Selective amylin inhibition of the glucagon response to arginine is extrinsic to the pancreas.

Amylin, a peptide hormone from pancreatic beta-cells, is reported to inhibit insulin secretion in vitro and in vivo and to inhibit nutrient-stimulated glucagon secretion in vivo. However, it has been reported not to affect arginine-stimulated glucagon secretion in vitro. To resolve if the latter resulted from inactive peptide (a problem in the early literature), those experiments were repeated here with well-characterized peptide and found to be valid. In isolated perfused rat pancreas preparations, coperfusion with 1 nM amylin had no effect on arginine-, carbachol-, or vasoactive intestinal p

Extracellular vesicles in inflammatory bowel disease: from immune regulation and barrier repair to precise transformation.

Extracellular vesicles (EVs) have been shown to be key regulators of intestinal homeostasis and inflammatory responses, offering new insights into the pathogenesis of inflammatory bowel disease (IBD) and the development of targeted therapeutic strategies. EVs in the intestine can be secreted by intestinal epithelial cells (IECs), mesenchymal stem cells (MSCs), immune cell subsets, the gut microbiota, and even dietary sources. Its vesicle internal load contains a variety of biologically active molecules, including proteins, miRNAs, and microbial signals, which mediate immune regulation, intesti

Chicken-derived R-spondin1, Wnt3a, Nrg1 and human FGF1 allow branched growth of chicken intestinal organoid cultures.

Intestinal organoids are widely used in mammalian studies to mimic and study intestinal function and host-pathogen interactions without using of whole animal studies. For chicken, this was not yet feasible. Recently, we have shown that embryonal intestinal organoids can grow for 10-15 passages (approximately 6 weeks) when using chicken-derived Rspo1 and Wnt3. In this paper, we show that replacing Wnt3 with chicken Wnt3a and adding chicken Nrg1 and human FGF1 results in efficient and long lasting growth, over 40 weeks, of embryonal chicken intestinal organoids. Moreover, the typical branching p

Conserved B cell signaling, activation, and differentiation in porcine jejunal and ileal Peyer's patches despite distinct immune landscapes.

Peyer's patches (PPs) are B cell-rich sites of intestinal immune induction, yet PP-associated B cell signaling, activation, and differentiation are poorly defined. Single-cell and spatial transcriptomics were completed to study B cells from porcine jejunum and ileum containing PPs. Intestinal locations had distinct immune landscapes, including more follicular B cells in ileum and increased MHC-II-encoding gene expression in jejunal B cells. Despite distinct landscapes, conserved B cell dynamics were detected across intestinal locations, including B cell signaling to CD4+ macrophages that are p

Shared expression of mucin12 in Ascaris lumbricoides and the human small intestine.

This study focuses on the host-parasite relationship of human Ascaris lumbricoides, which is a parasite of the small intestine and is also one of the commonest parasites worldwide. As part of this investigation, we examined the host-parasite relationship assuming that there is a common antigenicity, shared protein between A. lumbricoides and human small intestinal mucosa, using molecular techniques. We obtained three DNA clones from human colon cDNA library by screening for anti-A. lumbricoides polyclonal antibodies. The transmembrane mucin12 gene was identified after sequencing analysis of th

TLR9 Regulates the NF-κB-NLRP3-IL-1β Pathway Negatively in Salmonella-Induced NKG2D-Mediated Intestinal Inflammation.

TLRs are key sensors for conserved bacterial molecules and play a critical role in host defense against invading pathogens. Although the roles of TLRs in defense against pathogen infection and in maintaining gut immune homeostasis have been studied, the precise functions of different TLRs in response to pathogen infection in the gut remain elusive. The present study investigated the role of TLR signaling in defense against the Gram-negative bacterial pathogen Salmonella typhimurium The results indicated that TLR9-deficient mice were more susceptible to S. typhimurium infection compared with wi

Interplay between microbial d-amino acids and host d-amino acid oxidase modifies murine mucosal defence and gut microbiota.

L-Amino acids are the building blocks for proteins synthesized in ribosomes in all kingdoms of life, but d-amino acids (d-aa) have important non-ribosome-based functions(1). Mammals synthesize d-Ser and d-Asp, primarily in the central nervous system, where d-Ser is critical for neurotransmission(2). Bacteria synthesize a largely distinct set of d-aa, which become integral components of the cell wall and are also released as free d-aa(3,4). However, the impact of free microbial d-aa on host physiology at the host-microbial interface has not been explored. Here, we show that the mouse intestine

Coordinated regulation of colonic fluid and mucus secretion.

The human colon, unlike the small intestine which is relatively sterile, contains a diverse microbiome which contributes to host metabolism. The luminal environment is constantly changing and responds to epithelial absorption and secretion which occurs as motility is regulated by longitudinal and circular smooth muscle. Mucous gels are crucial to lubrication and maintenance of an unstirred layer which separate the epithelium from the lumen. Gel-forming mucins are produced and released by goblet cells and become hydrated, although the source of water is not definitively understood. The purpose

Recent developments in the discovery of FFA1 receptor agonists as novel oral treatment for type 2 diabetes mellitus.

Despite the availability of established medication for treatment of type 2 diabetes mellitus (T2DM) there still remains a significant unmet need for new effective, oral antidiabetic agents that improve glycemic control while maintaining an excellent safety profile. In this regard the FFA1 receptor has emerged as an attractive target in recent years. Activation of the FFA1 receptor has been shown to not only amplify glucose induced insulin secretion from pancreatic beta cells but also to stimulate incretin secretion from intestinal endocrine cells. The current review highlights on the latest de

Carbohydrate histochemistry of the alimentary canal of the shi drum, Umbrina cirrosa L.

Histochemical staining techniques, which differentiate the main categories of carbohydrates, were applied to sections from different segments of the alimentary canal of the shi drum Umbrina cirrosa L. to study patterns of distribution of epithelial glycoconjugates. In the oesophagus, mucous cells contained sulphomucins, together with a small amount of sialomucins. Stomach epithelial cells secreted neutral and acidic glycoconjugates, while gastric glands only produced small quantities of sialomucins. Goblet cells showed the presence of sialo and sulphomucins in the pyloric caeca, whereas intest

Neuropeptide levels in murine liver and biliary pathways.

Neuropeptide concentrations in the liver, gallbladder and bile duct of 30 male mice were determined by radioimmunoassay of tissue extracts. Vasoactive intestinal peptide (VIP), substance P, somatostatin, neurotensin, neuropeptide Y (NPY), galanin, gastrin-releasing peptide (GRP) and enkephalin were found in extracts of liver, gallbladder and bile duct. The level of enkephalin was low in all tissues investigated. The most predominant neuropeptide in liver and bile duct was VIP and in the gallbladder, NPY. The function of these neuropeptides in the liver and biliary pathways is unclear. One can

Shengu granules ameliorate ovariectomy-induced osteoporosis by the gut-bone-immune axis.

INTRODUCTION: Postmenopausal osteoporosis (PMOP) is a common chronic disease, and the loss of bone density and bone strength after menopause are its main symptoms. Effective treatments for PMOP are still uncertain, but Chinese medicine has some advantages in slowing down bone loss. Shengu granules are often used clinically to treat PMOP. It has been shown to be an effective prescription for the treatment of PMOP, and there is evidence that gut flora may play an important role. However, whether Shengu granules attenuate PMOP by modulating gut flora and related mechanisms remains unclear. METHO

Early host-parasite interaction models reveal a key role for fibrinolysis in Fasciola hepatica intestinal migration.

BACKGROUND: Fasciola hepatica is the most common etiologic agent of fasciolosis, a parasitic disease that affects millions of ruminants worldwide and a zoonotic human infection of public health concern. Upon ingestion of infective metacercariae, F. hepatica newly excysted juveniles (FhNEJ) emerge in the duodenum and cross the intestinal wall to initiate a migration route that culminates with their establishment within the hepatic bile ducts. The ability of FhNEJ to exploit the broad-spectrum activities of host plasmin, the central protease of the fibrinolytic system, has been proposed as a str

Effect of somatostatin infusion on jejunal water and electrolyte transport in a patient with secretory diarrhea due to malignant carcinoid syndrome.

The effect of intravenous somatostatin infusion was investigated in a patient with severe secretory diarrhea due to malignant carcinoid syndrome. Continuous somatostatin infusion for 48 hr (4 microgram/kg/hr) reduced stool volume from 2.2 to 0.7 liters/day. Intestinal perfusion studies before intravenous somatostatin infusion revealed decreased water and sodium absorption and jejunal chloride secretion. Somatostatin infusion for 2 hr (8 microgram/kg/hr) reversed chloride secretion to absorption and markedly enhanced water and sodium absorption. These results suggest that somatostatin inhibited

METTL9 tests Candida's mettle by limiting metal acquisition.

The gut mucosal immune system orchestrates diverse defense mechanisms against fungal pathogens. In this issue of Cell Host & Microbe, Bao and Yang et al.1 demonstrate that intestinal epithelial cells secrete an anti-zincophore protein, METTL9, that limits fungal access to the essential micronutrient zinc, thereby diminishing colonization and growth.

Motilin and erythromycin stimulate pepsinogen secretion by chief cells isolated from guinea pig stomach.

BACKGROUND: Motilin and erythromycin bind the same receptor in the gastrointestinal muscle cells. Motilin stimulates pepsinogen secretion in humans and dogs. However, it is unclear whether it acts directly on gastric chief cells or via other neurotransmitters. METHODS: Isolated gastric chief cells were obtained from guinea pig stomach by collagenase digestion and calcium chelation with ethylene glycol-bis(B-amynomethyl ether)-N,N,N',N'-tetraacetic acid. RESULTS: Significant stimulation of pespinogen secretion induced by motilin occurred at 1 pmol/L, half maximum at 10 pmol/L, and maximum at

Role of serine/threonine phosphatases 1 and 2A in pancreatic acinar fluid and electrolyte secretion.

Serine/threonine phosphatase 1 (PP1) and phosphatase 2A (PP2A) play important roles in mediating cellular signaling in different tissues to different stimuli, including in protein synthesis, growth, cell cycle regulation, and secretion. However, their roles in various pancreatic exocrine functions, such as pancreatic acinar fluid/electrolyte secretion, is still unclear. Therefore, in the present study, we examined the ability of vasoactive intestinal peptide (VIP) and secretin, which stimulate cAMP generation in pancreatic acini, to activate serine/threonine phosphatase 1 (PP1) and phosphatase
