[Preparation of antibodies against zip product and the distribution of zip protein in Drosophila embryos].

Zip gene is required during the late neurogenesis of Drosophila. Molecular analysis of zip gene revealed that it encodes an integral membrane glycoprotein, functioning as a cellular recognition and/or adhesion molecule. It has been cloned into an expression plasmid pWR 590. By expression of this plasmid in E. coli, a fusion protein of zip with lacZ has been purified and used to prepare polyclonal antibodies from rabbits. After being identified by West blotting, antibodies were used to react with the whole mount embryos of Drosophila. The results of this antibody labelling experiment showed tha

Inactivation of Sebol Induces Male Infertility in Spodoptera exigua via Arrested Meiosis: A Potential Molecular Target for Genetics-Based Sterile Insect Technique.

The beet armyworm, Spodoptera exigua, is a significant agricultural pest managed primarily through chemical insecticides, which present environmental concerns and promote resistance. Genetics-based sterile insect technique (gSIT) provides an alternative by inducing male sterility without irradiation. In this study, we cloned the S. exigua boule gene (Sebol), a 741-nucleotide open reading frame encoding a 246-amino-acid protein with an RNA recognition motif. Phylogenetic analysis confirmed its conservation across Lepidoptera and high similarity to orthologs in Spodoptera litura and Helicoverpa

Gene expression changes in honey bees induced by sublethal imidacloprid exposure during the larval stage.

Honey bee larvae exposed to sublethal doses of imidacloprid show behavioural abnormalities as adult insects. Previous studies have demonstrated that this phenomenon originates from abnormal neural development in response to imidacloprid exposure. Here, we further investigated the global gene expression changes in the heads of newly emerged adults and observed that 578 genes showed more than 2-fold changes in gene expression after imidacloprid exposure. This information might aid in understanding the effects of pesticides on the health of pollinators. For example, the genes encoding major royal

Synchronous expression of individual metacyclic variant surface glycoprotein genes in Trypanosoma brucei.

One distinctive feature of the Trypanosoma brucei life cycle is the presence of two discrete populations that are based on differential expression of variant surface glycoproteins (VSGs). Both are adapted to the environmental pressures they face and more importantly, both contribute directly to transmission. Metacyclics in the tsetse fly enable transmission to a new mammalian host, whereas bloodstream trypanosomes must avoid immune destruction to the extent that sufficient numbers are available for transmission, when the insect vector takes a blood meal. At present, there are few investigation

Detection of the Wolbachia protein WPIP0282 in mosquito spermathecae: implications for cytoplasmic incompatibility.

Cytoplasmic incompatibility (CI) is a conditional sterility induced by the bacterium Wolbachia pipientis that infects reproductive tissues in many arthropods. Although CI provides a potential tool to control insect vectors of arthropod-borne diseases, the molecular basis for CI induction is unknown. We hypothesized that a Wolbachia-encoded, CI-inducing factor would be enriched in sperm recovered from spermathecae of female mosquitoes. Using SDS-PAGE and mass spectrometry, we detected peptides from the 56 kDa hypothetical protein, encoded by wPip_0282, associated with sperm transferred to femal

Co-Option and Conflict: The Deep Evolutionary History of ZP-Domain Proteins from ECMs to Species Barriers.

The Zona Pellucida (ZP) and its structural analogs are evolutionarily ancient extracellular matrix components. These are essential for oocyte protection, species-specific gamete recognition, and prevention of polyspermy across Metazoa. Defined by the conserved ZP-domain-comprising ZP-N and ZP-C subdomains-these glycoproteins self-assemble into fibrillar matrices through tightly regulated polymerization. Mechanisms of the regulated polymerization involve furin cleavage, disulfide bonding, and hydrophobic interactions. Once considered a vertebrate innovation, the canonical ZP-domain-defined by i

Aspergillus fumigatus G-Protein Coupled Receptors GprM and GprJ Are Important for the Regulation of the Cell Wall Integrity Pathway, Secondary Metabolite Production, and Virulence.

G-protein coupled receptors (GPCRs) are extracellular signaling receptors that sense environmental cues. Fungi sense their environment primarily through GPCR-mediated signaling pathways, which, in turn, regulate fungal development, metabolism, virulence, and mycotoxin biosynthesis. Aspergillus fumigatus is an important human pathogen that causes aspergillosis, a heterogeneous group of diseases that present a wide range of clinical manifestations. Here, we investigate in detail the role of the GPCRs GprM and GprJ in growth and gene expression. GprM and GprJ are important for melanin production

Recombinant Zika Virus Subunits Are Immunogenic and Efficacious in Mice.

Following the 2015 Zika virus (ZIKV) outbreaks in the South Pacific, Caribbean, and Americas, ZIKV has emerged as a serious threat due to its association with infantile microcephaly and other neurologic disorders. Despite an international effort to develop a safe and effective vaccine to combat congenital Zika syndrome and ZIKV infection, only DNA and mRNA vaccines encoding the precursor membrane (prM) and envelope (E) proteins, an inactivated-ZIKV vaccine, and a measles virus-based ZIKV vaccine are currently in phase I or II (prM/E DNA) clinical trials. A ZIKV vaccine based on a nonreplicatin

Immunological control of ectoparasites: past achievements and future research priorities.

Recombinant vaccines are available for the control of the tick Boophilus microplus, while progress has been made in the development of vaccines against Lucilia cuprina and Chrysomya bezziana. Literature suggests that the control of other ectoparasites is feasible, either through the duplication in a vaccine of naturally acquired immunity or through 'concealed' antigen vaccines. Major deficiencies in our current knowledge however point to possible research opportunities for the future. The identification of protective antigens from all species is proceeding slowly, particularly for the antigens

Phosphorylated retinoblastoma protein stimulates DNA polymerase alpha.

Human retinoblastoma (Rb) protein, immunopurified from an extract of recombinant baculovirus infected cells, stimulated 10-100-fold the activity of DNA polymerase alpha from calf thymus or human HeLa cells. Purified Rb protein is composed of two electrophoretically distinguishable forms, i.e., partially phosphorylated and under-phosphorylated forms. Dephosphorylation of Rb protein by protein phosphatase 2A largely diminished its stimulatory effect. On the other hand, a hyperphosphorylated Rb protein, obtained from insect cells overexpressing Rb protein, cyclin E and cyclin-dependent kinase 2 s

Durability and effectiveness of insecticide-treated nets in Mali: a longitudinal gSG6-P1 biomarker-based assessment of children's exposure to Anopheles bites.

BACKGROUND: Insecticide-treated nets (ITNs) remain a key intervention in malaria prevention. However, their protective effectiveness may decline with physical deterioration, even when usage remains high. This study assessed the impact of ITN physical integrity on children's exposure to Anopheles mosquito bites over a three-year period in Mali, using gSG6-P1 biomarker as an innovative immuno-epidemiological indicator of human exposure. METHODS: A three-year prospective cohort study was conducted from 2018, 2019, and 2020 in two rural health districts of Mali: Kéniéba using Yorkool® ITNs and Ki

Salivary antigens rPagSP02 and rPagSP06 are a reliable composite biomarker for evaluating exposure to Phlebotomus argentipes in Sri Lanka.

Phlebotomus argentipes is the established vector of leishmaniasis in the Indian sub-continent. Antibodies to sand fly salivary antigens are biomarkers for vector-host exposure in leishmaniasis-endemic regions. Ph. argentipes transmits Leishmania donovani in Sri Lanka, primarily causing cutaneous leishmaniasis (CL). Our study compared the performance of salivary gland homogenate (SGH) from a lab-reared local strain of Ph. argentipes females to a composite recombinant salivary biomarker (rPagSP02 + rPagSP06) in a CL-endemic population. Sera from 546 healthy individuals, 30 CL patients, and 15 no

A knockout screen of genes expressed specifically in Ae. aegypti pupae reveals a critical role for stretchin in mosquito flight.

Aedes aegypti is a critical vector for transmitting Zika, dengue, chikungunya, and yellow fever viruses to humans. Genetic strategies to limit mosquito survival based upon sex distortion or disruption of development may be valuable new tools to control Ae. aegypti populations. We identified six genes with expression limited to pupal development; osi8 and osi11 (Osiris protein family), CPRs and CPF (cuticle protein family), and stretchin (a muscle protein). Heritable CRISPR/Cas9-mediated gene knockout of these genes did not reveal any defects in pupal development. However, stretchin-null mutati

Investigation of the population structure of the tick vector of Lyme disease Ixodes scapularis (Acari: Ixodidae) in Canada using mitochondrial cytochrome C oxidase subunit I gene sequences.

Genotyping of Ixodes scapularis Say (Acari: Ixodidae) ticks could enhance understanding of the occurrence and genotypes of I. scapularis-borne pathogens. We investigated the utility of mitochondrial (mt) Cytochrome C Oxidase subunit I gene (cox1) sequences as a tool for understanding the population structure of I. scapularis collected in Canada, where we also investigated the geographic occurrence of different cox1 haplotypes. Sequences obtained from 414 ticks were one of 55 unique haplotypes, most of which grouped into one of six clades. Demographic analysis suggested that cox1 sequences have

Locusts increase carbohydrate consumption to protect against a fungal biopesticide.

There is growing evidence to suggest that hosts can alter their dietary intake to recoup the specific resources involved in mounting effective resistance against parasites and pathogens. We examined macronutrient ingestion and disease-resistance in the Australian plague locust (Chortoicetes terminifera), challenged with a fungal pathogen (Metarhizium acridum) under dietary regimes varying in their relative amounts of protein and digestible carbohydrate. Dietary protein influenced constitutive immune function to a greater extent than did carbohydrate, indicating higher protein costs of mounting

Purification and renaturation of Japanese encephalitis virus nonstructural glycoprotein NS1 overproduced by insect cells.

The nonstructural protein NS1 of Japanese encephalitis virus is a major immunogen produced during flavivirus infection. However, the function of this protein has not been identified. To analyze its biochemical properties and evaluate its potential activity in the virus life cycle, the protein was produced in Spodoptera frugiperda insect cells (Sf9), using a recombinant baculovirus, and purified. As described previously by M. Flamand, V. Deubel, and M. Girard (1992, Virology 191, 826-836), a small fraction of the synthesized recombinant protein could mature into a dimer, whereas the major part

Rasputin/G3BP mediates subversion of antiviral immunity by o'nyong-nyong virus in Anopheles coluzzii.

Cellular G3BP proteins are essential for alphavirus infection in both vertebrate and mosquito hosts, but the underlying mechanism of their proviral activity is poorly understood in any host. Whether the mosquito G3BP ortholog, Rasputin (Rin), interacts with host immunity to influence alphavirus infection has not been investigated, and anopheline mosquito interactions with arboviruses have been little studied. Here, we find that Rin silencing in Anopheles mosquitoes results in decreased ONNV infection levels, indicating a proviral activity for Anopheles Rin. We find that Rin function is require

Cloning, expression and purification of AsSGU protein and its immunogenic response analysis as target candidate for transmission blocking vaccine against malaria.

AsSGU (Anopheles stephensi Secretory Glycoconjugate of Unknown function) is a GPI-anchored (Glycosyl-phosphatidyl-inositol) protein that is expressed in An. stephensi midgut after blood feeding. The role of midgut specific SGU protein in ookinetes invasion and transmission-blocking activity of the Plasmodium parasite has been confirmed and reported in An. sgambiae. It is a highly conserved protein in mosquito species Anopheles, Culex, and Aedes. It has critical role in ookinetes invasion; hence AsSGU protein is investigated for its biological functions in An. stephensi. The assgu gene expressi

Plasmodium vivax infection-driven modulation of sterol carrier protein reveals a metabolic link to reproductive physiology in Anopheles stephensi.

Mosquitoes rely exclusively on blood-derived sterols, transported by sterol carrier protein (SCP), for reproduction and metabolic regulation. SCP expression is strongly induced after blood feeding and dynamically modulated during Plasmodium vivax infection-upregulated in the midgut and salivary glands but suppressed in hemocytes-indicating that parasite infection modulates the host sterol transport and metabolism. RNAi-mediated knockdown of SCP significantly reduced mosquito fecundity, underscoring its vital role in reproductive physiology. These findings highlight SCP as a key metabolic regul

Heat Shock Proteins expression in malaria and dengue vector.

The survival of mosquitoes under changing climatic conditions particularly temperature, is known to be supported by Heat Shock Proteins (HSPs). In view of climate change, it is imperative to know whether the mosquito vectors will be able to withstand the increased temperatures or perish. Therefore, the present study was undertaken on the expression of HSPs' gene in An. stephensi and Ae. aegypti by exposing them to temperatures ranging from 5 to 45°C for 15-180-minutes for once and continuously or with rest in between. We compared the temperature-tolerance of both the vectors in terms of expres
