Acute Toxoplasma gondii infection alters the number of neurons and the proportion of enteric glial cells in the duodenum in Wistar rats.

BACKGROUND: Toxoplasma gondii infection can occur through the ingestion of raw meat that contains tissue cysts or food that contains oocysts. Through the ingestion of oocysts, the parasite crosses the intestinal barrier, where the enteric nervous system is located. The objective was to investigate the kinetics of neuronal and glial responses during acute T. gondii infection. METHODS: We used 45 Wistar rats that were divided into a control group and infected groups that were evaluated at 6, 12, 24, 48, 72 hours, 7 days, 10 days, and 15 days after infection. The rats received 5000 sporulated oo

How the Emotional Motor System Controls the Pelvic Organs.

INTRODUCTION: The brain has two goals: survival of the individual and survival of the species. It ensures that the body resides in safe circumstances and can obtain sufficient drink and food. It also has to produce and protect offspring. Its most important tool is its motor system, which consists of the voluntary and emotional motor systems. AIM: To explain how the brain uses its emotional motor system to control the pelvic organs. METHODS: Anatomic and physiologic data in cats and humans are used to find out how this motor system works and what parts of the brain and brainstem are involved.

Postnatal sulfur dioxide exposure reversibly alters parasympathetic regulation of heart rate.

Perinatal sulfur dioxide exposure disrupts parasympathetic regulation of cardiovascular activity. Here, we examine the relative risks of prenatal versus postnatal exposure to the air pollutant and the reversibility of the cardiovascular effects. Two groups of animals were used for this study. For prenatal exposure, pregnant Sprague-Dawley dams were exposed to 5 parts per million sulfur dioxide for 1 hour daily throughout gestation and with their pups after birth to medical-grade air through 6 days postnatal. For postnatal exposure, dams were exposed to air, and after delivery along with their

Catalase inhibition into the fourth cerebral ventricle affects bradycardic parasympathetic response to increase in arterial pressure without changing the baroreflex.

Exogenous catalase influences neural control of cardiovascular system; however, we do not know yet if its inhibition into the fourth cerebral ventricle (4(th) V) influences baroreflex regulation. We evaluated the effects of central catalase inhibition on baroreflex in conscious Wistar rats. We used males Wistar rats (320-370 g), which were implanted with a stainless steel guide cannula into 4(th) V. The femoral artery and vein were cannulated for mean arterial pressure (MAP) and heart rate (HR) measurement and drug infusion, respectively. After basal MAP and HR recordings, the baroreflex was t

Altered plasticity of the parasympathetic innervation in the recovering rat submandibular gland following extensive atrophy.

Adult rat submandibular glands have a rich autonomic innervation, with parasympathetic and sympathetic nerves working in synergy rather than antagonistically. Ligation of the secretory duct rapidly causes atrophy and the loss of most acini, which are the main target cell for parasympathetic nerves. Following deligation, there is a recovery of gland structure and function, as assessed by autonomimetic stimulation. This study examines whether the parasympathetic nerves reattach to new target cells to form functional neuro-effector junctions. Under recovery anaesthesia, the submandibular duct of

Role of parasympathetic overactivity in water immersion stress-induced gastric mucosal lesion in rat.

Stress ulcer is clinically prevalent, but the underlying mechanisms are not well understood. This study aimed to investigate the role of sympathovagal imbalance in the development of water immersion restraint stress (WRS)-induced gastric mucosal lesion. Wistar rats were subjected to either restraint stress (RS) (n = 7) or WRS (n = 7) for 5 h. Linear parameters of heart rate variability and Poincaré plot were analyzed on the basis of the surface ECGs. Gastric mucosal lesion was evaluated by gross anatomy and histology. Mean R-R intervals significantly increased (P < 0.001) in a time-dependent m

Postprandial variations in ST-segment in a patient with Brugada syndrome and partial gastrectomy.

A 74-year-old man with a history of partial gastrectomy presented with an electrocardiogram consistent with Brugada syndrome and marked meal related fluctuations in the ST segment. ST-segment elevation was prominently attenuated at 30 minutes and increased at 120 minutes after meals. Analysis of heart rate variability revealed a relationship between postprandial heightened parasympathetic activity and increase in Brugada-type ECG abnormality. A rapid postprandial increase in blood glucose may initially stimulate sympathetic nervous activity and secondarily increase parasympathetic tone. Food i

Treatment of vascular dementia--evidence from clinical trials with cholinesterase inhibitors.

Cerebrovascular disease (CVD), as well as secondary ischemic brain injury from cardiovascular disease, are common causes of dementia and cognitive decline in the elderly. In addition, CVD frequently contributes to cognitive loss in patients with Alzheimer's disease (AD). Progress in understanding the pathogenetic mechanism involved in vascular cognitive impairment (VCI) and vascular dementia (VaD) has resulted in promising treatments of these conditions. Cholinergic deficits in VaD are due to ischemia of basal forebrain nuclei and of cholinergic pathways and can be treated with the use of the

Direct parasympathetic pathway from midbrain to ciliary muscles in cats and monkeys.

In cats and monkeys, we examined the parasympathetic component of the oculomotor complex, which directly innervates the ciliary muscle, using horseradish peroxidase (HRP). Labeled neurons of varying form and size were found in the Edinger-Westphal(EW) and the Perlia nuclei of the cat and in the anteromedian, EW, and Perlia nuclei of the monkey. Our study confirmed that a direct parasympathetic pathway exists from the midbrain to the ciliary muscles, and that accommodation is controlled in part by this direct link from the midsagittal region via a parasympathetic neuron of the oculomotor nuclea

Apical secretion of rat submandibular tissue kallikrein continues in the absence of external stimulation: evidence for a constitutive secretory pathway.

Sequential samples of saliva evoked by low frequency parasympathetic nerve stimulation were collected from cannulated submandibular ducts of anaesthetized rats following periods of varying length without stimulation. After such rest periods the first samples of saliva were found to contain much higher levels of tissue kallikrein (rK1) activity and protein than the ensuing samples from the same stimulation period and the latter contained levels similar to those found previously in parasympathetically-evoked saliva. rK1 activity in first samples increased with the length of the preceding interva

Mechanisms of substance P-induced pulmonary oedema in the rabbit: interactions between parasympathetic and excitatory NANC nerves.

The pharmacological mechanisms involved in the substance P (SP)-induced pulmonary oedema were studied in isolated perfused rabbit lungs. Substance P induced a dose-dependent increase in the capillary filtration coefficient (Kf,c), responsible for oedema. Atropine, hemicholinium-3 and ruthenium red pretreatment partly protected the lungs against SP effects, while tetrodotoxin and hexamethonium did not significantly modify them. (+/-)CP96,345, a NK1 receptor antagonist, completely inhibited the SP-induced increase in the Kf,c. Like SP, acetylcholine (ACh) and capsaicin also increased the Kf,c. A

CNS cell groups projecting to the submandibular parasympathetic preganglionic neurons in the rat: a retrograde transneuronal viral cell body labeling study.

The retrograde transneuronal viral tracing method was used to study the CNS nuclei that innervate the parasympathetic preganglionic neurons controlling the submandibular gland in the rat. A genetically engineered beta-galactosidase expressing Bartha strain of pseudorabies virus (PRV) was injected into the submandibular gland of rats. After 4 days, PRV infected tissues were reacted with the Bluo-Gal substrate (halogenated indolyl-beta-D-galactoside) and labeled cell bodies were identified throughout the brain. In the medulla oblongata, cell body labeling was seen in the superior salivatory nucl

The role of substance P in parasympathetic nerve-induced secretion in the rat submandibular gland.

Fluid secretion from rat submandibular saliva evoked by electrical stimulation of the chorda tympani (parasympathetic secretory stimulation) was greatly reduced in the presence of atropine. However, some secretion was always evident at frequencies of 5 Hz and higher. The substance P-antagonist ([D-Arg1, D-Pro2, D-Trp7,9, Leu11]-substance P) also reduced the chorda-evoked saliva. Salivary secretion induced by the electrical stimulation of the superior cervical ganglion (sympathetic secretory nerve) was not affected by either atropine or substance P-antagonist. Continuous chordal stimulation red

The mechanism of acid-induced increases in canine lower esophageal sphincter pressure.

Acidification of the gastric cardia has been shown to increase lower esophageal sphincter pressure (LESP). The mechanism by which this phenomenon occurs remains unknown. This study was undertaken to examine the effect and mechanism of action of proximal gastric acidification on LESP in the dog model. In long-term studies, acidification resulted in a significant increase in mean LESP (23.2 cm H2O). Pretreatment with either topical lidocaine or subcutaneous atropine blocked the sphincteric response to acidification. Neither truncal vagotomy and pyloroplasty, proximal gastric vagotomy, antral vag

Cholinergic bronchomotor tone in COPD. Estimates of its amount in comparison with that in normal subjects.

The use of anticholinergic bronchodilators in COPD is based on the reversal of cholinergic bronchomotor tone. There is little information about the magnitude of cholinergic tone in patients with COPD as compared with normal subjects. As an index of the amount of cholinergic tone we measured the maximum increase in FEV1 following administration of an optimal dose of the anticholinergic agent atropine methonitrate. The study included nine normal nonsmoking subjects, ten normal smokers and 22 subjects with mild to moderately severe COPD. We found that normal nonsmokers had smallest increases in F

Synaptic potentials induced by postganglionic stimulations in cat bladder parasympathetic neurones.

Intracellular recording techniques were used to examine and compare synaptic potentials evoked by stimulating pre- and postganglionic nerve trunks in cat bladder parasympathetic ganglia. In the 76 ganglion cells examined, two types of responses were recorded on stimulating the postganglionic nerve: an antidromic action potential (type PostNS1; n = 30) or a fast excitatory postsynaptic potential (f-EPSP; type PostNS2; n = 46) which resulted in an orthodromic-like action potential. In some of the cells exhibiting a PostNS1 response (n = 19), a fast depolarization was superimposed on the antidrom

Effects of adenosine on contractile response of circular muscle in electrically stimulated guinea-pig ileum.

The action of adenosine on the electrically induced mechanical response of circular muscle in isolated guinea-pig ileum has been investigated. Electrical stimulation (0.1 Hz) elicited the twitch response, which was completely abolished by tetrodotoxin (0.2 microM), morphine (1 microM) and atropine (0.1 microM). Adenosine (0.1-100 microM) markedly depressed the twitch response in a concentration-dependent manner, and the concentration-depression curve for adenosine was significantly shifted to the right in the presence of theophylline (30 microM). On the other hand, the contractile responses in

[Mechanism of changes in secretory cell polarization in submandibular salivary glands during experimental botulism].

The polarization level (membrane potential-MP) of varied cells from the submandibular salivary gland was studied in different stages of botulinic poisoning in rats. The MP level of the cells decreased only in the late period (30 days) of local injury to the gland. However, pilocarpine stimulation of the secretory activity over the same period of time made MP return to normal. The authors believe this to be related to the phenomenon of gland decentralization since botulism is likely to entail an injury to the preganglionic parasympathetic neuron innervating the salivary gland, whereas the postg
