The Role of Extracellular Histones in Influenza Virus Pathogenesis.

Although exaggerated host immune responses have been implicated in influenza-induced lung pathogenesis, the etiologic factors that contribute to these events are not completely understood. We previously demonstrated that neutrophil extracellular traps exacerbate pulmonary injury during influenza pneumonia. Histones are the major protein components of neutrophil extracellular traps and are known to have cytotoxic effects. Here, we examined the role of extracellular histones in lung pathogenesis during influenza. Mice infected with influenza virus displayed high accumulation of extracellular his…

Cellular Transmigration Increases Nanoparticle Penetration Into Solid Tumors.

Delivering nanoparticles to cancer cells requires the penetration of nanoparticles into solid tumors. Nanoparticles predominantly enter solid tumors through blood vessel endothelial cells but then become trapped in the dense extracellular matrix layer. There are two known mechanisms that govern nanoparticle escape from this layer. Smaller nanoparticles slowly diffuse through the matrix into the tumor microenvironment. Larger nanoparticles trapped in the extracellular matrix are internalized and transported deeper into the tumor by macrophages. Here, we determined a third mechanism of nanoparti

Advances in the study of low-density neutrophils in rheumatic diseases.

Rheumatic diseases are characterized by immune dysfunction, chronic inflammation and tissue damage, in which neutrophils play a pivotal role … functional features and exert their pathogenic effects primarily through the release of neutrophil extracellular traps (NETs) and pro-inflammatory cytokines, as well as their involvement in the immune regulatory network. LDNs are implicated in the pathogenesis of multiple rheumatic diseases. Targeting LDNs or their key pathogenic pathways may provide novel insights …

Periodontal immune homeostasis: neutrophils as guardians and betrayers, and strategies for precision modulation.

The health of periodontal tissues fundamentally depends on a delicate, dynamic immune homeostasis; disruption of this balance constitutes the pathological basis driving the initiation and progression of chronic periodontitis. Due to their high heterogeneity and phenotypic plasticity, neutrophils play multifaceted roles in maintaining periodontal homeostasis and driving disease progression. Under homeostatic conditions, through precisely regulated mechanisms including chemotaxis, phagocytosis, and extracellular trap formation, neutrophils actively clear microorganisms and sustain tissue toleran

The Relationship Between NETosis and Biofilm Formation in Chronic Infections.

Chronic infections pose significant clinical challenges due to their persistent nature, heightened resistance to conventional therapies, and association with biofilm formation. Neutrophil extracellular traps (NETs), released through a unique form of cell death known as NETosis, serve as an innate immune defense mechanism by trapping and neutralizing pathogens. However, accumulating evidence reveals a complex and paradoxical relationship between NETs and microbial biofilms. While NETs can immobilize and kill planktonic microbes, the extracellular DNA and associated proteins often contribute to …

Editor's Highlights-July 2025.

Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease characterized by painful nodules, abscesses, and tunnels. This issue highlights key advances in understanding its pathogenesis and improving clinical care. New data suggest that the haptoglobin 2-2 genotype may be linked to familial HS and increased flare frequency, pointing toward potential applications in risk stratification. Another study explores the role of neutrophil extracellular traps (NETs) in HS, identifying their presence in both lesional and perilesional skin, and correlating serum NET markers with disease severit…

The Roles of Neutrophil Cells in the Pathogenesis of Fibrosis: Mechanisms for Disease Progression.

Fibrosis is a dysregulated tissue repair process with dynamic reorganization of extracellular matrix. Neutrophils have traditionally been viewed as short-lived, first-responder immune cells primarily involved in acute inflammation and pathogen clearance. However, accumulating evidence reveals that they also play multifaceted roles in the pathogenesis of fibrosis across multiple organs. Through the release of cytokines, granule proteins, neutrophil extracellular traps, and metabolic mediators, as well as through interactions with fibroblasts, macrophages, and other immune cells, neutrophils act…

NETosis of psoriasis: a critical step in amplifying the inflammatory response.

NETosis, a regulated form of neutrophil death, is crucial for host defense against pathogens. However, the release of neutrophil extracellular traps (NETs) during NETosis can have detrimental effects on surrounding tissues and contribute to the pro-inflammatory response, in addition to their role in controlling microbes. Although it is well-established that the IL-23-Th17 axis plays a key role in the pathogenesis of psoriasis, emerging evidence suggests that psoriasis, as an autoinflammatory disease, is also associated with NETosis. The purpose of this review is to provide a comprehensive unde…

Targeting NETosis in Acute Brain Injury: A Systematic Review of Preclinical and Clinical Evidence.

Acute brain injury (ABI) remains one of the leading causes of death and disability world-wide. Its treatment is challenging due to the heterogeneity of the mechanisms involved and the variability among individuals. This systematic review aims at evaluating the impact of anti-histone treatments on outcomes in ABI patients and experimental animals and defining the trend of nucleosome levels in biological samples post injury. We performed a search in Pubmed/Medline and Embase databases for randomized controlled trials and cohort studies involving humans or experimental settings with various cause

Platelets in the NETworks interweaving inflammation and thrombosis.

Platelets are well characterized for their indispensable role in primary hemostasis to control hemorrhage. Research over the past years has … a sufficient immune response. In particular, platelets can activate neutrophils to form neutrophil extracellular traps (NETs). This specific neutrophil effector function is characterized by neutrophils expelling chromatin fibres decorated with histones and antimicrobial proteins into the extracellular space where they serve to trap and kill pathogens. Until now, the …

Hypoxia Promotes Neutrophil Survival After Acute Myocardial Infarction.

Phagocytosis, degranulation, and neutrophil extracellular traps (NETs) formation build the armory of neutrophils for the first line of defense against invading pathogens. All these processes are modulated by the microenvironment including tonicity, pH and oxygen levels. Here we investigated the neutrophil infiltration in cardiac tissue autopsy samples of patients with acute myocardial infarction (AMI) and compared these with tissues from patients with sepsis, endocarditis, dermal inflammation, abscesses and diseases with prominent neutrophil infiltration. We observed many neutrophils infiltrat…

[The role of neutrophil density in neutrophils-mediated inflammatory response induced by monosodium urate crystals].

Objective: To study the role of neutrophil density and molecular mechanism in neutrophils-mediated inflammatory response induced by monosodium urate (MSU) crystals. Methods: Polymorphonuclear neutrophils (PMNs) isolated from healthy human peripheral blood were treated with MSU crystals at different density (5×10(6)/ml, 20×10(6)/ml, 100×10(6)/ml) in vitro. The mean fluorescence intensity (MFI) of PMNs and production of reactive oxygen species (ROS) were detected by flow cytometry. The distribution of MSU crystals was observed by polarized light microscopy. The neutrophil extracellular traps (NE…

Induction of Ocular Surface Inflammation and Collection of Involved Tissues.

Ocular surface diseases include a range of disorders that disturb the functions and structures of the cornea, conjunctiva, and the associated ocular surface gland network. Meibomian glands (MG) secrete lipids that create a covering layer that prevents the evaporation of the aqueous part of the tear film. Neutrophils and extracellular DNA traps populate MG and the ocular surface in a mouse model of allergic eye disease. Aggregated neutrophil extracellular traps (aggNETs) formulate a mesh-like matrix composed of extracellular chromatin that occludes MG outlets and conditions MG dysfunction. Here…

Advances in research on the role of neutrophils in organ transplant rejection.

Organ transplantation is an effective treatment for end-stage organ failure, but rejection remains a major obstacle to transplant success. Neutrophils play a key role in organ transplant rejection, participating not only in early immune responses but also exacerbating graft injury through mechanisms such as the release of neutrophil extracellular traps (NETs). Therefore, in-depth exploration of the immunological role of neutrophils in transplant rejection and their interactions with other immune cells is highly important. This article reviews the latest research progress on the mechanisms of a…
