Blastic Plasmacytoid Dendritic Cell Neoplasm With De Novo TET2 and KDM6A Mutations.

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare, highly aggressive hematologic malignancy characterized by cutaneous involvement with potential dissemination to peripheral blood, bone marrow, lymph nodes, or extranodal sites. We present the first documented case of BPDCN exhibiting complex epigenetic dysregulation in a 54-year-old female who developed rapidly progressive purpuric lesions on the trunk, diagnosed via skin biopsy and immunohistochemistry. Cytogenetic analysis revealed triple molecular aberrations including a t(1;6) (p31;q25) translocation, a TET2 frameshift deletio

Electroacupuncture alleviates obesity-associated inflammatory responses by inhibiting dendritic cell migration.

BACKGROUND: Obesity is closely associated with chronic low-grade inflammation, particularly within visceral adipose tissues (VAT). Dendritic cells (DCs), as key antigen-presenting cells, bridge innate and adaptive immunity and play a pivotal role in regulating adipose tissue immune homeostasis. This study aims to investigate the effects and underlying mechanisms of electroacupuncture (EA) on obesity-associated inflammation, with a particular focus on DCs migration. METHODS: A high-fat diet (HFD) was used to induce obesity in C57BL/6 mice. Electroacupuncture (EA) was administered to obese mice

Ultrastructural Evidence of Altered Dendritic Morphology in the Prefrontal Cortex of Alzheimer's Model Rats.

Alzheimer's disease (AD) is a neurodegenerative disorder with synaptic pathology as a core theme in aging-related research. Conventional imaging and limited tools fail to resolve AD-relevant nanoscale structures. Here, we present an automated pipeline integrating volume electron microscopy (vEM) with deep learning to analyze synapses and subcellular components, enabling 3D reconstruction of neuronal architecture in ~130,000 μm3 of rat prefrontal cortex (PFC). Preliminary results from small animal samples show that, vs. controls, AD rats have fewer mushroom spines but more thin/stubby spines. S

Actomyosin rings constrain CD40 mobility to organize the dendritic cell immunological synapse.

BACKGROUND: The interaction between dendritic cell CD40 and T cell CD40L is critical for dendritic cell licensing and the initiation of adaptive immunity. The spatial organization of CD40 on the dendritic cell surface is thought to dictate its signaling output, yet the underlying mechanisms are poorly understood. METHODS: Fluorescently tagged BMDCs were generated via lentiviral transduction of CD40-mGFP and electroporation of LifeAct-eGFP constructs. BMDCs, derived from C57BL/6 J mice and matured with LPS, were co-cultured with naïve OT-II CD4⁺ T cells or functionalized beads to model immunol

STING Activation in Dendritic Cells Enhances Antigen Presentation by Live Tumor Cells.

The stimulator of interferon genes (STING) pathway is a key regulator of antitumor immunity. Activation of STING by the natural agonist 2'3'-cyclic GMP-AMP (cGAMP) induces type I interferon and promotes CD8+ T cell responses against tumors. However, how type I interferon modulates tumor cell recognition by T cells remains unclear. Here, using a co-culture model and virus-like particles encapsulating cGAMP (cGAMP-VLP), we show that STING activation in dendritic cells enhances CD8+ T cell recognition of live tumor cells through type I interferon signaling. This effect is independent of enhanced

Mechanical licensing of functional dendritic cell states for enhanced T cell priming.

The plasticity of dendritic cell (DC) functional state is a major hurdle in DC therapy, yet how DCs acquire distinct states independent of ontogeny remains poorly understood. Here, we demonstrate that changes in matrix stress relaxation mechanically educate DCs to adopt distinct, persistent functional states even after the removal of mechanical cues. Stem cell-derived DCs cultured in a fast-relaxing environment exhibited enhanced antigen presentation, faster migration, and higher expression of T cell-recruiting chemokines. Slow-relaxing DCs, biased towards pro-inflammatory cytokine secretion,

Sleep deprivation alters hippocampal dendritic spines in a contextual fear memory engram.

Sleep is critically involved in strengthening memories. However, our understanding of the morphological changes underlying this process is still emerging. Recent studies suggest that specific subsets of dendritic spines are strengthened during sleep in specific neurons involved in recent learning. Contextual memories associated with traumatic experiences are involved in post-traumatic stress disorder (PTSD) and represent recent learning that may be strengthened during sleep. We tested the hypothesis that dendritic spines encoding contextual fear memories are selectively impacted by sleep depri

Crosslinked ionizable lipids reprogram dendritic cell metabolism for potent mRNA vaccination.

Modulating metabolism in immune cells is an effective approach to induce desired immune responses. Here we develop a lipid nanoparticle (LNP) capable of metabolic reprogramming of dendritic cells for mRNA vaccine applications. Using imidoester-based conjugation chemistry, we design a crosslinked ionizable lipid, C12-2aN, which possesses intrinsic metabolic modulatory properties. This multifunctional ionizable lipid not only promotes effective mRNA expression by facilitating endosomal escape but also stimulates glycolysis through mTORC2 pathway activation. As both an mRNA carrier and a metaboli

Dendritic cells in cancer immunotherapy: functional barriers, reprogramming strategies and translational challenges.

Cancer immunotherapy depends on effective antigen presentation and T cell activation. Antigen-presenting cells (APCs), especially dendritic cells (DCs), play a central role in this process by capturing tumor antigens, processing them, and presenting antigenic peptides to T cells through major histocompatibility complex molecules. However, APC function is often impaired within the tumor microenvironment. Reduced antigen presentation, weak co-stimulatory signaling, suppressive cytokines, metabolic stress, and inhibition by myeloid-derived suppressor cells and regulatory T cells all limit effecti

25 Years of Cancer Immunoediting: Dendritic Cells and Macrophages Filled the Missing Gap.

In 1909, Paul Ehrlich first suggested that the immune system is critical for suppressing carcinogenesis and cancer growth, which led to the introduction of cancer immunosurveillance by Sir MacFarlane Burnet in 1960. Burnet's cancer immunosurveillance was further supported and elaborated by Thomas Lewis in 1982, who noted, for example, the limitations of investigating cancer immunosurveillance in available experimental models, except for virus-induced cancers. The concept of cancer immunosurveillance in 2001 was revised to cancer immunoediting (the immunoediting term was introduced by Dr. Rober

Peripheral immune-inducer dendritic cells drive early-life allergic inflammation.

Atopic diseases associated with allergens, as well as allergic diseases, frequently arise early in life; however, the age-dependent mechanisms governing immune responses to allergens remain poorly understood1. Here we find that in early life, exposure to common allergens triggers a distinct bifurcated immune response, simultaneously triggering type 17 inflammation in the skin and initiating canonical T helper 2 sensitization in the lymph nodes. This early-life γδ type 17-mediated dermatitis primes the exaggerated allergic lung inflammation upon secondary allergen exposure. Mechanistically, we

Concurrent follicular lymphoma and follicular dendritic cell sarcoma: composite neoplasm or transdifferentiation?

Follicular dendritic cell sarcoma (FDCS) is a rare neoplasm, and its synchronous occurrence with follicular lymphoma (FL) has not been reported. Whether composite tumors arise independently or through clonal evolution remains unclear. A 52-year-old woman presented with night sweats, weight loss, and an obstructing hepatic flexure colonic mass with mesenteric disease. Biopsy showed two tumor populations: an atypical B cell infiltrate positive for CD20, CD10, focal BCL6, and weak/focal MUM1, with kappa predominance and Ki-67 of 20-50%, consistent with follicular large B cell lymphoma/FL grade 3B

Immunostimulant-induced maturation of monocyte-derived dendritic cells in respiratory infection prophylaxis.

BACKGROUND: Immunostimulants were established many years ago to improve the capacity of the immune system to react to exogenous noxae. Immunostimulants can be synthesized in the lab or extracted from bacterial cultures. For example, bacterial lysates have been shown to have a significant effect on the number of infectious episodes, duration of fever and use of antibiotics in patients affected by recurrent respiratory tract infections. The capacity of bacterial lysates to induce the maturation of human monocyte-derived dendritic cells (DCs) has been indicated as the first and fundamental effect

Dendritic Cell-Derived Extracellular Vesicles Mediate Inflammation in Egg Allergy Patients.

Atopic allergy is rising globally and placing a significant strain on healthcare systems, yet the understanding of the underpinning mechanisms of allergic sensitization remains incomplete. Extracellular vesicles (EVs) have recently emerged as important mediators of immune modulation, due to their diverse cargo, and therefore may play a mechanistic role in allergic sensitization development. Thus, this study investigated whether EVs released by activated dendritic cells (DCs) contribute to allergic sensitization of the common egg allergen, ovalbumin (OVA). DCs were generated from human monocyte

Generation of Regulatory Dendritic Cells From Living Donor Liver Transplant Perfusates.

BACKGROUND: Safety and immunological activity of donor blood monocyte-derived regulatory dendritic cells (DCreg) generated ex vivo have been demonstrated recently in living donor liver transplantation. A potential alternative/additional source of DCreg precursors is the allograft perfusion fluid. Our goal was to establish the extent to which DCreg could be generated from CD14 + myeloid precursors in living donor liver perfusate (LD-LP) and to assess the phenotype and immunoregulatory properties of the perfusate-derived DCreg (LP-DCreg). METHODS: Presumptive DCreg were generated from CD14 + pr

Effects of repetitive scanning ultrasound on the intraneuronal dendritic signalling of CA1 pyramidal neurons.

INTRODUCTION: Neural development is regulated by several spatiotemporally changing factors, which are essential for enabling neurons to develop functional networks, during early developmental stages. Additionally, external physical stimuli, like scanning prenatal ultrasound (US) examination, may influence neural development. Aim of our study is to examine potential consequences of diagnostic level ultrasound exposure during the early phase of hippocampal CA1 neural network development by using computational models. METHODS: Using in silico modelling, this study explores and compares multiple

Tailoring Zn2+ Solvation for Dendrite-Free and High-Efficiency Aqueous Zinc Batteries.

Aqueous zinc-metal batteries (ZMBs) offer high capacity, intrinsic safety, and low cost but remain challenged by dendrite growth, hydrogen evolution, and interfacial instability. Here, we develop a solvation-engineered hybrid electrolyte comprising 3 M Zn(OTf)2 in water, methanol, and dimethyl sulfoxide (3:1:1, v/v/v). Methanol disrupts the hydrogen-bond network, while DMSO stabilizes Zn2+ coordination, collectively forming a unique solvation structure as confirmed by isothermal titration calorimetry, density functional theory, and Raman spectroscopy. This environment promotes preferential tri

Dendritic cell-driven immune rebalancing as a systemic regulator of tumor and neurodegenerative microenvironments.

Dendritic cells (DCs) critically regulate immune dynamics within the tumor microenvironment (TME), yet the mechanisms governing the balance between effective anti-tumor immunity and immune tolerance remain incompletely defined. Existing reviews have largely examined DC biology, tumor immunology and neuroinflammation within disease-specific frameworks, leaving insufficient attention to how DC dysfunction may be compared across chronic pathological microenvironments. Here, we propose DC-driven immune rebalancing as a context-dependent framework for understanding how antigen-presenting cell state

Gbx2 and Robo receptors control amacrine cell dendrite stratification in the mammalian retina.

Within the neuronal classes of the retina, amacrine cells (ACs) exhibit the greatest neuronal diversity in morphology and function. We show that the selective expression of the transcription factor Gbx2 is required for dendritic stratification of an individual AC subtype in the mouse retina. We identify Robo1 and Robo2 as downstream effectors that when deleted, phenocopy the dendritic misprojections seen in Gbx2 mutants. Slit1 and Slit2, the ligands of Robo receptors, are localized to the upper layers of the inner plexiform layer where we observe the dendritic misprojections in both Gbx2 and R
