A Common Eurasian Fish Tapeworm, Caryophyllaeides fennica (Cestoda), in Western North America: Further Evidence of 'Amphi-Pacific' Vicariance in Freshwater Fish Parasites.

The freshwater fish faunas of the Palaearctic and Nearctic regions show similarities but also notable differences, resulting in diverse distributions of their parasites. Relatively few parasite species occur in both regions and fewer still have been examined using molecular data. We report a rare example of 'amphi-Pacific' distribution, involving the common Palaearctic parasite, the caryophyllidean cestode Caryophyllaeides fennica (Schneider, 1902), in the chiselmouth Acrocheilus alutaceus Agassiz and Pickering, 1855, an endemic cyprinid in northwestern Nearctic, Oregon. Circulation of Dirofilaria repens and Dirofilaria immitis in Moldova.

BACKGROUND: Over the last two decades, a significant spread of dirofilariasis has been observed in eastern and central Europe. However, data on the circulation of Dirofilaria spp. in Moldova were absent although direct neighbor states reported high incidence rates of human dirofilariasis. METHODS: Daily mean temperature data were used to calculate Dirofilaria spp. development units, which were used to estimate the potential for complete extrinsic development in the mosquitoes (= sum of potential Dirofilaria spp. transmission days). In addition, 4,481 adult female mosquitoes were collected fro

Sequence variability in four mitochondrial genes among rabbit pinworm (Passalurus ambiguus) isolates from different localities in China.

Passalurus ambiguus is a common pinworm which parasitizes in the caecum and colon of rabbits. This study examined genetic variability among P. ambiguus isolated from naturally infected rabbits in four different provinces in China. The partial mitochondrial (mt) cytochrome c oxidase subunit 1 (pcox1), cytochrome b (pcytb) and NADH dehydrogenase subunits 1 and 5 (pnad1 and pnad5) were amplified separately from individual nematodes by PCR and sequenced. The results showed that pcox1, pcytb, pnad1 and pnad5 were 714, 663, 645 and 546 bp in length, respectively. The intra-specific sequence variatio

A case of Fasciola hepatica infection mimicking cholangiocarcinoma and ITS-1 sequencing of the worm.

Fascioliasis is a zoonotic infection caused by Fasciola hepatica or Fasciola gigantica. We report an 87-year-old Korean male patient with postprandial abdominal pain and discomfort due to F. hepatica infection who was diagnosed and managed by endoscopic retrograde cholangiopancreatography (ERCP) with extraction of 2 worms. At his first visit to the hospital, a gallbladder stone was suspected. CT and magnetic retrograde cholangiopancreatography (MRCP) showed an intraductal mass in the common bile duct (CBD) without proximal duct dilatation. Based on radiological findings, the presumed diagnosis

Molecular characterization of Taenia multiceps isolates from Gansu Province, China by sequencing of mitochondrial cytochrome C oxidase subunit 1.

A total of 16 Taenia multiceps isolates collected from naturally infected sheep or goats in Gansu Province, China were characterized by sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. The complete cox1 gene was amplified for individual T. multiceps isolates by PCR, ligated to pMD18T vector, and sequenced. Sequence analysis indicated that out of 16 T. multiceps isolates 10 unique cox1 gene sequences of 1,623 bp were obtained with sequence variation of 0.12-0.68%. The results showed that the cox1 gene sequences were highly conserved among the examined T. multiceps isolates

Anisakis species (Nematoda: Anisakidae) of Dwarf Sperm Whale Kogia sima (Owen, 1866) stranded off the Pacific coast of southern Philippine archipelago.

Anisakid nematodes in the Pacific region of the Philippine archipelago still remain unexplored. This study was carried out to identify anisakid species from one of their final hosts, the Kogiid whale (Dwarf Sperm Whale, Kogia sima) stranded off the southern part (Davao Gulf) of the Philippine archipelago. Anisakid worms were initially identified morphologically using light and scanning electron microscopy, whereas identification to species level was carried out molecularly using PCR-RFLP and sequencing of the ITS (ITS1-5.8s rRNA-ITS2) and mtDNA cox2 regions. Parasitological study revealed new

Genetic heterogeneity of Fasciola hepatica isolates in the northwest of Spain.

The liver fluke Fasciola hepatica is a worldwide distributed trematode parasite of great veterinary importance. In Spain, the prevalence of the infection is up to 56% in sheep. Its control is mainly based on the use of anthelmitics, although some resistant populations have been described. Genetic studies of the genus Fasciola have been focused on the 28S ribosomal deoxyribonucleic acid (rDNA) gene, with no intraspecific variations and a few interspecific nucleotide differences. We have designed a specific polymerase chain reaction to amplify and sequence a 510-bp-long fragment of the gene to c

Occurrence of brachylaimid trematodes, alive and shell-trapped, in sympatric species of terrestrial gastropods in France.

Following the detection of Brachylaima spp., which are potentially pathogenic for domestic animals and humans, in Cornu aspersum and Cepaea nemoralis at a rural site in France, we investigated trematode distribution among sympatric gastropod species to assess their role in the parasite life cycle. Eight species were investigated (Cepaea nemoralis, Cernuella neglecta, Clausilia bidentata, Cochlicella acuta, Cornu aspersum, Lauria cylindracea, Pomatias elegans, and Theba pisana). Four species were parasitised by trematodes: C. aspersum (prevalence 56%), C. nemoralis (10%), and T. pisana (2%) as

Faecal DNA detection and molecular identification of nematodes of veterinary relevance in invasive raccoons and raccoon dogs in Central Europe.

Invasive carnivores such as the raccoon (Procyon lotor) and raccoon dog (Nyctereutes procyonoides) are widespread in Europe and may contribute to the circulation of nematodes of veterinary and zoonotic relevance at the wildlife-domestic animal interface. Molecular data on nematode taxa infecting these hosts in Central Europe remain scarce, particularly those derived from faecal DNA. We analysed 212 post-mortem faecal samples collected in western Poland (97 raccoons, 115 raccoon dogs) using PCR-based screening followed by mitochondrial cytochrome c oxidase subunit 1 (cox1) sequencing for molecu

On misidentification of metacercariae of Posthodiplostomum minimum (MacCallum, 1921) Dubois, 1936, in Nearctic minnows.

Posthodiplostomum minimum (MacCallum, 1921) Dubois, 1936 (Digenea, Diplostomidae) was described from adult worms from a great blue heron (Ardea herodias; Ardeidae) in New York. Metacercariae have been widely reported from diverse freshwater fishes, despite early experimental work and recent molecular data indicating exclusive infection of minnows (Leuciscidae) in North America. We compared the type specimens of P. minimum to adults taken from domestic chicks (Gallus gallus domesticus) fed metacercariae from fathead minnow (Pimephales promelas; Leuciscidae), as well as adults in naturally infec

Optimisation and analytical assessment of a TaqMan™ probe-based real-time PCR assay designed to diagnose infection with Schistosoma japonicum.

BACKGROUND: Intestinal schistosomiasis, caused by infection with Schistosoma japonicum, remains a zoonotic neglected tropical disease (NTD) of significant public health importance in parts of China, the Philippines, and Indonesia. Ongoing schistosomiasis control programmes in these areas have achieved significant reductions in disease prevalence, transmission, and morbidity associated with infection; however, they have also presented new challenges, particularly in detecting and monitoring low-intensity and low-prevalence S. japonicum infections. Highly sensitive and highly specific diagnostic

Frequency, Geospatial Distribution, and Subspecies of Echinococcus granulosus Identified in Dog Stool in Urban Public Spaces of an Endemic Area in Cusco, Peru.

The impact of environmental contamination with Echinococcus granulosus eggs on the transmission to humans remains unknown. The control of free-roaming and stray dogs is a public health problem in urban and periurban areas of the Cusco region, such as the city of Sicuani in the Canchis province. Nearly one in four human cystic echinococcosis cases referred to tertiary hospitals in the Cusco region of Peru is referred from the Sicuani district. This study sought to determine the frequency and spatial distribution of E. granulosus DNA in dog stool collected within urban public spaces of Sicuani.

Accurate species discrimination of Fasciola hepatica from Türkiye based on the fatty acid binding protein type I (FABP type I) gene and phylogenetic analysis using mitochondrial DNA.

Accurate species discrimination based on a robust nuclear protein-coding gene marker is essential for Fasciola spp. because of the presence of F. hepatica, F. gigantica, and hybrid Fasciola flukes that originate from the interspecific hybridization between the two species in Asia. Molecular identification of Fasciola species uses multiplex polymerase chain reaction (PCR) targeting phosphoenolpyruvate carboxykinase (pepck) and PCR-restriction fragment length polymorphism (RFLP) targeting DNA polymerase delta (pold). However, these methods have demonstrated limitations, including misidentificati

Ambient temperature storage in DESS supports molecular studies of benzimidazole resistance from canine hookworm eggs.

Emerging benzimidazole resistance in canine hookworms poses a growing concern for veterinary and public health. Molecular diagnostics targeting β-tubulin gene mutations are essential for resistance surveillance but traditionally rely on refrigerated faecal samples. This study evaluates dimethyl sulfoxide, EDTA, and saturated NaCl (DESS) as a room-temperature preservation medium for canine faeces. Using ITS-2 rDNA and isotype-1 β-tubulin gene (tubb-1) amplicon metabarcoding, we compared DNA integrity and diagnostic performance of DESS-preserved samples (4, 28, and 106 days) to refrigerated cont

First molecular characterization and phylogenetic analysis of Echinococcus granulosus (sensu stricto) genotype 1 in human in Saudi Arabia.

Echinococcosis is a neglected zoonotic parasitic disease caused by Echinococcus granulosus. Currently, little information is available on the genetic figure of Echinococcus species among human in Saudi Arabia. In the present study, 60 formalin-fixed paraffin-embedded tissue (FFPT) hydatid cysts that were confirmed and collected from Taif hospitals between 2019 and 2023. Deparaffinization was followed by DNA extraction. PCR protocol was used to amplify the mitochondrial 12S rRNA gene. The amplified PCR products were sequenced, and subsequent phylogenetic tree was constructed and analyzed. Fifty

Taenia saginata Infection in a 25-Year-Old Immigrant in Malaysia: A Case Study.

Taeniasis is a parasitic infection in humans by tapeworm species typically acquired through the ingestion of undercooked pork or beef leading to infections with Taenia solium, Taenia asiatica or Taenia saginata. We reported a case of taeniasis in a young man of Chinese nationality who presented with recurrent episodes of expelling proglottids. He was at risk due to his preference for consuming undercooked beef. He remained asymptomatic and his physical examination was unremarkable. A 330 cm tapeworm was recovered; each mature proglottid contained 12-30 primary lateral uterine branches. Stool e

A RAPID DIAGNOSTIC PCR ASSAY FOR THE DETECTION OF SCHISTOSOMA MANSONI IN THEIR SNAIL VECTORS.

Accurate detection of schistosome infections in snails is vital for epidemiologic and laboratory studies. Traditional microscopy methods to detect schistosomes in snails are hindered by long prepatent periods and snail survivorship, leading to inaccurate assessment of infections. A rapid, multiplexed PCR assay targeting Biomphalaria sudanica or Biomphalaria glabrata (internal control) and Schistosoma mansoni DNA is described. The method takes less than 90 min starting from extracted snail DNA and is successful at amplifying schistosome DNA in snail tissue as soon as 30 min following exposure.

Metagonimus yokogawai Ancient DNA Recovered from 16th- to 17th-Century Korean Mummy Feces of the Joseon Dynasty.

Metagonimiasis is foodborne intestinal parasitism occurring by the definitive hosts' ingestion of raw or undercooked fish, mostly commonly sweetfish. Most Metagonimus infection is caused by Metagonimus yokogawai but also rarely by Metagonimus takahashii as well as Metagonimus miyatai. Despite recent molecular work on Metagonimus spp., there are still insufficient data to reveal the genetic characteristics of ancient M. yokogawai in a wide geo-historical scope. In this study, we were successful in the analysis of M. yokogawai ancient DNA (aDNA) using coprolite samples retrieved from 16th- to 17
