Channel activities of skeletal muscle ryanodine receptor (RyR1) are activated by micromolar Ca2+ and inactivated by higher (∼1 mM) Ca2+ To gain insight into a mechanism underlying Ca2+-dependent inactivation of RyR1 and its relationship with skeletal muscle diseases, we constructed nine recombinant RyR1 mutants carrying malignant hyperthermia or centronuclear myopathy-associated mutations and determined RyR1 channel activities by [3H]ryanodine binding assay. These mutations are localized in or near the RyR1 domains which are responsible for Ca2+-dependent inactivation of RyR1. Four RyR1 mutati

T-type calcium channels (TTCCs), including the Cav3.1 and Cav3.2 subtypes, play a critical role in regulating cardiac electrical activity. Their abnormal expression is closely associated with arrhythmias and cardiac dysfunction caused by myocardial infarction (MI). Dronedarone, a multi-target antiarrhythmic drug, is widely used to treat atrial fibrillation and atrial flutter but has shown adverse effects under pathological conditions such as MI. This study aims to investigate the direct effects and underlying mechanisms of dronedarone on TTCCs. Using neonatal rat cardiomyocytes, HEK293T cells …

Disrupted calcium (Ca2+) homeostasis is a hallmark of neurodegenerative diseases, yet the mechanisms driving excessive Ca2+ entry at the neuronal plasma membrane remain poorly understood. Here, we show that α-synuclein pre-formed fibrils trigger a reorganization of voltage-gated calcium (CaV) channels in cultured mouse cortical neurons, increasing their clustering at the soma and dendrites. We find that α-synuclein fibrils promote cyclin-dependent kinase 5-mediated phosphorylation of KV2.1 at serine 603, enhancing its scaffolding capacity for CaV channels. Disrupting CaV-KV2.1 coupling with a

Amyotrophic lateral sclerosis (ALS) is a late-onset, fatal neurodegenerative disease affecting upper and lower motor neurons in the central nervous system. Drugs like riluzole, edaravone, and tofersen treat disease symptoms or are designed for a specific pathological mutation (e.g., SOD1), but they cannot prevent or halt the disease. For this reason, the search for new therapeutic strategies continues. The voltage-gated calcium channel CaV2.2 might be a novel target in ALS treatment as the channel was shown to be overexpressed in murine SOD1*G93A cortical neurons, resulting in higher mortality

Despite its prevalence, bladder cancer (BC) remains an unsolved pathogenesis. It is believed that TRPC6 channels have unique electrophysiological properties that contribute to intracellular Ca2+ signalling and tumorigenesis in cells. However, the mechanism by which TRPC6 contributes to BC progression and intracellular Ca2+ homeostasis remains unclear. In this study, TRPC6 expressions in paired BC and adjacent normal tissues were measured by immunohistochemistry. A KEGG pathway enrichment analysis was conducted to determine TRPC6’s potential contribution to BC. Ca2+ imaging analysis was perfo

Autoimmune encephalitis is an immune-mediated inflammatory disorder of the brain with diverse neurological and psychiatric manifestations. Voltage-gated calcium channel (VGCC) antibody-associated autoimmune encephalitis is rare and has been predominantly described in paraneoplastic settings. Emerging reports suggest a broader range of central nervous system involvement, but presentations dominated by psychiatric symptoms remain uncommon and diagnostically challenging. A 39-year-old man with no prior medical illness presented with subacute progressive cognitive decline, behavioral changes, and

The chloride channel regulator, calcium-activated-1, CLCA1, acts as a multifunctional secreted glycoprotein in anion channel modulation, mucus homeostasis, immune regulation, and other functions. So far, it has been described in goblet and other mucus-producing cells in mucous membranes, primarily of the respiratory, alimentary, and urogenital tracts. Here, we identify the expression of CLCA1 in fibroblast-like synoviocytes and superficial as well as, to a lesser extent, intermediate zone chondrocytes of diarthrodial articular joints. The expression pattern was found to be conserved in major j

The 1,4-dihydropyridines, drugs with well-established bioavailability and toxicity profiles, have proven efficacy in treating human hypertension, peripheral vascular disorders, and coronary artery disease. Every 1,4-dihydropyridine in clinical use blocks L-type voltage-gated calcium channels. We now report our development, using selective optimization of a side activity (SOSA), of a class of 1,4-dihydropyridines that selectively and potently inhibit the intermediate-conductance calcium-activated K+ channel KCa3.1, a validated therapeutic target for diseases affecting many organ systems. One of…

BACKGROUND: Adalimumab, a TNF-α inhibitor, is widely used clinically. Recent studies suggest Adalimumab can improve muscle damage after ischemic stroke (IS), but its protective mechanisms remain unclear. This study investigates the effect of adalimumab on muscle structure post-IS and the role of calcium balance in muscle strength, while validating their synergistic effect.
METHODS: This study investigates the effects of adalimumab and GV-58 on muscle structure and function in male middle cerebral artery occlusion (MCAO) rat models using behavioural, imaging, pathological and WB experiments. I

Mammalian sperm are characterized as specialized cells, as their transcriptional and translational processes are largely inactive. Emerging researches indicate that Ca2+ serves as a crucial second messenger in the modulation of various sperm physiological processes, such as capacitation, hyperactivation, and the acrosome reaction. Specifically, sperm-specific calcium channels, including CatSper, voltage-gated calcium channels (VGCCs), store-operated calcium channels (SOCCs), and cyclic nucleotide-gated (CNG) channels, are implicated in the regulation of calcium signaling in mammalian sperm. Ca…

Acute pancreatitis is initiated within pancreatic exocrine cells and sustained by dysregulated systemic inflammatory responses mediated by neutrophils. Store-operated Ca2+ entry (SOCE) through ORAI1 channels in pancreatic acinar cells triggers acute pancreatitis, and ORAI1 inhibitors ameliorate experimental acute pancreatitis, but the role of ORAI1 in pancreatitis-associated acute lung injury has not been determined. Here, we showed mice with pancreas-specific deletion of Orai1 (Orai1ΔPdx1, ∼70% reduction in the expression of Orai1) are protected against pancreatic tissue damage and immune cel

BACKGROUND: L-type Ca2+ channel CaV1.2 is essential for cardiomyocyte excitation, contraction and gene transcription in the heart, and abnormal functions of cardiac CaV1.2 channels are presented in diabetic cardiomyopathy. However, the underlying mechanisms are largely unclear. The functions of CaV1.2 channels are subtly modulated by splicing factor-mediated alternative splicing (AS), but whether and how CaV1.2 channels are alternatively spliced in diabetic heart remains unknown.
METHODS: Diabetic rat models were established by using high-fat diet in combination with low dose streptozotocin.
