Role of the duplicated CCAAT box region in gamma-globin gene regulation and hereditary persistence of fetal haemoglobin.

Hereditary persistence of fetal haemoglobin (HPFH) is a clinically important condition in which a change in the developmental specificity of the gamma-globin genes results in varying levels of expression of fetal haemoglobin in the adult. The condition is benign and can significantly alleviate the symptoms of thalassaemia or sickle cell anaemia when co-inherited with these disorders. We have examined structure-function relationships in the -117 HPFH gamma promoter by analysing the effect of mutating specific promoter elements on the functioning of the wild-type and HPFH promoters. We find that

Selective muscle imaging reveals myopodia clustering in multiple ventral muscles during Drosophila neuromuscular development.

Precise neuromuscular connectivity depends on coordinated interactions between motor axons and their muscle targets during development. In the Drosophila embryo, dynamic muscle protrusions termed myopodia arise during early phases of muscle innervation and have been proposed to contribute to synaptic partner matching. However, whether similar structures occur across different muscle fibers and how they are developmentally regulated remain unclear. Here, we developed a genetic imaging toolkit based on GAL4 drivers that label defined subsets of embryonic muscle fibers or enable stochastic single

Spatial Transcriptomics of Early Tooth Morphogenesis in Formalin-fixed Paraffin-embedded Mouse Embryonic Tissue.

The developing tooth comprises diverse and highly specialized cell populations that work together to maintain proper form and function. Elucidating the interactions among these cells and their surrounding microenvironment is critical for understanding the regulatory mechanisms underlying normal tooth development. Perturbations in these processes can result in congenital disorders such as tooth agenesis, dentinogenesis imperfecta, and amelogenesis imperfecta. Despite the substantial progress enabled by single-cell RNA sequencing (scRNA-seq) in revealing cellular heterogeneity, it does not prese

Segmental Vitiligo and Somatic Mosaicism: From Pathogenesis to Therapeutics.

Segmental vitiligo (SV) is a distinct clinical subtype of vitiligo characterized by unilateral, sharply demarcated depigmentation, rapid progression and early stabilization, and frequent leukotrichia. Compared with non-segmental vitiligo (NSV), SV is generally less strongly associated with systemic autoimmunity, suggesting potential differences in pathogenic mechanisms, although overlap phenotypes and mixed presentations have been reported. Accumulating clinical, epidemiological, and mechanistic evidence supports the concept that SV arises from somatic mosaicism, in which post-zygotic genetic

Applications and limitations of pluripotent stem cell-derived liver organoids.

Liver disease is one of leading causes of death worldwide. However, current liver models have limited research progress. Therefore, models that accurately reflect the physiological functions of the human liver are urgently needed in both clinical and laboratory research. Over the past decade, liver organoids have emerged as valuable research tools offering significant breakthroughs and demonstrating great promise as advanced liver models. Liver organoids derived from pluripotent stem cells (PSCs), including embryonic and induced pluripotent stem cells, have shown significant potential for mode

A New Vision for Ocular Disease: Stem Cell Strategies in Regenerative Ophthalmology.

Degenerative ocular diseases such as age-related macular degeneration (AMD), glaucoma, cataracts, retinitis pigmentosa (RP), dry eye disease (DED), and limbal stem cell deficiency (LSCD) are major contributors to global visual impairment and blindness. These conditions not only affect vision but also significantly reduce patients’ quality of life. Conventional therapies largely aim to manage symptoms or slow disease progression, with limited ability to regenerate or restore damaged ocular tissues. In contrast, stem cell-based therapies have emerged as a promising strategy, offering the potenti

Multiple Pseudo-Placentational Endometrial Hyperplasia (PEH) as a Rare Uterine Disorder in the Bitch.

The reproductive cycle in domestic dogs involves prolonged phases of hormonal activity, making the endometrial tissue vulnerable to degenerative changes. One common result of these changes is cystic endometrial hyperplasia (CEH), which often leads to pyometra. Another, though rarer, condition is pseudo-placentational endometrial hyperplasia (PEH), characterized by abnormal proliferation of the endometrial glands and stroma, forming placental-like structures without the presence of pregnancy. Case presentation: An 8-year-old female Golden Retriever, healthy and intact, was presented for a routi

A Tousled-like kinase (TLK) homolog required for male germline stem cell homeostasis in the planarian Schmidtea mediterranea.

Tousled-like kinases (TLKs) are serine/threonine kinases that are conserved across metazoan life and involved in DNA stability and cell division. Mammalian TLKs are most highly expressed in the testes, but their role in spermatogenesis has remained elusive because mutations result in embryonic lethality. Here, we identify two TLK homologs in the planarian Schmidtea mediterranea (Smed-TLK1a and Smed-TLK1b) and reveal a testis-specific requirement for Smed-TLK1a. Preferential expression of Smed-TLK1a was detected in testes of S. mediterranea hermaphrodites by whole-mount in situ hybridization, a

Sigmoid gallbladder in a 38-year-old female with gallstone disease: a case report.

BACKGROUND: The sigmoid gallbladder is a rare anatomic variant characterized by an S-shaped curve in its structure, often stemming from abnormal embryonic development during gestation. This anomaly complicates surgical procedures like cholecystectomy, primarily due to its convoluted shape and potential for misdiagnosis. Case studies indicate that preoperative imaging techniques, including ultrasound and MRCP, are essential for accurate diagnosis and preventing bile duct injuries. Although sigmoid gallbladders are typically asymptomatic unless associated with gallstones, thorough imaging and su

Neural stem cell-derived exosomes regulate cell proliferation, migration, and cell death of brain microvascular endothelial cells via the miR-9/Hes1 axis under hypoxia.

BACKGROUND: Our previous study found that mouse embryonic neural stem cell (NSC)-derived exosomes (EXOs) regulated NSC differentiation via the miR-9/Hes1 axis. However, the effects of EXOs on brain microvascular endothelial cell (BMEC) dysfunction via the miR-9/Hes1 axis remain unknown. Therefore, the current study aimed to determine the effects of EXOs on BMEC proliferation, migration, and death via the miR-9/Hes1 axis. METHODS: Immunofluorescence, quantitative real-time polymerase chain reaction, cell counting kit-8 assay, wound healing assay, calcein-acetoxymethyl/propidium iodide staining

The Notch-mediated circuitry in the evolution and generation of new cell lineages: the tooth model.

The Notch pathway is an ancient, evolutionary conserved intercellular signaling mechanism that is involved in cell fate specification and proper embryonic development. The Jagged2 gene, which encodes a ligand for the Notch family of receptors, is expressed from the earliest stages of odontogenesis in epithelial cells that will later generate the enamel-producing ameloblasts. Homozygous Jagged2 mutant mice exhibit abnormal tooth morphology and impaired enamel deposition. Enamel composition and structure in mammals are tightly linked to the enamel organ that represents an evolutionary unit forme

Human brain microphysiological systems in the study of neuroinfectious disorders.

Microphysiological systems (MPS) are 2D or 3D multicellular constructs able to mimic tissue microenvironments. The latest models encompass a range of techniques, including co-culturing of various cell types, utilization of scaffolds and extracellular matrix materials, perfusion systems, 3D culture methods, 3D bioprinting, organ-on-a-chip technology, and examination of tissue structures. Several human brain 3D cultures or brain MPS (BMPS) have emerged in the last decade. These organoids or spheroids are 3D culture systems derived from induced pluripotent cells or embryonic stem cells that conta

Generation and Characterization of Native and Sialic Acid-Deficient IgE.

Efficient characterization of IgE antibodies and their glycan structures is required for understanding their function in allergy and in the emerging AllergoOncology field for antibody immunotherapy. We report the generation, glyco-profiling and functional analysis of native and sialic acid-deficient glyco-engineered human IgE. The antibodies produced from human embryonic kidney cells were purified via a human IgE class-specific affinity matrix and structural integrity was confirmed by SDS-PAGE and size-exclusion chromatography (SEC). Purified IgEs specific for the tumor-associated antigens Cho

Eyes on CHARGE syndrome: Roles of CHD7 in ocular development.

The development of the vertebrate visual system involves complex morphogenetic interactions of cells derived from multiple embryonic lineages. Disruptions in this process are associated with structural birth defects such as microphthalmia, anophthalmia, and coloboma (collectively referred to as MAC), and inherited retinal degenerative diseases such as retinitis pigmentosa and allied dystrophies. MAC and retinal degeneration are also observed in systemic congenital malformation syndromes. One important example is CHARGE syndrome, a genetic disorder characterized by coloboma, heart defects, choa

Kidney organoids generated from erythroid progenitors cells of patients with autosomal dominant polycystic kidney disease.

BACKGROUND: Kidney organoids have been broadly obtained from commercially available induced pluripotent stem cells (iPSCs); however, it has been a great challenge to efficiently produce renal organoid models from patients with autosomal dominant polycystic kidney disease (ADPKD) that recapitulate both embryogenesis and the mechanisms of cystogenesis. METHODS: Blood erythroid progenitors (EPs) from two ADPKD patients and one healthy donor (HC) was used as a comparative control to normalize the many technical steps for reprogramming EPs and for the organoids generation. EPs were reprogrammed by

A 3D ontogenetic atlas of Alligator mississippiensis cranial nerves and their significance for comparative neurology of reptiles.

Cranial nerves are key features of the nervous system and vertebrate body plan. However, little is known about the anatomical relationships and ontogeny of cranial nerves in crocodylians and other reptiles, hampering understanding of adaptations, evolution, and development of special senses, somatosensation, and motor control of cranial organs. Here we share three dimensional (3D) models an of the cranial nerves and cranial nerve targets of embryonic, juvenile, and adult American Alligators (Alligator mississippiensis) derived from iodine-contrast CT imaging, for the first time, exploring anat

Ocular coloboma combined with cleft lip and palate: a case report.

BACKGROUND: Ocular coloboma is an excavation of ocular structures that occurs due to abnormal fusion of the embryonic optic fissure. Further, cleft lip/palate (CL/P), a congenital midline abnormality, is caused by a defect in the fusion of the frontonasal, maxillary, and mandibular prominences. No study has reported the association between these two phenotypes in the absence of other systemic abnormalities. We present a case of ocular coloboma along with CL/P and without other neurological abnormalities. CASE PRESENTATION: A 5-year-old Asian boy presented with decreased visual acuity in his r

SQSTM1L341V variant that is linked to sporadic ALS exhibits impaired association with MAP1LC3 in cultured cells.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are genetically, pathologically and clinically-related progressive neurodegenerative diseases. Thus far, several SQSTM1 variations have been identified in patients with ALS and FTD. However, it remains unclear how SQSTM1 variations lead to neurodegeneration. To address this issue, we investigated the effects of ectopic expression of SQSTM1 variants, which were originally identified in Japanese and Chinese sporadic ALS patients, on the cellular viability, their intracellular distributions and the autophagic activity in cultur

MSH homeobox 1 polymorphisms and the risk of non-syndromic orofacial clefts: a meta-analysis.

MSH homebox 1 (MSX1) is a susceptibility gene for non-syndromic orofacial clefts (NSOCs). Here, a meta-analysis was conducted to assess their associations. A systematic search of PubMed to 1 September 2017, was performed to retrieve all eligible studies. Odds ratios (ORs) were used to calculate the associations. The stability of the results was evaluated by sensitivity analysis. Publication bias was assessed using Begg's funnel plots and the Egger test. In silico Msx1 expression during early mouse craniofacial development was evaluated by the Gene Expression Omnibus. In the overall analysis, M

Foxc2 is required for proper cardiac neural crest cell migration, outflow tract septation, and ventricle expansion.

BACKGROUND: Proper development of the great vessels of the heart and septation of the cardiac outflow tract requires cardiac neural crest cells. These cells give rise to the parasympathetic cardiac ganglia, the smooth muscle layer of the great vessels, some cardiomyocytes, and the conotruncal cushions and aorticopulmonary septum of the outflow tract. Ablation of cardiac neural crest cells results in defective patterning of each of these structures. Previous studies have shown that targeted deletion of the forkhead transcription factor C2 (Foxc2), results in cardiac phenotypes similar to that d
