Human early-onset dementia caused by DAP12 deficiency reveals a unique signature of dysregulated microglia.

The TREM2-DAP12 receptor complex sustains microglia functions. Heterozygous hypofunctional TREM2 variants impair microglia, accelerating late-onset Alzheimer's disease. Homozygous inactivating variants of TREM2 or TYROBP-encoding DAP12 cause Nasu-Hakola disease (NHD), an early-onset dementia characterized by cerebral atrophy, myelin loss and gliosis. Mechanisms underpinning NHD are unknown. Here, single-nucleus RNA-sequencing analysis of brain specimens from DAP12-deficient NHD individuals revealed a unique microglia signature indicating heightened RUNX1, STAT3 and transforming growth factor-β

A Protective Role of Tumor Necrosis Factor Superfamily-15 in Intracerebral Hemorrhage-Induced Secondary Brain Injury.

Destabilization of blood vessels by the activities of vascular endothelial growth factor (VEGF) and matrix metalloproteinases (MMPs) following intracerebral hemorrhage (ICH) has been considered the main causes of aggravated secondary brain injury. Here, we show that tumor necrosis factor superfamily-15 (TNFSF15; also known as vascular endothelial growth inhibitor), an inhibitor of VEGF-induced vascular hyper-permeability, when overexpressed in transgenic mice, exhibits a neuroprotective function post-ICH. In this study, we set-up a collagenase-induced ICH model with TNFSF15-transgenic mice and

X-linked thrombocytopenia with thalassemia displays bone marrow reticulin fibrosis and enhanced angiogenesis: comparisons with primary myelofibrosis.

X-linked thrombocytopenia with thalassemia (XLTT) is caused by the mutation 216R > Q in exon 4 of the GATA1 gene. Male hemizygous patients display macrothrombocytopenia, splenomegaly, and a β-thalassemia trait. We describe two XLTT families where three males were initially misdiagnosed as having primary myelofibrosis (PMF) and all five investigated males showed mild-moderate bone marrow (BM) reticulin fibrosis. Comparative investigations were performed on blood samples and BM biopsies from males with XLTT, PMF patients and healthy controls. Like PMF, XLTT presented with high BM microvessel den

[Pathology of soft tissue sarcomas].

Soft tissue sarcomas comprise around 1% of all malignant tumors, but they are relatively more frequent in childhood and adolescence. This latter fact emphasizes the need that timely diagnosis should be established for optimal treatment. The very recent WHO classification (2013) lays down the following main categories: adipocyte tumors, fibroblast/myofibroblast tumors, so-called fibrohistiocyte tumors, smooth-muscle tumors, pericyte tumors, skeletal-muscle tumors, vascular tumors, chondro-osseous tumors, gastrointestinal stromal tumors, nerve sheath tumors, tumors of uncertain differentiation a

Endometrial bleeding in postmenopausal women: with and without hormone therapy.

The aim of this study was to present a review of the potential mechanisms involved in the occurrence of endometrial bleeding in postmenopaus … dysfunction in vasculogenesis that could result in blood vessel fragility and bleeding. The role of pericytes in maintaining vessel morphology and integrity is discussed. Endometrial leukocytes and metalloproteinases are involved in normal menstruation, but their role in postmenopausal bleeding is not clear suggesting involvement of mechanisms in the bleedi …

Vascularization of cutaneous melanoma involves vessel co-option and has clinical significance.

This study was undertaken to determine the role and the fate of the peritumoural vascular plexus during the vascularization of human malignant melanoma (hMM) and in an appropriate murine melanoma model system. The prognostic significance of the vascularity of different tumour areas was also evaluated. Despite morphometry revealing several-fold higher microvessel densities (MVDs) in the peritumoural tissue than at the centre of the tumour, the development of visceral metastases of hMM was exclusively correlated with the MVD of the tumour centre. Furthermore, the 5-year survival of the patient g

3D Bioprinted Blood-Brain-Tumor Barrier Spheroid-Based In Situ Evaluation of Anticancer Compounds Against Glioblastoma: ROS-Producing Ruthenium as a Potential Therapeutic Candidate.

The blood-brain-tumor barrier (BBTB) penetration by anticancer drugs and their effects on glioblastoma (GBM) are assessed using 3D bioprinted-spheroids. Comparative analysis reveals prominent cytoplasmic mixing between pericytes and the human GBM cell line U-87MG, expressions of tight junction proteins ZO-1, occludin, and claudin-5, which act as a barrier to the paracellular transport across BBTB, and the expression of ABC transporter such as P-glycoprotein (P-gp) in the vascular endothelial cell layer of BBTB spheroids. Anticancer drug-induced U-87 MG cell death is quantified through in situ …

Development of a Human 3D Immune-Competent Neurovascular Model Enabling Time-Resolved Monitoring of Neuroinflammatory Dynamics and Neuroimmune Interactions.

Neuroinflammation disrupts the blood-brain barrier (BBB) and drives neurological disease, but current in vitro MPS and animal models fail to capture its dynamic, human-specific immune complexity. Here, we introduce a 3D human immune-competent BBB (3D-HIC-BBB) platform that reconstitutes the neurovascular unit (NVU) by integrating primary human brain microvascular endothelial cells, astrocytes, brain vascular pericytes, human microglia, and dopaminergic neurospheroids with embedded transepithelial electrical resistance (TEER) microsensors for continuous functional monitoring. Our platform desig…

In situ mapping of activated PDGFRβ defines a prognostic discrepancy between histological subtypes of NSCLC.

BACKGROUND: Increased stromal Platelet-derived growth factor receptor beta (PDGFRβ) expression is a hallmark of the desmoplastic tissue reaction in cancer and marks subsets of cancer-associated fibroblasts, pericytes, and smooth muscle cells. However, its functional status in situ has been anticipated from static expression measures, which cannot determine whether high receptor abundance reflects active signaling. METHODS: We established two second-generation proximity ligation assays (PLAs) to quantify PDGFRβ activation in the in situ environment of human lung cancer by detecting either phosp…

Resection of multiple intramedullary spinal cord cavernous malformations in infancy: illustrative case.

BACKGROUND: Cavernous malformations (CMs) are vascular malformations characterized by enlarged, thin-walled blood vessels composed of dysplastic endothelial cells and pericytes within a loose extracellular matrix. Endothelial cell dysplasia leads to increased permeability and portends an increased risk of hemorrhage within these malformations, and hemorrhage often manifests with acute-onset neurological deficits due to mass effect and irritation of surrounding neural tissue within the brain and spinal cord. OBSERVATIONS: A 5-month-old male presented with concern for possible tethered cord syn…

Protective Effects of Extracts from Green Leaves and Rhizomes of Posidonia oceanica (L.) Delile on an In Vitro Model of the Human Blood-Brain Barrier.

Posidonia oceanica (L.) Delile, a Mediterranean seagrass, is rich in bioactive compounds with anti-inflammatory potential. While marine-derived molecules are increasingly studied, their direct effects on blood-brain barrier (BBB) integrity under inflammatory conditions remain largely unexplored. This study evaluated the ability of aqueous extracts from its green leaves (GLEs) and rhizomes (REs) to protect the BBB using a human in vitro model consisting of brain-like endothelial cells co-cultured with brain pericytes. The model was exposed to TNFα, with or without GLEs or REs. We assessed NO pr…
