Puff adder venom shock: a model of increased vascular permeability.

The circulatory, respiratory, metabolic, lethal and tissue permeability effects of an i.v. infusion (30 min; 1.5, 2.0, 2.5 and 3.0 mg/kg) of puff adder (Bitis arietans) venom were studied in Sprague-Dawley rats (n = 35, 275-325 g). Venom (2.5 mg/kg) produced circulatory failure with arterial hypotension, hemodilution, hypoproteinemia, lactacidemia and marked hyperventilation by +4 hr. In a separate test (n = 20, 282-325 g) blood volume was measured at end venom (2.5 mg/kg) infusion (0 time) and at +3 hr with radioiodinated (125I) human serum albumin and 51Cr-labeled rat red blood cells. Venom

Continuous negative external chest pressure decreases transvascular lung water transport in sheep after Pseudomonas bacteremia.

We studied the effects of continuous negative external chest pressure (CNECP) produced by a cuirass appliance on lung water and protein transport in sheep with chronic lung lymphatic fistulas. We compared data obtained during periods of mechanical ventilation (base line) to period of CNECP, using identical ventilatory support. Three groups were studied: six sheep were studied before and after application of CNECP for 1 h (control) and again after induction of a pulmonary vascular permeability defect (PVPD) by infusing live Pseudomonas bacteria (group I); another six sheep were studied under co

Permeability of blood-ocular barriers of neonatal and adult cat to sodium fluorescein.

The permeability of the ocular blood vessels to sodium fluorescein (NaFl) was evaluated in neonatal and adult cats by fluorescence microscopy. The iris, ciliary body, and choroidal vessels were markedly permeable, whereas the mature and immature retinal vessels were impermeable. Since there is no apparent barrier to NaFl at the level of the iris vessels, the role of those vessels in aqueous formation is possibly significant. The fact that the immature retinal vessels are impermeable suggests that abnormal permeability to NaFl in retinal neovascularization is a consequence of pathology rather t

Release of inflammatory mediators into the gastric wall of rats with acute acetic acid induced ulcer.

An acute, acetic acid induced gastric ulcer in rats is associated with increased capillary permeability and albumin leakage into the gastric wall surrounding the ulcer. This is an inflammatory reaction, and the present experiment was undertaken in an attempt to identify inflammatory mediators in the gastric wall around the ulcer. Rats were pretreated with inflammatory antagonists, and the protein leakage was estimated by means of 125I-labelled albumin 40--60 min and 4 h after ulcer induction. We found no effect on the protein leakage of mepyramine (antihistamine) deseril (antiserotonin) or ind…

Dietary Compound Chrysin Inhibits Retinal Neovascularization with Abnormal Capillaries in db/db Mice.

Diabetic retinopathy (DR) develops in a significant proportion of patients with chronic diabetes, characterized by retinal macular edema and abnormal retinal vessel outgrowth leading to vision loss. Chrysin, a naturally-occurring flavonoid found in herb and honeycomb, has anti-inflammatory, antioxidant, and anti-cancer properties. This study sought to determine the protective effects of chrysin on retinal neovascularization with abnormal vessels and blood-retinal barrier (BRB) breakdown in 33 mM glucose-exposed human retinal endothelial cells and in db/db mouse eyes. High glucose caused retina

Effects of Incretin-Based Therapies on Neuro-Cardiovascular Dynamic Changes Induced by High Fat Diet in Rats.

BACKGROUND AND AIMS: Obesity promotes cardiac and cerebral microcirculatory dysfunction that could be improved by incretin-based therapies. However, the effects of this class of compounds on neuro-cardiovascular system damage induced by high fat diet remain unclear. The aim of this study was to investigate the effects of incretin-based therapies on neuro-cardiovascular dysfunction induced by high fat diet in Wistar rats. METHODS AND RESULTS: We have evaluated fasting glucose levels and insulin resistance, heart rate variability quantified on time and frequency domains, cerebral microcirculati

Preparation and application of hydrophobic hybrid monolithic columns containing polyhedral oligomeric silsesquioxanes for capillary electrochromatography.

Hydrophobic organic-inorganic hybrid monolithic columns were synthesized via thermally initiated free radical polymerization with the confines of 75 μm id capillary using a polyhedral oligomeric silsesquioxane (POSS) reagent containing eight or more methacrylate groups as the crosslinker. Three organic functional monomers, butyl methacrylate (BuMA), lauryl methacrylate (LMA) and methacrylic acid (MAA), were selected and copolymerized with the POSS in the presence of 1-propanol and 1,4-butanediol to prepare the poly(POSS-co-BuMA), poly(POSS-co-LMA), and poly(POSS-co-MAA) monoliths, respectively

Mechanisms and consequences of cell activation in the microcirculation.

Cells undergo activation in response to a wide range of stimuli. In vascular cells (leukocytes, endothelial cells, and platelets), the different forms of activation include degranulation, oxygen free radical formation, expression of membrane adhesion proteins, and biophysical changes such as pseudopod formation and increased cytoplasmic viscosity. Cell activation and low flow are common features of many cardiovascular diseases. There is evidence that plasma from patients contains an activating factor for neutrophils as well as other vascular cells. Activated neutrophils have the ability to imp

Thallium-201-SPECT and 99Tc-HM-PAO SPECT imaging to study functionally cerebral supratentorial neoplasms: the biological basis of the functional imaging interpretation.

Nineteen patients with histologically diagnosed supratentorial cerebral neoplasms carried out CT and/or MRI, 201T1 SPECT and 99mTc-HM-PAO studies preoperatively. 99mTc-HM-PAO SPECT images revealed information about both tumoral perfusion and intracellular concentration of mediators converting 99mTc-HM-PAO to hydrophilic derivates (glutathione and other yet unknown factors) within viable tumoral cells while 201T1 SPECT images about permeability, extension of tumoral capillary network and viable tumoral cells presence. Basing on the different mechanisms of the tracer uptake, cerebral supratentor

Albumin 20% in surgical and critically ill patients; a comprehensive review.

BACKGROUND: Hyper-oncotic (20%) albumin has long been used during surgery and intensive care, but its pharmacology and relevant clinical use have not been adequately summarized. MAIN BODY: Hyper-oncotic albumin expands the plasma volume by twice the infused volume due to colloid pressure-induced recuitment of interstitial fluid, which primarily occurs via the lymphatic route. The interstitium and also the whole body is dehydrated, which helps maintaining the intravascular space well filled also at low arterial pressures. Albumin reduces the capillary permeability by volume exclusion and by tra…

A Secreted Glutamate-Specific Serine Protease from Staphylococcus aureus Activates NF-κB Signaling and Enhances Vascular Leakage.

Staphylococcus aureus secretes multiple proteases that contribute to tissue damage and modulation of host immune responses. The glutamate-specific serine endopeptidase (VSPase) is secreted by S. aureus strain C-66; however, its effects on inflammatory signaling and vascular permeability remain poorly understood. In this study, we investigated the pro-inflammatory and vascular permeability-enhancing activities of VSPase and compared them with those of its catalytically inactive S237L mutant. In RAW 264.7 macrophages, VSPase induced the mRNA expression of TNF-α, IL-1β, IL-12β, MIP-2, cyclooxygen

Insights into the Cellular and Molecular Mechanisms behind the Antifibrotic Effects of Nerandomilast.

The quest for innovative pharmacologic interventions in idiopathic pulmonary fibrosis (IPF) is a challenging journey. The complexity of the disease demands a comprehensive approach that targets multiple cell types and pathways. This study examined the antifibrotic properties of nerandomilast, a preferential phosphodiesterase 4B inhibitor, focusing on its effects on myofibroblasts (MFs) and endothelial cells. Using cytokine-stimulated human IPF lung fibroblasts and RNA sequencing, we assessed the effects of nerandomilast on MF contractility, MF markers, and differentiation mechanisms. In additi

Blood nerve barrier permeability enables nerve targeting of circulating nanoparticles in experimental autoimmune neuritis.

Guillain-Barré syndrome (GBS) is a devastating autoimmune disease of the peripheral nervous system (PNS) with limited treatment options. Several studies have shown attenuation of the well-characterized GBS preclinical experimental autoimmune neuritis (EAN) model with systemically administered therapeutic compounds via anti-inflammatory or immunomodulatory mechanisms. Despite this, clinical advancement of these findings is limited by dosing that is not translatable to humans or is associated with off-target and toxic effects. This is due, in part, to the blood-nerve barrier (BNB), which restric

Orthoflavivirus omskense NS1
