[Olfactory dysfunction in Parkinson's disease: its role as a new cardinal sign in early and differential diagnosis].

Olfactory dysfunction is a prominent symptom in Parkinson's disease (PD) and found in about 70-100% of patients. In earlier studies significant loss of olfactory function seemed to be unrelated to disease duration, did not correlate with motor function, and was uninfluenced by antiparkinsonian medication. We suggest that the increase of dopaminergic cells in the olfactory bulb is responsible for the hyposmia in PD patients. Interestingly, this olfactory dysfunction is not found in progressive supranuclear palsy or corticobasal degeneration. In multiple system atrophy, the deficit is mild and i…

Evidences of copper nanoparticle exposure in indoor environments: Long-term assessment, high-resolution field emission scanning electron microscopy evaluation, in silico respiratory dosimetry study and possible health implications.

A variety of appliances operated by brush electric motors, widely used in indoor environments, emit nanoparticles (NPs). Due to electric arc discharge during the operation of such motors, some NPs contain copper (Cu). Their dimensions are the same of those found in brain tissue samples by other authors who speculated their possible translocation to brain through olfactory bulb. Cu has been reported to play an important role in the etiopathogenesis of Alzheimer's disease. Thus, the present study was performed to 1. estimate by means of Multiple-Path Particle Dosimetry model the doses of NPs rel…

[Study on the significance of mercury accumulation in the brain from dental amalgam fillings through direct mouth-nose-brain transport].

The transport of mercury (Hg) from the oro-nasal to the cranial cavity via a direct route was investigated. In 55 deceased persons, Hg concentrations were measured in the olfactory bulb and the trigeminal ganglion, and the number of dental amalgam fillings was assessed. For the purpose of comparison, Hg concentrations were also determined in the occipital lobe cortex, the pituitary gland and the kidney cortex. Quantitative Hg analysis was performed by cold vapor atomic absorption spectroscopy after acid digestion using high pressure microwave treatment. In the olfactory bulb (geom. mean 17.4 m…

Triploidy alters brain morphology in pre-smolt Atlantic salmon Salmo salar: possible implications for behaviour.

Total brain mass and the volumes of five specific brain regions in diploid and triploid Atlantic salmon Salmo salar pre-smolts were measured using digital images. There were no significant differences (P > 0·05) in total brain mass when corrected for fork length, or the volumes of the optic tecta or hypothalamus when corrected for brain mass, between diploids and triploids. There was a significant effect (P < 0·01) of ploidy on the volume of the olfactory bulb, with it being 9·0% larger in diploids compared with triploids. The cerebellum and telencephalon, however, were significantly larger, 1…

Rapid Improvement of COVID-Related Anosmia Following Electroconvulsive Therapy: A Report of 2 Cases.

We present 2 cases of patients who received electroconvulsive therapy (ECT) for psychotic depression and catatonia, respectively. Both patients had previously been diagnosed with COVID-related anosmia. Following the first ECT session, the patients reported a significant improvement in their sense of smell, preceding the resolution of depressive and catatonic symptoms. There is a growing body of literature regarding the relationship between psychiatric illness and olfactory dysfunction. Furthermore, considering the neurogenesis hypothesis of ECT, the olfactory bulb is the only brain structure a…

Early development of the olfactory and terminalis systems in baleen whales.

The development of the olfactory and terminalis systems was studied in tissue from eight embryonic and early fetal specimens belonging to th … material in the embryonic period, and they form compact masses medial and caudal to the developing olfactory bulb. As in most prenatal toothed whales, there are two large intrameningeal terminalis ganglia. These are connected with the telencephalic wall by central rootlets and with the septal mucosa by fiber bundles running through the level of the future cribr …

Mapping of class I and class II odorant receptors to glomerular domains by two distinct types of olfactory sensory neurons in the mouse.

The repertoire of approximately 1200 odorant receptors (ORs) is mapped onto the array of approximately 1800 glomeruli in the mouse olfactory bulb (OB). The spatial organization of this array is influenced by the ORs. Here we show that glomerular mapping to broad domains in the dorsal OB is determined by two types of olfactory sensory neurons (OSNs), which reside in the dorsal olfactory epithelium. The OSN types express either class I or class II OR genes. Axons from the two OSN types segregate already within the olfactory nerve and form distinct domains of glomeruli in the OB. These class-spec…

Transmission and integration of biologically meaningful olfactory information after bilateral transection of the lateral olfactory tract in the rat.

The contribution of the olfactory inputs conveyed by the lateral olfactory tract (LOT) to the central olfactory areas in the onset of behavioral and electrophysiological responses i.e. multiunit activity of mitral cells was studied in rats which were submitted to a bilateral transection of LOT. The characteristic emotional reactions elicited in sham-operated rats by biologically meaningful odorants--odors of predator or of conspecific--were no longer observed after such lesion. At the olfactory bulb level, the differential habituation of mitral cell electrical responses according to the biolog…

Differential distribution of cAMP-specific phosphodiesterase 7A mRNA in rat brain and peripheral organs.

We investigated the regional distribution and cellular localization of mRNA coding for the cAMP-specific phosphodiesterase 7A (PDE7A) in rat brain and several peripheral organs by in situ hybridization histochemistry. The regional expression of two splice variants, PDE7A1 and PDE7A2, was examined by RT-PCR using RNA extracted from several brain regions. PDE7A mRNA was found to be widely distributed in rat brain in both neuronal and nonneuronal cell populations. The highest levels of hybridization were observed in the olfactory bulb, olfactory tubercle, hippocampus, cerebellum, medial habenula …

Postmortem Assessment of Olfactory Tissue Degeneration and Microvasculopathy in Patients With COVID-19.

IMPORTANCE: Loss of smell is an early and common presentation of COVID-19 infection. Although it has been speculated that viral infection of olfactory neurons may be the culprit, it is unclear whether viral infection causes injuries in the olfactory bulb region. OBJECTIVE: To characterize the olfactory pathology associated with COVID-19 infection in a postmortem study. DESIGN, SETTING, AND PARTICIPANTS: This multicenter postmortem cohort study was conducted from April 7, 2020, to September 11, 2021. Deceased patients with COVID-19 and control individuals were included in the cohort. One infa…

The olfactory nerve: a shortcut for influenza and other viral diseases into the central nervous system.

The olfactory nerve consists mainly of olfactory receptor neurons and directly connects the nasal cavity with the central nervous system (CNS). Each olfactory receptor neuron projects a dendrite into the nasal cavity on the apical side, and on the basal side extends its axon through the cribriform plate into the olfactory bulb of the brain. Viruses that can use the olfactory nerve as a shortcut into the CNS include influenza A virus, herpesviruses, poliovirus, paramyxoviruses, vesicular stomatitis virus, rabies virus, parainfluenza virus, adenoviruses, Japanese encephalitis virus, West Nile vi…

Stimulation of olfactory ensheathing cell motility enhances olfactory axon growth.

Axons of primary olfactory neurons are intimately associated with olfactory ensheathing cells (OECs) from the olfactory epithelium until the final targeting of axons within the olfactory bulb. However, little is understood about the nature and role of interactions between OECs and axons during development of the olfactory nerve pathway. We have used high resolution time-lapse microscopy to examine the growth and interactions of olfactory axons and OECs in vitro. Transgenic mice expressing fluorescent reporters in primary olfactory axons (OMP-ZsGreen) and ensheathing cells (S100ß-DsRed) enabled…

Olfactory-auditory sensory integration in the lateral entorhinal cortex.

Multisensory integration plays an important role in animal cognition. Although many studies have focused on visual-auditory integration, studies on olfactory-auditory integration are rare. Here, we investigated neural activity patterns and odor decoding in the lateral entorhinal cortex (LEC) under uni-sensory and multisensory stimuli in awake, head-fixed mice. Using specific retrograde tracing, we verified that the LEC receives direct inputs from the primary auditory cortex (AC) and the medial geniculate body (MGB). Strikingly, we found that mitral/tufted cells (M/Ts) in the olfactory bulb (OB…

Functional maturation of the first synapse in olfaction: development and adult neurogenesis.

The first synapse in olfaction undergoes considerable anatomical plasticity in both early postnatal development and adult neurogenesis, yet we know very little concerning its functional maturation at these times. Here, we used whole-cell recordings in olfactory bulb slices to describe olfactory nerve inputs to developing postnatal neurons and to maturing adult-born cells labeled with a GFP-encoding lentivirus. In both postnatal development and adult neurogenesis, the maturation of olfactory nerve synapses involved an increase in the relative contribution of AMPA over NMDA receptors, and a decr…

Imaging of olfaction and gustation.

Positron emission tomographic (PET) scanning can be used to assess the cortical processing of responses to different odorants in man. The olfactory bulb receives the first projections on an ipsilateral basis from the nose, but then the cortex and the limbic system respond bilaterally. More irritating, burning odors (e.g., acetone) project via the trigeminal rather than the olfactory nerve and induce changes in the brain stem and the cortex. Sweet and salty tastes are readily distinguishable, but a bitter or painful stimulus leads to a deactivation of the primary gustatory cortex, which is in k…
