Neutralization of acyl-CoA-binding protein attenuates glucocorticoid-mediated suppression of cancer immunosurveillance.

Glucocorticoids (GCs) are potent immunosuppressive agents that compromise anticancer immune responses, yet the molecular mediators of this effect remain incompletely understood. Here, we identify the acyl-CoA-binding protein/diazepam-binding inhibitor (ACBP/DBI) as a critical effector of the GC-induced suppression of tumor immunosurveillance and immunotherapy efficacy. Using orthotopic murine models of breast cancer, non-small cell lung cancer, and cutaneous fibrosarcoma, we show that corticosterone (CORT) accelerates tumor progression and abrogates therapeutic responses to immunogenic chemoth

Rosiglitazone hepatotoxicity resulting from hepatic protein covalent binding derived from its aldehyde metabolite.

Rosiglitazone (RGZ), a thiazolidinedione insulin sensitizer, is widely used in the treatment of type 2 diabetes mellitus. However, concerns regarding its dose-dependent hepatotoxicity have emerged, and the mechanisms of the toxic action remain elusive. This study aimed to investigate the mechanism by which an aldehyde metabolite generated through metabolic activation of RGZ forms covalent adduction with protein lysine residues and its contribution to hepatotoxicity. Using in vitro liver microsomal incubations coupled with LC-MS/MS, the reactive aldehyde metabolite and its corresponding lysine

Cell-specific Y-box-binding protein-1 drives liver-kidney injury in cholestasis.

Acute kidney injury is a frequent, life-threatening complication of liver disease. The stress-responsive DNA- and RNA-binding protein Y-box-binding protein-1 (YB-1) regulates gene expression and modulates liver-kidney cross talk, with partial systemic YB-1 deficiency protecting the liver but worsening kidney injury in a murine bile duct ligation (BDL) model. To further define cell-specific roles of YB-1, its expression was selectively reduced in the kidney and immune cells, and the effects on primary liver and secondary kidney injury were evaluated. Conditional Ybx1 knockout mice targeting tub

Probiotics mitigate non-antibiotic drug-induced dysbiosis via protein homology-driven competitive binding.

While the impact of non-antibiotic drugs on gut bacteria is well-known, their mechanisms of action remain poorly characterized, and effective mitigation strategies for drug-induced dysbiosis are still limited. Here, we screened bacteria-derived drug-target protein homologs (BDTPHs) mapped to 63 target proteins and 107 associated drugs to quantify "drug-BDTPH-bacterium" interactions. These interactions were validated by co-culture experiments using 10 drugs and 25 strains, enzyme assays, and genetic perturbations in Escherichia coli. Ex vivo and in vivo testing with six drugs showed that over 5

Sulfoximines as Secondary Sulfonamide Isosteres: Paradoxical Impact on Solubility and Plasma Protein Binding.

We report paradoxical findings in properties when secondary sulfonamides are replaced with isosteric secondary sulfoximines. Despite the reduction in polar surface area and the elimination of a hydrogen bond acceptor and a hydrogen bond donor in the RS(O)2NHR to RS(O)(Me) = NR transformation, the resulting sulfoximines generally had increased aqueous solubility and decreased albumin binding, with experimental logP/D values substantially lower than predicted. Despite these characteristics of increased hydrophilicity of the sulfoximine motif, permeability and metabolic stability were similar to

Salt-Dependent Modulation of the RNA Chaperone Activity of RNA-Binding Protein La.

It is well established that the RNA-binding protein La has RNA chaperone activity. Recent work suggests that the La protein has two distinct RNA chaperone domains (RCD-A and RCD-B), assisting structural changes in diverse groups of RNA molecules such as RNA Polymerase III transcripts (e.g., pre-tRNA, U6 snRNA), cellular messenger, and viral RNAs. In this protocol, we focus on the RNA chaperone domain RCD-B, which is located in the carboxy-terminal domain of La. It has been shown that this RNA chaperone domain assists structural changes in predicted RNA hairpins folded in the 5'-untranslated re

RNA binding protein DDX3X drives pancreatic cancer progression via the TLE2-MYL9 axis.

Current treatments for pancreatic ductal adenocarcinoma (PDAC) fall short of meeting clinical needs, highlighting the urgent need for a comprehensive understanding of PDAC progression, which involves not only biochemical signals but also essential biomechanical cues. Here, we used a CRISPR-Cas9 screen in an orthotopic xenograft model to explore PDAC dynamics. The RNA binding protein DEAD-box helicase 3X-linked (DDX3X) was identified as a pivotal oncogene and biomechanical checkpoint. Specifically, DDX3X up-regulation in PDAC promoted tumorigenesis and metastasis, primarily through the transcri

Nonstructural Protein 1 of Influenza A (NS1A) Demonstrates Strain-Specific dsRNA Binding Capabilities.

Nonstructural protein 1 of influenza A (NS1A) is a key virulence factor produced inside host cells infected with Influenza A Virus (IAV) and consists of an N-terminal dsRNA binding domain (RBD) and a C-terminal effector domain (ED), joined by a flexible linker. While NS1A is a highly promiscuous protein with a number of intracellular functions, its primary function is nonspecific dsRNA binding that enables influenza to evade our innate immune system. For this reason, NS1A has long been proposed as a potential drug target. Previous research in the field has demonstrated the necessity of dimer f

Dual-channel hierarchical interactive learning for the prediction of Protein-Ligand binding affinity.

Protein-ligand binding affinity (PLBA) is a crucial metric in drug screening for identifying potential candidate compounds. In recent years, deep learning-based methods have used representation learning to model interactions within protein-ligand complexes, demonstrating great promise in affinity prediction tasks. Existing studies have considered both intramolecular (covalent) and intermolecular (non-covalent) interactions to some extent. However, these interactions are often treated as independent features, lacking explicit hierarchical dependency modeling, which may lead to insufficient repr

Lipopolysaccharide-Binding Protein (LBP) and Inflammatory Biomarkers in SARS-CoV-2 Hospitalized Patients.

Background/Objectives: Infection with the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused a global pandemic with far-reaching impacts on human activities. Moreover, direct viral damage and uncontrolled inflammation have been proposed as contributing factors to the severity of SARS-CoV-2 disease. Lipopolysaccharide binding protein (LBP) is also well recognized for its capability to trigger and modulate the host's innate immune system by attaching to bacterial substances. Nevertheless, the pandemic has further emphasized the critical role of an effective host immune response

The RNA-binding protein HuR impairs adipose tissue anabolism in pancreatic cancer cachexia.

BACKGROUND: Cachexia is defined by chronic loss of fat and muscle, is a frequent complication of pancreatic ductal adenocarcinoma (PDAC), and negatively impacts patient outcomes. Nutritional supplementation cannot fully reverse tissue wasting, and the mechanisms underlying this phenotype are unclear. This work aims to define the relative contributions of catabolism and anabolism to adipose wasting in PDAC-bearing mice. Human antigen R (HuR) is an RNA-binding protein recently shown to suppress adipogenesis. We hypothesize that fat wasting results from a loss of adipose anabolism driven by incre

Cold Inducible RNA-Binding Protein: A Novel Therapeutic Target for Ventricular Arrhythmia Control.

Ventricular arrhythmias following myocardial infarction (MI) remain a leading cause of sudden cardiac death, yet therapeutic options are limited by incomplete understanding of the molecular mechanisms governing post-infarction arrhythmogenesis. While RNA-binding proteins have emerged as critical regulators of cardiovascular pathophysiology, their role in cardiac electrophysiology remains largely unexplored. Here, we demonstrate that cold- inducible RNA-binding protein (CIRP) functions as a critical regulator of ventricular arrhythmia susceptibility through post-transcriptional control of cardi

Fatty acid-binding protein 5 aggravates psoriasis and psoriasis-like disease through ferroptosis.

Psoriasis (Ps) is a chronic inflammatory skin disease with systemic manifestations, such as psoriatic arthritis (PsA), cardiovascular and psychiatric complications, and subsequent negative effects on patients' quality of life. Although biologics targeting specific disease mediators have become a mainstay in Ps treatment, exploration of new disease targets to improve treatment is still needed. Here we show that fatty-acid binding protein 5 (Fabp5) promotes skin inflammation through a therapeutically relevant modulation of the ferroptotic response. Altered mRNA transport and local translation in i3Neurons with RNA-binding protein knockdown.

Neurons rely on messenger RNA (mRNA) transport and local translation to facilitate rapid protein synthesis in processes far from the cell body. These processes allow precise spatial and temporal control of translation and are mediated by RNA-binding proteins (RBPs), including those associated with neurodegenerative diseases. Here, we use proteomics, transcriptomics, and microscopy to investigate the impact of RBP depletion on mRNA transport and local translation in induced pluripotent stem cell-derived neurons. We find thousands of transcripts enriched in neurites and that many of these transc

A divergent cyclic nucleotide binding protein promotes Plasmodium ookinete infection of the mosquito.

Colonisation of mosquitos by the malarial parasite is critically reliant on the invasive ookinete stage. Ookinete invasion of mosquito is coordinated by the apical complex, a specialised parasite structure containing components for secretion, attachment and penetration. While studies have investigated cytoskeletal and secretory elements, it is currently unknown if signalling modules are present or functional at the apical complex. Here we elucidate the role of a cryptic cyclic nucleotide-binding protein which we name CBP-O. PbCBP-O showed a marked localisation to the ookinete apex and disrupti

Periplasmic Binding Protein YiuA Enables Yersinia pestis to Scavenge Fe(III)-Catechol Siderophores.

Yersinia pestis, the pathogen causing plague, requires iron to grow. Y. pestis employs several uptake pathways for iron, including the siderophore yersiniabactin, as well as hemin and inorganic iron. The Y. pestis iron assimilation repertoire further harbors the uncharacterized YiuRABC pathway, presumed to transport an unidentified Fe(III)-siderophore(s). Through intrinsic fluorescence quenching of the periplasmic binding protein YiuA, we discovered that YiuA displays high affinity toward Fe(III) complexes of the hydrolysis products of enterobactin, Fe(III)-[di(DHB-LSer)] and Fe(III)-[DHB-LSer
