapt and change compared to "normal" conditions. Some of these changes, like biofilm formation, can impact astronaut health and spacecraft integrity in a negative way, while others, such as a propensity for plastic degradation, can promote self-sufficiency and sustainability in space. With the next era of space exploration upon us, which will see crew

Human gene and microbial analyses in rectal cancer complete responses to radiotherapy.

BACKGROUND: The gold standard treatment for locally advanced rectal cancer is total mesorectal excision after preoperative chemoradiotherapy. Response to chemoradiotherapy varies, with some patients completely responding to the treatment and some failing to respond at all. Identifying biomarkers of response to chemoradiotherapy could allow patients to avoid unnecessary treatment-associated morbidity rate. While previous studies have attempted to identify such biomarkers, none have reached clinical utility, which may be due to heterogeneity of the cancer. In this study, potential human gene and
