Too much of a good thing? Elevated baseline sleep spindles predict poor avoidance performance in rats.

Sleep spindles may be involved in synaptic plasticity. Learning-dependent increases in spindles have been observed in both humans and rats. In humans, the innate (i.e., baseline) number of spindles correlate with measures of academic potential such as Intelligence Quotient (IQ) tests. The present study investigated if spindles predict whether rats are able to learn to make avoidance responses in the two-way shuttle task. Baseline recordings were taken continuously for 24h prior to training on the two-way shuttle task for 50trials/day for two days followed by a 25 trial re-test on the third day

Prolonged TASER use on exhausted humans does not worsen markers of acidosis.

OBJECTIVE: There are safety concerns about TASER conducted electrical weapon (CEW) use on humans, and there have been media reports of adverse human outcomes after CEW exposure. Conducted electrical weapons are often used on physically exhausted subjects. A single CEW application of a CEW is generally accepted to be 5 seconds of exposure. Some exposures in reality involve more than 5 seconds. We sought to determine if a prolonged (15 seconds) CEW exposure on exhausted humans caused acidosis, hyperkalemia, serum lactate change, or troponin change. METHODS: This was a prospective study of gener

The alterations in adenosine nucleotides and lactic acid levels in striated muscles following death with cervical dislocation or electric shock.

In this study, changes in adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP) and lactic acid levels in masseter, triceps, and quadriceps muscles obtained from right and left sides of Spraque-Dawley rats following two different types of death were investigated. The samples were taken immediately and 120 minutes after death occurred either by cervical dislocation or electric shock. ATP concentrations in the muscles of masseter, triceps, and quadriceps were lower in samples obtained 120 minutes after death than that of samples obtained immediately after death

Conditioned fear attenuates light-induced suppression of melatonin release in rats.

Male rats were given 5 min of intermittent footshock, or were not shocked, for 3 or 5 consecutive days in a novel context at the midpoint of the dark phase of a 12:12-h light:dark cycle. Six days later, animals were reexposed to the context without footshock and received either a 5-min light pulse or were not disturbed. Reexposure to the context significantly increased plasma corticosterone in animals previously shocked there. Prior context-shock pairings significantly attenuated the suppression of melatonin by light, but did not affect basal levels of melatonin. These results suggest that the

Stressor- or drug-induced sensitization of the corticosterone response is not critically involved in the long-term expression of behavioural sensitization to amphetamine.

Repeated exposure to drugs of abuse induces long-lasting behavioural sensitization, which is thought to play a role in the persistence of drug-seeking behaviour. Recently, we showed that repeated exposure of rats to cocaine resulted in a long-lasting (weeks) sensitization of the hypothalamus-pituitary-adrenal axis, i.e. hypersecretion of adrenocorticotropic hormone and of the glucocorticoid corticosterone. Moreover, we found that the administration of a glucocorticoid receptor antagonist abolished the expression of psychostimulant-induced behavioural sensitization. In the present study we test

A role for brain glucocorticoid receptors in contextual fear conditioning: dependence upon training intensity.

We studied the possible involvement of corticosteroids in the establishment and long-term expression of contextual fear conditioning and questioned whether a corticosteroid action might be dependent upon stimulus intensity at training. Experiments included: (i) the intracerebroventricular administration of specific antagonists for the two types of intracellular corticosteroid receptors to rats trained at either 1 mA or 0.4 mA shock intensity at conditioning; and (ii) the administration of corticosterone after conditioning rats to 0.2 mA shocks. The results showed that the administration of a t

N-methyl-D-aspartate receptor antagonist MK-801 impairs learning but not memory fixation or expression of classical fear conditioning in goldfish (Carassius auratus).

The amnestic effects of the noncompetitive antagonist MK-801 on visually mediated, classic fear conditioning in goldfish (Carassius auratus) was examined in 5 experiments. MK-801 was administered 30 min before the training session on Day 1 to look for anterograde amnestic effects, immediately after training to look for retrograde amnestic effects, and before the training or test session, or both, to look for state-dependence effects. The results showed that MK-801 produced anterograde amnesia at doses that did not produce retrograde amnesia or state dependency and did not impair the expression

Influence of various acute stressors on the activity of adult male rats in a holeboard and in the forced swim test.

The effects of various acute stressors on the activity of adult male rats in a holeboard and in the forced swim test were studied. When tested immediately or 24 h after 1 h exposure to noise, restraint in tubes or tail shock, no changes in either defecation rate or activity in the holeboard were observed. In contrast, immediately after 1 h immobilization in wood-boards, a reduction of the number of areas crossed and the number of head-dips was found. The inhibitory effect of immobilization on head-dips persisted 24 h later. The behavior of the rats in the forced swim test was classified into t

Benzodiazepines and their antagonists interfere with opioid-dependent stress-induced analgesia.

Timing or intensity of shocks significantly modify the characteristics of the analgesia induced by footshock, and conditioning to footshock induces analgesia, independently from the time and shock parameters used for conditioning. However, whatever the parameters of shock, and the presence of conditioning or not, the stress has to be inescapable in order to produce an increase in pain thresholds. This observation suggests that anxiety plays a major role in the development of stress-induced analgesia. In order to test this hypothesis we investigated the effects of the benzodiazepine agonists di

Passive avoidance deficit following intracerebroventricular administration of cholecystokinin tetrapeptide amide in rats.

The effect of cholecystokinin tetrapeptide amide (CCK-4) injected into the lateral cerebral ventricle on memory processes was examined by a one-trial passive avoidance test in the rat. CCK-4 injection 30 and 60 min before the first retention test caused a shortened latency to response, and its chronic infusion into the lateral ventricle at a rate of 2 micrograms/day shortened the latency of the response to the level of almost complete amnesia. CCK-4 also reduced arginine-vasopressin effect on memory processes when administered simultaneously 30 min before the first retention test, but its amne

A role for histamine and histamine H2-receptors in non-opiate footshock-induced analgesia.

Scrambled DC current applied to the hind paws of rats caused an analgesic response that was inhibited by the histamine H2-receptor antagonists cimetidine, ranitidine and oxmetidine, but not by high doses of naloxone (the opiate antagonist), or other transmitter receptor antagonists. In contrast, AC current applied to all paws produced analgesia that was blocked by naloxone, but not cimetidine, showing the independence of these systems. These findings indicate a specific role for histamine and H2-receptors as mediators of endogenous non-opiate analgesia. In addition, a combination of cimetidine

Adjunctive drinking and the pituitary-adrenal response: effects of prior aversive stimulation (preshock).

Two groups of rats were exposed to an intermittent food schedule which induced polydipsia. One group of rats had previously been exposed to inescapable shock. This group showed reduced amounts of polydipsia, and the polydipsia was ineffective in reducing arousal levels (relative to those of the control group), as indexed by plasma corticosterone levels.

[Comparative evaluation of memory-regulating neuropeptides before and after electric shock in albino rats].

Antiamnestic properties of memory neuropeptide--modulators and their effect on learning were studied as well as consolidation of oligopeptide ACTH4-7, vasopressin, oxytocin, leu enkephalin and the melanocyte inhibiting factor. Effect of these neuropeptides on the rat higher nervous activity is bath strong and specific: ACTH4-7 and leu enkephalin speed up the primary learning whereas vasopressin and oxytocin improve long--term memory. Peptides are able to reverse retrograde as well as anterograde amnesia and to preserve previously formed habits from disturbance by electric shock. Vasopressin an

Thresholds of response to electric shock in previously undernourished rats.

1. Rats were undernourished in early life by feeding their mothers a restricted quantity of a good-quality diet during pregnancy and lactation. All offspring were fed ad lib. from weaning. Behavioural tests were done using adult males. 2. Thresholds of aversion to electric shock were determined in a rectangular box, one side of which was designated 'live' and the other 'safe' (o V side) for any particular shock level. Three 15 min sessions were given at each of five shock levels. Previously undernourished rats had a lower aversion threshold than controls. They spent more time than controls on
