4-Aminopyrazolo[3,4-d]pyrimidine (4-APP) as a novel inhibitor of the RNA and DNA depurination induced by Shiga toxin 1.

Shiga toxin 1 (Stx1) catalyses the removal of a unique and specific adenine from 28S RNA in ribosomes (RNA-N-glycosidase activity) and the release of multiple adenines from DNA (DNA glycosylase activity). Added adenine behaves as an uncompetitive inhibitor of the RNA-N-glycosidase reaction binding more tightly to the Stx1-ribosome complex than to the free enzyme. Several purine derivatives and analogues have now been assayed as inhibitors of Stx1. Most of the compounds showed only minor differences in the rank order of activity on the two enzymatic reactions catalysed by Stx1. The survey highl

Both hydroxylamine and nitroxide protect cardiomyocytes from oxidative stress.

The unique anti-oxidative activity of nitroxide radicals protecting against reactive oxygen-derived species (ROS) has been recently demonstrated in several model systems. The present study focuses on the activity of nitroxide and of its reduced form in cultured rat ventricular cardiomyocytes exposed to O2.- and H2O2 generated by hypoxanthine (HX) and xanthine oxidase (XO). To evaluate cell injury, spontaneous beating, leakage of lactate dehydrogenase (LDH), and depletion of cellular ATP were determined. The protective effect of 4-OH-2,2,6,6-tetramethyl-piperidine-N-oxyl (TPL) was compared with

Effects of ketoconazole on ovulatory changes in the rat: implications on the role of a meiosis-activating sterol.

In-vitro studies on mouse oocytes have shown that human follicular fluid and bull testes contain an activity which partially overrides the inhibitory action of hypoxanthine on meiosis. This activity was ascribed to two closely related sterols, subsequently named meiosis-activating sterols (MAS). We have used a potent inhibitor of sterol synthesis, ketoconazole, in order to test in vivo and in vitro whether MAS play a necessary physiological role in the resumption of meiosis in the rat. When administered systemically, ketoconazole (8.3-16.6 mg/rat) suppressed ovulation by 40%. Local unilateral

Induction of mutations in V79-4 mammalian cells under hypoxic and aerobic conditions by the cytotoxic 2-nitroimidazole-aziridines, RSU-1069 and RSU-1131. The influence of cellular glutathione.

Incubation of the 2-nitroimidazole-aziridine, RSU-1069 [1-(2-nitro-1-imidazolyl)-3-(1-aziridinyl)-2-propanol], and its monomethylaziridine analogue, RSU-1131 [1-(2-nitro-1-imidazolyl)-3-(1-(2-methylaziridinyl))-2-propanol], with V79-4 mammalian cells for 2 hr under aerobic or hypoxic conditions induces mutations as measured at the hypoxanthine phosphoribosyl transferase locus. The ability of these agents to induce mutations is increased by a factor of 12-14 under hypoxic conditions. The increased cytotoxicity of these agents under hypoxic conditions was confirmed following a 2 hr incubation pe

Protection by alpha-thalassaemia against Plasmodium falciparum malaria: modified surface antigen expression rather than impaired growth or cytoadherence.

We have attempted to determine the cellular mechanism by which alpha-thalassaemia may protect against Plasmodium falciparum malaria. Invasion and development of P. falciparum in the microcytic red cells of two-gene deletion forms of alpha-thalassaemia when measured morphologically or by [3H]hypoxanthine incorporation were normal compared to controls. Normal invasion rates were also observed following schizogony in thalassaemic red cells. Neither the addition of the oxidant menadione, 30% oxygen, nor modified medium, produced differential damage to parasites within thalassaemic cells. Furthermo

Adenylate deaminase deficiency in a mutant murine T cell lymphoma cell line.

From a population of wild type S49 cells, a clone, DTB6, was isolated in a single step from selective medium containing thymidine and dibutyryl cyclic AMP that exhibited a 60% deficiency in AMP deaminase (AMP-D) activity. The AMP-D deficiency conferred to the DTB6 cells a striking susceptibility to killing by low concentrations of either adenine or adenosine, the latter in the presence of an inhibitor of adenosine deaminase activity. This growth supersensitivity of DTB6 cells toward adenine could be ameliorated by the addition of hypoxanthine to the culture medium. Immunoprecipitation of AMP-D

Isolation of the human chromosomal band Xq28 within somatic cell hybrids by fragile X site breakage.

The chromosomal fragile-site mapping to Xq27.3 is associated with a frequent form of mental retardation and is prone to breakage after induced deoxyribonucleotide pool perturbation. The human hypoxanthine phosphoribosyltransferase (HPRT) and glucose-6-phosphate dehydrogenase (G6PD) genes flank the fragile X chromosome site and can be used to monitor integrity of the site in human-hamster somatic cell hybrids deficient in the rodent forms of these activities. After induction of the fragile X site, negative selection for HPRT and positive enrichment for G6PD resulted in 31 independent colonies o

Retention of cyclic AMP response to TSH in a cloned human thyrocyte/T cell hybridoma (HY2-15).

Our observation of a human T cell leukemia cell (Molt 4) demonstrating low affinity thyroid-stimulating hormone (TSH) responses, as evidenced by generation of cyclic AMP, led us to test Molt 4 cells as a suitable partner for immortalizing high affinity TSH receptors present on human thyroid cells. Therefore, we generated a hybridoma (HY2-15) by a fusion between thyroid monolayer cells from a patient with Graves' disease, and a hypoxanthine-aminopterin-thymidine (HAT)-sensitive variant of this human T cell leukemia line, Molt 4-8AGR. The hybrid nature of HY2-15 was confirmed by DNA histograms u

Effects of asbestos fibers on cell division, cell survival, and formation of thioguanine-resistant mutants in Chinese hamster ovary cells.

The ability of crocidolite fibers to induce point mutations and mitotic abnormalities in Chinese hamster ovary (CHO) cells was examined in cell cultures. The purpose has been to study the possibilities for establishing in vitro test methods to quantify genetic damage induced by asbestos and other mineral fibers. Results obtained with the CHO/hypoxanthine guanine phosphoribosyl transferase system indicated that crocidolite fibers per se do not significantly increase the number of thioguanine-resistant mutants. Crocidolite fibers also failed to potentiate the mutagenicity of benzo[a]pyrene. Time

Purine oversecretion in cultured murine lymphoma cells deficient in adenylosuccinate synthetase: genetic model for inherited hyperuricemia and gout.

Alterations in several specific enzymes have been associated with increased rates of purine synthesis de novo in human and other mammalian cells. However, these recognized abnormalities in humans account for only a few percent of the clinical cases of hyperuricemia and gout. We have examined in detail the rates of purine production de novo and purine excretion by normal and by mutant (AU-100) murine lymphoma T cells (S49) 80% deficient in adenylosuccinate synthetase [IMP:L-aspartate ligase (GDP-forming), EC 6.3.4.4]. The intracellular ATP concentration of the mutant cells is slightly diminishe

Carbon monoxide oxidation by Clostridium thermoaceticum and Clostridium formicoaceticum.

Cultures of Clostridium formicoaceticum and C. thermoaceticum growing on fructose and glucose, respectively, were shown to rapidly oxidize CO to CO(2). Rates up to 0.4 mumol min(-1) mg of wet cells(-1) were observed. Carbon monoxide oxidation by cell suspensions was found (i) to be dependent on pyruvate, (ii) to be inhibited by alkyl halides and arsenate, and (iii) to stimulate CO(2) reduction to acetate. Cell extracts catalyzed the oxidation of carbon monoxide with methyl viologen at specific rates up to 10 mumol min(-1) mg of protein(-1) (35 degrees C, pH 7.2). Nicotinamide adenine dinucleot

Targeted metabolomics of nucleotide intermediates for biomarker discovery in acute kidney injury.

Acute kidney injury (AKI) is a common syndrome among critically ill patients with high incidence and high mortality, characterized by elevated serum creatinine (SCr) and uric acid (UA) levels. Given that xanthine is a key intermediate in nucleotide metabolism and the direct precursor of UA, profiling of nucleotide metabolic dysregulation in AKI remains largely unexplored. Using ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS), we simultaneously quantified 29 nucleotide intermediates both in plasma and matched urine samples from 58 propensity score-matched pai

Unravelling the effect of autochthonous scotta-derived vs. commercial starter cultures in pilot-scale hard sheep milk cheese production: A multi-omics and sensory combined approach.

Autochthonous whey-based (scotta-innesto) starters are increasingly recognized as a valuable resource to preserve microbial biodiversity and sensory identity in hard sheep milk cheeses produced under PDO-type specifications. In this study, two indigenous scotta-innesto cultures collected in Sardinia (Italy) in the 1960s were compared with a widely used commercial starter in pilot-scale hard sheep milk cheese manufacture produced according to Pecorino Romano PDO specifications. To this end, an integrated multidisciplinary approach was employed by combining starter metagenomics, culture-dependen

Dynamic Triangulation of Resting-State Networks and CSF Metabolites Reveals Bidirectional Causal Pathways in Multiple Sclerosis: A Mendelian Randomization Analysis.

Emotional disorders associated with multiple sclerosis (MS) are key drivers of disease progression. Recently, resting-state functional magnetic resonance imaging (rs-fMRI) and cerebrospinal fluid (CSF) metabolites have been found to play a crucial role in elucidating this process, but the causal relationship between them remains to be clarified. Exploring underlying mechanisms is critical for developing targeted therapies, especially combined multidimensional neuromodulation. This study employed bidirectional two-sample Mendelian randomization (MR) analysis to investigate the causal relationsh

Investigation of 6-thioguanine as a strategy to overcome methotrexate resistance in a mouse model of leptomeningeal carcinomatosis.

PURPOSE: Leptomeningeal carcinomatosis (LC), the dissemination of malignant cells into the cerebrospinal fluid, occurs in 3-5% of patients with solid tumors and is being recognized more frequently due to prolonged survival with systemic therapies. The prognosis remains dismal, with a median survival of 4-8 weeks. Methotrexate (MTX), the current standard treatment, is often compromised by resistance through dihydrofolate reductase (DHFR) upregulation and by neurotoxicity at high doses, underscoring the need for alternative therapeutic approaches. METHODS: An MTX-resistant subline (R-MM46) was

Secoisolariciresinol diglucoside (SDG) from flaxseed meal alleviates hyperuricemia in mice by regulating uric acid metabolism and intestinal homeostasis.

Hyperuricemia (HUA) is a metabolic disorder with an increasing prevalence rate in last decade. Although secoisolariciresinol diglucoside (SDG) from flaxseed was reported to have anti-inflammatory effects, its role and action mechanism in alleviating HUA remain unclear. This study investigated the effect of SDG on reducing uric acid (UA) in mice with HUA induced by potassium oxonate (PO) and hypoxanthine. The results showed that oral administration of SDG at 300 mg/kg significantly reduced UA levels by 75.37 ± 4.05 % and suppressed hepatic xanthine oxidase (XOD) gene expression by 50.99 ± 2.72

Maternal obesity changes the small intestine endocannabinoid system and fecal metabolites of weanling rats associated with reduced intestinal permeability and impaired glucose homeostasis.

The small intestine, including the endocannabinoid system (ECS), regulates the energy homeostasis. If maternal obesity modifies the intestinal ECS of the offspring favoring metabolic disorders throughout life is unexplored. Regardless maternal insults, overaction of the ECS has been related to obesity, mainly via type 1 cannabinoid receptor (CB1) signaling, while type 2 cannabinoid receptor (CB2) signaling and the endocannabinoid-like compounds, such as oleoylethanolamide (OEA) and palmitoylethanolamide (PEA), have been associated with anti-inflammatory effects. We hypothesized that maternal o
