Dual inhibition of neutrophil superoxide generation and elastase release via JNK, ERK and FAK pathways: blockade by methoxy-chalcone derivatives.

A series of chalcone-based derivatives were synthesized and assessed for their ability to target neutrophil-driven inflammation. Strategic modifications on the 1,3-diaryl-prop-2-en-1-one scaffold, all bearing a methoxylated ring B, revealed two distinct anti-inflammatory profiles: inhibitors of both superoxide (SO) generation and elastase release, and selective SO inhibitors. Compound 17, featuring 3-methoxy and 2,4-dichlorophenyl substitutions, demonstrated the most potent dual inhibition (IC50 = 1.17 µM for SO and 2.60 µM for elastase), while compound 15 selectively suppressed SO production

Activity-guided native-PAGE electroelution coupled with DIA proteomics uncovers the superoxide dismutase and antioxidant network in mature peanut seeds.

Mature peanut (Arachis hypogaea L.) seeds accumulate abundant superoxide dismutase (SOD), but the high abundance of storage proteins masks trace antioxidant enzymes and prevents accurate profiling of endogenous SOD isoforms by conventional proteomics. Here we report an activity-guided workflow that combines native-PAGE NBT zymography, low-temperature dialysis-bag electroelution and DIA-based nano-LC-MS/MS to enrich native SOD complexes from seed crude extracts. The two SOD-active gel bands, B1 and B2, were excised, embedded in agarose and electroeluted to recover intact proteins before proteom

Goethite-mediated enhancement of microbial Mn(II) oxidation and BaP degradation by Pseudomonas putida: The driving mechanism of extracellular superoxide radical.

Microbial functions and activities of Mn(II)-oxidizing microorganisms are influenced by minerals in the environment; however, the role of environmental minerals in mediating microbial Mn(II) oxidation and degradation of pollutants remains poorly understood. Here, the effect of a typical environmental mineral (goethite, Goe) on microbial Mn(II) oxidation by Pseudomonas putida MnB1 and degradation of benzo [a]pyrene (BaP) was systematically investigated. The addition of goethite significantly increased the bacterial oxidation rate of Mn(II) by MnB1 from 0.59 mg/L/h to 1.18 mg/L/h. Extracellular

Triclosan induces DNA damage in human ovarian granulosa cells via mitochondrial superoxide overproduction mediated by suppressed glutathione synthesis.

Triclosan (TCS) is a broad-spectrum antimicrobial agent that has raised public health concerns due to its continuous environmental release and human exposure. Previous studies suggest that TCS may disrupt endocrine function. Nonetheless, the toxicological effects on the female reproductive system, notably on ovarian granulosa cells, and the underlying pathways remain poorly understood. This study was designed to examine the toxic effects of TCS on human granulosa cells and the related mechanisms. To this end, human granulosa cell lines (KGN and SVOG) were treated in vitro with varying doses of

Targeted Proteomics upon Treatment with Tofersen Identifies Novel Response Markers for Superoxide Dismutase 1-Linked Amyotrophic Lateral Sclerosis.

OBJECTIVE: Tofersen is the first effective and approved therapy for superoxide dismutase 1 (SOD1)-associated amyotrophic lateral sclerosis (ALS [SOD1-ALS]). Following treatment with tofersen, neurofilament levels in patients' cerebrospinal fluid (CSF) and serum seem to respond earlier than clinical parameters. This evidence prompted us to hypothesize that this novel treatment could provide an opportunity to identify additional biomarkers responsive to therapy in SOD1-ALS. METHODS: We investigated a panel of 120 neural, glial, and inflammatory markers in CSF and serum samples longitudinally co

The effects of transcranial direct current stimulation on serum superoxide dismutase levels in ambulatory ischaemic stroke patients: A randomised pilot study.

INTRODUCTION: Stroke is one of the leading causes of disability worldwide, imposing a significant public health and economic burden. Oxidative stress, which results from an imbalance between free radical production and antioxidant defences, exacerbates stroke outcomes via its role in promoting neuroinflammation and tissue damage. Superoxide dismutase (SOD), a critical antioxidant enzyme, plays a key role in mitigating oxidative stress. This study evaluates the effect of transcranial direct current stimulation (tDCS) on serum SOD levels in ambulatory ischaemic stroke patients with mild cognitiv

Concentration-dependent effects of bacterial melanin on new superoxide-producing associates in rat tissues: a rotenone neurotoxic model of parkinson's disease.

BACKGROUND: Oxidative stress plays a central role in the degeneration of dopaminergic neurons and is a key factor in the pathogenesis of Parkinson's disease (PD). Therefore, antioxidant therapy represents a promising therapeutic strategy. This process involves numerous enzymes and signaling pathways that contribute to disease progression. In the present study, bacterial melanin (BM), known for its high biological activity and biostimulatory effects, was applied as a potential antioxidant. We examined quantitative and qualitative alterations in total fractions of biomembrane-associated componen

Parkinson-like wild-type superoxide dismutase 1 pathology induces nigral dopamine neuron degeneration in a novel murine model.

Atypical wild-type superoxide dismutase 1 (SOD1) protein misfolding and deposition occurs specifically within the degenerating substantia nigra pars compacta (SNc) in Parkinson disease. Mechanisms driving the formation of this pathology and relationship with SNc dopamine neuron health are yet to be fully understood. We applied proteomic mass spectrometry and synchrotron-based biometal quantification to post-mortem brain tissues from the SNc of Parkinson disease patients and age-matched controls to uncover key factors underlying the formation of wild-type SOD1 pathology in this disorder. We als

Electrochemical biosensing platform based on AuNWs/rGO-CMC-PEDOT:PSS composite for the detection of superoxide anion released from living cells.

Detection of superoxide anion (O2·-) levels holds significant importance for the diagnosis and even clinical treatments of oxidative stress-related diseases. Herein, we prepared a composite electrode material to encapsulate copper-zinc superoxide dismutase (SOD1) for biosensing of O2·-. The sensing material consists of gold nanowires (AuNWs), reduced graphene oxide (rGO), carboxymethyl cellulose (CMC) and PEDOT:PSS. CMC provides abundant -COOH to bind SOD1, with a high adsorption coverage of 1.499 × 10-9 mol cm-2 on the sensor surface. rGO and PEDOT endow the composite with significant conduct

Regulatory effect of bacterial melanin on the isoforms of new superoxide-producing associates from rat tissues in rotenone-induced Parkinson's disease.

According to recent research, selective neuronal vulnerability in Parkinson's disease (PD) results from several phenotypic traits, including calcium-dependent, feed-forward control of mitochondrial respiration leading to elevated reactive oxygen species and cytosolic calcium concentration, an extensive axonal arbor, and a reactive neurotransmitter. Therefore, antioxidant therapy is a promising direction in the treatment of PD. In vitro studies have indicated the survival-promoting activity of bacterial melanin (BM) on midbrain dopaminergic neuron cultures. It has been established that BM has a

Exercise induces superoxide and NOX4 contribution in endothelium-dependent dilation in coronary arterioles from a swine model of chronic myocardial ischemia.

Exercise training is an effective, nonpharmacologic therapy and preventative measure for ischemic heart disease. While recent studies have examined reactive oxygen species (ROS) as mediators of exercise training-enhanced coronary blood flow, specific oxidants and their sources have yet to be fully elucidated. We investigated the hypothesis that NADPH oxidase (NOX)-derived superoxide anion would contribute to vasodilation effects in the coronary microcirculation of swine and that these effects would be impaired by chronic ischemia and rescued with exercise training. Measurement of Mitochondrial (Dys)Function in Cellular Systems Using Electron Paramagnetic Resonance (EPR): Oxygen Consumption Rate and Superoxide Production.

The oxygen consumption rate (OCR) and superoxide production are crucial when assessing mitochondrial function and/or dysfunction. EPR spectroscopy allows the measurement of both components either independently or simultaneously in a same cellular or mitochondrial preparation. OCR determination using EPR oximetry is based on the change in EPR linewidth of a paramagnetic oxygen sensing probe (a perdeuterated nitroxide) in the presence of oxygen consuming cells in a closed system. Superoxide production can be monitored by the oxidation of cyclic hydroxylamines into nitroxides. The contribution of

Clinically Meaningful Improvements in Sperm DNA Fragmentation Severity in Infertile Men Treated with Superoxide Dismutase Supplementation: A Single-Center Experience.

Background. Antioxidants are commonly used for the treatment of idiopathic male infertility. Previous studies have shown that antioxidants are able to improve sperm quality, but little is known about their impact on sperm DNA fragmentation (SDF). Preliminary findings showed that superoxide-dismutase (SOD)-based antioxidant plus hydroxytyrosol and carnosol (FertiPlus® SOD) therapy was associated with SDF improvement in a small cohort of infertile men. Therefore, we aimed to assess rates of and predictors of semen parameters and SDF improvements in infertile men treated with FertiPlus® SOD thera

Global Reach 2018: Nitric oxide-mediated cutaneous vasodilation is reduced in chronic, but not acute, hypoxia independently of enzymatic superoxide formation.

We tested the hypotheses that 1) cutaneous microvascular function is impaired by acute normobaric and chronic hypobaric hypoxia and 2) that the superoxide free radical (via NADPH oxidase or xanthine oxidase) contributes to this impairment via nitric oxide (NO) scavenging. Local heating-induced cutaneous hyperemia (39 °C) was measured in the forearm of 11 male lowlanders at sea level (SL) and following 14-18 days at high altitude (HA; 4340 m in Cerro de Pasco, Peru), and compared to 11 highlanders residing permanently at this elevation. The Effect of Superoxide Dismutase on Inhibition of Acute Kidney Injury Induced by Sepsis Based on Kidney Tissue Histology and Murine Sepsis Score.

Sepsis is one of the leading causes contributing to the incidence of acute kidney injury (AKI). Oxidative stress can be used as the main approach against sepsis-induced AKI. One of the primary antioxidants that plays a role in warding off oxidative stress is superoxide dismutase (SOD). This research aimed to observe the effect of antioxidant SOD in inhibiting sepsis in AKI based on kidney tissue histopathology. The research method was an experimental laboratory with a post-test-only control group design. Twenty-five adult male rats aged 12-16 weeks, weighing between 200 and 250 g, were randoml
