pH and Glucose Dual-Responsive Hydrogels Promoted Diabetic Wound Healing by Remodeling the Wound Microenvironment.

The microenvironment of diabetic wounds is complicated and characterized by hyperglycemia, hyperinflammation, persistent infection, hypoxia, and ischemia, making wound restoration and healing extremely challenging. Therefore, functional hydrogel dressings with the ability to regulate the microenvironment of diabetic wounds are a promising strategy for the treatment of diabetic wounds. In this study, a pH/glucose dual-responsive hydrogel based on phenylboric acid-modified carboxymethyl chitosan (CMCSPBA), aldehyde-terminated polyethylene glycol (PEGCHO), and polyvinyl alcohol (PVA) has been dev

Simultaneous modulation of chirality and mechanical properties in supramolecular hydrogels by varying alkyl spacer length.

Mimicking the extracellular matrix's (ECM's) integration of chiral and mechanical cues remains challenging in synthetic hydrogels. Here, we demonstrate that alkyl spacer engineering in C2-symmetric benzene-para-dicarboxamide phenylalanine derivatives enables simultaneous control of supramolecular handedness and mechanics: helical handedness follows spacer odd-even effects, while spacer elongation (n = 2→4) drives a 17-fold elastic modulus enhancement (0.45→7.64 kPa). This dual regulation provides a versatile strategy for designing biomimetic hydrogels.

Peptide Hydrogels for Renal Carcinoma Therapy via Synergistic Inhibition of Glycolysis and Mitochondrial Metabolism Reprogramming.

The pathogenesis of renal cell carcinoma (RCC) is intricately associated with metabolic reprogramming as a key characteristic of cancer malignancy. This study presents a peptide-based hydrogel platform to disrupt tumor metabolic plasticity by simultaneously targeting oxidative phosphorylation (OXPHOS) and glycolysis. The hydrogels were synthesized through the self-assembly of 9-fluorenylmethoxycarbonyl-modified diphenylalanine (Fmoc-FF), followed by electrostatic complexation with glycol chitosan (GCS). The hydrophobic OXPHOS inhibitor Oligomycin A (Oligo) and the hydrophilic glycolysis inhibi…

Self-contracting thermo-adhesive hydrogels with artesunate-loaded chitin/lignin micelles for accelerated wound healing.

Due to the skin's limited self-repair capacity after injury, developing effective wound dressings is crucial to accelerate wound closure and enhance tissue regeneration. In the work, a thermo-sensitive and tissue-adhesive hydrogel with an interpenetrating polymer network (QGNPI) was fabricated by using N-isopropylacrylamide (NIPAM), galactomannan, and chitin-lignin micelles. The abundant hydroxyl groups within galactomannan conferred enhanced adhesion properties to the hydrogel. To improve the stability of artesunate, it was encapsulated within the chitin-lignin micelles. Moreover, the QGNPI h

Self-Assembling Peptide Hydrogels Support Stromal Vascular Fraction Viability to Promote In Vivo Nerve Regeneration.

Autograft, the gold standard in nerve reconstruction, outperforms the alternatives of acellular allograft/nerve conduits likely due to the transplanted Schwann cell population within. The stromal vascular fraction (SVF) is a heterogenous cell isolate, that may improve nerve regeneration outcomes when transplanted at the site of a nerve defect. Self-Assembling Peptide hydrogels (SAPH) are synthetic materials derived from short chains of biological amino acids. They are safe, injectable hydrogels whose charge and mechanical properties are easily tuned. Our hypothesis is that SVF, when transplant…

Metabolic microenvironment-forming porous hydrogels for the protection of phenylalanine ammonia-lyase in the intestine.

Biological therapeutics drive biomedical innovation, yet their oral delivery is hindered by gastrointestinal instability. Phenylalanine ammonia-lyase (PAL), a promising enzyme for phenylketonuria management, exemplifies this challenge. Here, we present ultraporous template-based hydrogels (TGels) that protect and deliver PAL in the small intestine. PAL-TGels are obtained via UV crosslinking of poly(ethylene glycol) diacrylamide in the presence of sacrificial CaCO3 templates, followed by absorption-based loading of PAL and a protease inhibitor. The interconnected porous matrix retains PAL while…

Evaluation of Carboxymethyl Chitosan-Genipin Hydrogels as Reservoir Systems for Suramin Delivery in Epithelial Tissues.

Hydrogels (HDs) offer a promising platform for localized and sustained drug delivery. In this study, carboxymethyl chitosan (CMC)-based hydrogels were crosslinked with genipin and evaluated for the controlled release and tissue retention of suramin, a polyanionic drug with anti-inflammatory and antifibrotic properties. The influence of crosslinking density (1%, 3%, and 5%) on drug release, permeation kinetics, and retention was investigated using in vitro synthetic membranes and reconstructed human epithelial tissue models. The 1% genipin HD exhibited the highest cumulative release and drug re…

Photoactivated growth factor release from bio-orthogonally crosslinked hydrogels for the regeneration of corneal defects.

In situ-forming hydrogels are an attractive option for corneal regeneration, and the delivery of growth factors from such constructs have the potential to improve re-epithelialization and stromal remodeling. However, challenges persist in controlling the release of therapeutic molecules from hydrogels. Here, an in situ-forming bio-orthogonally crosslinked hydrogel containing growth factors tethered via photocleavable linkages (PC-HACol hydrogel) was developed to accelerate corneal regeneration. Epidermal growth factor (EGF) was conjugated to the hydrogel backbone through photo-cleavable (PC) s…

Temperature-Governed Microstructure of Poly(vinyl alcohol) Hydrogels Prepared through Mixed-Solvent-Induced Phase Separation.

The formation of phase-separated structures in hydrogels plays a crucial role in determining their optical and mechanical properties. Traditionally, phase-separated hydrogels are prepared through a two-step process involving initial hydrogel synthesis followed by post-treatment. In this study, we present an approach for temperature-governed phase separation microstructure modulation in hydrogels, harnessing the cononsolvency effect. This method allows the phase-separated structure to develop during hydrogel synthesis, significantly simplifying the preparation process. Importantly, we found tha…

Fabrication, characteristics and properties of α-lactalbumin fibril-derived hydrogels: Effects of metal ions type.

The effects of different valence metal ions on the formation of hydrogels with α-lactalbumin fibrils (ALAF) were comprehensively examined in this study. The properties of hydrogel were generally characterized with water holding capacity (WHC), rheology, texture, DSC and ICP tests. Except FeCl3, it was shown that KCl, NaCl, CaCl2, MgCl2, NiCl2, and AlCl3 at 90 mM could induce the formation of hydrogels with ALAF (40 mg/mL), and hydrogels formed by high valence metal salts had more good properties (viscoelasticity, WHC, and thermal stability), and the amounts of metal ions released from hydrogel…

Rheological and Injectability Evaluation of Sterilized Poloxamer-407-Based Hydrogels Containing Docetaxel-Loaded Lipid Nanoparticles.

Nanostructured lipid carriers (NLCs) have the potential to increase the bioavailability and reduce the side effects of docetaxel (DTX). However, only a small fraction of nanoparticles given intravenously can reach a solid tumor. In situ-forming gels combined with nanoparticles facilitate local administration and promote drug retention at the tumor site. Injectable hydrogels based on poloxamer 407 are excellent candidates for this hybrid nanoparticle-hydrogel system because of their thermoresponsive behavior and biocompatibility. Therefore, this work aimed to develop injectable poloxamer hydrog…

Block Sequence Effects on the Self-Assembly Behaviors of Polypeptide-Based Penta-Block Copolymer Hydrogels.

Peptide-based hydrogels have great potential for applications in tissue engineering, drug delivery, and so on. We systematically synthesize, characterize, and investigate the self-assembly behaviors of a series of polypeptide-based penta-block copolymers by varying block sequences and lengths. The copolymers contain hydrophobic blocks of poly(γ-benzyl-l-glutamate) (PBG, Bx) and two kinds of hydrophilic blocks, poly(l-lysine) (PLL, Ky) and poly(ethylene glycol) (PEG, EG34), where x and y are the number of repeating units of each block, where PBG and PLL blocks have unique functions for nerve re…

Enhanced ROS scavenging and tissue adhesive abilities in injectable hydrogels by protein modification with oligoethyleneimine.

Postsurgical treatment comprehensively benefits from the application of tissue-adhesive injectable hydrogels, which reduce postoperative complications by promoting wound closure and tissue regeneration. Although various hydrogels have been employed as clinical tissue adhesives, many exhibit deficiencies in adhesive strength under wet conditions or in immunomodulatory functions. Herein, we report the development of reactive oxygen species (ROS) scavenging and tissue-adhesive injectable hydrogels composed of polyamine-modified gelatin crosslinked with the 4-arm poly (ethylene glycol) crosslinker…
