Kinetics of simultaneous photocatalytic degradation of phenolic compounds and reduction of metal ions with nano-TiO2.

The simultaneous photocatalytic degradation of phenol and 4-nitrophenol and reduction of metal ions like copper (Cu2+) and chromium (Cr6+) was studied with solution combustion synthesized nanoanatase TiO2 (CS TiO2) and commercial titania, Degussa P-25. The presence of metal ion reduces the rate of degradation of phenol and 4-nitrophenol. It was found that Cu2+ reduction to Cu+ is accelerated in the presence of phenol. In the case of Cr6+, CS TiO2 enhances the initial adsorption of Cr6+ and complete reduction is achieved within the first 10 min of UV irradiation. The presence of phenol or 4-nit…

Combined cytotoxicity and genotoxicity of a marine toxin and seafood contaminant metal ions (chromium and cadmium).

Algal bloom with consequent production of marine toxins contaminating bivalves is increasing in costal regions worldwide because of sea water quality worsening. Contamination of seafood by diarrheic shellfish poisoning toxins (DSP) together with metals is frequently reported, a phenomenon not fully explained yet. In this context, metal ions were assayed in clams collected from the banned area of Boughrara, Tunisia, contaminated by Gymnodinium and other algae such as Dinophysis sp, accumulated by these bivalves. The presence of toxic metals ions such as Chromium (Cr) and Cadmium (Cd) in meat, s…

The role of calcium and other ions in sorting and delivery in the late endocytic pathway.

The passage of endocytosed receptor-bound ligands and membrane proteins through the endocytic pathway of mammalian cells to lysosomes occurs … th in the trafficking of endocytosed ligands and in the membrane fusion events. In particular, H(+) ions play a role in sorting processes and providing an appropriate environment for the action of lysosomal acid hydrolases. Na(+)/H(+) exchangers in the endosomal membrane have been implicated in the formation of MVBs and sorting into luminal vesicles. C …

Metal ions differentially influence the aggregation and deposition of Alzheimer's beta-amyloid on a solid template.

The abnormal deposition and aggregation of beta-amyloid (Abeta) on brain tissues are considered to be one of the characteristic neuropathological features of Alzheimer's disease (AD). Environmental conditions such as metal ions, pH, and cell membranes are associated with Abeta deposition and plaque formation. According to the amyloid cascade hypothesis of AD, the deposition of Abeta42 oligomers as diffuse plaques in vivo is an important earliest event, leading to the formation of fibrillar amyloid plaques by the further accumulation of soluble Abeta under certain environmental conditions. In o…

Regulation of tubular cell MCP-1 production by intracellular ions: a role for sodium and calcium.

Proximal tubule cells (PTC) in chronic renal disease produce chemokines which cause renal interstitial inflammation, and also transport more Na+ than normal. To investigate whether these two events might be related, monocyte chemoattractant protein-1 (MCP-1) production was examined in rat PTC in primary culture. Amiloride reduced, while ouabain increased levels of MCP-1 mRNA and protein. Amiloride reduced MCP-1 in cells stimulated with ouabain, lipopolysaccharide (LPS) or albumin. Intracellular Na+ rose with ouabain, but not LPS or albumin. Effects of amiloride, ouabain, LPS and albumin were a

Zinc finger proteins as potential targets for toxic metal ions: differential effects on structure and function.

Zinc finger structures are frequently found in transcription factors and DNA repair proteins, mediating DNA-protein and protein-protein binding. As low concentrations of transition metal compounds, including those of cadmium, nickel, and cobalt, have been shown to interfere with DNA transcription and repair, several studies have been conducted to elucidate potential interactions of toxic metal ions with zinc-binding protein domains. Various effects have been identified, including the displacement of zinc, e.g., by cadmium or cobalt, the formation of mixed complexes, incomplete coordination of …

Lipopolysaccharide enhances oxidative modification of low density lipoprotein by copper ions, endothelial and smooth muscle cells.

The effect of lipopolysaccharide (LPS, endotoxin) on low density lipoprotein (LDL) oxidative modification by copper ions, endothelial and smooth muscle cells was studied by determination of the level of lipid peroxidation products (thiobarbituric acid reactive substances or TBARS), the diene level and the electrophoretic mobility of the LDL particle. LPS 25-75 microg/ml induced a dose-dependent increase in LDL oxidation by copper ions, endothelial and smooth muscle cells. At 75 microg LPS/ml, the TBARS content was 1.9, 1.6, and 1.8-fold increased, respectively. The LDL degradation by J774 macr…

[Effect of heavy metal ions on superprecipitation and ATPase activity of uterine smooth muscle actomyosin activity].

The inhibiting effect of Pb2+, Zn2+ and Cd2+ on Mg(2+)-dependent superprecipitation and ATPase activity of myometrium actomyosin. The inhibiting effect of heavy metals cations on the both processes satisfies the succession: Pb2+ > Zn2+ > Cd2+. Cadmium and zinc ions in concentration of 1 mM stimulate the initial velocity (v0) of Mg(2+)-dependent superprecipitation by 25% and 80%, respectively, while the lead ions under the same concentration inhibit the initial velocity by 40%. It is possible that these results evidence for the direct effect of ions of heavy metals on active-myosin interaction …

Probing the role of metal ions in the mechanism of inositol monophosphatase by site-directed mutagenesis.

Since inhibition of myo-inositol monophosphatase (EC 3.1.3.25) by lithium ions and the resulting attenuation of phosphatidylinositol cycle activity may be the mechanism by which lithium exerts its therapeutic effect in the treatment of manic depression, it is of great interest to understand the mechanism of the enzyme and how lithium and other metals interact with it. Divalent magnesium is essential for enzyme activity, whereas Li+ and high concentrations of Mg2+ act as uncompetitive inhibitors with respect to substrate. From the recently solved crystal structure of the human enzyme, several a…

Modulation of mu, delta and kappa opioid receptors in rat brain by metal ions and histidine.

The effect of zinc (Zn2+) and several other trace elements was studied on the binding of the opioid receptor agonists [3H] DAGO [( ([Tyr-D-Ala-Gly-Methyl-Phe-Glyol]-enkephalin)a, [3H] DSTLE ([Tyr-D-Ser-Gly-Phe-Leu-Thr]-enkephalin) and [3H] EKC (ethylketocyclazocine), which are specific for the mu, delta and kappa opioid receptors, respectively, in the cerebral cortex of the rat. Physiological concentrations of zinc were inhibitory to mu receptor binding, whereas the delta and kappa receptors were relatively insensitive to this inhibition. Scatchard analysis, using these opioid agonists, reveal…

Biphasic Ca2+ response of adenylate cyclase. The role of calmodulin in its activation by Ca2+ ions.

The Ca2+-dependent regulation of human platelet membrane adenylate cyclase has been studied. This enzyme exhibited a biphasic response to Ca2+ within a narrow range of Ca2+ concentrations (0.1-1.0 microM). At low Ca2+ (0.08-0.3 microM) adenylate cyclase was stimulated (Ka = 0.10 microM), whereas at higher Ca2+ (greater than 0.3 microM) the enzyme was inhibited to 70-80% control (Ki = 0.8 microM). Membrane fractions, prepared by washing in the presence of LaCl3 to remove endogenous calmodulin (approximately equal to 70-80% depletion), exhibited no stimulation of adenylate cyclase by Ca2+ but di…

Dual effects of internal n-alkyltrimethylammonium ions on the sodium current of the squid giant axon.

The actions of members of the homologous series of alkyl cations CH3 (CH2)n-1 N+ (CH3)3 (Cn TMA) on the sodium current in giant axons of Loligo forbesi have been investigated. The substances tested correspond to n = 6, 8, 10, 12, 14 and 16. These cations only produced significant sodium current suppression when applied inside the axon. Actions on first-pulse sodium currents and use-dependent effects were separately studied. The shorter members of the series (C6TMA and C8TMA) produced suppression of first-pulse sodium currents without causing significant use dependence. The first-pulse suppress…

Thiol groups and reduced acidogenicity of dental plaque in the presence of metal ions in vivo.

Metal ions are known to influence the cariogenicity of dental plaque. Inhibition of acid metabolism in plaque may be of importance in this respect. Metal ions inhibit the acidogenicity of dental plaque to a different extent and it has been suggested that an enzyme inhibition based on oxidation of thiol groups may explain this observation. The aim of the present study was to evaluate the significance of oxidation of thiol groups in the inhibition of acid production in plaque by silver, tin and zinc salts. Nine subjects with 3-d sucrose induced plaque received topical applications of the metal i…

The in vitro effect of metal ions on the activity of erythrocyte delta-aminolevulinic acid dehydratase.

The effects of a total of 15 metals on the activity of erythrocyte delta-aminolevulinic acid dehydratase (ALA-D) were investigated in vitro. The degree of inhibition of the ALA-D activity by copper, tin, mercury and silver was almost equal to that by lead, when the metal was added to the hemolysate of whole blood at the concentrations of 0.1 to 1.0 mmol/l in the ALA-D assay. Magnesium, manganese and beryllium had no effect on the ALA-D activity at the same concentrations. Zinc had a mild activating effect on ALA-D at the optimum concentrations and it eliminated the inhibitory effect of lead to…
