Role of endogenous opioids and central opioid receptors in cerebral cortical blood flow autoregulation.

To examine the role of endogenous opioids in autoregulatory maintenance of cerebral cortical blood flow (CoBF), CoBF was measured continuously by laser-Doppler flowmetry during changes in arterial pressure. Experiments were conducted on pentobarbital sodium-anesthetized adult mongrel dogs. Mean arterial pressure (MAP) was decreased or increased by inflating a perivascular occluder placed around the inferior vena cava or the thoracic descending aorta, respectively. To exclude the influence of the baroreceptor reflex on the autoregulatory maintenance of CoBF, all experiments were conducted on do

Edward E. Smissman-Bristol-Myers Squibb Award Address. The role of concepts in structure-activity relationship studies of opioid ligands.

Various concepts have served as the basis for the development of models to explore the relationship between molecular structure and biological activity. In this presentation I have outlined five concepts that have been useful in our investigation of opioid receptor multiplicity and in the design of selective opioid receptor antagonists. The first of these, the multiple binding modality concept, led to our application of four other concepts in the development of opioid receptor probes. Some of these probes are now standard tools in opioid research. These include the mu-selective affinity label

Anorexia in the elderly.

Weight loss and anorexia occur commonly in the elderly. While in many cases the anorexia can be attributed to associated disease processes, it does appear that a true anorexia of aging exists. Animal studies have suggested that older rodents have an excessive satiety effect of cholecystokinin and a decreased opioid feeding drive. Other older persons develop anorexia in association with depression. In these subjects, excess corticotropin-releasing factor may be the neurotransmitter involved in the pathogenesis of the anorexia. In Alzheimer's disease, decreases in norepinephrine and neuropeptide

Functional dyspepsia and chronic idiopathic gastric stasis. Role of endogenous opiates.

Chronic idiopathic gastric stasis can be responsible for unexplained dyspepsia. Because exogenous opiates inhibit gastric emptying and endogenouslike substances are present in the gastrointestinal tract, we tested the hypothesis that increased endogenous opiate activity may be responsible for chronic idiopathic gastric stasis. Eighteen patients with chronic idiopathic gastric stasis and ten healthy volunteers were studied by gastrointestinal manometry. Scintigraphic technique also was used, during which either intravenous saline or naloxone hydrochloride were infused. Manometry showed gastric

The neuronal basis of compulsive behaviour in anorexia nervosa.

The relationship between arousal and efficiency of the brain is shown by the inverted U of the Yerkes-Dodson curve. Measuring arousal has been difficult because the three types of arousal (EEG, behaviour and autonomic) do not change in unison. From Magoun's work, arousal can be stimulated via the reticular formation or from parts of the cortex. Kyotorphin (Tyr-Arg) causes widespread excitation when applied to the cortex and may represent this mechanism: it is then inhibited only by noradrenaline. The hippocampus causes stimulation of arousal to persist after the exciting stimulus stops and can

Pulsatile secretion of gonadotropins and prolactin in male marathon runners. Relation to the endogenous opiate system.

We tested the hypothesis that sustained, strenuous physical training alters the neuroendocrine regulation of pulsatile gonadotropin and/or prolactin secretion in men. Blood was sampled at 20-minute intervals over 8 hours in five endurance-trained men after a 10-15 mile run in the middle of the active training season, and in 11 nonendurance trained normal controls. In these two groups, basal patterns of physiologically pulsatile secretion of LH, FSH, and prolactin (PRL) were not significantly different in relation to the following parameters: mean serum concentration of each of the three hormon

Opioid peptides and central control of blood pressure.

The role of endogenous opioids in central modulation of baroreceptor reflex function has been assessed in rabbits and in man using stable enkephalin analogues and synthetic opiates with a range of mu, delta and kappa opiate receptor agonist specificity. In addition the effects of naloxone, a mu opiate receptor antagonist, have been studied. In rabbits descarboxy-leu-enkephalin (RX783016) given by intracisternal injection reduced baroreflex sensitivity as assessed by heart rate responses to phenylephrine, sodium nitroprusside and controlled haemorrhage. These effects were prevented by intraveno

Ventral tegmental analgesia in two strains of rats: effects of amphetamine, naloxone and parachlorophenylalanine.

Pain sensitivity and analgesia induced by the stimulation of the ventral tegmentum (VT) were studied in 72 male rats of two lines, LC2-Hi and LC2-Lo, genetically selected for high and low rates of lateral hypothalamic self-stimulation, respectively. LC2-Lo rats were more sensitive to acute peripheral pain and developed a stronger analgesia than their LC2-Hi counterparts. In order to assess the pharmacological substrate of ventral tegmental stimulation-induced analgesia (VT-SIA), the effects of amphetamine (AMP, 21 animals), naloxone (NX, 24 animals) and parachlorophenylalanine (PCPA, 27 animal

Effects of enkephalins versus opiates on locomotor activity of the horse.

The enkephalins are small, pentapeptide neurotransmitter molecules which have reportedly been used in racing horses. In our experiments, D-Ala2-Metenkephalinamide and leucine enkephalin were administered to horses intravenously (IV) and intracisternally (IC). Leucine enkephalin had little effect on locomotor activity by either route at doses of 0.01 mg/Kg or less. Methionine enkephalinamide, an enzyme resistant enkephalin analog, had no significant effect when given IV (0.002 and 0.008 mg/kg). Other experiments involving intracisternal dosing with this long acting form at higher levels (0.005-

Dynorphin immunocytochemical localization in brain and peripheral nervous system: preliminary studies.

Using antisera specific for the opioid peptide dynorphin, we have carried out immunocytochemical studies of the distribution in rat brain and periphery. In the central nervous system, cells that stain positively for dynorphin are found in the supraoptic nucleus, with less-well-stained cells in the paraventricular nucleus of the hypothalamus. Few positive fibers were detected in brain, suggesting problems with fixation and preservation of antigenicity. In pituitary no staining was seen in the anterior and intermediate lobes but heavy staining was detected in the posterior lobe. In the guinea pi

[Measurement of methionine-enkephalin-like substance in CSF from normal subjects and patients with pituitary adenoma (author's transl)].

Methionine-enkephalin-like substance was measured in CSF by the radioreceptor-assay established by Furui et al. Samples were obtained from preoperative 20 cases, in which were included 11 cases of pituitary adenoma, 3 cases of craniopharyngioma, 2 cases of pseudtumor cerebri and 4 normal cases, by lumbar puncture. Also postoperative measurement of this substance and pre- and postoperative measurement of ACTH in plasma were performed in 5 cases of Cushing's disease. Five ml of CSF was chromatograpied on two successive columns, lyophilized and assayed for opiate receptor affinity against 3H-dihy

[Opiate receptors and endogenous morphines: a new approach to the study of the brain].

The paper deals with a range of problems connected with a study of a nature and a function of specific neuronal receptors and endogenous morphines. It is demonstrated that a further development of studies in the given area will be of a principal significance in solving the cardinal problems of psychopharmacology and a search for biological bases of mental diseases. The most perspective ways of studying functional opiate receptors and endogenous morphines in schizophrenia are discussed.

C-fragment of lipotropin--an endogenous potent analgesic peptide.

1 A series of peptides derived from porcine lipotropin was examined for analgesic and other morphine-like properties on infusion into the cannulated third ventricle of cats.2 Lipotropin (LPH 1-91) itself produced no analgesia or other morphine-like effects when infused in a dose of 150 mug.3 C-fragment (LPH 61-91) produced strong long-lasting analgesia when infused in a dose of 10 or 20 mug; on a molar basis the potency was between 90 and 180 times that of morphine. The following morphine-like effects were also produced: shivering leading to fever, vasodilatation of the pinnae, mydriasis, open

[Place of the opioid system in biology and treatment of Alcohol Use Disorder].

While the DSM 5 has formalized the terminology "Alcohol Use Disorders" (AUD) or "disorders of the use of alcohol" (UAW French translation in progress), the term "alcohol dependence" still used in ICD-10, apriority in the future ICD-11 and above in clinical practice. Addiction to alcohol is the cause of mortality and major morbidity. In terms of therapeutic strategies for its management, alongside the maintenance of abstinence after withdrawal (with a high rate of relapse), the reduction of alcohol consumption below certain thresholds of intake is emerging in order to reduce risk, improve healt

Pathological gambling. A comprehensive review.

Due to recent changes of gambling laws, accessibility to gambling has become more widespread and thus, there has also been an increase in the prevalence of pathological gambling (PG). The wide range of social, economic, and psychological problems associated with PG are well known. There is a need for better understanding of PG and this review attempts to do so. Literature searches using the Medline and PsycINFO databases were used. Critical examining of the literature showed that familial/genetic, sociological, and individual factors (e.g., an individual's personality, biochemistry, psychologi
