[Desoxyfructo-serotonin: its therapeutic effect in the treatment of leprosy].

Desoxyfructo-serotonin (DFS) has shown good results in clinical trials of LL patients. After clinical trials in Bamako (Mali) reported in three articles, clinical trials began in India, at Bombay. Acute toxicity tests done in Paris and chronic toxicity tests done in India had shown absence of side effects. This was also confirmed after pre-clinical pharmacology. In vitro tests show that DFS enhances cellular immune response. Receptors for anti-erythrocyte antibody on LL macrophages are demonstrated by erythrocyte rosetting. Infection with M. leprae markedly reduces rosetting. But in the presen…

Optimization of the extraction conditions and dermal toxicity of oil body fused with acidic fibroblast growth factor (OLAF).

INTRODUCTION: Oil body (OB), a subcellular organelle that stores oil in plant seeds, is considered a new transdermal drug delivery system. With the increasing understanding of the OB and its main protein (oleosin), numerous studies have been conducted on OB as "carrier" for the expression of exogenous proteins. In our previous study, oil body fused with aFGF (OLAF) was obtained using a plant oil body expression system that had been preliminarily proven to be effective in accelerating the healing of skin wounds. However, no dermal toxicological information on OLAF is available. OBJECTIVE: To e

Neurotoxic effects of representative pharmaceutical mixtures from Mediterranean rivers impacted by waste water treated effluents.

Mixture toxicity studies on neuroactive chemicals remain scarce and often rely on simplified combinations of compounds with known modes of action (MoA), despite the frequent non-monotonicity of neuroactive responses. Furthermore, real environmental mixtures are far more complex, containing chemicals with diverse MoAs, concentrations, and effect directions. This study identified environmentally relevant neuroactive mixtures in Mediterranean rivers by integrating behavioural and cardiovascular assays in Daphnia magna and zebrafish embryos with mixture-based laboratory tests and field sample comp

Disentangling micropollutant toxicity from major ion effects in treated water effluents: Advancing invertebrate biotests for enhanced ecotoxicological assessment.

Water treatment systems generate effluents with varying compositions of major geochemical ions and diverse micropollutants. Well-established procedures exist to assess the toxicity of effluents as a complete and unique matrix. Although informative, these approaches may fail to identify the toxicity specifically attributable to micropollutants. This review provides a bibliographic overview of the physicochemical and ecotoxicological characterization of 181 effluents reported across 88 publications. It highlights a general lack of information on effluent ionic composition while assessments are p

Subchronic Inhalation of TiO2 Nanoparticles Leads to Deposition in the Lung and Alterations in Erythrocyte Morphology in Mice.

TiO2 nanoparticles (NPs) are extensively used in various applications, highlighting the importance of ongoing research into their effects. This work belongs among rare whole-body inhalation studies investigating the effects of TiO2 NPs on mice. Unlike previous studies, the concentration of TiO2 NPs in the inhalation chamber (130.8 μg/m3) was significantly lower. This 11-week study on mice confirmed in vivo the presence of TiO2 NPs in lung macrophages and type II pneumocytes including their intracellular localization by using the electron microscopy and the state-of-the-art methods detecting NP

Hepatitis A Virus Infection in Cynomolgus Monkeys Confounds the Safety Evaluation of a Drug Candidate.

In a 3-month toxicity study in cynomolgus monkeys at a European contract laboratory, animals were infected with HAV, initially resulting in hepatic injury being incorrectly attributed to the test compound. Elevated serum ALT/AST/GLDH (5- to 10-fold) were noted in individual animals from all groups including controls, with no apparent dose, exposure, or time-related relationship. Liver histopathology revealed minimal to slight inflammatory cell accumulation in periportal zones of most animals, and minimal to slight hepatocyte degeneration/necrosis in 10/42 animals from all groups. As these find

Short exposure to ethyl and methylmercury prompts similar toxic responses in Drosophila melanogaster.

Methylmercury (MeHg) and ethylmercury (EtHg) are important mercury organic forms in terms of human poisoning. Since the comparative effects of compounds are mainly in vitro, this study was designed to investigate the toxicities induced by MeHg and EtHg in an in vivo study using adult Drosophila melanogaster (D. melanogaster). Firstly, we performed a survival curve, where the flies were fed on a medium containing MeHg and EtHg at concentrations ranging from 2.5 to 200 μM, until the end of their lifespan. After that, the concentrations 25 and 200 μM of MeHg and EtHg were chosen to be tested in a

[Current Issues in Photosafety Evaluation].

Phototoxicity is a toxic response elicited by topically applied or systemically administered photoreactive chemicals after exposure to light and can be broadly categorized into photoirritation, photoallergy, photogenotoxicity, and photocarcinogenicity. The need in the 21st century for accurate evaluation of photosafety has led to the publication of a number of guidelines from government agencies in Europe and the U.S.A. as well as the Organisation for Economic Co-operation and Development (OECD). In this review, we first discuss the mechanisms of phototoxicity and how they can be evaluated. We

Metabolic adaptability in liver and gastrocnemius muscle of mice following subacute lead toxicity.

Lead (Pb) is one of several environmental pollutants that adversely affect human health by producing toxicity at the tissue level. The aim of the study was to understand the effects of Pb on the metabolic profiles of liver and gastrocnemius muscle of mice in relation to carbohydrate and fat metabolisms. Swiss albino mice were chosen and divided into two groups, control and Pb-treated. The Pb-treated animals were exposed to Pb at a dose of 5 mg/kg body weight for 30 days orally, which resulted in hypoglycemia, glycosuria, and increased glycogenolysis in the liver and gastrocnemius muscle of tre

Evaluation of Pulmonary Toxicity of Nanoparticles by Bronchoalveolar Lavage.

Nanoparticles can enter the body via the lungs, the digestive tract, and the skin. The pulmonary effects of nanoparticles and the underlying mechanisms may be investigated by histopathological examination, immunostaining, bronchoalveolar lavage (BAL), and so on. BAL is a simple, fast, and inexpensive method that is commonly used to investigate the immune cells and acellular components in the lungs, which reflect the health state of the lungs. In this chapter, we describe the detailed procedures of performing BAL on mouse lungs. In a typical procedure, a catheter is inserted in the exposed trac

Ultra-morphology of the scale as an indicator of the stress of Acid Black-1 (AB-1, CI: 20470) and zinc (Zn).

High contents of azo dyes and heavy metals enter surface waters with the wastewater from dying and dye-manufacturing industries and pose serious threat to fish. In the present study, changes in the ultra-morphological features of the scale have been evaluated as indicators of the stress of lethal and sublethal concentrations of an azo dye Acid Black-1 (AB-1, CI 20470), zinc (Zn), and their mixture AB-1 + Zn to Labeo rohita. Fish were exposed for 96 h to lethal concentration (LC) causing 0-70% mortality, i.e., LC0, LC20, LC50, and LC70 of AB-1 (4, 6, 8, and 10 mg/L respectively) and Zn (25, 50,

Preclinical safety evaluation of ET-26 hydrochloride, a novel intravenous anesthetic agent, in beagle dogs.

ET-26 hydrochloride (ET-26HCl) is a novel etomidate analogue, approved for clinical trials, which has an effective sedative-hypnotic effect, a stable myocardial performance, and milder adrenocortical suppression than etomidate in rats and beagle dogs. Additionally, ET-26HCl showed similar hemodynamic stability as etomidate in the rat uncontrolled hemorrhagic shock model. Furthermore, ET-26HCl, in the rat lipopolysaccharide-induced sepsis model, was found to have a higher survival rate, a lower inflammatory reaction, and less organ injury. In the present study, we measured the potential adverse

Critical assessment and integration of separate lines of evidence for risk assessment of chemical mixtures.

Humans are exposed to multiple chemicals on a daily basis instead of to just a single chemical, yet the majority of existing toxicity data comes from single-chemical exposure. Multiple factors must be considered such as the route, concentration, duration, and the timing of exposure when determining toxicity to the organism. The need for adequate model systems (in vivo, in vitro, in silico and mathematical) is paramount for better understanding of chemical mixture toxicity. Currently, shortcomings plague each model system as investigators struggle to find the appropriate balance of rigor, repro

BotAF, a new Buthus occitanus tunetanus scorpion toxin, produces potent analgesia in rodents.

This work reports the purification of new potent scorpion neuropeptide, named BotAF, by an activity-guided screening approach. BotAF is a 64-residue long-chain peptide that shares very high similarity with the original β-like scorpion toxin group, in which several peptides have been characterized to be anti-nociceptive in rodents. BotAF administration to rodents does not produce any toxicity or motor impairment, including at high doses. In all models investigated, BotAF turned out to be an efficient peptide in abolishing acute and inflammatory (both somatic and visceral) pain in rodents. It pe

Chronic effects of atrazine exposure and recovery in freshwater benthic diatoms from two communities with different pollution histories.

Diffuse agricultural runoff into rivers can result in contamination with herbicides for prolonged periods of time. Chronic exposure to herbicides has the potential to alter toxic impacts in primary producers such as benthic diatoms. Determining how individual diatom taxa respond to herbicide exposure over varied exposure durations is essential for assessing herbicide impacts. This study investigated the responses of various benthic diatom taxa and effects at the community level over 12days of atrazine exposure. Diatom communities were collected from two sites with differing exposure histories;

Dermal exposure to weathered MC252 crude oil results in echocardiographically identifiable systolic myocardial dysfunction in double-crested cormorants (Phalacrocorax auritus).

During the Deepwater Horizon Natural Resource Damage Assessment, gross morphologic cardiac abnormalities, including softer, more distensible musculature, were noted upon gross necropsy in hearts from laughing gulls and double-crested cormorants exposed to weathered MC252 crude oil. A species specific, echocardiographic technique was developed for antemortem evaluation of function that was used to evaluate and better characterize cardiac dysfunction. Control (n=12) and treated (n=13) cormorant groups of similar sex-ratio and ages were dermally treated with approximately 13ml of water or weather
