Early life exposure to per- and polyfluoroalkyl substances (PFAS) and latent health outcomes: A review including the placenta as a target tissue and possible driver of peri- and postnatal effects.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous drinking water contaminants of concern due to mounting evidence implicating adverse health outcomes associated with exposure, including reduced kidney function, metabolic syndrome, thyroid disruption, and adverse pregnancy outcomes. PFAS have been produced in the U.S. since the 1940s and now encompass a growing chemical family comprised of diverse chemical moieties, yet the toxicological effects have been studied for relatively few compounds. Critically, exposures to some PFAS in utero are associated with adverse outcomes for both mothe

Microplastic trafficking in maternal-fetal system: a systematic review and quantitative profiling linking particle characteristics to developmental biology.

Microplastics (MPs) have been detected across multiple maternal and neonatal biological matrices, suggesting widespread exposure during critical developmental stages. However, the mechanisms shaping their distribution and accumulation within the maternal-fetal system remain poorly understood. This systematic review and quantitative synthesis provides a comprehensive quantitative profiling of MP burden across six maternal and neonatal biological matrices placenta, breast milk, endometrium, amniotic fluid, meconium, and feces. Our analysis reveals three critical insights: (1) Tissue‑specific MP

Multimatrix Biomonitoring and Transplacental Transfer of Legacy and Emerging PFAS in Korean Pregnant Women.

Maternal-fetal exposure to per- and polyfluoroalkyl substances (PFAS) cannot be adequately characterized using single biological matrices because compound-specific distribution patterns vary according to physicochemical properties and elimination pathways. We measured 33 legacy, precursor, and emerging PFAS in maternal serum, plasma, urine, and cord serum from Korean singleton pregnancies (n = 151) to characterize matrix-specific exposure profiles, transplacental transfer, and associations with maternal and neonatal factors. Matrix-specific detection patterns distinguished predominantly protei

Arsenic and the placenta: A review with emphasis on the immune system.

Chronic arsenic exposure affects over 140 million people globally. While arsenic easily crosses the placenta, the specific mechanisms impacting placental immune cell populations and fetal health are unclear. Maternal arsenic exposure is epidemiologically linked to increased infection risk, mortality, and cancer susceptibility in offspring, emphasizing the importance of understanding placentally-mediated immune effects. This review explores the potential role of the placenta, a key organ for immune transfer to the developing fetus, in mediating chronic low-dose arsenic exposure effects. Examini

Prenatal environmental risk factors for autism spectrum disorder and their potential mechanisms.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder that is globally increasing in prevalence. The rise of ASD can be partially attributed to diagnostic expansion and advocacy efforts; however, the interplay between genetic predisposition and modern environmental exposures is likely driving a true increase in incidence. A range of evidence indicates that prenatal exposures are critical. Infection during pregnancy, gestational diabetes, and maternal obesity are established risk factors for ASD. Emerging areas of research include the effects of maternal use of selective serotonin reu

Prenatal nitrate and nitrite intake in mammals: A scoping review of effects and associations with pregnancy and maternal and child health.

Nitrate and nitrite are present in drinking water, vegetables and processed meats. While they may offer health benefits in some contexts, high levels, especially during pregnancy, can be harmful. This scoping review investigates prenatal nitrate/nitrite exposure and maternal, child, pregnancy and birth outcomes, in animal models and humans. Six databases were searched systematically for peer-reviewed publications relating to nitrate and/or nitrite exposure in pregnancy. 3834 unique papers were identified, with 133 meeting inclusion criteria. In human studies (n = 66), high processed meat nitri

Periconceptional maternal alcohol consumption and neural tube defects.

BACKGROUND: Neural tube defects (NTD)s, which occur when the neural tube fails to close during early gestation, are some of the most common birth defects worldwide. Alcohol is a known teratogen and has been shown to induce NTDs in animal studies, although most human studies have failed to corroborate these results. Using data from the National Birth Defects Prevention Study, associations between maternal reports of periconceptional (1 month prior through 2 months postconception) alcohol consumption and NTDs were examined. METHODS: NTD cases and unaffected live born control infants, delivered

Craniosynostosis and maternal smoking.

BACKGROUND: Several previous studies suggested increased risk of craniosynostosis among infants born to women who smoked. METHODS: This study used data from the National Birth Defects Prevention Study, a multi-state, population-based case-control study of infants delivered from 1997-2003. Nonmalformed, liveborn controls were selected randomly from birth certificates or birth hospitals. Data from maternal telephone interviews were available for 531 cases and 5008 controls. RESULTS: Smoking during the first month of pregnancy was not associated with craniosynostosis. Smoking later in pregnancy

Aflatoxin exposure during pregnancy or infancy and its effect on infant health: a narrative review.

Pregnant women are exposed to various contaminants through foods, with environmental toxicants and aflatoxin (AF) being among the major food contaminants. Therefore, this review was conducted for a better perspective on the AF exposure during pregnancy or infancy, highlighting how exposure through the mother (via placenta and breast milk) and directly through infant foods ultimately affects infant health. The literature suggests that AF exposure during pregnancy may lead to maternal anaemia, premature delivery, pregnancy loss or decreased number of live births. AF crosses through the placenta

Nanoplastic toxicology following gestational and lactational exposure.

Nanoplatics (NPs), particularly polystyrene (PS)-NPs, can traverse the placental barrier upon maternal exposure, leading to bioaccumulation in both dam and offspring organs, and inducing widespread transplacental toxicity. The distribution and toxicity of NPs are influenced by a variety of factors, including NP properties (type, size, and charge), exposure parameters (dose, route, and timing), and biological variables (model and co-exposures). Due to their minute size, NPs pose significant threats to multiple systems in animal models. In rodent studies, reproductive and endocrine toxicity prim…

Association between prenatal exposure to tetrachloroethylene and adverse birth outcomes: Systematic review and meta-analysis.

BACKGROUND: Tetrachloroethylene (PCE) is a ubiquitous chlorinated solvent with documented placental transfer. Despite widespread environmental and occupational exposure, no prior systematic review has synthesized evidence on prenatal PCE exposure and adverse birth outcomes. METHODS: We conducted a systematic review and meta-analysis of observational studies. PubMed, Web of Science, PsycINFO, EMBASE, and CINAHL were searched from inception to July 13, 2026. Eligible studies reported associations between prenatal PCE exposure (drinking water or inhalation) and adverse birth outcomes. Study qual

The fetal exposome and preterm birth: a systematic synthesis of environmental exposures and multi-omics evidence.

OBJECTIVES: Preterm birth (PTB), defined as delivery before 37 weeks of gestation, is a leading cause of neonatal mortality and long-term developmental impairment. Its complex etiology, spanning environmental, genetic, psychosocial, and socio-economic domains, limits effective prediction and prevention. We systematically synthesized evidence on how environmental exposures influence PTB risk through multi-omic disruptions within a fetal exposome framework. METHODS: A comprehensive literature search was conducted in major biomedical databases, following PRISMA guidelines. Ninety-five human stud

Novel biomarkers of fetal and neonatal environmental exposure, effect and susceptibility.

Rapid advancements in science and technology have allowed medical providers to treat wider ranges of diseases with safer and more effective therapies than ever before. One of the areas of health that has been consistently understudied, however, is one that affects us all: environmental health or the effects that the chemicals we are exposed to every day have on our acute and chronic health. This effect can be exacerbated during and shortly after pregnancy, as an individual exposure is often shared by both the mother and the fetus/neonate. The diagnosis and monitoring of chemical exposure can b

Acute and chronic sulphur dioxide (SO2) exposure: an overview of its effects on humans and laboratory animals.

Sulphur dioxide (SO2) is a common air pollutant found both in indoor and outdoor environments. Studies of controlled human exposure as well as epidemiological and animal investigations have documented several short- and long-term effects of SO2 exposure on the respiratory and other systems. Exercise, duration and other exposure factors may potentiate the pollutant's effects, especially in sensitive individuals such as children and asthmatics. Early postnatal somatic and behavioural alterations have been shown after maternal SO2 exposure, during pregnancy and neonatal exposure. Such exposure sh

Impact of endocrine disrupting chemicals on macrophages at the maternal-fetal interface.

Pregnancy is a complex and dynamic process, immune homeostasis at the maternal-fetal interface is one of the keys to a successful pregnancy and essential for fetal nutrient exchange and the establishment of immune tolerance. Healthy pregnant women with normally functioning immune systems can successfully maintain a semi-allogeneic fetus to full term without immune-mediated rejection, many immune cells including macrophages, NK cells, T cells, B cells and dendritic cells are involved in this process. In particular, macrophages play a vital role in the establishment of immune tolerance, infectio
