Development and experimental validation of an osteoporosis diagnosis model based on disulfidoptosis-related genes and immune infiltration analysis.

BACKGROUND: The role of disulfidoptosis, a novel form of programmed cell death, in osteoporosis remains unclear. This study aimed to identify disulfidoptosis-related molecular signatures for diagnosis, explore their link to immune infiltration, and validate their expression experimentally. METHODS: We conducted an integrative bioinformatics analysis utilizing microarray datasets from the Gene Expression Omnibus (GEO). Analyses included the identification of disulfidoptosis-related differentially expressed genes (DRDEGs), weighted gene co-expression network analysis (WGCNA), and functional enr

Decoding the grasping intention from electromyography during reaching motions.

BACKGROUND: Active upper-limb prostheses are used to restore important hand functionalities, such as grasping. In conventional approaches, a pattern recognition system is trained over a number of static grasping gestures. However, training a classifier in a static position results in lower classification accuracy when performing dynamic motions, such as reach-to-grasp. We propose an electromyography-based learning approach that decodes the grasping intention during the reaching motion, leading to a faster and more natural response of the prosthesis. METHODS AND RESULTS: Eight able-bodied subj

LIMK1 and PKM as Integratively Identified Transcriptomic Signatures in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) involves complex immunometabolic dysregulation. Mitochondria-associated endoplasmic reticulum membranes (MAMs) link metabolic adaptation and inflammatory signaling, but gene expression features associated with MAM-related transcriptomic states in IBD remain unclear. Public transcriptomic datasets were integrated with a curated MAM-related gene set and herbal target information. Differential expression analysis, weighted gene coexpression network analysis, and machine learning-assisted feature selection were used to identify candidate genes. A two-gene nomogram

NewRep: a classification model for online pattern recognition based on HD-EMG signals.

High density (HD) electromyography (EMG) signals have been widely used for pattern recognition (PR) in myoelectric control, particularly for gesture recognition. Although neural networks generally outperform traditional machine learning models in EMG PR, the limited accuracy in recognition remains a critical challenge. To further improve the online classification of neural networks in EMG PR, this study establishes a new neural network named NewRep by introducing the self-attention mechanism module into the existing RepViT, a lightweight convolutional neural network inspired by vision transfor

Individual voxel-based subtype prediction can differentiate progressive supranuclear palsy from idiopathic Parkinson syndrome and healthy controls.

Voxel-based morphometry (VBM) shows a differentiated pattern in patients with atypical Parkinson syndrome but so far has had little impact in individual cases. It is desirable to translate VBM findings into clinical practice and individual classification. To this end, we examined whether a support vector machine (SVM) can provide useful accuracies for the differential diagnosis. We acquired a volumetric 3D T1-weighted MRI of 21 patients with idiopathic Parkinson syndrome (IPS), 11 multiple systems atrophy (MSA-P) and 10 progressive supranuclear palsy (PSP), and 22 healthy controls. Images were

Multidomain Analysis of Clinical Cognitive Assessments and Imaging Data in Alzheimer's Disease Accurately Predicts Disease Stage and Grade Independent of Amyloid and Tau.

Background Individual clinical cognitive assessments (CCA) for Alzheimer's disease (AD) provide broad disease stratification but are limited in sensitivity and specificity, requiring integration of multiple CCA for optimal disease staging. Recent work from our lab suggests that neuro-metabolic and vascular dysregulation (MVD) occurs early in AD, prior to clinical symptoms, and may provide higher sensitivity and specificity than CCA alone. In this study, we combined three widely accepted CCA with MVD readouts and developed a multimodal ensemble machine learning approach across the AD spectrum t

Functional connectivity of large-scale brain networks in patients with anti-NMDA receptor encephalitis: an observational study.

BACKGROUND: In anti-NMDA receptor (NMDAR) encephalitis, antibody-mediated dysfunction of NMDARs causes severe neuropsychiatric symptoms, including psychosis, memory deficits, and movement disorders. However, it remains elusive how antibody-mediated NMDAR dysfunction leads to these symptoms, and whether the symptoms arise from impairment in specific brain regions and the interactions between impaired regions. METHODS: In this observational study, we recruited 43 patients with anti-NMDAR encephalitis from a tertiary university hospital and 43 age-matched and sex-matched healthy controls without

Multiscale Brain Structural Network Reorganization in Diabetic Retinopathy Using Regional Radiomics Similarity Networks.

BACKGROUND: Diabetic retinopathy (DR), a common microvascular complication of diabetes, is increasingly viewed as a systemic disorder involving the retina-brain axis. Although neuroimaging studies have reported brain abnormalities in DR, large-scale network reorganization and its biological substrates remain unclear. METHODS: This cross-sectional study included 47 patients with type 2 diabetes mellitus and non-proliferative diabetic retinopathy (T2DM-NPDR) and 45 matched healthy controls. Structural MRI data were used to construct individualized regional radiomics similarity networks (R2SNs).

Identification of distinct clinical phenotypes of cardiogenic shock using machine learning consensus clustering approach.

BACKGROUND: Cardiogenic shock (CS) is a complex state with many underlying causes and associated outcomes. It is still difficult to differentiate between various CS phenotypes. We investigated if the CS phenotypes with distinctive clinical profiles and prognoses might be found using the machine learning (ML) consensus clustering approach. METHODS: The current study included patients who were diagnosed with CS at the time of admission from the electronic ICU (eICU) Collaborative Research Database. Among 21,925 patients with CS, an unsupervised ML consensus clustering analysis was conducted. Th

Oscillatory edge connectivity in pain-related regions supports machine learning identification of migraine.

BACKGROUND: Objective and interpretable brain signatures for migraine diagnosis and phenotyping remain limited. Edge-centric connectivity treats interregional connections as primary analytical units and captures higher-order, frequency-resolved coordination patterns that may not be detected by conventional node-based functional connectivity approaches. This study investigated whether resting-state magnetoencephalography-derived oscillatory edge-connectivity features within pain-related regions may provide mechanistic insights into migraine neuropathology and inform future research on complemen

Integrated clinical and multi-omics analysis links composite inflammatory indices to macrophage-associated molecular programs in aortic dissection.

BACKGROUND: Aortic dissection (AD) is a life-threatening vascular disease characterized by high mortality and complex pathophysiology involving inflammation and vascular wall degeneration. Composite inflammatory indices derived from routine blood tests have shown prognostic value in AD; however, their role in disease assessment and their integration with molecular mechanisms remain unclear. METHODS: A total of 288 participants (144 AD patients and 144 controls) were enrolled for clinical analysis. Six composite inflammatory indices (NLR, MLR, SIRI, SII, PHR, and AISI) were calculated and eval

BACKGROUND: Schizophrenia (SC) is one of the most common mental illnesses. However, the underlying genes that cause it and its effective treatments are unknown. Programmed cell death (PCD) is associated with many immune diseases and plays an important role in schizophrenia, which may be a diagnostic indicator of the disease. METHODS: Two groups as training and validation groups were chosen for schizophrenia datasets from the Gene Expression Omnibus Database (GEO). Furthermore, the PCD-related genes of the 12 patterns were extracted from databases such as KEGG. Limma analysis was performed for

Deep behavioural phenotyping of the Q175 Huntington disease mouse model: effects of age, sex, and weight.

BACKGROUND: Huntington disease (HD) is a neurodegenerative disorder with complex motor and behavioural manifestations. The Q175 knock-in mouse model of HD has gained recent popularity as a genetically accurate model of the human disease. However, behavioural phenotypes are often subtle and progress slowly in this model. Here, we have implemented machine-learning algorithms to investigate behaviour in the Q175 model and compare differences between sexes and disease stages. We explore distinct behavioural patterns and motor functions in open field, rotarod, water T-maze, and home cage lever-pull
