Drinking motives and alcohol sensitivity mediate multidimensional genetic influences on alcohol use behaviors.

BACKGROUND: Genetic influences account for a substantial proportion of individual differences in alcohol use behaviors (AUBs). However, mult … relationship between PGSs and 22 potential mediators, including personality, alcohol expectancies, drinking motives, and alcohol sensitivity. Nominally significant effects were carried forward to a multiple mediation model to estimate direct and indirect effects on four measured AUBs (frequency, quantity, alcohol use disorder symptoms [AUDsx], and maximum …

Binge drinking and veteran status increase risk for suicide planning in U.S. adults.

OBJECTIVE: Suicide is a leading cause of preventable death. Problematic alcohol use and service in the U.S. military confer elevated risk for suicide, yet it is unknown whether their combination further heightens the risk. Extant literature is circumscribed to veterans enrolled in Veterans Affairs care, those with diagnosed alcohol use disorder, or suicide mortality, resulting in notable literature gaps on non-Veterans Affairs-enrolled veterans, nondiagnostic problematic drinking behaviors, and premortality suicide risk. METHOD: To address these gaps, this study included U.S. adults who parti…

Gray matter volume and its molecular profiles characteristic drinking severity in young adults.

BACKGROUND: Alcohol misuse is associated with decreased grey matter volume (GMV) and more rule-breaking behavior. It remains unclear how cortical structures, rule-breaking and alcohol misuse were inter-related. Here, we investigated how GMV are altered in association with the severity of drinking and clinical comorbidities and how men and women differ in this association. METHODS: We employed the Human Connectome Project data and included a total of 1053 subjects (484 men) in data analyses. The severity of alcohol use was quantified for individual subjects with the first principal component (…

Health Trade-Offs of Boiling Drinking Water with Solid Fuels: A Modeling Study.

BACKGROUND: Billions of the world's poorest households are faced with the lack of access to both safe drinking water and clean cooking. One solution to microbiologically contaminated water is boiling, often promoted without acknowledging the additional risks incurred from indoor air degradation from using solid fuels. OBJECTIVES: This modeling study explores the trade-off of increased air pollution from boiling drinking water under multiple contamination and fuel use scenarios typical of low-income settings. METHODS: We calculated the total change in disability-adjusted life years (DALYs) fr…

Alcohol's acute effects on emotion recognition and empathy in heavy-drinking young adults.

Alcohol intoxication can lead to both positive and negative social outcomes, and recent research suggests that alcohol-induced changes in emotion recognition and empathy may underlie these consequences. However, methodological limitations of prior studies make drawing firm conclusions difficult. This study addresses some of these limitations by investigating the acute effects of alcohol on emotion recognition and empathy in a large sample of young adults, using an intoxicating alcohol dose and more ecologically valid tasks. Participants (N = 156, mean age = 22.51 ± 2.16, 31.41% female, 72.44%

PEth Cut-Off Thresholds for Hazardous Alcohol Consumption Based on a Drinking Study.

This study aimed to explore how PEth and other commonly used alcohol biomarkers (CDT, AST, ALT, and GGT) respond to regular consumption of what has been generally considered to correspond to low to moderate amounts of alcohol over a 2-week period. A total of 21 voluntary participants (aged 31-69 years) took part in a 2-week drinking study. Group 1 (n = 11) consumed one glass of wine daily (16 g of alcohol), close to the present Swedish limit for hazardous alcohol consumption, while Group 2 (n = 10) consumed two glasses daily (32 g of alcohol). Alcohol biomarkers were measured at baseline and a…

Efficient removal of drugs of abuse from drinking water using metal-organic frameworks.

The presence of residues of drugs of abuse in potable water is a growing concern worldwide. Different studies have detected traces of opioids, cocaine, amphetamines, or cannabinoids in surface water, groundwater, and even treated drinking water, which is a clear indicator of insufficient removal during wastewater treatment processes. These substances may persist in the environment, posing potential long-term risks to human health and ecosystems, and consequently, making the quest for efficient decontamination technologies mandatory. Herein, we explore the use of a family of six eco-friendly wa…

Mu-opioid receptor knockout on Foxp2-expressing neurons reduces aversion-resistant alcohol drinking.

Mu-opioid receptors (MORs) in the amygdala and striatum are important in addictive and rewarding behaviors. The transcription factor Foxp2 is a genetic marker of intercalated (ITC) cells in the amygdala and a subset of striatal medium spiny neurons (MSNs), both of which express MORs in wild-type mice and are neuronal subpopulations of potential relevance to alcohol-drinking behaviors. For the current series of studies, we characterized the behavior of mice with genetic deletion of the MOR gene Oprm1 in Foxp2-expressing neurons (Foxp2-Cre/Oprm1fl/fl). Male and female Foxp2-Cre/Oprm1fl/fl mice w…

When the Going Gets Tough: Multimorbidity and Heavy and Binge Drinking Among Adults.

INTRODUCTION: Multimorbidity, the presence of two or more long-term health conditions in the same individual, is an emerging epidemic associated with increased morbidity and mortality. Continued drinking concurrent with alcohol-related chronic conditions, particularly with multimorbidity, is likely to further elevate health risk. This study aimed to examine the associations of multimorbidity among diabetes, hypertension, heart disease, and cancer with drinking, and moderation of these associations by age. METHODS: Logistic regression modeling was performed in 2023 using a nationally represent…

Adaptations in glutathione-based redox protein signaling pathways and alcohol drinking across species.

Alcohol use disorder (AUD) is the most prevalent substance use disorder but there is incomplete knowledge of the underlying molecular etiology. Here, we examined the cytosolic proteome from the nucleus accumbens core (NAcC) of ethanol drinking rhesus macaques to identify ethanol-sensitive signaling proteins. The targets were subsequently investigated using bioinformatics, genetic, and pharmacological manipulations in mouse models of ethanol drinking. Of the 1000+ cytosolic proteins identified in our screen, 50 proteins differed significantly between control and ethanol drinking macaques. Gene …

Do Earmarks Target Low-Income and Minority Communities? Evidence from US Drinking Water.

The quality and inequality of US drinking water investments have gained attention after recent environmental disasters in Flint, Michigan, and elsewhere. We compare the formula-based targeting of subsidized loans provided under the Safe Drinking Water Act with the targeting of congressional drinking water earmarks ("pork barrel" spending). Earmarks are often critiqued for potentially privileging wealthier and more politically connected communities. We find that earmarks target Black, Hispanic, and low-income communities, partly due to targeting water systems serving large populations. Earmark …

Sex differences in distributed error-related neural activation in problem-drinking young adults.

BACKGROUND: Detecting and responding to errors is central to goal-directed behavior and cognitive control and is thought to be supported by a network of structures that includes the anterior cingulate cortex and anterior insula. Sex differences in the maturational timing of cognitive control systems create differential periods of vulnerability for psychiatric conditions, such as substance use disorders. METHODS: We examined sex differences in error-related activation across an array of distributed brain regions during a Go/No-Go task in young adults with problem alcohol use (N=69; 34 females;

Peri-ictal Water Drinking in an Omani Patient with Bilateral Mesial Temporal Sclerosis.

Peri-ictal water drinking (PIWD) is a rare vegetative manifestation of temporal lobe epilepsy without a definite lateralisation value. We report a case of PIWD in a 22-year-old Omani male patient with post-concussion syndrome and epilepsy presented to a tertiary care hospital in Muscat, Oman, in 2021 for evaluation of paroxysmal events. His behaviour of PIWD was misinterpreted by his family until characterised in the epilepsy-monitoring unit as a manifestation of epilepsy that was treated medically. To the best of the authors' knowledge, this is the second reported case in the region.

Pulmonary Actinomycosis Induced by Total Gastrectomy and Heavy Alcohol Drinking: A Case Report.

Combined risk factors such as total gastrectomy, heavy alcohol consumption, smoking, and poor oral hygiene may contribute to the development of pulmonary actinomycosis. Here, we present a rare case of pulmonary actinomycosis triggered by total gastrectomy and heavy alcohol consumption. The patient presented with hemoptysis and a suspected lung mass. With a history of total gastrectomy and heavy alcohol consumption, the patient underwent imaging studies that initially suggested lung cancer. While imaging studies initially suggested lung cancer, multiple bronchoscopic examinations ultimately led

Oropharyngeal tularemia outbreak linked to drinking contaminated tap water in North-Western Iran.

Tularemia is a widespread bacterial disease caused by Francisella tularensis. Iran is an endemic country for this zoonosis. In this report, we present a 2020 tularemia outbreak in a village in northwestern Iran involving 15 patients with the oropharyngeal form of the disease. This outbreak was probably linked to the consumption of contaminated drinking water.

Somatostatin signaling modulates binge drinking behavior via the central nucleus of the amygdala.

Somatostatin (SST) is a neuropeptide widely expressed in the central nervous system with dense expression in limbic regions such as the exte … umption associated with health complications and the transition into alcohol dependence. We use the Drinking in the Dark (DID) model of binge intake in C57BL/6J male and female mice to examine: 1) the impact of 3 DID cycles on CeA SST expression; 2) the effect of intra-CeA SST injection on binge-like ethanol consumption; and 3) if the SST receptor 2 or 4 ( …

Non-consummatory behavior signals predict aversion-resistant alcohol drinking in head-fixed mice.

A key facet of alcohol use disorder is continuing to drink alcohol despite negative consequences (so called "aversion-resistant drinking"). In this study, we sought to assess the degree to which head-fixed mice exhibit aversion-resistant drinking and to leverage behavioral analysis techniques available in head-fixture to relate non-consummatory behaviors to aversion-resistant drinking. We assessed aversion-resistant drinking in head-fixed female and male C57BL/6 J mice. We adulterated 20% (v/v) alcohol with varying concentrations of the bitter tastant quinine to measure the degree to which mic…
