Dopaminergic mechanisms in self-inflicting biting behavior.

Dopaminergic mechanisms involved in self-inflicting biting behavior (SBB) were investigated in two animal models: monkeys with unilateral ventromedial tegmental (VMT) lesions of the brainstem and rats with unilateral 6-hydroxydopamine (6-OHDA) lesions of the nigrostriatal dopamine (DA) neurons. The administration of mixed D1/D2 DA agonists to some monkeys with unilateral VMT lesions of the brainstem elicits SBB of the forelimb digits contralateral to the lesion and spasticity of the contralateral hindlimb. This behavior is prevented by pretreatment with the selective D1 antagonist SCH 23390 an

Differential effects of fluphenazine-N-mustard, on calmodulin activity and on D1 and D2 dopaminergic responses.

Fluphenazine-N-mustard (FNM) has been shown to irreversibly block dopaminergic receptor sites and inhibit certain dopaminergically-mediated behaviors. In this study we measured whether FNM has any differential effects on D1 and D2 dopaminergic events. Accordingly, we examined the relative effects of FNM on rotational behavior induced by SKF 38393 (D1 agonist) and Ly 171555 (D2 agonist) in mice with unilateral, 6-hydroxydopamine-induced lesions of the striatum and the effects of FNM on the binding of [3H]Sch 23390 (D1 ligand) and [3H]spiroperidol (D2 ligand) to mouse striatal membranes. FNM inh

Chronic pharmacological manipulation of dopamine receptors in brain.

Dopamine receptors in the brain play an important role in the treatment of schizophrenia and in the development of tardive dyskinesia. In Parkinson's disease the loss of dopamine innervation and the use of chronic administration of L-DOPA or therapy with dopamine agonists also affects the function of dopamine receptors in brain. Subacute administration of neuroleptic drugs to rodents for a few weeks followed by the withdrawal of the drug induces supersensitivity of dopamine receptors in the striatum. However, the long-term administration of neuroleptic drugs to rodents shows that typical neuro

[3H]mazindol binding associated with neuronal dopamine and norepinephrine uptake sites.

[3H]Mazindol labels neuronal dopamine uptake sites in corpus striatum membranes (KD = 18 nM) and neuronal norepinephrine uptake sites in cerebral cortex and submaxillary/sublingual gland membranes (KD = 4 nM). The potencies of various inhibitors of biogenic amine uptake in reducing [3H]mazindol binding in striatal membranes correlate with their potencies for inhibition of neuronal [3H]dopamine accumulation, whereas their potencies in reducing [3H]mazindol binding to cortical and salivary gland membranes correlate with their potencies for inhibition of neuronal [3H]norepinephrine accumulation.

Pars compacta of the substantia nigra modulates motor activity but is not involved importantly in regulating food and water intake.

Precise, bilateral radio-frequency lesions of pars compacta of the substantia nigra in rats resulted in the immediate and sustained appearance of hyperactivity, but such lesions did not produce significant alterations in food or water intake. These behavioral effects were correlated with considerable, histochemically assessed loss of dopamine terminals in the caudate-putamen complex, but dopamine innervation in nucleus accumbens and other forebrain areas was only slightly affected. The magnitude of motor activity increase was positively correlated with the degree of pars compacta involvement.

Hyperactivity induced by stimulation of separate dopamine D-1 and D-2 receptors in rats with bilateral 6-OHDA lesions.

The effects of DA agonists and antagonists with different dopamine (DA) D-1 and D-2 receptor selectivity have been studied in rats with bilateral 6-OHDA lesions. The D-1 agonist SK & F 38393, the D-2 agonist pergolide and the mixed agonist apomorphine all induced marked hyperactivity in lesioned rats in doses which were without stimulant effect in sham-operated animals. The hyperactivity induced by SK & F 38393 was blocked by the DA D-1 antagonist SCH 23390, but unaffected by the D-2 antagonists spiroperidol or clebopride. Pergolide-induced hyperactivity showed the reverse selectivity. The mix

Late endocrine effects of L-dopa, 5-HTP, and 6-OH-dopa administered to neonatal rats.

Neonatal rats were injected with L-dopa, 5-hydroxytryptophan (5-HTP) or 6-hydroxy-dopa (6-OH-dopa) and allowed to mature. Eye opening, vaginal opening, endocrine organ weights and pituitary hormone concentration in the serum were measured. Treatment with either the dopamine (DA) precursor or the serotonin (5-HT) precursor caused an acceleration in eye opening, a significant decrease in serum TSH (males) an elevation of serum prolactin (Prl; males), and a lowering of serum growth hormone (females). After chemical sympathectomy (6-OH-dopa) vaginal opening was delayed, thyroid weights were increa

Gramicidin A attenuates oxidative stress, mitochondrial dysfunction, DNA damage, and inflammation in 6-OHDA-induced SH-SY5Y cells.

BACKGROUND: Oxidative stress and mitochondrial dysfunction are major contributors to neurodegenerative diseases, and this study investigated the protective potential of Gramicidin A in a 6-hydroxydopamine (6-OHDA)-induced SH-SY5Y cellular injury model relevant to Parkinson's disease (PD). METHODS AND RESULTS: Following the 3-(4,5-Dimethyl-2-thiazolyl)- 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay to determine the appropriate Gramicidin A concentration, cells were evaluated for oxidative stress parameters, including reactive oxygen species (ROS), total antioxidant status (TAS), total oxidan

Neural stem cells transplantation combined with ethyl stearate promotes the activation of quiescent NSCs and improves PD rats motor behavior.

BACKGROUND: This research aimed to examine the impact of co-transplantation of ethyl stearate (PubChem CID: 8122) and neural stem cells (NSCs) on endogenous NSCs in Parkinson's disease (PD) model rats, and also determine the impact of ethyl stearate on quiescent neural stem cells (qNSCs) and explore its possible mechanism. METHODS: The induction of PD rats was achieved through the injection of 6-hydroxydopamine (6-OHDA) into the right striatum (STR) and substantia nigra (SN). Transplant treatment was then followed for 6 weeks in the right STR. Apomorphine (APO)-induced rotation behavior and t

Extracellular vesicles released in vitro from a Parkinson's disease-like model: a combined biochemical and spectroscopic approach as proof of concept for the diagnosis of neurodegenerative diseases.

BACKGROUND: Extracellular vesicles (EVs) are stable carriers of molecular signals and can cross the blood-brain barrier, making them promising non-invasive biomarkers for diseases of the central nervous system. While their diagnostic potential is already established and is present in many clinical trials for brain tumors, applications in neurodegenerative disorders such as Parkinson's disease (PD) are still emerging. Although researchers have shown that EVs are involved in the intercellular diffusion of aggregated α-synuclein, a hallmark of PD pathology, the clinical use of EVs as disease biom

Long-term continuous theta burst stimulation ameliorates L-DOPA-induced dyskinesia in Parkinsonian rats through modulation of the cerebello-thalamo-striatal circuit.

Levodopa-induced dyskinesia (LID) is a debilitating complication of Parkinson's disease therapy. Emerging evidence suggests that the cerebellum is involved via cerebello -thalamo-striatal pathways. We first performed dual viral tracing to confirm cerebello-thalamo-striatal connectivity in a unilateral 6- hydroxydopamine rat model of LID. We then compared the efficacy of two cerebellar continuous theta burst stimulation (cTBS) protocols: a 2block protocol (14 days) and an intensified 3block protocol (10 days). Behavioral outcomes were assessed using the abnormal involuntary movement scale (AIMs)

TMBIM6 enhances dopaminergic neuron survival by modulating the IRE1a pathway in Parkinson's disease.

The core pathological hallmark of Parkinson's disease (PD) is the progressive degeneration of dopaminergic (DAergic) neurons in the substantia nigra pars compacta (SNpc), driven by misfolding and aggregation of a-synuclein (aSyn) into Lewy bodies. This triggers severe cellular dysfunction, including endoplasmic reticulum (ER) stress and the dysregulation of the unfolded protein response (UPR). TMBIM6, an anti-apoptotic ER protein, inhibits the UPR sensor IRE1a. Although TMBIM6 exhibits neuroprotective effects in neurological disorders, its role in PD-related DAergic neuron survival remains unk

Application of In Silico QSAR and Molecular Docking Studies to a Series of Xanthine-Based Analogues and Design, Synthesis and Pharmacological Evaluation of Identified New Potential Selective MAO-B Inhibitors.

Background/Objectives: Methylxanthines, such as caffeine, exhibit neuroprotective properties in neurodegenerative conditions, partly linked to modulation of monoamine oxidase B (MAO-B) and oxidative stress pathways. This work aimed to design, synthesize and functionally characterize new caffeine-8-methylthioglycolic acid derivatives as selective MAO-B inhibitors with neuroprotective potential. Methods: A QSAR model was built on 94 studies of xanthine derivatives to guide the design of ten new semi- and thiosemicarbazides (Jas1-Jas10), followed by molecular docking to human MAO-B (PDB: 2V5Z) us

Consensus Molecules Associated with Parkinson's Disease.

Parkinson's disease (PD) has been associated with some types of food and drugs. Here, we query PubMed for the association of PD with foods and drugs, using a list of 217,776 compounds derived from the Human Metabolome Database (HMDB). To calculate associations, a Python script was developed to query PubMed for co-citations of PD with each compound, and adjust this count for compound abundance. Notably, PD is found to be associated with small-molecule drugs, adjunctive therapies, contraindicated drugs, diagnostic agents, biomarkers, conditional essential molecules, and inducers. Drugs include L

Amantadine Modulates Action-Specific Neural Ensembles in Hypokinetic and Hyperkinetic Conditions.

BACKGROUND: In the classical model of basal ganglia circuitry, striatal spiny projection neurons of the direct and indirect pathways (dSPNs, iSPNs) promote and suppress movement, respectively, and exhibit unbalanced activity levels during hypokinetic or hyperkinetic conditions. Most therapies for these conditions are thought to work by rebalancing the relative activity of dSPNs and iSPNs toward normal levels. However, the mechanism of amantadine, which uniquely improves both hypokinetic and hyperkinetic conditions, is poorly understood. OBJECTIVE: Our aim was to determine whether amantadine r

Mechanistic insights into the neuroprotective effects of monoterpene-rich Eucalyptus citriodora essential oil: integrated in silico and in vitro analysis in 6-OHDA-treated N2a neuronal cells.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuronal loss driven by mitochondrial dysfunction, oxidative stress, and proteostasis collapse, for which current therapies remain symptomatic. This study investigated the neuroprotective effects of Eucalyptus citriodora essential oil (ECEO) on 6-hydroxydopamine (6-OHDA)-induced toxicity in N2a neuroblastoma cells. GC-MS analysis showed that ECEO was dominated by oxygenated monoterpenes, mainly citronellal (60.7%), isopulegol (16%), and citronellol (5.7%). In silico analysis of these constituents showed favorable ADMET profi

Genetically Encoded Constitutive Upregulation of β2 Subunit-Containing Neuronal Nicotinic Acetylcholine Receptors Is Neuroprotective in Female Parkinsonian Mice.

Parkinson's disease is projected to rise to pandemic proportions by 2050, which has resulted in an urgent need for disease-modifying treatments. We previously showed that in a mouse model of parkinsonism with unilateral 6-hydroxydopamine (6-OHDA) injection into the dorsolateral striatum, low doses of the neuronal nicotinic acetylcholine receptor (nAChR) partial agonist and smoking cessation drug, cytisine, exert neuroprotection in substantia nigra pars compacta (SNc) dopaminergic (DA) neurons of only female mice by reducing endoplasmic reticulum (ER) stress and that cytisine-mediated neuroprot
