The "Equilibrium Model" has provided new tools for describing and investigating enzyme thermal adaptation. It has been shown that the effect of temperature on enzyme activity is not only governed by deltaG(double dagger)(cat) and deltaG(double dagger)(inact) but also by two new intrinsic parameters, deltaH(eq) and T(eq), which describe the enthalpy and midpoint, respectively, of a reversible equilibrium between active and inactive (but not denatured) forms of enzyme. Twenty-one enzymes from organisms with a wide range of growth temperatures were characterized using the Equilibrium Model. Stati

Hepatitis E virus (HEV) is the sole member of the genus Hepevirus in the family Hepeviridae. HEV is transmitted primarily by the fecal-oral route, and water-borne epidemics are characteristic of hepatitis E in many developing countries in Asia, Africa and Latin America where sanitation conditions are suboptimal. Accumulating lines of evidence indicate that HEV-associated hepatitis also occurs domestically among individuals in industrialized countries, that there are animal reservoirs of HEV such as domestic pigs and wild boars, and that hepatitis E is a zoonosis. Based on the extensive genomic

Dengue virus transmission has increased dramatically in the past 2 decades, making this virus one of the most important mosquito-borne human pathogens. The emergence of dengue hemorrhagic fever in most tropical countries has made its control a public health priority, but no vaccines or treatments exist. Little is understood about dengue virus pathogenesis, because no other animals develop symptoms of disease, and research, therefore, has been limited to studies involving patients. Although epidemiologic and evolutionary studies have pointed to host and viral factors in determining disease outc

Variation in 815bp of mitochondrial DNA from two gene fragments (300bp of cytochrome b and 395-515bp of 12S rRNA) for 26 Malpolon monspessulanus, and cytochrome b for a further 21 individuals, indicates that this species originated in the Maghreb area of Northwest Africa. Here, an estimated 3.5-6Mya, it divided into the western M. m. monspessulanus, and an eastern clade including M. m. insignitus and M. m. fuscus. The very limited genetic differentiation between Maghreb and Southwest European populations of this form suggests that it arrived in the Iberian Peninsula only recently. Population g

Legionella pneumophila is a gram-negative facultative intracellular human pathogen that can cause fatal Legionnaires' disease. Polypeptide deformylase (PDF) is a novel broad-spectrum antibacterial target, and reports of inhibitors of PDF with potent activities against L. pneumophila have been published previously. Here, we report the identification of not one but three putative pdf genes, pdfA, pdfB, and pdfC, in the complete genome sequences of three strains of L. pneumophila. Phylogenetic analysis showed that L. pneumophila PdfA is most closely related to the commonly known gamma-proteobacte

BACKGROUND AND AIMS: Brassicaceae, with nearly 340 genera and more than 3350 species, anchors the low range of angiosperm genome sizes. The relatively narrow range of DNA content (0.16 pg < 1C < 1.95 pg) was maintained in spite of extensive chromosomal change. The aim of this study was to erect a cytological and molecular phylogenetic framework for a selected subset of the Brassicacae, and use this as a template to examine genome size evolution in Brassicaceae.
METHODS: DNA contents were determined by flow cytometry and chromosomes were counted for 34 species of the family Brassicaceae and fo

Recent studies have provided evolutionary explanations for much of the variation in mortality among human infectious diseases. One gap in this knowledge concerns respiratory tract pathogens transmitted from person to person by direct contact or through environmental contamination. The sit-and-wait hypothesis predicts that virulence should be positively correlated with durability in the external environment because high durability reduces the dependence of transmission on host mobility. Reviewing the epidemiological and medical literature, we confirm this prediction for respiratory tract pathog

The TEM-1 and SHV-1 beta-lactamases are important contributors to resistance to beta-lactam antibiotics in gram-negative bacteria. These enzymes share 68% amino acid sequence identity and their atomic structures are nearly superimposable. Extended-spectrum cephalosporins were introduced to avoid the action of these beta-lactamases. The widespread use of antibiotics has led to the evolution of variant TEM and SHV enzymes that can hydrolyze extended-spectrum antibiotics. Despite being highly similar in structure, the TEM and SHV enzymes have evolved differently in response to the selective press

Mitochondrial DNA is a small, well characterized chromosome which is transmitted across the generations in the maternal lineage, independently of nuclear DNA. mtDNA acts in effect as a robust, species specific biological clock and tracer which can be used to follow the evolution and spread by geographic migration of populations from their origins. Mutations in mtDNA cause specific maternally hereditable diseases, and can be used for forensic purposes. They are not specifically implicated in neoplasia, but they may provide clues as to the nature and origins of cancer susceptibility in various p

The family of human peroxidases described includes myeloperoxidase, eosinophil peroxidase, uterine peroxidase, lactoperoxidase, salivary peroxidase, thyroid peroxidase and prostaglandin H1/2 synthases. The chemical identity of the peroxidase compound I and II oxidation states for the different peroxidases are compared. The identities of the distal and proximal amino acids of the catalytic site of each peroxidase are also compared. The gene characteristics and chromosomal location of the human peroxidase family have been tabulated and their molecular evolution discussed. Myeloperoxidase polymor

The chromosomal speciation model invokes chromosomal rearrangements as the primary cause of reproductive isolation. In a heterozygous carrier, chromosomes bearing reciprocal translocations mis-segregate at meiosis, resulting in reduced fertility or complete sterility. Thus, chromosomal rearrangements act as a post-zygotic isolating mechanism. Reproductive isolation in yeast is due to post-zygotic barriers, as many species mate successfully but the hybrids are sterile. Reciprocal translocations are thought to be the main form of large-scale rearrangement since the hypothesized duplication of th

We present DNA-hybridization data on 21 amniotes and two anurans showing that discrimination is obtained among most of these at the class and lower levels. Trees generated from these data largely agree with conventional views, for example in not associating birds and mammals. However, the sister relationships found here of the monotremes to marsupials, and of turtles to the alligator, are surprising results which are nonetheless consistent with the results of some other studies. The Marsupionta hypothesis of Gregory is reviewed, as are opinions about the placement of chelonians. Anatomical and

This communication provides a multidisciplinary perspective on the contribution of nuclear medicine, liquid biopsy-derived molecular markers, and AI/ML-based approaches to evolve a stepped-care model in precision oncology. The subject is of significance, particularly in the current era of precision medicine, where individualized treatment strategies increasingly require the integration of molecular imaging, molecular profiling, and multidisciplinary decision-making. The authors also emphasize the growing role of nuclear medicine in personalized treatment selection and theranostic decision-maki

Phylogenetic networks and, more generally, directed acyclic graphs (DAGs) represent hierarchical structure beyond trees, for instance in the presence of reticulate evolutionary events such as hybridization or horizontal gene transfer. A central question is which parts of such graphs are essential with respect to leaf-observable information, and which parts can be removed without changing this information. Resolving this question can lead to principled simplification methods for phylogenetic networks, such as the recent normalization approach of Francis et al. In this paper, we study this quest

Concerted evolution is the model by which multicopy gene families are homogenized across genomes, with ribosomal DNA (rDNA) as its textbook example. However, the extent and efficiency of this process may vary depending on genomic factors. The plant family Asteraceae provides a natural comparative system to address the evolutionary dynamics of rDNA, as it is uniquely characterized by the coexistence of two contrasting genomic arrangements: a separate (S-type) and a linked (L-type) organization of 5S and 35S rRNA genes. Here we analyzed rDNA arrangements in 71 representative Asteraceae species (

Species barriers limit gene flow and maintain co-adapted genomes. Interspecific hybridization can break down species barriers, generating phenotypic variation and altering evolutionary potential. Although the phenotypic and genomic consequences of hybridization have been extensively studied in many systems, far less is known about the longer-term evolutionary dynamics of highly divergent, mosaic genomes and the extent to which genetic incompatibilities shape their adaptation. By suppressing anti-recombination genes in meiosis, we generate 20 haploid recombinant hybrids from a cross between Sac

Grasses in the Panicoideae, Aristidoideae, Chloridoideae, Micrairoideae, Arundinoideae, and Danthonioideae (PACMAD) clade independently colonized cold environments from warm-climate ancestors, but whether their molecular responses to freezing reflect shared evolutionary solutions or lineage-specific innovations remains poorly understood at the protein level. We used comparative proteomics to test whether protein-level cold responses show stronger cross-species conservation than previously observed at the transcript level. We quantified seasonal rhizome proteomes (winter vs summer) from five PA

De novo gene emergence refers to the process by which new genes arise from mutations in previously non-coding genomic regions. Investigating early stages of de novo gene emergence is essential for understanding the mechanisms that enable gene formation from scratch. Several workflows have been developed to detect and study the early stage of de novo gene emergence in specific species. However, no software currently exists that can automatically identify and characterize novel, unannotated open reading frames from a transcriptome, and analyse their mutations and fixation patterns within or acro

HIV-1 circulating recombinant form (CRF) 01_AE has driven major HIV epidemics across Southeast and East Asia for decades, yet the mechanisms underlying its long-term regional spread remain poorly resolved. Using near-complete genome sequences and Bayesian phylogeographic reconstruction, we traced the evolutionary history of CRF01_AE from its origin in Central Africa to its expansion across Asia. Our results uncover a decades-long transmission corridor linking Africa, Thailand, and China, in which CRF01_AE was introduced into Thailand in the early 1980s and subsequently disseminated across Sout
